The individual toxicities of five organophosphorus pesticides (dichlorvos, parathion, methyl parathion, malathion and dimethoate) to Daphnia magna were investigated in 24-h immobilization experiments. Using these toxicity data, their combined toxicities were measured in pesticide mixtures designed using either ‘equivalent-effect concentration ratios’ or ‘uniform-design concentration ratios’. The toxicities of mixtures of similarly or dissimilarly acting toxicants are often predicted from the individual toxicities of the component compounds, using one of two distinct biometric models: concentration addition (CA) or independent action (IA). The relative accuracies of the CA and IA models were assessed using the model deviation rate (MDR), which represents the difference between the effect predicted from the individual pesticide concentrations and the observed effect. The mean MDR value of CA was 0.93 (range 0.75–1.31) and the mean value obtained by IA was 3.13 (range 2.52–4.37). We conclude that the CA model is better able to predict the joint toxicities of mixtures of organophosphorus pesticides to D. magna.  相似文献

Phlorotannins have been proven to contain numerous bioactive compounds that have potential to be applied in variety industries, including cosmetics, functional foods, nutraceuticals, environmental management, and medicine. The larvicidal and growth-inhibiting properties of phlorotannins have been extensively studied in various organisms. However, the toxicity of the phloroglucinol oligomer of phlorotannin is unclear, especially in Artemia salina, Daphnia magna, Lactuca sativa, and Chlorella vulgaris, which are commonly used in many bioassays. Therefore, research using these four organisms should be designed to provide basic information about the toxic effects of phlorotannins and phloroglucinol. This study aimed to evaluate the larvicidal and inhibitory properties of phlorotannins and phloroglucinol on A. salina, D. magna, L. sativa, and C. vulgaris. Phlorotannin extract and phloroglucinol were administered at various concentrations to each test organism. The survival rate of A. salina nauplii and D. magna neonates was observed every 24 h to 72 h, whereas the L. sativa seed germination and inhibition rate of C. vulgaris were observed up to 96 h. The results showed that the 24 h LC50 of phlorotannin on A. salina and D. magna were 10.67 and 1.32 mg/mL, respectively. The germination inhibition of L. sativa was 53.3% with a seed growth of less than 4 mm after 96 h upon exposure to 1 mg/mL of phlorotannin. Freshwater and seawater C. vulgaris experienced yield inhibition of 39.47 and 43.46%, respectively, when 2 mg/mL of phlorotanin was added. These results indicate that phlorotannin affects the survival and growth of the test organisms, so its use as a pesticide, herbicide, and algaecide agent for environmental and aquaculture applications can be further studied.  相似文献

Concomitant releases of various engineered nanoparticles (NPs) into the environment have resulted in concerns regarding their combined toxicity to aquatic organisms. It is however, still elusive to distinguish the contribution to toxicity of components in NP mixtures. In the present study, we quantitatively evaluated the relative contribution of NPs in their particulate form (NP(particle)) and of dissolved ions released from NPs (NP(ion)) to the combined toxicity of binary mixtures of ZnO NPs and graphene oxide nanoplatelets (GO NPs) to three aquatic organisms of different trophic levels, including an alga species (Scenedesmus obliquus), a cladoceran species (Daphnia magna), and a freshwater fish larva (Danio rerio). Our results revealed that the effects of ZnO NPs and GO NPs were additive to S. obliquus and D. magna but antagonistic to D. rerio. The relative contribution to toxicity (RCT) of the mixture components to S. obliquus decreased in the order of RCTGO NP(particle) >?RCTZnO NP(particle)?>?RCTZnO NP(ion), while the RCT of the mixture components to D. magna and D. rerio decreased in the order of RCTZnO NP(particle)?>?RCTGO NP(particle)?>?RCTZnO NP(ion). This finding also implies that the suspended particles rather than the dissolved Zn-ions dictated the combined toxicity of binary mixtures of ZnO NPs and GO NPs to the aquatic organisms of different trophic level. The alleviation of the contribution to toxicity of the ionic form of ZnO NPs was caused by the adsorption of the dissolved ions on GO NPs. Furthermore, the ZnO NP(particle) and GO NP(particle) displayed a different contribution to the observed mixture toxicity, dependent on the trophic level of the aquatic organisms tested. The difference of the contributions between the two particulate forms was mainly associated with differences in the intracellular accumulation of reactive oxygen species. Our findings highlight the important role of particles in the ecological impact of multi-nanomaterial systems.  相似文献

