Objective  

CD4+CD25+ T regulatory (Treg) cells are a population of T cells which suppress an overactive immune system. CCR4 is a chemokine receptor involved in the recruitment of lymphocytes. Nasopharyngeal carcinoma (NPC) is resistant to immunosurveillance, owing to the increased number of tumor-infiltrating Treg cells which are recruited to the tumor by CCR4.  相似文献

肝癌微环境中CD4+CD25+Treg细胞与肿瘤免疫细胞的关系   总被引:2,自引:0,他引:2  
[目的]探讨肝癌微环境中Treg细胞的数量增多与HCC临床分期晚的可能原因。[方法]双重酶标免疫组织化学方法检测52例HCC组织中CD4^+CD25+Treg细胞以及CD4^+CD25-T(CD4^+T)细胞分布,免疫组化EnVision法检测20例HCC组织中CD8+T细胞分布。[结果]正常肝脏组织中未发现Treg细胞。HCC组织中Treg细胞数量较癌旁组织明显增多(P=0.000),且肝癌组织中Treg细胞的数量与其浸润性CD4+T淋巴细胞的数量以及CD4+T/CD8^+T比值呈显著负相关(r=-0.539,P=0.014;r=-0.545,P=0.000),而与浸润性CD8^+T淋巴细胞的数量无明显相关性(f=-0.403,P=0.078)。[结论]Treg细胞在体内可能通过细胞接触的方式抑制CD8^+T淋巴细胞的增殖来抑制肿瘤局部的免疫,去除或减少HCC微环境中浸润件Tree细胞数量可能提高肿瘤局部免疫治疗效果。  相似文献

 目的 分析CD4+CD25+ FOXP3+调节性T细胞(Treg)与CD4+T、CD8+T在结直肠癌(colorectal carcinoma, CRC)组织中的分布及其与临床病理特征之间的关系。方法 收集42例CRC新鲜手术标本,应用冰冻切片、免疫组织化学SP法检测肿瘤组织和癌旁组织中FOXP3+、CD4+T和CD8+T阳性细胞数。结果 CRC患者肿瘤组织中FOXP3表达水平显著升高,与癌旁组织相比差异有统计学意义(P<0.01);中低分化组Treg细胞数明显高于高分化组(P<0.01);淋巴结转移组Treg细胞数明显高于无淋巴结转移组(P<0.05);癌巢内CD4+、CD8+T细胞数及CD4+/CD8+值显著低于间质(P<0.01);Ⅲ+Ⅳ期、淋巴结转移组癌巢内CD4+/CD8+比值显著低于Ⅰ+Ⅱ期及无淋巴结转移组(P<0.05);CRC中Treg数量与癌巢内CD4+/CD8+比值显著负相关(r=-0.605, P<0.01)。结论 CRC的发生发展可能与其癌组织局部微环境中Treg数量变化相关,肿瘤局部Treg数量的增多与T淋巴细胞亚群比例失调可能成为肿瘤免疫逃逸的机制之一。  相似文献

目的 探讨贲门癌患者的免疫功能异常及其临床意义.方法 采用流式细胞术(FCM)和酶联免疫吸附(ELISA)方法分别检测56例贲门癌患者、15名健康人外周血中CD4+CD25hiCD127lowc调节性T细胞(Treg细胞)和血清IL-10、TGF-β1水平,结合临床资料进行分析.结果 56例贲门癌患者外周血中CD4+CD25hiCD127lowTreg细胞占CD4+淋巴细胞的比例为(5.73±1.56)%,与健康对照组的(4.45±1.06)%相比,差异有统计学意义(P<0.01);血清中IL-10和TGF-β1含量均明显高于健康对照(P<0.05).贲门癌患者外周血CD4+CD25hiCD127low Treg细胞水平与血清IL-10和TGF-β1含量呈正相关.贲门癌患者外周血CD4+CD25hiCD127low Treg细胞数量与患者临床分期、淋巴结转移有关.结论 贲门癌患者外周血中CD4+CD25hiCD127low Treg细胞表达增高,与临床分期有关,提示Treg水平异常与贲门癌的发生发展密切相关.  相似文献

