Background Correlations between the malignant potential and prognosis of cancer and abnormal control mechanisms of apoptosis have been discovered in a variety of cancers. Survivin is a member of the inhibiting apoptosis protein family that is abundant in embryonic and carcinoma tissues. We measured the expression of survivin mRNA in gastric cancer to determine whether levels of survivin mRNA expression could serve as an index of malignancy.Methods Expression of survivin mRNA was measured in samples of both gastric cancer and noncancerous tissue from 107 patients. Survivin mRNA was detected by the real-time polymerase chain reaction (PCR) method, and then the relationship between the survivin mRNA level and histological diagnosis was analyzed.Results Expression of survivin mRNA was observed in 105 of 107 cancerous tissues and in 101 of 107 noncancerous tissues. The Mean value of survivin mRNA expression in cancerous tissue was 5.18 ± 1.30, significantly higher (P 0.01) than that in noncancerous tissue, at 4.21 ± 1.48. No significant differences were found in the values of survivin mRNA expression according to histological classification or according to increasing depth of tumor invasion. However, survivin mRNA expression was significantly higher (P 0.01) in patients displaying lymph node metastasis (5.48 ± 1.01) than in patients without the metastasis (4.70 ± 1.55).Conclusions These results indicate that increased survivin mRNA expression begins in the early stages of gastric carcinogenesis. Moreover, the level of survivin mRNA expression may indicate the potential for lymph node metastasis in patients with gastric cancer.  相似文献

目的 探讨下调survivin基因对卵巢癌顺铂(DDP)耐药细胞株SKOV3/DDP细胞生长、凋亡及药物敏感性的影响.方法 构建survivin基因的小干扰RNA(siRNA)表达载体pSilencer-survivin,用脂质体方法转染SKOV3/DDP细胞株,另设未转染组和转染pSilencer-control组作为对照.显微镜下观察转染前后细胞的变化,逆转录聚合酶链反应(RT-PCR)和Western blot分别检测survivinmRNA及蛋白的表达,二苯基溴化四氮唑蓝(MTT)法检测细胞生长情况,流式细胞仪检测细胞凋亡及周期的变化.各组细胞加入DDP,检测其对DDP药物敏感性的影响.结果 survivin siRNA可明显下调SKOV3/DDP细胞中survivin mRNA及蛋白表达水平.SKOV3/DDP细胞生长受到明显抑制,生长曲线低平.转染48 h后,转染pSilencer-survivin组细胞凋亡率为19.1%,明显高于末转染组(2.6%)和转染pSilencer-control组(3.5%),G1/G0期细胞所占的比例增高,而G2/M期细胞所占的比例降低.转染pSilencer-survivin组细胞的IC50明显降低,为1.16 μg/mi.结论 survivin siRNA可下调SKOV3/DDP细胞中survivin的表达,使细胞生长减慢,凋亡增加,对DDP的药物敏感性增强.  相似文献

目的探讨survivinmRNA、caspase-3蛋白在食管鳞癌中的表达及其与细胞凋亡的关系。方法分别采用RT-PCR和免疫组化技术检测38例食管鳞癌及其对应的正常组织中survivinmRNA和caspase-3蛋白的表达,运用TUNEL方法检测细胞凋亡。结果73.68%食管鳞癌组织呈survivinmRNA阳性表达,显著高于正常组织,并与食管鳞癌的分化程度、临床分期有关,与年龄、性别、淋巴结转移无关;caspase-3蛋白在食管鳞癌和正常组织中的阳性表达率分别为26.32%和89.47%,差异有显著性,并与食管鳞癌的分化程度有关,与年龄、性别、淋巴结转移和临床分期无关;在食管鳞癌组织中,survivinmRNA和Caspase-3蛋白表达呈负相关;survivinmRNA阳性的食管鳞癌组中细胞凋亡指数明显低于survivinmRNA阴性组,(P<0.01),caspase-3蛋白阳性的食管鳞癌组中平均细胞凋亡指数明显高于caspase-3蛋白阴性组(P<0.01)。结论survivin、caspase-3参与了食管鳞癌的发生、发展,可以作为食管鳞癌预后的指标,survivin通过抑制caspase-3的活性而发挥其抑制细胞凋亡的作用是其主要的分子机制。  相似文献

