BACKGROUND: Although hepcidin is proposed as a regulator of iron absorption, this has not been assessed in humans. OBJECTIVE: Our objective was to assess the relation between serum or urinary prohepcidin and iron absorption in healthy premenopausal women. DESIGN: The subjects were 28 healthy women aged 22-51 y with normal hemoglobin concentrations (120-152 g/L). Absorption of 0.5 mg Fe with 0.2 microCi 59Fe tracer, both as FeSO4, was measured by whole-body scintillation counting 13 d after oral administration. Fasting blood and urine samples were collected the day of and 16 wk after the absorption measurement. Serum and urinary prohepcidin concentrations were measured by an enzyme-linked immunosorbent assay by using an antibody against amino acid residues 28-47 of the proregion. RESULTS: Mean (+/-SD) iron absorption was 36 +/- 19% (range: 4-81%), and serum ferritin (geometric x) was 27 microg/L (range: 4-122 microg/L), as commonly observed in healthy premenopausal women. Serum prohepcidin was 196 microg/L (range: 99-376 microg/L) and, in contrast with urinary prohepcidin, was relatively consistent for the women between 0 and 16 wk. Serum prohepcidin correlated directly with serum ferritin (R2 = 0.28, P < 0.01) but was unrelated to 59Fe absorption, in contrast to serum ferritin (R2 = 0.33, P < 0.01). CONCLUSIONS: Serum prohepcidin concentrations were relatively stable within subjects and correlated with serum ferritin. However, unlike serum ferritin, neither serum nor urinary prohepcidin concentrations were related to iron absorption in healthy women.  相似文献

The mammalian lignans (a form of phytoestrogens), metabolically derived by the intestinal microflora from dietary precursors, may have several health benefits. Information concerning their dietary sources and bioavailability is scarce. We assessed lignan intake via a 24-h dietary recall (n = 2852) and determined serum enterolactone (EL) concentration (n = 1784) in 25- to 64-y-old Finnish men and women participating in a national survey in 1997. Mean intake of lignans [sum of matairesinol (MAT) and secoisolariciresinol (SECO)] in men and women was 173 microg/d (19 microg/MJ) and 151 microg/d (23 microg/MJ), respectively. SECO made up over two thirds of the total lignan intake. The major sources of SECO were fruit, berries and cereals, whereas MAT derived almost exclusively from cereals. Lignan intake was positively associated with serum EL concentration (r = 0.19, P < 0.0001), i.e., the mean EL concentration in the highest quintile of lignan intake was 50% higher than that in the lowest quintile. We conclude that lignans are common components of the Finnish diet, although the mean daily intake is low (<0.2 mg). The main dietary sources of lignans, i.e., whole grain, vegetables and fruits, are foods commonly associated with lower risk of cardiovascular diseases and cancer. Serum EL concentration is a feasible biomarker of lignan intake.  相似文献

High DDE and DDT concentrations were found to be associated with shortened menstrual cycle length in Laotian immigrants to the United States. We examined this issue in a sample of young Chinese women. A total of 60 women aged 20-24 years were enrolled in three maternal and child health clinics (20 from urban, 20 from suburban, 20 from rural) in Shanghai, China, and vicinity, in 1998. Of these women, 47 who did not use hormonal contraceptives and had valid menstrual cycle characteristics were included in the analysis for associations among serum DDE and DDT concentration and menstrual cycle length, duration of menses, and heaviness of menstrual flow. In univariate analysis, higher p,p'-DDE concentration was associated with longer menstrual cycle length (0.66 day per 10 microg/L, 95% confidence interval [CI]: 0.21, 1.11 day). With adjustment for age, body mass index, education, occupation, and resident location, the estimate was 0.42 day (95% CI: -0.35, 1.19 day). p,p'-DDE was not associated with duration of menses or heaviness of menstrual flow. Neither p,p'-DDT nor o,p'-DDT were associated with menstrual cycle length, duration of menses or heaviness of menstrual flow. The study largely suggests no association between DDE and DDT concentrations and menstrual cycle characteristics in young Chinese women, though the weak-to-no correlation of DDE with menstrual cycle length merits further study.  相似文献

Purpose

Increased iron storage, as measured by circulating ferritin, has been linked to an increased risk of various diseases including diabetes. We examined the association of circulating ferritin with serum adiponectin, leptin, resistin, plasminogen activator inhibitor-1 (PAI-1), and visfatin levels.

Methods

We conducted a cross-sectional study among 429 Japanese employees (284 men and 145 premenopausal women, mean age: 42.5 ± 10.5 years). Serum adipokines were measured using Luminex suspension bead-based multiplexed array, and serum ferritin was determined using a chemiluminescence immunoassay. Multivariable regression analysis was performed to calculate mean concentrations of adipokine according to the tertile of ferritin concentrations with adjustment for potential confounders.