目的探讨结直肠癌患者外周血CD4+CD25high调节性T细胞(Treg)的作用。方法采用流式细胞仪测定36例结直肠癌患者及30例健康对照者外周血中CD4+CD25highFoxP3 Treg、CD4+CD25highTreg、CD4+CTLA-4 Treg及CD4+T细胞水平。结果与健康对照组比较,结直肠癌组患者外周血CD4+CD25highFoxP3 Treg显著增多(P<0.01),CD4+CD25high Treg和CD4+CTLA-4 T细胞也增多(P<0.05)。结论CD4+CD25highFoxP3 Treg可能在结直肠癌发展过程中起着重要作用。  相似文献

目的:探讨DC-CIK对胃癌合并腹水患者外周血CD4+CD25+调节性T胞(Treg细胞)比例及功能的影响。方法:60例胃癌合并腹水患者,于输注DC-CIK前1天及DC-CIK治疗结束后1周分别采集外周血。流式细胞术检测外周血Treg细胞的比例,RT-PCR法检测其Foxp3mRNA表达情况;将分选出的Treg细胞和CD4+CD25-T细胞分为单纯Treg细胞组(A组)、1∶1混合培养(B组)、单纯CD4+CD25-细胞组(C组)进行培养,3H-TdR掺入法检测Treg细胞抑制CD4+CD25-细胞增殖的能力。结果:治疗后外周血Treg细胞占CD4+T细胞的比例较疗前显著下降[(6.21±1.37)% vs (9.38±1.06)%,P<0.05]。治疗后Treg细胞Foxp3mRNA表达水平较治疗前显著下降[(56.18±13.25)% vs (85.26±11.58)%,P<0.05]。治疗后Treg对CD4+CD25-T细胞抑制增殖能力较治疗前明显下降[(37.31±4.16)% vs (48.92±5.25)%,P<0.05]。结论:输注DC-CIK免疫治疗,可显著降低胃癌合并腹水患者外周血Treg细胞比例,下调Foxp3mRNA表达水平,降低Treg细胞免疫抑制功能,有利于诱导抗肿瘤免疫效应。  相似文献

Patients with squamous cell carcinoma of the head and neck (SCCHN) have depressed antitumour immunity. The presence of CD4+CD25+ (Treg) cells in these patients might be, in part, responsible for downregulation of antitumour immune responses. To evaluate the frequency and characteristics of Treg in the peripheral circulation of patients with SCCHN, we used multicolour flow cytometry. Expression of CCR7, CD62L, zeta chain and Annexin V binding to Treg and non-Treg CD4+ lymphocyte populations were evaluated. Treg were confirmed to be Foxp3+ and GITR+. The Treg frequency was significantly elevated in patients with active disease and those with no evidence of disease (NED) following curative therapies. Both Treg and non-Treg CD4+ T cells in patients were significantly enriched in CCR7- and CD62L- cell subsets. Although Treg in patients contained a higher proportion of double negative (CCR7-CD62L-) cells, the majority of Tregs were CCR7-CD62L+. The proportion of Annexin V+CD4+ T cells was higher in patients (P<0.00005) than normal controls (NC), and Treg were significantly more sensitive to apoptosis than non-Treg in patients and NC. Expression of zeta was reduced in all subsets of CD4+ T cells obtained from patients vs NC. The data suggest that Treg in patients with SCCHN largely contain T cells with the 'effector' phenotype, which bind Annexin V and have low zeta expression, consistent with their activation state and a rapid turnover in the peripheral circulation.  相似文献