目的探讨p21活化激酶4(PAK4)在人胃癌组织和胃癌细胞系中的表达及与临床病理因素的关系。方法免疫组化法检测95例人胃癌组织中PAK4蛋白的表达;Westernblot法检测40例新鲜胃癌组织和相应的癌旁正常胃黏膜组织以及7种人胃癌细胞系中PAK4蛋白的表达。结果95例人胃癌组织中,PAK4蛋白的阳性表达率为67.4%(64/95),其中62例(96.9%)阳性染色位于细胞浆中,仅有2例(3.1%)位于细胞膜上。PAK4蛋白的阳性表达率与胃癌TNM分期(Χ^2=10.426,P〈0.01)和淋巴结转移(Χ^2=4.912,P〈0.05)有关。新鲜胃癌组织中PAK4蛋白的表达量显著高于癌旁正常胃黏膜组织(t=-5.978,P〈0.01),且随胃癌TNM分期的进展,其表达量逐渐增加。7种人胃癌细胞系中均有PAK4蛋白的表达,与GES-1细胞系相比,PAK4蛋自在ACTS、BGC823、MKN45和N87细胞系中呈高表达,而在MGC803、MKN1和SGC7901细胞系中呈低表达。结论PAK4蛋白的过度表达可能与人胃癌的发生发展及预后有关。  相似文献

目的 探讨胃癌患者外周血中Survivin mRNA水平与幽门螺杆菌(Hp)感染的关系。方法 应用实时荧光定量qPCR技术检测胃炎及化疗前胃癌各50例患者外周血中Survivin mRNA表达,并采用快速尿素酶试验与Giemsa 染色2种方法检测Hp感染情况。结果 胃炎患者外周血中未检测到Survivin mRNA表达;胃癌组患者Survivin表达检测率为84.0%(42/50),外周血Survivin mRNA表达水平的中位数为0.46。Survivin表达水平与胃癌患者的年龄、性别及淋巴结转移均无关(P>0.05),而与TNM分期有关(P<0.0001)。胃癌患者Hp感染率为80.0%(40/50),显著高于胃炎患者的58.0%(29/50),差异有统计学意义(P<0.05)。Hp阳性的胃癌患者外周血Survivin mRNA表达水平为0.578±0.507,显著高于Hp阴性患者的0.194±0.419,差异有统计学意义(P<0.05)。经Spearman相关分析,胃癌患者Survivin mRNA表达与Hp感染呈正相关(r=0.464, P<0.05)。结论 Hp感染及Survivin与胃癌发生可能有关,Survivin可能参与了Hp感染性胃癌的发展。  相似文献

Survivin, an anti-apoptosis gene that is abnormally overexpressed in a variety of human tumors, may play an important role in the carcinogenesis and drug resistance of cancer. This study was designed to explore the effects of liposome-survivin antisense oligonucleotide (Lip-ASODN) on the growth and apoptosis of human ovarian cancer cell lines, A2780 and SKOV3. To investigate the use of survivin as a therapeutic target on ovarian cancer, we carried out transfections with Lip-ASODN to induce apoptosis in ovarian cancer cell lines, A2780 and SKOV3. The expression of survivin mRNA and relative protein were evaluated separately by quantitative real-time RT-PCR and Western blot analysis. Cell proliferation inhibition was determined by methyl thiazolyl tetrazolium (MTT) assay, and the induced cell apoptosis was examined using flow cytometry (FCM) after Lip-ASODN transfection. Our results showed that the overexpression of survivin led to infinite carcino-proliferation, and survivin expression in the survivin-positive ovarian cancer cell line A2780 and SKOV3 cells was significantly and gradually reduced when transfected with Lip-ASODN at concentrations of 200, 400 and 600 nM by degrees. Lip-ASODN transfection induced greater apoptosis rates in the human ovarian cancer cell lines A2780 and SKOV3 (p<0.05). The growth inhibition and apoptotic rates of tumor cells change when treated with different concentrations of Lip-ASODN. The cell growth inhibition peak rate was reached when increasing Lip-ASODN concentration to 600 nM. Furthermore, time course evaluation showed that survivin protein expression was inhibited by Lip-ASODN within 12 h after transfection. We concluded that down-regulation of survivin by a targeted antisense oligonucleotide appears to be an effective gene therapy approach in the treatment of ovarian cancer.  相似文献

目的检测膀胱癌组织中Survivin、VEGF表达的相关性;观察Survivin反义寡核苷酸(ASODN)对人膀胱癌的作用研究。方法采用免疫组化和蛋白印迹技术,检测41例膀胱癌组织及癌旁非癌组织中Sur-vivin和VEGF的表达;应用SurvivinASODN作用后收集各组细胞。观察细胞形态变化及超微结果变化,检测各组Survivin表达情况、AI和PI及ASODN对细胞的生长抑制率。结果41例患者中29例(67.4%)表达Survivin蛋白,其中25例(61.0%)VEGF表达阳性或弱阳性;在癌周非癌组织中未检测到Survivin蛋白的表达。Survivin与膀胱癌的转移有关,P=0.002;Survivin蛋白的表达与VEGF蛋白的表达有相关性,P=0.001。电镜下ASODN组可见典型凋亡样改变;各ASODN转染组细胞Survivin表达减弱,呈时间与剂量依赖性;ASODN转染组AI和PI与对照组相比差异有统计学意义,P=0.012,而AI和PI在各对照组间差异无统计学意义,P〉0.05。结论Survivin在膀胱癌组织中的表达能促进膀胱癌的发生与发展,并可能通过上调VEGF的表达而促进膀胱癌发展与转移。Survivin ASODN转染后能下调Survivin蛋白表达,诱导癌细胞凋亡,抑制癌细胞增殖。  相似文献

