Abstract – The aim of this study was to evaluate the rat subcutaneous tissue reaction to implanted polyethylene tubes filled with mineral trioxide aggregate (MTA) FILLAPEX® compared to the reaction to tubes filled with Sealapex® or Angelus MTA®. These materials were placed in polyethylene tubes and implanted into the dorsal connective tissue of Wistar rats for 7, 15, 30, 60, and 90 days. The specimens were stained with hematoxylin and eosin or Von Kossa or left unstained for examination under polarized light. Qualitative and quantitative evaluations of the reaction were performed. All materials caused moderate reactions after 7 days, which decreased with time. The reactions were moderate and similar to that evoked by the control and Sealapex® on the 15th day. MTA FILLAPEX® and Angelus MTA caused mild reactions beginning after 15 days. Mineralization and granulation birefringent to polarized light were observed with all materials. It was concluded that MTA FILLAPEX® was biocompatible and stimulated mineralization.  相似文献

Reactions of connective tissue to mineral trioxide aggregate and amalgam   总被引:4,自引:0,他引:4  
The purpose of this study was to evaluate the subcutaneous connective tissue reactions to ProRoot, mineral trioxide aggregate (MTA; Dentsply), and Oralloy, high-copper amalgam (Coltene). These materials were placed in polyethylene tubes and implanted into dorsal connective tissue of Wistar albino rats, and tissue biopsies were collected and histologically examined 7, 15, 30, 60, and 90 days after the implantation procedure. The presence of inflammation, predominant cell type, calcification, and thickness of fibrous connective tissue were recorded. Scores were defined as follows: 0, none or few inflammatory cells, no reaction; 1, < 25 cells, mild reaction; 2, 25 to 125 cells, moderate reaction; 3, > or = 125 cells, severe reaction. Fibrous capsule was categorized as "thin" when thickness was < 150 microm and "thick" at > 150 microm. Necrosis and formation of calcification were recorded. Both materials were well tolerated by the tissues in a 90-day evaluation period. One notable finding is the presence of dystrophic calcification in connective tissue adjacent to MTA; this finding is consistent with the hypothesis of hard tissue induction by this material.  相似文献

The purpose of this paper was to study the reaction of rat subcutaneous connective tissue to the implantation of dentin tubes filled with white mineral trioxide aggregate (MTA), a material that will be marketed. The tubes were implanted into rat subcutaneous tissue and the animals were sacrificed after 7 and 30 days. The undecalcified pieces were prepared for histological analysis with polarized light and von Kossa technique for mineralized tissues. Granulations birefringent to polarized light and an irregular structure like a bridge were observed next to the material; both were von Kossa positive. Also, in the dentin wall tubules a layer of birefringent granulations was observed. The results were similar to those reported for gray MTA, indicating that the mechanisms of action of the white and gray MTA are similar.  相似文献

Objectives: The purpose of this study was to evaluate and compare the cytotoxic effects of ProRoot MTA and DiaRoot BA, a bioceramic nanoparticulate cement, on subcutaneous rat tissue. Study Design: Fifty Sprouge Dawley rats were used in this study. Polyethylene tubes filled with ProRoot MTA and DiaRoot BioAggregate, along with a control group of empty, were implanted into dorsal connective tissue of rats for 7, 15, 30, 60, and 90 days. After estimated time intervals the rats were sacrificed. The specimens were fixed, stained with hematoxylin and eosin, and then evaluated under a light microscope for inflammatory reactions and mineralization. Results: All groups evoked a severe to moderate chronic inflammatory reaction at 7 and 15 days, which decreased with time. Both the MTA and BioAggregate groups showed similar inflammatory reactions, except at 90 days when MTA showed statistically significant greater inflammation (p>0.05). The MTA group showed foreign body reaction at all times. Compared to BioAggregate, MTA showed significantly more foreign body reaction at 60 and 90 days (p<0.0001). After 30 days foreign body reaction of BioAggregate decreased significantly. Both MTA and BioAggregate groups showed similar necrosis at 7 and 15 days (p=0.094 and p=0.186 respectively). No necrosis was observed after 15 days. Similarly there was no fibrosis after 30 days for both MTA and BioAggregate groups (p>0.05). Conclusions: Since DiaRoot BioAggregate showed significantly better results than MTA, we can conclude that it is more biocompatible. However, further studies are required to confirm this result. Objective