Results

Leptin and visfatin concentrations increased with increasing ferritin concentrations in men after multivariable adjustment of physical activity, smoking, alcohol use, and body mass index (P for trend = 0.02 and 0.01 for leptin and visfatin, respectively). Serum ferritin concentrations were inversely and significantly associated with adiponectin in women (P for trend = 0.01). Resistin and PAI-1 were not appreciably associated with ferritin concentration.

Conclusions

Increased iron storage may be associated with higher circulating concentrations of leptin and visfatin in men and with lower concentrations of adiponectin in women.

We investigated whether weekly iron supplementation was as effective as the national daily iron supplementation program in Indonesia in improving iron status at near term in pregnancy. In addition, we examined whether weekly vitamin A and iron supplementation was more efficacious than weekly supplementation with iron alone. One group of pregnant women (n = 122)was supplemented weekly with iron (120 mg Fe as FeSO4) and folic acid (500 microg); another group (n = 121) received the same amount of iron and folic acid plus vitamin A [4800 retinol equivalents (RE)]. A third ("daily") group (n = 123), participating in the national iron plus folic acid supplementation program, was also recruited. Data on subjects with complete biochemical data are reported (n = 190). At near term, hemoglobin concentrations increased, whereas serum ferritin concentrations decreased significantly in the weekly vitamin A and iron group, suggesting that vitamin A improved utilization of iron for hematopoiesis. Iron status in the weekly iron group was not different from that of the "daily" group. However, iron status decreased with daily supplementation if <50 iron tablets were ingested. Serum transferrin receptor concentrations increased in all groups (P < 0.01). Serum retinol concentrations were maintained in the weekly vitamin A and iron group, but decreased in the other two groups (P < 0.01). Thus, delivery of iron supplements on a weekly basis can be as effective as ona daily basis if compliance can be ensured. Addition of vitamin A to the supplement improved hemoglobin concentration.  相似文献

The percent absorption of iron from four dietary sources was compared in 2018 human subjects with three indicators of iron status, serum ferritin concentration, percent saturation of plasma transferrin and iron absorption from a reference dose of ferrous sulfate. Higher correlation coefficients (r) were obtained by comparing dietary iron absorption with the reference dose absorption rather than with serum ferritin; for example, r = +0.61 and r = -0.38, respectively, for a meat and vegetable meal. However, in practice serum ferritin is almost as efficient as the reference dose absorption in estimating dietary iron absorption, because the 95% confidence limits calculated from the regression equations were very similar. The values of r calculated for iron absorption versus transferrin saturation were comparable to those obtained with the other indicators only in the range of transferrin saturation values below 25%, whereas in more iron-replete subjects (transferrin saturation greater than 25%), this correlation virtually disappeared. This indicates that, although both serum ferritin and transferrin saturation reflect iron status in iron-depleted subjects, the control of iron absorption in iron-replete subjects is more dependent on iron stores as reflected in the serum ferritin concentration than the percent saturation of transferrin.  相似文献

To evaluate the effects of breakfast habits on the quality of life in young women in Japan, the relation between breakfast habits and menstrual and intestinal disorders was estimated by responses to a questionnaire. Subjects were recruited from young Japanese female students of Ashiya College ranging in age from 18 to 20 years. Breakfast habits were classified into three groups (group I, having breakfast every morning, n=289; group II, having breakfast one to six times per week, n=106; group III, having breakfast less than once a week, n=44). The intensity of dysmenorrhea was classified into three grades (score 1, free of pain or painful, but without need for analgesic; score 2, painful, requiring analgesic; score 3, painful, not relieved by analgesic). The grades of bowel movement were classified into three groups (score 1, no more than once a week; score 2, two to six times a week; score 3, every day). Group II and group III had significantly higher scores for dysmenorrhea compared with group I. Students in group III showed a tendency toward constipation. Since dysmenorrhea is a risk factor for psychological disorders as well as gynecological diseases, the present study suggests the importance of eating breakfast to the quality of life of young women.  相似文献