目的探讨CD4+CD25+T细胞在乳腺癌发生、发展中的作用。方法将取自35例乳腺癌患者的105个肿大淋巴结,制备成单细胞悬液,应用流式细胞仪检测CD4+CD25+T细胞比例及CD4+CD25-、CD8+T细胞、NK细胞的相对水平。采用定量RT-PCR法,检测IL-2、IL-10、TGF-β1和IFN-γ的细胞因子水平。结果乳腺癌患者淋巴结中的CD4+CD25+T细胞水平(在CD4+T细胞中的百分含量)与淋巴结转移相关,转移淋巴结中其水平明显高于未转移淋巴结。乳腺癌患者淋巴结中CD4+CD25+T细胞与CD4+CD25-、CD8+T细胞和NK细胞的水平呈负相关关系。乳腺癌患者淋巴结中CD4+CD25+T细胞水平与TGF-β1呈正相关,与IL-2、IL-10、IFN-γ无相关性。乳腺癌患者淋巴结中TGF-β1、IL-10、IFN-γ水平与淋巴结转移相关,转移淋巴结中TGF-β1、IL-10含量高而IFN-γ含量较低。IL-2与淋巴结转移无相关性。结论乳腺癌患者转移淋巴结中的CD4+CD25+T细胞水平高于未转移淋巴结。  相似文献

目的研究化疗药物对肺癌患者外周血中Treg(CD4+ CD25+调节性T细胞)的影响及意义。方法采集60例肺癌术后患者化疗前1天及化疗后第10天外周静脉血,应用流式细胞技术检测外周血中Treg细胞以及CD3+、CD4+、CD8+T、NK细胞占T淋巴细胞百分比,CD4+T/CD8+T比值。结果化疗前NSCLC患者外周血CD4+ CD25+调节T细胞比率明显高于健康对照组(P〈0.05);且Ⅳ期患者调节T细胞比率明显高于Ⅲ期患者(P〈0.05)。化疗后NSCLC患者外周血CD4+ CD25+调节T细胞较化疗前显著降低(P〈0.05)。化疗前后不同病理分型患者外周血中CD4+CD25+细胞变化差异无统计学意义。化疗后CD8+T细胞占T淋巴细胞比例(28.129±10.900)%较化疗前(24.876±6.631)%升高(P〈0.05)。化疗后CD4+/CD8+(1.506±0.691)较化疗前(1.680±0.704)降低(P〈0.05)。结论肿瘤负荷可显著促进肺癌患者外周血Treg细胞分化,化疗后肺癌患者Treg细胞比例下降。  相似文献

The CD4+CD25+ regulatory T cell (Treg) is a special kind of T cell subset. Studies have showed that Tregcells are involved in a number of physiological processes and pathologic conditions such as autoimmune diseases,transplantation tolerance and cancer. Tregs with unique capacity for immune inhibition can impair anti-tumourimmunity and help tumor cells to escape from immune surveillance. The aim of our study was to investigatewhether Tregs are involved in hepatocellular carcinoma (HCC). A BABL/C mouse with HCC in situ model wasestablished to evaluate the Treg existence in carcinoma tissues and the changes of Tregs in spleen using flowcytometry and immunohistochemistry methods. Granzyme B expression in carcinoma tissues was analyzedby immunohistochemistry to investigate the tumor local immune status.The proportion of CD4+CD25+/CD4+spleen lymphocytes of tumor bearing mice (18.8%±1.26%) was found to be significantly higher than that innormal mice (9.99%±1.90%) (P<0.01 ). Immunohistochemistry of spleen tissue also confirmed that there wasan increase in Treg in tumor-bearing mice, while in carcinomas it showed Treg cells to be present in tumorinfiltrating lymphocyte areas while Granzyme B was rarely observed. Anti-tumour immunity was suppressed,and this might be associated with the increase of Tregs. Our observations suggest that the CD4+CD25+Treg/CD4+ proportion in spleen lymphocytes can be a sensitive index to evaluate the change of Tregs in hepatocellularcarcinoma mice and the Treg may be a promising therapeutic target for cancer.  相似文献