The continued widespread use of larvicides in Aedes aegypti control programs is still a necessary strategy, since there are no apparent efficient vaccines against arboviruses. However, chemical approaches may affect nontarget organisms and produce detrimental effects to environmental health. Therefore, the aim of this study was to conduct toxicity testing for pyriproxyfen at different concentrations using Daphnia magna and Artemia salina as model organisms to evaluate the ecotoxicological parameters. This study describes the toxicological effects of pyriproxyfen on both microcrustaceans, which are widely used in bioassays because of their sensitivity to changes in hydrosphere. Data demonstrated that the calculated EC50-48h value of pyriproxyfen was 2.5 μg/for D. magna and A. salina; the no-observed-effect concentration (NOEC) and the lowest-observed-effect concentration (LOEC) of pyriproxyfen were found to be 0.63 and 1.25 μg/L for Artemia salina and Daphnia magna, respectively. In chronic toxicity and reproduction tests on D. magna, a calculated CL50-7day (lethality on 50% of daphnids after 7 days of chronic test) and an EC50-21day (50% reduction in the reproductive output of parental daphnids after 21 days of exposure) higher than 1.25 μg/L pyriproxyfen were observed. The time of first reproduction was significantly increased in D. magna after exposure to environmentally relevant concentrations of pyriproxyfen, but other reproduction parameters were not markedly altered. Environmental risk assessment revealed that pyriproxyfen is highly toxic for both branchiopods. Data demonstrated that pyriproxyfen may produce adverse effects on the aquatic ecosystem at concentrations required to control Ae. aegypti.  相似文献

Pyrethroid pesticides are applied to both agricultural and aquacultural industries for pest control. However, information of their impact on the commercial important freshwater crayfish, Procambarus clarkii is scarce. Therefore, the present study aimed to characterize to effects of a commonly used pyrethroid pesticide, deltamethrin on DNA damage, immune response, and neurotoxicity in P. clarkii. Animals were exposed to 7, 14, and 28 ng/L of deltamethrin, which correspond to 1/8, 1/4, and 1/2 of the LC50 (96 hours) of this pyrethroid to P. clarkii. Significant increase of olive tail moment (OTM) and 8‐hydroxy‐2′‐deoxyguanosine (8‐OHdG) was found after deltamethrin exposure in a dose‐dependent way. Total hemocyte counts (THC) and activities of immune‐related enzymes including acid phosphatase (ACP), lysozyme (LZM), and phenoloxidase (PO) were all decreased and significantly lower than control at concentration of 28 ng/L after 96 hours exposure. Acetylcholinesterase (AChE) activity, an indicator of neurotoxic effect was investigated and it was decreased significantly in muscles at 14 and 28 ng/L after 24 hours exposure. The level of intracellular reactive oxygen species (ROS) in hemocytes was also measured and the significant increase of ROS was found at 14 and 28 ng/L concentrations. The results revealed that deltamethrin induced DNA damage, immunotoxicity, and neurotoxicity in P. clarkii by excessive generation of ROS. Because of the dose‐dependent responses of all parameters under exposure of deltamethrin at environmentally realistic concentrations, these parameters could be used as sensitive biomarkers for risk assessment of deltamethrin in aquaculture area.  相似文献

Dengue transmitted by mosquitoes of the genus Aedes, species aegypti, is a major public health concern in Brazil. The chemical control of the mosquito larvae has been performed with the larvicide temephos since 1967. However, vector resistance was reported to temephos in several Brazilian states, and the Ministry of Health ordered the replacement of this larvicide by diflubenzuron (DFB), an inhibitor of chitin synthesis. Both insecticides are diluted in water with larvae and are able to reach aquatic environments in which they subsequently adversely damage nontarget organisms. The aims of this study were to (1) determine the acute toxicity (EC50) and environmental risk (RQ) of DFB and temephos to the microcrustacean Daphnia magna, and (2) evaluate the chronic toxicity (no-observed-effect concentration [NOEC] and lowest-observed-effect concentration [LOEC]) of these larvicides to D. magna. The experiments were performed according to a completely randomized design. The estimated 48-h EC50 of temephos was 0.15 μg/L (lower limit = 0.1 and upper limit = 0.2 μg/L) and the 48-h EC50 of DFB was 0.06 μg/L (lower limit = 0.03 and upper limit = 0.1 μg/L). RQ values were 4.166.7 to DFB and 6.666.6 to temephos. NOEC and LOEC values were respectively 2.5 and 5 ng/L for DFB, and respectively 6.2 and 12.5 ng/L for temephos. Thus, temephos and DFB are classified as highly toxic to Daphnia magna and pose a high environmental risk to this species. Mortality of D. magna was observed at concentrations lower than those used in the field to control A. aegypti larvae.  相似文献