The aim of this study was to compare two methodologies used in the evaluation of tissue response to root-end filling materials in rats.

Material and Methods

Forty rats were divided into 4 groups: in Groups I and II (control groups), empty polyethylene tubes were implanted in the extraction site and in the subcutaneous tissue, respectively; in Groups III and IV, polyethylene tubes filled with ProRoot MTA were implanted in the extraction site and in the subcutaneous tissue, respectively. The animals were killed 7 and 30 days after tube implantation, and the hemi-maxillas and the capsular subcutaneous tissue, both with the tubes, were removed. Specimens were processed and evaluated histomorphologicaly under light microscopy. The scores obtained were analyzed statistically by the Kruskal-Wallis test (p<0.05).

Results

There were no statistically significant differences between the implantation methods (p=0.78033, p=0.72039). It was observed that the 30-day groups presented a more mature healing process due to smaller number of inflammatory cells.

Conclusion

The present study showed no differences in tissue responses as far as the implantation site and the studied period were concerned. Alveolar socket implantation methodology represents an interesting method in the study of the biological properties of root-end filling endodontic materials due to the opportunity to evaluate bone tissue response.  相似文献

Introduction

The present in vitro study was conducted with the aim of evaluating and comparing the cytotoxic effects of three root-end filling materials, ProRoot mineral trioxide aggregate (ProRoot MTA; Dentsply Tulsa Dental, Memphis, TN), MTA Angelus (Angelus, Londrina, Brazil), and a modified zinc oxide-eugenol cement (Super-EBA; Bosworth Co, Skokie, IL) on human periodontal ligament (PDL) fibroblasts.

Methods

PDL cells were cultured in an mineral trioxide aggregate (MTA)- or a Super-EBA-conditioned medium to assess the viability as determined by the trypan blue exclusion assay. The proliferation of the cells was recorded, and the cellular morphology was observed by confocal microscopy. Moreover, PDL cell aggregates were cultured on the substrate surfaces to assess cell adhesion.

Results

ProRoot MTA was found to be the most biocompatible material, whereas Super-EBA was found to be the most cytotoxic material because it significantly inhibited the cell growth and adherence on its. In the presence of ProRoot MTA, the PDL cell proliferation was almost unaltered. MTA Angelus was found to be more cytotoxic than ProRoot MTA, offering, however, excellent scaffold properties for the adhesion of cell aggregates.

Conclusions

Under the conditions of the present study, it seems that commercially available forms of MTA may behave in different ways regarding their proliferative effect on human PDL fibroblasts. ProRoot MTA appears to be the most biocompatible of the three tested materials when considering use for root-end endodontic microsurgery.  相似文献

Introduction

Hydraulic silicate cements such as mineral trioxide aggregate (MTA) have many clinical advantages. Newer hydraulic silicate materials have been developed that improve on the limitations of mineral trioxide aggregate such as the long setting time and difficult handling characteristics. The purpose of this study was to examine the effect of saline and fetal bovine serum (FBS) on the setting and compressive strength of the following hydraulic silicate cements: ProRoot MTA (white WMTA; Dentsply International, Tulsa Dental Specialties, Johnson City, TN), EndoSequence Root Repair Material (Brasseler USA, Savannah, GA), MTA Plus (MTAP; Avalon Biomed Inc, Bradenton, FL), and QuickSet (QS; Avalon Biomed Inc, Bradenton, FL).