The n-3 series is considered as anti-inflammatory, and some studies have reported increased plasma n-3 polyunsaturated fatty acid pattern in chronic inflammatory conditions. Studying a stratified random sample of university women (n = 869) we investigated the correlates of proficiency (i.e., the correctness or competence) of breast self-examination (BSE). A 19-item BSE quality index score was developed based on the recommended steps a woman should perform when doing the exam in the three BSE positions. A comprehensive set of independent variables including knowledge, attitudes and perceptions, personal characteristics, and environmental factors were studied. While proficiency scores for both undergraduates and graduates were relatively low, the mean score for graduates was significantly higher than for undergraduates. This study corroborates the few recent studies that indicate that frequency of performance is not highly correlated with proficiency (relationships were r = .14 and .04, respectively, for undergraduate and graduate women). Bivariate and multivariate regression analyses demonstrate that the relevance of such factors varies by age (undergraduate and graduate). Results of the regression model indicate that the skill knowledge variable regarding time of month BSE should be performed, awareness of BSE, perceived effect of cancer detection, and perceived benefit of BSE were predictors of more proficient practice for undergraduates. Knowledge of time of month to do BSE and mammogram as a detection technology and discussion of BSE with others were important for graduate women. Little is known about the relation of dietary factors to circulating C-reactive protein (CRP) concentrations in young adults and non-Western populations. We cross-sectionally examined associations between dietary intake and serum CRP concentrations in young Japanese women. The subjects were 443 female Japanese dietetic students aged 18 to 22 years. Dietary intake was assessed with a validated, self-administered, comprehensive, diet history questionnaire. Serum CRP concentrations were measured by highly sensitive nephelometry. The prevalence of elevated CRP (≥1 mg/L) was 5.6%. After adjustment for possible confounding factors including body mass index, a significant inverse association was seen between total n-3 polyunsaturated fatty acid intake and elevated CRP. The multivariate adjusted odds ratios of elevated CRP for women with intake below and above the median (1.1% of energy) were 1.00 and 0.33 (95% confidence interval, 0.13-0.82; P = .02), respectively. Intake of eicosapentaenoic acid + docosahexaenoic acid and α-linolenic acid was not associated with elevated CRP concentrations (P = .62 and P = .27, respectively). Vitamin C intake was independently inversely associated with elevated CRP, although the association was nonsignificant (P = .10). No clear associations were observed for other dietary factors examined including total fat, saturated fatty acids, monounsaturated fatty acids, polyunsaturated fatty acids, total dietary fiber, soluble dietary fiber, insoluble dietary fiber, and magnesium; fruits, vegetables, and fish and shellfish; and dietary glycemic load (P = .27 to P = .99). In conclusion, total n-3 polyunsaturated fatty acid intake showed an independent inverse association with elevated serum CRP concentration in a group of young Japanese women.  相似文献

Meat is a well-known enhancer of iron absorption, yet the molecular entity mediating the effect remains obscure. Recently published data indicate that highly acidic sulfated glycosaminoglycans (GAG) from fish and chicken muscle are effective stimulants of iron uptake in Caco-2 cells. Two fully randomized stable isotope studies with crossover design were performed in a group of 16 apparently healthy young women to assess the effect of purified sulfated and unsulfated GAG on human iron absorption. Iron absorption was measured on the basis of erythrocyte incorporation of (57)Fe or (58)Fe 14 d after the administration of labeled semisynthetic meals (SSM) based on egg albumin, corn oil, maltodextrin, and water. The meals were consumed with or without added sodium hyaluronate (NaH, 300 mg) or chondroitin sulfate (CS, 360 mg) as representative unsulfated and sulfated GAG, respectively. The level of GAG added was 3 times (NaH) to about 10 times (CS), the amount expected to be present in 150 g beef muscle. Geometric mean iron absorption from SSM containing NaH (21.2%) or CS (19.4%) did not differ from that of SSM without GAG (19.5 and 20.3%, respectively). NaH and CS at those levels do not affect human nonheme iron absorption.  相似文献

BACKGROUND: Dietary calcium and milk intakes at specific ages may influence bone mineral measures at specific sites during development of peak bone mass. OBJECTIVE: Relations of previous milk intake and current calcium intake to current bone mineral measures were investigated in young women. DESIGN: A food-frequency interview and recall of previous milk intake from early childhood to 12 y of age and during adolescence (13-19 y) were completed in a cross-sectional analysis in young women (age 18-31 y; n = 224). Three levels of previous milk intake were defined: 1) infrequently or never, 2) sometimes, and 3) at every or almost every meal. Total body (TB), femoral neck, radius (R), and spine (S) bone mineral density (BMD) and bone mineral content (BMC) were determined by using dual-energy X-ray absorptiometry. RESULTS: Childhood and adolescent milk intakes were positively correlated (r = 0.66). Childhood and adolescent milk intakes correlated with current calcium intakes (r = 0.26 and 0.33, respectively). Adolescent milk intake correlated with RBMD (r = 0.16). When weight was controlled for, adolescent milk intake correlated with TBBMD (r = 0.16), TBBMC (r = 0.21), SBMC (r = 0.16), RBMD (r = 0.18), and RBMC (r = 0.15). Current calcium intakes correlated with SBMC (r = 0.17). Regression analyses supported these results. CONCLUSIONS: Results were consistent with the hypothesis that higher milk intake during adolescence is associated with greater total body, spine, and radial bone mineral measures during development of peak bone mass, whereas current calcium intakes may influence SBMC. In addition, milk intake at a younger age may contribute to similar habits of milk intake later in life.  相似文献

