目的 对42例深静脉拴塞(DVT)的诊断和溶拴治疗进行初步临床研究。方法 从DVT患者的远端静脉慢滴入尿激酶(UK)和中药三七总甙(血塞通),并在拴塞部位上方用皮条阻滞浅静脉,以减少静脉的回流,治疗前后均作深静脉造影对照。结果 本组深静脉造影的再通率为82%。结论 从患肢远端静脉滴入溶栓剂进行溶栓治疗优于全身用药。本方法简单易行,易于推广应用,再通率与时限、病因有关。  相似文献

<正> 绿化有良好的环境效应为人所共知,如对于调节微小气候、防尘降尘、吸收有害气体、减弱噪声等已有不少报导。除此之外,树木还有杀菌作用,也有过少量报导。为了观察亚热带地区城镇绿化的杀菌作用,我们对百色城绿化进行综合考察之后,于一九八二年四月又对百色城不同绿化地区的空气含菌量进行了监测,初步发现了定的效应。  相似文献

自从索绪尔在其普通语言学课程讲义中提出“语言符号是任意的”以来,语言学家对这一提法的“是与否”的论证就从没停息过。在这一过程中,人们加深了对语言本质的认识,促进了与之相关的其他学科的发展。本文试图对语言符号的任意性提出自己的一些看法。并非索绪尔最先指出语言中的音义关系带有约定性,不过,却是索绪尔首先为音义关系的任意性确定了在理论体系中头等重要的地位。他认为语言不是分类命名集,不是给已经存在的概念范畴命名,相反是在语言系统中创造出概念范畴,这就从根本上否定了孤立的符号单位具有外在于系统的价值。  相似文献

以纯化嗜麦芽假单胞菌的绒毛膜促性腺激素(CG)受体为抗原,融合得到4株(ED490,DG390,AB890,GE590)对细菌CG受体特异的单克隆抗体杂交瘤细胞,其腹水和培养上清中的抗体滴度分别为10~(-2)~10~(-6)和1~10~(-2);亚类鉴定显示:单抗GE590为IgGl,单抗ED490、DG390和AB890为IgG2b。研究发现,ED490、AB890和DG390与细菌CG受体作用后,~(125)I-HCG仍能结合该受体,揭示其作用于受体不同位点。GE590与CG受体结合实验发现,随着单抗浓度不断增加,~(125)I-HCG与受体结合逐渐减少,推测该单抗与~(125)I-HCG结合受体同一位点。  相似文献

We reported the production of monoclonal antibodies (McAb) against chorionic gonadotropin (CG) receptor by fusing spleen cells of BALB/c mice immunized with purified pseudomonas maltophilia CG receptor with mouse myeloma line (SP 2/0). Four hybridoma cell lines secreting CG receptor McAbs were obtained (ED 490, DG 390, AB 890, GE 590). GE 590 is IgG 1; ED 490, DG 390, AB 890 are IgG2b. I125-HCG and ED 490, DG 390. AB 890 recognized CG receptor different antigenic determinant. Increased concentrations of GE 590 were in inverse proportion to the amount of I125-HCG binding to human ovarian tissues showing that normal ovarian tissues and ovarian malignant tumors have different HCG receptor antigenic determinant. This study indicated that these McAbs may be useful in studying the structure of CG receptor and in providing a valuable evidence for early diagnosis and treatment of ovarian malignant tumors.  相似文献

本文报道了应用淋巴细胞杂交瘤技术将羊垂体催乳素(oPRL)免疫过的BALB/c小鼠脾细胞与NS-O骨髓瘤细胞融合。制备了5株分泌抗oPRL单克隆抗体的杂交瘤细胞株。其中1种单克隆抗体属IgG1,4种属IgG2a。ELISA检测小鼠腹水抗体稀释度均在5×10~4倍以上。用固相夹心免疫放射测定法测出5种单克隆抗体可识别oPRL分子上3个不同的抗原决定簇,本文还对5种单克隆抗体与和oPRL结构相似的生长激素进行交义反应以鉴别其特异性。  相似文献

Hebdomadis is one of the major serogroups found in China. In Sichuan, this serogroup appears to have a close relationship with local outbreak of leptospirosis. BALB/c mice were immunized intrasplenically with outer envelope of serogroup Hendomadis serovar hebdomadis strain 245. Spleen cells were fused with SP2/0 myeloma cells, two monoclonal antibodies Af2 and Bb2 were produced by hybridoma technique. McAb Af2 and McAb Bb2 were identified to be IgG1 and IgG3 by immunodiffusion respectively. Specificities of these two McAbs were determined by MAT; both reacted to 8 serovars (hebdomadis, nona, kambale, kremastos, worsfoldi, jules, maruborincana) of Hebdomadis serogroup. The agglutination titres of McAb Af2 and McAb Bb2 were 1:640-1:2,500,000 and 1:320-1:2,500,000, respectively. The two McAbs did not agglutinate with serovar kabura of Hebdomadis serogroup, they did not agglutinate with 11 serovars of Sejroe serogroup, 4 serovars of Mini serogroup, 18 representive serovars of L. interrogans in 18 serogroups, L. biflexa strain Patoc I and Leptonema illini. So it was found that McAb Af2 and McAb Bb2 showed partial serogroup specificity for Hebdomadis by agglutination.  相似文献