目的 探讨Treg及Th1/Th2类细胞因子在晚期肺癌肿瘤免疫抑制中的作用.方法 选取100例初治晚期肺癌患者及50例健康自愿者.采用流式细胞术检测其外周血中Treg、Th1类细胞因子(IFN-γ、IL-2、TNF-a)、Th2类细胞因子(IL-4、IL-6、IL-10)水平,同时分析CD4+CD25+Treg与Th1/Th2类细胞因子之间的相关性.结果 ①晚期肺癌患者外周血中Treg为(11.12±5.83)%,高于健康对照组(7.46±3.07)%,差异有统计学意义(P=0.003);②化疗前肺癌患者外周血中Treg为(11.12±5.83)%,明显高于化疗后(6.45±3.74)%,差异有统计学意义(P<0.001);③晚期肺癌患者与正常对照组Th1/Th2类细胞因子水平分别为:IFN-γ(8.56±3.62 vs 10.79±3.27,P=0.049)、IL-2(8.48±2.87 vs 10.22±4.03,P=0.03)、TNF-a(6.18±2.67vs8.14±2.87,P=0.007)、IFN-γ/IL-4(3.33±1.44 vs 4.09±1.00,P=0.028)、IL-4(3.17±1.19 vs 2.45±0.43,P<0.001)、IL-6(3.88±2.08 vs 2.33±0.88,P<0.001)、IL-10(3.64±1.73 vs 2.54±1.08,P=0.008),其中Th2类因子水平明显升高,差异有统计学意义(P均<0.05);④CD4+CD25+Treg与Th1类细胞因子IFN-γ、TNF-a、IL-2及IL-6无相关性(P均>0.05);与Th1/Th2(γ=-0.273,P=0.003)呈负相关;与Th2类细胞因子IL-4(γ=0.237,P=0.009)、IL-10(0.626,P<0.001)呈正相关(P均<0.05).结论 晚期肺癌患者CD4+CD25+Treg、Th2类细胞因子水平显著升高,Th1类细胞因子水平下降,它们共同导致肿瘤患者免疫抑制及肿瘤进展,监测其水平变化有助于判断肺癌患者疗效、预后,有效调控CD4+CD25+Treg及负性细胞因子水平可能是治疗肺癌的一个新策略.  相似文献

Objective: To study the frequency of the CD4+CD25+ regulatory T cells (Tregs) in the patients with ovarian carcinoma and its possible mechanism. Methods: The percentages of CD4+CD25+ Tregs in the peripheral blood lymphocytes (PBLs), tumor infiltrating lymphocytes (TILs) and tumor associated lymphocytes (TALs) from 13 patients with ovarian carcinoma and in the PBLs from 14 healthy women were determined by flow cytometry. The expression of CD69 on CD4+PBLs from the patients was detected. PBLs from healthy women were cultured in complete RPMI 1640 containing the supernatant from SKOV3 cell line with or without PHA (phytohemagglutinin) stimulation for 72 hours, then the percentage of CD4+CD25+ T cells was detected. Results: CD4+CD25+ Tregs in the PBLs from patients with ovarian carcinoma were significantly increased compared with those from the control. The percentage of CD4+CD25+ Tregs in TILs was higher than that in PBLs and TALs from the patients, but not significantly. The expression of CD69 on CD4+PBLs from the patients was negative. The percentages of CD4+CD25+ T cells in PBLs cultured with SKOV3 supernatant elevated significantly compared with those without supernatant whether PHA was added or not (P = 0.001 and 0.001, respectively). Conclusion: There is an increasing of the proportion of CD4+CD25+ Tregs in PBLs, TILs and TALs of the patients with ovarian carcinoma, which probably results from up-regulation of soluble factor secreted by ovarian carcinoma cells.  相似文献

探讨CD4+CD25+CD127low调节性T细胞(Tregs)在原发性肝细胞性肝癌(HCC)患者外周血中的变化及其临床意义。方法:采集40例乙型肝炎病毒(HBV)相关的HCC患者[巴塞罗那临床肝癌(BCLC)分期A期患者7例、B期患者8例、C期患者20例、D期患者5例]、35例慢性乙型肝炎(CHB)患者及28例正常健康人的外周抗凝血,应用CD4(PE-CY5)、CD25(FITC)、CD127(PE)三种特异性荧光抗体标记后,通过流式细胞术对CD4+CD25+CD127lowTregs水平进行三色荧光抗体检测。结果:HCC患者外周血CD4+CD25+CD127lowTregs占CD4+T细胞的百分比显著高于正常健康人(P<0.001)和CHB患者(P=0.017),CHB患者外周血CD4+CD25+CD127lowTregs占CD4+T细胞的百分比高于正常健康人(P=0.035);HCC中BCLC分期为C期的患者外周血CD4+CD25+CD127lowTregs占CD4+T细胞的百分比显著高于A期患者(P=0.020)和B期患者(P=0.019)。结论:CD4+CD25+CD127lowTregs水平异常增高可能是HCC免疫逃逸的一个重要机制,且其变化水平与临床病情的进展存在一定的相关性。  相似文献