Methods

Samples of root-end filling materials were compacted into polyethylene molds. Samples were exposed to FBS or saline for 7 days. A universal testing machine was used to determine the compressive strengths.

Results

QS had significantly lower compressive strength than all other materials (P < .001). White MTA and MTAP mixed with liquid had lower compressive strengths after exposure to FBS compared with saline (P = .003). ERRM, MTAP mixed with gel, and QS were not affected by the exposure to FBS.

Conclusions

New silicate-based root-end filling materials, other than QS, have compressive strength similar to MTA. Within the limits of this study, premixed materials and those mixed with antiwashout gel maintain their compressive strength when exposed to biological fluids.  相似文献

Objective

The aim of this study was to investigate the effects of mineral trioxide aggregate (MTA), Sealapex, and a combination of Sealapex and MTA (Sealapex Plus) on the reaction of subcutaneous connective tissue of rats, and on cell viability and cytokine production in mouse fibroblasts.

Material and Methods

The tissue reaction was carried out with dentin tubes containing the materials implanted in the dorsal connective tissue of rats. The histological analysis was performed after 7 and 30 days. Millipore culture plate inserts with polyethylene tubes filled with materials were placed into 24-well cell culture plates with mouse fibroblasts to evaluate the cell viability by MTT assay. ELISA assays were also performed after 24 h of exposure of the mouse fibroblasts to set material disks.

Results

Histopathologic examination showed Von Kossa-positive granules that were birefringent to polarized light for all the studied materials at the tube openings. No material inhibited the cell viability in the in vitro test. It was detected IL-6 production in all root-end filling materials. MTA and Sealapex Plus induced a slight raise of mean levels of IL-1ß.

Conclusions

The results suggest that Sealapex Plus is biocompatible and stimulates the mineralization of the tissue.  相似文献

The aim of this investigation was to compare the quantity of inflammatory cells in response to white and gray mineral trioxide aggregate (MTA) in subcutaneous connective tissue of rats. Fifty Wistar rats were used in this study. Polyethylene tubes were filled with gray or white MTA and empty ones serving as the control group were implanted into subcutaneous tissue and harvested after 7-, 15-, 30-, 60-, and 90-day intervals. Sections of 5 microm were stained with hematoxylin and eosin and observed under a light microscope. Inflammatory reactions were categorized as 0, none (without inflammatory cells); 1, mild (inflammatory cells < or = 25); 2, moderate (25-125 inflammatory cells); and 3, severe (more than 125 inflammatory cells). Statistical analysis was performed with the Kruskal-Wallis test. Both kinds of MTA provoked severe inflammatory reaction after 7 days, which significantly differed from control group (p < 0.05). Our aim was to test whether the presence of three hydraulic calcium silicate dental cements – MTA Angelus, MTA Fillapex, and Theracal LC – in the dental extraction socket of an in vivo model, would affect the levels of aluminium (Al) in the plasma and liver. Following anesthesia, the right upper incisor of each male Wistar albino rat was extracted and polyethylene tubes filled with MTA Angelus, MTA Fillapex, or Theracal LC were inserted into the depth of the extraction socket and gingival tissue was sutured. The rats were killed 7, 30, or 60 d after the operation. Blood and liver samples were obtained from the rats before they were killed, and the levels of Al were measured by atomic absorption spectrometry. Plasma Al levels were higher in the rats in which the mineral trioxide aggregate (MTA) cements were implanted, especially MTA Angelus and MTA Fillapex, compared with control rats. In liver samples, however, the differences in Al level were not statistically significant. Our results show that Al might have been released into the circulation from the three dental cements tested, especially MTA Angelus and MTA Fillapex. Further research should be carried out on the possible biological effects of Al liberated from dental cements.  相似文献

Introduction

The aim of this study was to evaluate the rat alveolar bone response after the implantation of experimental light-cured mineral trioxide aggregate (MTA) or Angelus MTA (Angelus, Londrina, Paraná, Brazil) by histological and fluorescence analysis.