One focus of integrated pest management (IPM) is the use of biological and chemical control in an optimal way. The availability of selective pesticides is important as is information about both lethal and sublethal effects of pesticides on biocontrol agents. Acute and sublethal effects of imidacloprid and fenpyroximate exposure were studied on adult stage of egg parasitoid Trichogramma cacoeciae Marchal and the emergence rate and life table parameters were determined. The adult wasps were exposed to field recommended concentration (FRC) of the pesticides on glass plates. Field rates of imidacloprid and fenpyroximate caused 100 and 32% adult mortality, respectively. Based on concentration–response experiments, the LC50 values of imidacloprid and fenpyroximate were 6.25 and 1,949 ppm, respectively. The effect of imidacloprid and fenpyroximate on larvae, prepupae and pupae of the parasitoid was tested by exposing parasitized eggs of Sitotroga cerealella Olivier or Cydia pomonella L. to the FRC. Imidacloprid and fenpyroximate reduced adult emergence by 10.7 and 29%, respectively, when S. cerealella eggs were used as the host and 10.9 and 24.9%, respectively, when C. pomonella eggs were used as the host. Population parameters of emerged adults from treated pre-imaginal stages by FRC of the pesticides were also studied. The parameters were longevity and progeny production of emergent adults and also intrinsic rate of increase (r m ), generation time (T) and doubling time (DT). Longevity and progeny production of the emergent adults was not affected by pesticide exposure in comparison to the control. In addition, none of population parameters such as r m , T and DT were affected by pesticide exposure. The intrinsic rate of increase for the control, fenpyroximate and imidacloprid exposed populations were 0.388, 0.374, and 0.372 female offspring per female per day, respectively. Overall, results of this study suggest a relative compatibility between fenpyroximate and T. cacoeciae, but imidacloprid showed deleterious effects on adults of the parasitoid.  相似文献

Oxidative stress and osmoregulatory system damage-inducing potential of binary mixtures of neonicotinoid insecticide imidacloprid (IMI) and Cd2+ in Daphnia magna were evaluated. Animals were subjected to subchronic (7 days) and acute (48 h) of IMI and Cd2+ effects with single and binary mixtures. ATPase and antioxidant enzyme activities with lipid peroxidation were measured. Morphometric characteristics were also evaluated. Response patterns showed variability due to the duration, concentration, and toxicant type. While the enzyme activities mostly showed a decreasing trend upon the subchronic IMI effect, there was an increasing trend after the Cd2+. Declined enzyme activities were more pronounced with the acute higher IMI+Cd2+ exposure. Ca2+-ATPase and CAT were the most sensitive biomarkers in the toxicity response. IMI+Cd2+ exposures are appeared to increase their toxic effects due to their oxidative potential. ATPase inhibition and antioxidant enzyme alterations with a decrease in morphometric characteristics in Daphnia even at their low concentrations of IMI and Cd2+ show evidence of their toxicities on aquatic life. It was emphasized that investigating the combined effects of toxicants at their environmental level based on the multi-biomarker approach is essential in toxicity evaluation.  相似文献