The mechanism by which homocysteine (Hcy) causes endothelial dysfunction is probably mediated by oxidative stress. The aim of this study was to evaluate the effect of oxidative stress on endothelial function in young and elderly hyperhomocysteinemic (HHcy) men. A total of 35 HHcy (Hcy > 15 micro mol/L), young (n = 15; 20-40 y) and elderly men (n = 20; > 65 y) and 33 normohomocysteinemic (NHcy; controls) young (n = 14) and elderly (n = 19) men (Hcy < 13 micro mol/L), without classic cardiovascular risk factors were recruited. Serum Hcy, folate, and vitamin B-12, whole-blood glutathione, plasma total antioxidants status, TBARS, and 8-F(2alpha) isoprostanes were determined. Noninvasive ultrasound measurements of endothelium-dependent (EDVR) and -independent dilatation (EIVR) were performed. EDVR, EIVR, and markers of oxidative stress did not differ among the groups. Folate concentrations were higher in elderly than in young men (P < 0.001), independent of Hcy concentrations. Vitamin B-12 concentrations were lower in HHcy than in NHcy elderly men (P < 0.045). EDVR was correlated with folate concentrations in young men (r = 0.40, P = 0.04) and negatively with BMI in elderly men (r = -0.52, P = 0.002). In the present study, HHcy with normal serum folate concentrations was not associated with poor EDVR or oxidative stress in healthy young and elderly men.  相似文献

1. A combination of a mineral balance and a radioactive technique has been used to study the effects of increased levels of dietary calcium and phosphorus on the Ca and P metabolism of young and mature wether sheep previously made Ca-deficient. 2. Ca absorption was directly related to Ca intake until an absorption maximum was reached. Young animals absorbed dietary Ca more efficiently than mature animals and had a much higher maximum rate of absorption. Apparent P absorption was directly related to P intake and maximum P absorption was also higher for the young animals. 3. Excretion of Ca in the urine and into the intestine was not altered by changes in Ca intake. Excretion of P in the urine, however, varied considerably and it is suggested that this process may be largely responsible for P homoeostasis. 4. Ca retention was directly related to Ca absorption. Maximum retention was considerably higher for the young animals than for the mature animals and occurred for each group of animals at exactly the same dietary intake as maximum Ca absorption. 5. Ca and P were retained in a constant ratio of 1-25:1 and the results suggested that P retention was controlled by the rate of Ca retention. 6. Bone turnover was higher for the young than for the mature animals. Animals mobilized skeletal Ca when receiving the Ca-deficient diet and replaced their Ca losses when the dietary Ca was plentiful by changing the relative rates of bone resorption and bone accretion. Although bone resorption was mainly responsible for Ca homoeostasis, bone accretion appeared to play some part in the young animals. 7. Maximum retention of Ca occurred just as the rate of bone resorption became zero and was equal to the rate of bone accretion. The lower maximum retention of Ca for mature animals was due to a lower rate of bone accretion. 8. Results suggested that when bone resorption ceased, Ca absorption was regulated by homoeostatic mechanisms at a level just sufficient to maintain maximum retention, and that the maximum rates of absorption measured may not have been true maximum rates. 9. The consequences of a relationship between bone accretion and maximum Ca retention are discussed with reference to older, pregnant and lactating animals.  相似文献

OBJECTIVES: High glycemic index (GI) carbohydrates may increase brain serotonin, which in turn acts to alleviate premenstrual symptoms, because, although the main determinant of brain serotonin concentration is a high plasma ratio of tryptophan to other large neutral amino acids, a high-GI diet has been shown to increase this ratio. In this observational cross-sectional study, we investigated associations between dietary GI and other dietary carbohydrates and premenstrual symptoms. METHODS: Subjects were 640 female Japanese dietetic students 18-22 y of age. Dietary carbohydrates were assessed using a validated, self-administered, comprehensive diet history questionnaire. Menstrual cycle symptoms were assessed using the retrospective version of the Moos Menstrual Distress Questionnaire (MDQ). Independent associations of dietary GI and glycemic load and intake of available carbohydrate and dietary fiber with the MDQ total score and subscale scores (pain, concentration, behavioral change, autonomic reactions, water retention, and negative affect) in the premenstrual phase (expressed as percentages relative to those in the intermenstrual phase) were examined. RESULTS: Dietary GI was independently inversely associated with total MDQ score in the premenstrual phase (P for trend = 0.02). Dietary GI also showed independent and inverse associations with several MDQ subscale scores in the premenstrual phase, including concentration, autonomic reactions, and water retention (P for trend < 0.05). Conversely, dietary glycemic load and intake of available carbohydrate and dietary fiber were not associated with any of the MDQ scores in the premenstrual phase. CONCLUSION: Dietary GI was independently associated with decreased premenstrual symptoms in a group of young Japanese women.  相似文献