目的探讨健择联合顺铂对非小细胞肺癌患者免疫耐受的调控作用。方法38例经病理学或细胞学确诊的NSCLC患者,采用健择、顺铂联合化疗方案,应用流式细胞仪检测化疗前后外周血CD4+CD25+Foxp3+调节T细胞(Treg)占CD4+T细胞的百分率。结果化疗前NSCLC患者外周血CD4+CD25+Foxp3+调节T细胞(Treg)占CD4+T细胞的比率明显高于健康对照组(P<0.05);化疗后NSCLC患者外周血CD4+CD25+Foxp3+调节T细胞(Treg)占CD4+T细胞的百分率较化疗前显著降低(P<0.05);但鳞癌、腺癌及腺鳞癌3组间化疗前、化疗后各项指标之间差异无统计学意义(P>0.05)。结论健择联合顺铂化疗可调控晚期非小细胞肺癌机体的肿瘤免疫耐受,改善患者的免疫功能。  相似文献

目的 探讨原发性肝癌患者外周血中调节性T细胞(Treg)占CD4+T细胞(Treg/CID4+)比值和肝癌组织中Treg数量及其临床意义.方法 63例原发性肝癌患者均行根治性手术切除,采用流式细胞术检测患者手术前外周血中Treg/CD4+比值,用免疫组织化学方法检测肿瘤组织中Treg的表达.结果 原发性肝癌患者外周血中Treg/CD4+比值显著高于乙型肝炎患者(P<0.01)及正常对照组(P<0.01).癌组织中Treg数量为(15.69±13.29)个/mm2,癌旁肝组织、10例乙型肝炎肝组织、10例正常肝组织中未见或极少见Treg.外周血中Treg/CD4+比值与癌组织中Treg数量呈正相关(P=0.024).外周血和癌组织中Treg数量高者,术后5年生存率低(P值分别为0.042、0.019);癌组织中Treg数量高者,术后5年无瘤生存率低(P=0.001).结论 原发性肝癌患者外周血和组织中调节性T细胞水平升高;外周血中Treg/CD4+比值及癌组织中Treg数量可作为预测患者根治术后预后的免疫学指标.  相似文献

Circulating human CD4(+)CD25(high)Foxp3(+) T cell populations (Treg) may contain activated CD4(+)CD25(+) T cells interfering with Treg evaluation. To gain insights into the phenotypic and functional characteristics of Treg in patients with cancer, we have analyzed CD4(+)CD25(high) populations at the clonal level. Single-cell sorted (SCS) CD4(+)CD25(high) T cells obtained from PBMC of normal controls (NC) or patients with squamous cell carcinoma of the head and neck (HNSCC) were plated at 1 cell/well in 96 well plates and expanded with anti-CD3/anti-CD28 Abs and 1,000 IU IL-2/mL in the presence or absence of rapamycin (1 nM). All generated clones were evaluated for the phenotype by flow cyometry and suppressor function in CFSE-based proliferation assays. Clones had heterogeneous CD25 expression levels. Cloning efficiency of CD4(+)CD25(high) T cells was low. CD25(high) clones expressed CTLA-4, Foxp3, CD62L, but little GITR and suppressed proliferation of autologous CD4(+)CD25(-) responder cells. Clones of activated CD4(+)CD25(interm./low) cells expressed intermediate to high levels of GITR and HLA-DR and did not suppress proliferation of responder cells.