Chronic toxicity of chlordane, an organochlorine insecticide, was assessed on Ceriodaphnia dubia under standardized conditions of testing. Results were compared to Daphnia magna to determine the sensitivity of the two freshwater cladoceran species to this persistent organic pollutant (POP) and to explore the possibility of using the 7‐day C. dubia test as an alternative to the 21‐day D. magna test in chronic toxicity assessment of POPs. The NOEC‐7d value of chlordane on reproduction of C. dubia (2.9 μg/L) was much higher than the NOEC‐21d value of D. magna (0.18 μg/L), attesting that the 7‐day test on C. dubia was less sensitive than the 21‐day reproduction test on D. magna to chlordane. However, extending the period of exposure of C. dubia to chlordane from 7 to 14 days led to a NOEC‐14d value similar to the NOEC‐21d value in D. magna (0.18 μg/L). This study highlights the usefulness of prolonging the exposure time of the reproduction test in C. dubia from 7 to 14 days to increase the performances of the reproduction test on C. dubia for assessing chronic toxicity of POPs. © 2010 Wiley Periodicals, Inc. The acute toxicity of 21 parabens and their chlorinated derivatives was investigated by means of two toxicity bioassays: Daphnia magna immobilization test and the inhibition of bioluminescence of Vibrio fischeri. The median effective concentration (EC50) values of the tested parabens ranged from 2.2 to 62 mg l?1 in the D. magna test and from 0.0038 to 5.9 mg l?1 in the V. fischeri test at 15 min after exposure. The toxicity of dichlorinated methyl‐ and n‐propylparaben, the most commonly used preservatives in cosmetics, toward D. magna was 3.9‐ and 2.8‐fold that of their corresponding parent compounds. Toxicity toward D. magna showed a linear relationship with log P, indicating that toxicity increases with increasing hydrophobicity. On the other hand, the correlations of toxicity toward V. fischeri with hydrophobicity and with the degree of chlorination were poor. In addition, the results of the present study indicated that the V. fischeri test was more sensitive than the D. magna test for the determination of the acute toxicity of parabens. A complete assessment of the ecological and toxicological risks of parabens may require the examination of chlorinated parabens as well as the parent pollutants, as described in the present study. We exposed Japanese medaka (Oryzias latipes) to environmentally relevant concentrations of azinphos-methyl, chlorothalonil, endosulfan, and mixtures of all three to determine if combinations of these pesticides result in additive, less-than-additive, or more-than-additive effects. Medaka were exposed from fertilization until 7 days posthatching, and end points included survival, time to hatch, size at 7 days posthatching, activity level (as measured by distance swam) and foraging ability at 3 weeks posthatching, and adult size, liver size, and sex ratio at 5 months posthatching. Although exposure to individual pesticides or pesticide mixtures did not affect survival, hatching time, or foraging ability, fry exposed to azinphos-methyl were significantly smaller at 1 week of age, and those exposed to chlorothalonil and a combination of the chemicals showed reduced activity. Adult sex ratios were biased toward females in all groups exposed to pesticides, with those exposed to azinphos-methyl, chlorothalonil, and the pesticide mixture departing significantly from an even sex ratio. There was no evidence of additive or synergistic effects of pesticide mixtures.  相似文献

Imidacloprid is a systemic insecticide that belongs to the neonicotinoid class of chemicals that act on the central nervous system of insects. Imidacloprid is used to control sucking insects, chewing insects such as termites, soil insects, and fleas on pets, as well as to treat structures, crops, soil, and seeds. As a result of these factors, this pesticide may end up in the aquatic environment via municipal discharges and runoff. Although the presence of imidacloprid in aquatic environments has been underreported as widespread, the toxic effects of this pesticide may have serious implications on aquatic organisms, particularly at environmentally relevant concentrations and demand more attention. Given this knowledge, the goal of this study was to investigate the effects of imidacloprid on Clarias gariepinus embryonic development. Clarias gariepinus embryos (3?h post-fertilization) were exposed to environmentally relevant concentrations of imidacloprid (10, 30, 100, and 500?µg/L) until 48?h post-fertilization using a modified fish embryo acute toxicity test (OECD TG 236). A stereomicroscope was used to assess hatchability, deformity, heart rate, and swimming speed as endpoints. According to our results of the developmental acute toxicity test, imidacloprid significantly reduced the hatching rate and heartbeats of C. gariepinus embryos. It also influenced the swimming kinematics of exposed embryos and caused teratogenic effects such as yolk sac rupture, pericardial oedema, lordosis, an abnormally shaped head, and altered epiboly. Our results allow us to conclude that imidacloprid is a toxic pesticide in the early life stages of C. gariepinus due to its high teratogenic potential.

In the present study, we evaluated the individual and combined toxic effects of herbicide atrazine and three insecticides (chlorpyrifos, lambda-cyhalothrin and imidacloprid) on the earthworm, Eisenia fetida. Results from 48-h filter paper test indicated that imidacloprid had the highest toxicity to E. fetida with an LC50 of 0.05 (0.041–0.058) μg a.i. cm?2, followed by lambda-cyhalothrin and atrazine with LC50 values ranging from 4.89 (3.52–6.38) to 4.93 (3.76–6.35) μg a.i. cm?2, while chlorpyrifos had the least toxicity to the worms with an LC50 of 31.18 (16.22–52.85) μg a.i. cm?2. Results from 14-days soil toxicity test showed a different pattern of toxicity except that imidacloprid was the most toxic even under the soil toxicity bioassay system. The acute toxicity of atrazine was significantly higher than that of chlorpyrifos. In contrast, lambda-cyhalothrin was the least toxic to the animals under the soil toxicity bioassay system. The binary mixture of atrazine–lambda-cyhalothrin and ternary mixture of atrazine–chlorpyrifos–lambda-cyhalothrin displayed a significant synergistic effect on the earthworms under the soil toxicity bioassay. Our findings would help regulatory authorities understand the complexity of effects from pesticide mixtures on non-target organisms and provide useful information of the interaction of various pesticide classes detected in natural environment.  相似文献