Four McAbs (1A7E7, 2F9D4, 2G3H5, 1A7H12) against outer membrane antigens of L. interrogans serovar Lai strain 017 were produced by hybridoma technique. McAb 1A7E7 was identified to be IgG2a, the rest IgG1 by immunodiffusion. Outer membrane antigens of three pathogenic leptospiral strains (017, 601, 156) and two non-pathogenic leptospiral strains (Patoc I, 3055) were analysed by SDS- polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting with McAbs. It was found that McAb 1A7E7 recognized specifically the 42kd antigenic band of strain 017; McAb 2F9D4 the 42kd antigenic bands of strain 017, 601, 156, and McAbs 2G3H5 and 1A7H12 the 31 kd antigenic bands of 017, 601, 156. Further study of antigenic properties possessed by pathogenic leptospira may provide some new clues to look for specific diagnostic and protective antigens.  相似文献

目的:用分段表达纯化的SARS冠状病毒(SARS-CoV)的核衣壳蛋白(nucleocapsid N蛋白)制备针对该蛋白不同区域抗原表位的高特异性单克隆抗体(McAb),初步定位单克隆抗体识别表位所在区域。方法:用分段表达纯化的SARS~CoV的N蛋白(分别记为N1蛋白、N2蛋白)分别免疫Balb/c小鼠制备McAb,通过检测其亚类、效价及相对亲和力鉴定McAb的生物学特性,以Western blot鉴定单克隆抗体特异性,并对McAb结合表位进行初步分析。结果:筛选出7株抗SARS-CoV N1蛋白及2株抗SARS-CovN2蛋白的单克隆抗体杂交瘤细胞株,IgG亚类鉴定6株为IgG1,2株为IgG2b,1株为IgG3,腹水效价10^5以上;亲和常数达10^8以上。Western blot证实所获的单克隆抗体可与SARS-CoVN蛋白发生特异性反应。ELISA相加实验结果显示2株N1 McAb识别相同的抗原表位,其余均识别不同的抗原表位。结论:通过分段表达纯化的SARS-CoVN蛋白而制备的特异性针对SARS-CoV N蛋白不同区域抗原表位的单克隆抗体中,N1单抗识别N蛋白N端(1-549bp),N2单抗识别N蛋白C端(496~1269bp),可初步定位单克隆抗体识别表位所在区域。  相似文献

人生长激素(hGH)是垂体前叶分泌的重要激素,但有关其结构与功能的关系、抗原决定簇的数目及位置、受体结合位点以及不均一性的来源和意义等问题至今尚未解决。抗hGH单克隆抗体(McAb)不仅是研究上述问题的有用工具,而且还可用于建立更为快速、灵敏,特异和准确的GH新免疫分析法,以提高有关疾患的诊治水平。本实验用微量免疫法获得四种抗整分子hGH McAb,并对其特性进行了较详细的研究。  相似文献

Seven monoclonal antibodies (McAbs) specific for Plasmodium falciparum (Pf), humanimmune sera of three individuals, two adults and one child, living in a malaria endemic area inHainan Island of China and immune sera of BALB/c mice immunized with the erythrocytic stages ofPF were used to identify the target antigens of asexual blood stages of Pf. The ~(125)I-labeled surfaceantigens with apparent molecular weights of 125, 115, 83, 74 and 67 KDa were mainly recognizedby McAbs 93A3, 94B5, 92D4 and 93D4, but the ~(125)I-labeled surface antigens of 200 and 19 KDawere recognized by McAb 94Dl alone. In addition, the ~(35)S-methionine- labeled target antigens of183, 125, 83 and 55 KDa were also recognized by McAbs 93A3, 94B5 and 94C3. The McAb21E3 against the culture supernatant antigen of Pf only recognized the antigen of 125 KDa labeledwith ~(15)S-methionine. The human immune sera from the adulst living in a malaria endemic area andmouse immune sera not only immunoprecipitated the target antigens which were recognized by theMcAbs but also immunoprecipitated the polypeptides of 140, 100 and some lower - molecularweight. But these target antigens recognized by McAbs, adult immune sera of and mouse immunesera could not be immunoprecipitated by human immune serum of the child with a primary infec-tion. With the antigen competition assay, the unlabeled antigens of Pf could strongly inhibit the im-mune reaction between the McAbs and the ~(125)I-labeled antigens of Pf. It suggested that the antigensrecognized by McAbs and polyvalent immune sera were specific target antigens.  相似文献

作者以嗜麦芽假单胞菌(简称细菌)hCG结合蛋白抗体为探针,对比研究细菌hCG结合蛋白和大鼠睾丸细胞膜LH/CG受体的免疫特性。结果前者特异沉淀[~(125)I]hCG与hCG结合蛋白结合复合物;后者亦获得相同的结果。以上结果表明,细菌hCG结合蛋白与大鼠睾丸LH/CG受体蛋白有相同抗原决定簇,抗体识别该受体hCG结合位点以外的抗原决定簇。  相似文献