OBJECTIVE: Little is known about the relation of modifiable dietary factors to circulating leptin concentrations, particularly in young adults and non-Western populations. We examined cross-sectional associations between nutrient and food intake and serum leptin concentration in young Japanese women. METHODS: Subjects were 424 female Japanese dietetic students 18-22 y of age. Intake of macronutrients (protein, total fat; saturated, monounsaturated, and polyunsaturated fatty acids; and carbohydrate), dietary fiber, and 12 food groups was assessed with a validated, self-administered, comprehensive, diet history questionnaire. Fasting blood samples were collected, and serum leptin concentrations were measured by radioimmunoassay. RESULTS: For nutrients, only dietary fiber was a significant determinant of serum leptin concentration. Increasing dietary fiber intake was associated with lower serum leptin concentration independent of potential confounding factors, including body mass index (mean serum leptin concentrations in the lowest and highest quintiles of dietary fiber intake were 8.6 and 7.5 ng/mL, respectively; P for trend = 0.026). Vegetables and pulses were the only foods significantly associated with serum leptin concentration, with higher intakes independently associated with lower concentrations (mean serum leptin concentrations in the lowest and highest quintiles of intake were 8.1 and 7.0 ng/mL, P for trend = 0.007, for vegetables and 8.8 and 7.6 ng/mL, P for trend = 0.019, for pulses, respectively). CONCLUSION: Intake of dietary fiber, vegetables, and pulses showed an independent inverse association with serum leptin concentration in a group of young Japanese women.  相似文献

A study is described of influenza A anti-neuraminidase antibodies in the sera of young people of three different groups. Each serum was individually absorbed with viruses containing the N2 neuraminidases of 1957, 1968 and 1972. Rabbit antisera prepared against the viruses were similarly absorbed. Results obtained with the animal sera suggested that these neuraminidases were antigenically distinct, but the human sera had a broader range of anti-neuraminidase activity and gave indication of asymmetric antigenic relationships. Earlier workers who surveyed anti-haemagglutinin antibodies reported that the virus of primary infection absorbed all antibodies, and the virus of secondary infection only those directed against itself. We too found that the virus of secondary infection absorbed only homologous anti-neuraminidase antibody. However, although the primary infecting virus did absorb some secondary antibody, this absorption was incomplete and it lessened with the lengthening of the time interval between the primary and secondary infecting viruses. A similar pattern was seen with anti-haemagglutinin antibodies. Absorption of anti-neuraminidase antibodies from human sera proved much more difficult than absorption of anti-haemagglutinin antibodies particularly after repeated influenza virus infections. The relative rarity of antigenic shift in the neuraminidase subunit also creates problems in the interpretation of results of serum neuraminidase antibody surveys.  相似文献

The purpose of this study was to assess nutrition knowledge during early and late postpartum in a sample of low-income and minority women, and to determine if that knowledge had any relationship to weight retention at 1-year postpartum. A questionnaire was developed and validated in a sample of 151 low-income new mothers. This instrument was then administered to a separate sample of mothers (n=140) of similar demographics to assess nutrition knowledge at 0 to 1 days and 12 months postpartum. In addition, a survey of nutrition information sources was administered at both time points. Nutrition knowledge was compared with 12-month postpartum weight retention and demographic variables. Women who retained less than 5% of weight gained during pregnancy had greater knowledge of nutrition at 0 months (53% vs 49%, P<0.05) and 12 months (55% vs 51%, P<0.05) than those who retained 5% or more of weight gained during pregnancy. Whites had higher nutrition knowledge scores than non-Hispanic blacks and Hispanics. Women who lactated 6 months or more had more knowledge than those who lactated less than 6 months. Higher knowledge test scores were observed among women who used the Internet and books/magazines as their source for information. These results indicate that assessment of nutrition knowledge in early postpartum can identify women at risk for significant weight retention.  相似文献