PURPOSE: Survivin is a member of the inhibitor-of-apoptosis gene family and is known to be overexpressed in a number of tumor types. The aim of this study was to evaluate the prognostic value of survivin protein expression in tumor tissue extracts in a group of well-characterized soft-tissue sarcoma (STS) patients. EXPERIMENTAL DESIGN: In this investigation, malignant tissue samples from 63 STS patients as well as from a panel of tumor cell lines were investigated, with nonmalignant tissues serving as controls. The survivin protein level was quantified by a novel ELISA and by Western blot analysis. Results obtained by both methods were compared with clinicopathological parameters regarding tumor grade and tumor entity, and they were then correlated to survival in a multivariate Cox regression model. RESULTS: High survivin levels were detected by ELISA and Western blot analysis in tumor tissue extracts and in lysates of tumor cell lines. None or only weak expression of survivin protein was found in nonmalignant cells and tissues. When comparing survivin values obtained by ELISA or Western blot, we found a significant correlation between both methods (P = 0.013, Pearson test). Our findings revealed that, in multivariate Cox regression analyses, survivin levels measured by ELISA and Western blot were significantly associated with tumor-related death in STS patients (P = 0.001, RR = 19.8, and P = 0.004, RR = 5.1, respectively). However, in a direct comparison of both survivin protein detection assays, we found a higher sensitivity and a stronger correlation to prognosis in survivin ELISA as compared with the Western blot assays. Furthermore, a higher tumor grade and more aggressive STS entity showed an elevated survivin protein expression level. CONCLUSION: Altogether, an elevated survivin content in tumor tissue extracts has a significant and independent negative predictive value on the survival-rate of STS patients. This finding corresponds well to data obtained for the mRNA level of survivin, as shown previously (M. Kappler et al., Int. J. Cancer, 95: 360-363, 2001).  相似文献

目的:构建靶向存活素(survivin)的干扰RNA(siRNA)质粒,观察其对A549细胞增殖、凋亡以及顺铂敏感性的影响。方法:应用pSilencerU6质粒构建survivinsiRNA干扰质粒,RTPCR和Western blotting检测survivin mRNA和蛋白的表达,DAPI染色法检测细胞的凋亡,MTT法检测细胞的增殖。结果:成功构建了survivinsiRNA干扰质粒。SurvivinsiRNA质粒转染可明显下调A549细胞中survivin mRNA和蛋白的表达、抑制A549细胞的增殖、促进细胞的凋亡、增强A549细胞对顺铂的敏感性。结论:SurvivinsiRNA沉默survivin在肺癌细胞中的表达能够抑制肿瘤细胞的增殖、促进细胞凋亡,并能增强肿瘤细胞对化疗药物顺铂的敏感性,survivin 可作为肺癌治疗的潜在靶点。  相似文献

PURPOSE: Survivin, as an inhibitor of apoptosis, is undetectable in normal tissues but expressed in tumors. Survivin expression in rectal cancer patients who have undergone preoperative radiotherapy (RT) alone has not been studied. We analyzed the relationships of survivin expression to RT, clinicopathologic variables, apoptosis, and p53 expression in rectal cancer patients who participated in a trial of preoperative RT. METHODS AND MATERIALS: Survivin was immunohistochemically examined in 98 rectal tumors (74 had adjacent normal mucosa). Of 98 patients, 57 underwent surgery alone and 41 underwent RT before surgery. RESULTS: Survivin positivity was related to worse survival, independent of Dukes' stage, local and distant recurrence, differentiation, gender, age, apoptosis, and p53 expression (p = 0.02). Survivin was not associated with survival in the patients without (p = 0.08) or with (p = 0.19) RT. After RT, survivin tended to be increased in adjacent normal mucosa (p = 0.057) but not in tumors (p = 0.71). CONCLUSION: Survivin was independently related to survival in rectal cancer patients who participated in a trial of preoperative RT, but not in either treatment group (surgery alone or surgery plus RT). Whether the effect of survivin on tumors is associated with RT and further related to patient survival needs to be investigated in a larger number of patients.  相似文献