Methods

Thirty Wistar Albino rats were divided into three groups. In the control group, empty polyethylene tubes were inserted into the rat alveolar sockets immediately after extraction. In the other groups, the tubes were filled with light-cured MTA or Angelus MTA. Five animals from each group were injected with calcein on day 7, alizarin on day 14, and oxytetracycline on day 21. On day 30, these animals were killed, and the right hemimaxillas were removed and histologically processed. Half of the maxillas were processed and stained with hematoxylin and eosin. The remaining maxillas were processed for fluorescence analysis and stained with Stevenel blue and alizarin red. New bone was histomorphometrically evaluated using a Merz grid.

Results

The light-cured MTA presented a similar response when compared with Angelus MTA; it was characterized by a mild inflammatory response and complete bone healing. In the light-cured MTA group, the fluorescence areas were more evident at 21 days, showing an increase in bone formation. However, dystrophic mineralization was observed only with Angelus MTA.

Conclusions

It was concluded that both materials present a similar inflammatory response and bone healing, but dystrophic mineralization was observed only with Angelus MTA.  相似文献

AIM: To compare the major constituents present in ProRoot mineral trioxide aggregate (MTA), ProRoot MTA (tooth coloured formula), ordinary Portland cement and white Portland cement using powder X-ray diffractometery. METHODOLOGY: X-ray diffractometery of the four materials was carried out with the divergence and scatter slits set at 1 degree and the receiving slit at 0.10 mm. The scan range was set at 5-70 degrees and continuous scans for the theta-2theta range were run with a scan speed of 2 degrees min(-1). The patterns obtained were then compared with the Powder Diffraction Files (PDF) found in the International Centre for Diffraction Data database. The three strongest peaks were used for the identification of the constituents. The relative intensities were plotted against the angle 2theta and compared with the plots in the PDF. RESULTS: The main constituents were found to be tricalcium silicate, tricalcium aluminate, calcium silicate, and tetracalcium aluminoferrite in all the four cements with the additional presence of Bi2O3 in ProRoot MTA and ProRoot MTA (tooth coloured formula). CONCLUSIONS: The four cements had similar major constituents. Data on Portland cement may be used for the further development or modification of ProRoot MTA in order to improve its physical characteristics and expand its scope of clinical applications.  相似文献

The aim was to evaluate histologically the inflammatory reactions and tissue responses to an experimental tricalcium phosphate cement (TCP) and mineral trioxide aggregate (MTA) when used as repair materials in furcation perforations in dogs. Perforations were performed in 24 mandibular premolars of six anaesthetised dogs and filled either with ProRoot MTA (grey) or TCP. The root canals were subsequently shaped and filled, and the access cavities were closed with a bonded composite resin. The animals were killed at 12 weeks. After radiological examination, the teeth and surrounding structures were processed for light microscopy. Concerning the grades of inflammation, MTA exhibited significantly better results than TCP (chi-square test according to Pearson). No furcation was free of inflammatory cells. Mild inflammation was observed in nine of twelve cases with MTA and only twice in those with TCP. No significant differences were revealed between MTA and TCP in terms of bone reorganization or deposition of fibrous connective tissue (Mantel-Haenszel chi-square test). The grade of radiological examination corresponded with the grade of inflammation or differed by only one grade plus or minus. Perforations located in the furcation of teeth remain an endodontic and a periodontal problem with an uncertain prognosis, in spite of the promising modern materials applied.  相似文献