The 48-, 72- and 96-h acute toxicity and 14-d chronic toxicity of the sludge of treatment plant No. 1, 3 and 5 were examined based on the survival percentage and fertility changes of Daphnia magna and Eisenia fetida. The highest percentage of D. magna death in the acute toxicity test appeared in treatment plant No. 1 sludge sample. The 48-h and 72-h LC50 of treatment plant No. 1 sludge on D. magna were calculated to be 86.81 and 44.307% volume of the sludge, respectively. The release of untreated sludge into the environment could adversely affect the life of organisms.  相似文献

The present investigation was aimed to assess the effects of imidacloprid on the survival, genetic materials, hepatic transaminase activity and histopathology of loach (Misgurnus anguillicaudatus). The values of LC50 (24, 48, 72 and 96 h) of imidacloprid were 167.7, 158.6, 147.9 and 145.8 mg/L, respectively, and the safety concentration was 42.55 mg/L. The erythrocyte micronuclei assays and the comet assay results showed that imidacloprid had genetic toxic effect on the loach erythrocytes. To assess the physiological and biochemical damage caused by imidacloprid, the activities of hepatic glutamic-pyruvic transaminase (GPT) and glutamic-oxalacetic transaminase (GOT) were measured and their values declined in treatment groups. Histological examination of testis revealed that imidacloprid treatment resulted in disorganized lobules and cysts structures. In the present work, we also investigated the joint toxicity of pesticides commonly used in paddy fields (imidacloprid and lambda-cyhalothrin) on M. anguillicaudatus, and confirmed that a synergistic effect existing in the binary mixtures. The results of our study provide relevant and comparable toxicity information that are useful for safety application of pesticides.  相似文献

Severe deterioration of water quality occurs during jute retting in ponds, canals, floodplain lakes, and other inland water bodies in the rural areas of West Bengal in India. Attempts were made to evaluate changes in the physicochemical parameters of water caused by jute retting, and their impact on the survival of two species of freshwater fish (Labeo rohita and Hypophthalmicthys molitrix) and two species of freshwater invertebrate (Daphnia magna, a Cladocera, and Branchiura sowerbyi, an Oligochaeta). Results showed that jute retting in a pond for 30 days resulted in a sharp increase in the BOD (>1,000 times) and COD (>25 times) of the water, along with a sharp decrease in dissolved oxygen (DO). Free CO2, total ammonia, and nitrate nitrogen also increased (three to five times) in water as a result of jute retting. Ninety-six-hour static bioassays performed in the laboratory with different dilutions of jute-retting water (JRW) revealed that D. magna and B. sowerbyi were not susceptible to even the raw JRW whereas fingerlings of both species of fish were highly susceptible, L. rohita being more sensitive (96 h LC50 37.55% JRW) than H. molitrix (96 h LC50 57.54% JRW). Mortality of fish was significantly correlated with the percentage of JRW.  相似文献

Multigenerational exposure of Daphnia magna to tetracycline was carried out through four consecutive generations. The effects of tetracycline on the survival, reproduction and growth of D. magna were assessed over a period of 21 days per generation. The evaluated endpoints were overall fecundity (total mean neonates per female over 21 days), time to first reproduction, longevity, molting number and somatic growth, such as body weight and body length. Using the results obtained for reproduction and survival rates, the intrinsic population growth rate (PGR) was calculated and compared throughout the generations. Reproductive impairment was observed in all generations and magnified with increasing generation number. The value of no observed effect concentration on D. magna also markedly decreased with increasing generation number. This subsequently resulted in a reduction of the PGR value. In addition, the PGR value was decreased with increasing exposure concentration, decreasing by about 30 and 60% at 0.1 and 5.0 mg/L tetracycline, respectively. On the other hands, somatic growth increased with increasing generation number, because the remaining input energy from the reduced reproduction was mainly used for body maintenance. As a result, the somatic growth and reproduction showed reversed trends on continuous exposure of tetracycline to four generations. In conclusion, multigenerational exposure of tetracycline can induce overall responses on reproduction and the somatic growth of D. magna. Moreover, the PGR value of D. magna exposed to tetracycline was reduced with increasing generation number; thereby, inhibiting the long term propagation of D. magna.  相似文献