The aim of this study was to histopathologically examine the biocompatibility of the high-copper amalgam, intermediate restorative material (IRM), mineral trioxide aggregate (MTA), and MTA mixed with chlorhexidine (CHX). This study was conducted to observe the rat subcutaneous connective tissue reaction to the implanted tubes filled with amalgam, IRM, MTA, and MTA mixed with CHX. The animals were sacrificed 15, 30, and 60 days after the implantation procedure. The implant sites were excised and prepared for histological evaluation. Sections of 5 to 6 microm thickness were cut by a microtome and stained with hemotoxylin eosin and examined under a light microscope. The inflammatory reactions were categorized as weak (none or few inflammatory cells < or =25 cells), moderate (>25 cells), and severe (a lot of inflammatory cells not to be counted, giant cells, and granulation tissue). Thickness of fibrous capsules measured five different areas by the digital imaging and the mean values were scored. Amalgam, IRM, and MTA mixed with CHX caused a weak inflammatory response on days 15, 30, and 60. MTA provoked an initial severe inflammatory response that subsided at the 30 and 60 day study period. A clear fibrous capsule was observed beginning from the 15 days in all of the groups. Within the limits of this study, amalgam, IRM, MTA, and MTA mixed with CHX materials were surrounded by fibrous connective tissue indicated that they were well tolerated by the tissues, therefore, MTA/CHX seemed to be biocompatible.  相似文献

Ortho mineral trioxide aggregate (MTA) is a mineral aggregate newly developed for perforation repair, root end filling and pulp capping. The aim of this study was to investigate the levels of cadmium (Cd), copper (Cu), iron (Fe), manganese (Mn), nickel (Ni) and zinc (Zn) in Ortho MTA and ProRoot MTA. A total of 0.2 g of each MTA was digested using a mixture of hydrochloric and nitric acids and filtered. Six heavy metals in the resulting filtrates were analyzed by inductively coupled plasma–optical emission spectrometry (n = 5). The results were statistically analyzed using the Mann–Whitney U‐test. The concentrations of Cd, Cu, Fe, Mn, Ni and Zn in Ortho MTA were 0.10, 7.73, 49.51, 2.58, 0.82 and 10.09 p.p.m., respectively. The concentrations of Cd, Cu, Fe, Mn, Ni and Zn in ProRoot MTA were 0.16, 9.38, 1438.11, 74.51, 18.98 and 4.05 p.p.m., respectively. In conclusion, Ortho MTA had lower levels of Cd, Cu, Fe, Mn and Ni than ProRoot MTA.  相似文献

AIM: To compare solubility, microhardness and radiopacity of ProRoot mineral trioxide aggregate (MTA) with two Portland cements (PC: CEM I and CEM II). METHODOLOGY: Solubility: for standardized samples (n = 12/group) ring moulds were filled with the cements. These samples were immersed in double-distilled water for 1 min, 10 min, 1 h, 24 h, 72 h, and 28 days. Mean loss of weight was determined. Microhardness: five samples of each cement were produced. All samples were loaded with a diamond indenter point with a weight of 100 g for 30 s. Radiopacity: five samples per cement were produced. These samples were tested according to the ISO standards to compare their radiodensity to that of an aluminium step wedge (1-9 mm). Differences between the three materials with respect to their solubility, microhardness and radiopacity were analysed using ANOVA and Student-Newman-Keuls. RESULTS: After 28 days MTA was of low solubility (0.78%) compared with CEM I (31.38%) and CEM II (33.33%). At exposure times >1 min the two PCs were significantly more soluble than MTA (P < 0.05). The microhardness for MTA was significantly higher (39.99 HV; P < 0.001) compared with the two PC (CEM I: 16.32 HV; CEM II: 13.51 HV). MTA was significantly more radiopaque (5.34 mm Al) than CEM I (3.32 mm Al) and CEM II (2.52 mm Al) (P < 0.05), whereas CEM I was significantly more radiopaque than CEM II (P < 0.05). CONCLUSIONS: Mineral trioxide aggregate displayed superior material properties than both Portland cements.  相似文献

Introduction

Procedures used in single-visit or multiple-visit approaches to apical barrier creation were used with an experimental apexification model to test the surface hardness of 3 materials. The purpose of this study was to examine the microhardness of the materials after setting in moist or dry conditions.