Abstract

Nigella sativa L. belonging to Ranunculaceae family is an important medicinal spice which has been utilised to treat various chronic diseases. Lipid nanoemulsions containing oil from medicinal plants have shown to enhance drug dissolvability, diminish symptoms of different powerful medications and enhance the bioavailability of medications, in contrast with conventional formulations. In the present study, aqueous titration method was used to prepare nanoemulsion. The optimised formulation (NE11) with the mean particle size of 37.47?nm showed a minimum viscosity of 0.547 cps and maximum drug release (98.2%) in 24?h. The stability study showed considerably stable formulations at refrigerator temperature as compared to room temperature. The cancer cell line studies confirmed that 5d sprout extract of N. sativa nanoemulsion reduced the cell viability (p?<?.05) and increased colony formation, ROS intensity and chromatin condensation. All data such as colony formation, ROS intensity and chromatin condensation are represented as mean?±?SD (p?<?.001) treated cells for 48?hours. Our results concluded that the development of nanoemulsion could be an efficient carrier for drug delivery.  相似文献

Canadian Water Quality Guidelines (CWQG) are numerical or narrative limits that protect designated water uses. Development of CWQG is based on review of deltamethrin's properties, uses, fate in the environment, ambient levels, accumulation in biota, and toxicity. Deltamethrin is a synthetic pyrethroid insecticide used against agricultural pests. Concentrations of deltamethrin in Canadian agricultural areas range between 10 and 1000 ng m−3 in air, 0.04 and 24.0 μg L−1 in water, 3 and 5 ng g−1 in sediments, 281 and 1375 ng g−1 in plants, and 3.0 and 50.0 ng g−1 in animals. In nature, deltamethrin can undergo volatilization, isomerization, and hydrolysis. Estimates of toxicity of deltamethrin to freshwater life range from 0.0001 μg L−1 (48- and 96-h EC0 for immobilization of Daphnia magna) to 1700 mg L−1 (EC50 for photoinhibition of Selanastrum capricornutum). Terrestrial life is affected by concentrations of deltamethrin ranging from 1.0 mg kg−1 (retardation of fetus development in mice) to 5000 mg kg−1 (LD50 for a rat). A full guideline of 0.4 μg L−1 is recommended for the protection of freshwater life. A full guideline of 2.5 μg L−1 is recommended for water used for livestock production. Context: Leishmaniasis is a complex disease with a broad spectrum of clinical presentations.

Objective: We evaluated the anti-leishmanial effects of Nigella sativa L. (Ranunculaceae) against Leishmania tropica and Leishmania infantum with an in vitro model.

Materials and methods: Antileishmanial effects of essential oil and methanolic extract of N. sativa (0–200?µg/mL) and thymoquinone (0–25?µg/mL) on promastigotes of both species and their cytotoxicity activities against murine macrophages were evaluated using the MTT assay at 24, 48, and 72?h. Moreover, their leishmanicidal effects against amastigotes were investigated in a macrophage model, for 48 and 72?h.

Results: The findings showed that essential oil (L. tropica IC50 9.3?μg/mL and L. infantum IC50 11.7?μg/mL) and methanolic extract (L. tropica IC50 14.8?μg/mL and L. infantum IC50 15.7?μg/mL) of N. sativa, particularly thymoquinone (L. tropica IC50 1.16?μg/mL and L. infantum IC50 1.47?μg/mL), had potent antileishmanial activity on promastigotes of both species after 72?h. In addition, essential oil (L. tropica IC50 21.4?μg/mL and L. infantum IC50 26.3?μg/mL), methanolic extract (L. tropica IC50 30.8?μg/mL and L. infantum IC50 34.6?μg/mL), and thymoquinone (L. tropica IC50 2.1?μg/mL and L. infantum IC50 2.6?μg/mL) mediated a significant decrease in the growth rate of amastigote forms of both species. Thymoquinone (CC50 38.8?μg/mL) exhibited higher cytotoxic effects against murine macrophages than the other extracts.

Conclusion: N. sativa, especially its active principle, thymoquinone, showed a potent leishmanicidal activity against L. tropica and L.infantum with an in vitro model.  相似文献