Methods

A simulated open apex and periapical environment model was created using polyethylene tubes placed into a porous block filled with phosphate-buffered saline. White ProRoot Mineral Trioxide Aggregate (MTA; Dentsply Tulsa Dental, Tulsa, OK), EndoSequence Root Repair Material (ESRRM; Brasseler USA, Savannah, GA), and Biodentine (BD; Septodont, Louisville, CO) were mixed and placed into the apical 4 mm of the tubes (N = 15). The moist group had a damp cotton pellet above the test materials (mineral trioxide aggregate or ESSRM) with Fuji II LC (GC America, Alsip, IL) sealing the coronal segment. The dry group had gutta-percha placed directly against the test materials with amalgam sealing the coronal segment. After 10 days of storage in 100% humidity at 37°C, samples were sectioned, and microhardness was independently measured at 2 mm and 4 mm from the apical end. Differences were assessed using analysis of variance and a Tukey post hoc test (α = .05).

Results

Analysis of variance analyses showed no significant effect of wet or dry conditions on resultant material hardness. A Tukey post hoc test showed that using ESRRM and BD would not result in a significant difference in hardness, but using MTA would result in statistically significant different hardness values when compared with ESRRM or BD.

Conclusions

Either a moist or dry environment could allow hardening of materials; thus, both methods could be acceptable for clinical treatment procedures.  相似文献

Introduction

The aim of this study was to evaluate tooth discoloration after the use of mineral trioxide aggregate (MTA) and to examine the effect of internal bleaching on discoloration associated with MTA.

Methods

Thirty-two teeth were endodontically treated. Three-millimeter plugs of MTA, ProRoot, Angelus, or Endocem were placed on the access cavities of 24 teeth. Eight teeth served as the control group. After 24 hours, the access cavities were restored, and the tooth color was recorded at baseline and at 1, 2, 4, 8, and 12 weeks. After 12 weeks, the MTA materials were removed under a microscope, and an internal bleaching treatment was performed. After removal of the MTA materials and after a 1-week bleaching treatment, the color changes were measured, and the MTA-dentin interfaces were observed under a microscope.

Results

The ProRoot and Angelus groups displayed increasing discoloration during a period of 12 weeks. The discoloration associated with ProRoot and Angelus was observed at the MTA-dentin interface and on the interior surface of the dentin. However, the Endocem groups demonstrated no significant discoloration (P < .05). No marginal discoloration was observed around the material in the Endocem group. Removal of the discolored MTA was effective for resolving the discoloration in all of the experimental groups (P < .05). However, a subsequent internal bleaching treatment was not significantly effective compared with the removal of MTA.

Conclusions

ProRoot and Angelus caused tooth discoloration. However, Endocem did not affect the contacting dentin surface. Removing the discolored MTA materials contributed more to resolving the tooth discoloration than post-treatment internal bleaching.  相似文献

The purposes of this investigation were 2-fold: to study the physical properties and sealing ability of Viscosity Enhanced Root Repair Material (VERRM); and, to compare them with Mineral Trioxide Aggregate (MTA). VERRM has a composition similar to mineral trioxide aggregate, with handling characteristics and consistency similar to commercially available materials such as IRM and Super EBA. The pH, setting times, solubility, radiopacity, dimensional change upon setting, and apical sealing ability of VERRM were evaluated and compared to that of ProRoot MTA (GMTA) and ProRoot MTA (Tooth Colored Formula) (WMTA). The results showed that VERRM had physical properties similar to WMTA. VERRM and WMTA showed significantly greater dye penetration than GMTA (p < 0.05) when used as a root-end filling material. There was no significant difference in depth of dye penetration between VERRM and WMTA. Further development of VERRM is indicated to produce a biocompatible root-end filling material with superior handling characteristics.  相似文献
