目的 探讨碱性成纤维细胞生长因子(bFGF)在反复肺炎支原体肺部感染导致肺间质纤维化的发病机制中的作用。方法 给大鼠于24周内反复9次经超声雾化吸入肺炎支原体以复制其慢性肺部感染模型,随后用bFGF单克隆抗体按SABC法行免疫组织化学染色,定量图像分析,以观察肺组织bFGF蛋白质水平表达的变化。结果(1)感染组动物(n=4)支气管肺泡灌洗液肺炎支原体-PCR检测均为阳性,而对照组(n=4)和感染加红霉素治疗组动物(n=4)均为阴性(均为P<0.05);三组动物的支气管和肺组织常规细菌培养结果均为阴性;感染组动物透射电镜检查见肺泡间隔增宽,其中有较多胶原纤维堆积。其余两组则未见明显异常。(2)感染组动物支气管上皮细胞胞浆内、支气管和肺小动脉肌层以及肺泡巨噬细胞胞浆内可见较多棕黄色细颗粒状bFGF阳性表达产物,其积分光密度为45.17±5.09(n=4),显著高于对照组者(10.77±3.66,n=4,P<0.05)和感染加红霉素治疗组者(16.32±3.58,n=4,P<0.05)。结论 反复肺炎支原体肺部感染时bFGF蛋白质水平表达增加,可能参与致肺间质纤维化的发病过程。  相似文献

目的 检测肝细胞癌(hepatocellular carcinoma,HCC)患者术前血清碱性成纤维细胞生长因子(basic fibroblast growth,bFGF)水平,确定其与HCC根治性术后转移复发的关系。方法 采用Cox比例风险模型,对影响预后的临床指标进行分析,确定影响HCC切除术后转移复发的临床指标。同时应用ELISA方法,检测HCC患者术前血清bFGF水平,western blot方法对24例相应肿瘤组织bFGF表达水平进行定量分析。结果 Cox比例风险模型显示,肿瘤大小及肿瘤是否存在肝内播散,是影响HCC切除术后患者生存时间的显著因素(分别为P=0.022和P=0.040),并以此划分为高和传转移复发倾向组。根治性切除术后高转移复发倾向组,HCC患者术前血清bFGF显著高于传转移复发倾向组(P<0.01)。western blot显示,HCC患者血清bFGF水平变化与其对应肿瘤组织bFGF表达变化相一致。结论 HCC患者术前血清bFGF是预测HCC根治性切除术后转移复发的潜在指标。HCC患者血清bFGF水平升高是其对应肿瘤组织高表达的结果。  相似文献

目的:成纤维细胞生长因子(FGF)是一种结构相关的肽生长因子家族,具有很强的促细胞生长作用。本文探讨老年冠心病患者碱性成纤维细胞生长因子(bFGF)、纤维连接蛋白(Fn)的变化及临床意义。方法:对象共42例,冠心病非心绞痛组12例,心绞痛组15例,急性心肌梗死(AMI)组5例,陈旧性心肌梗死组5例,缺血心肌病组5例。用ELISA法测定bFGF、Fn。结果:冠心病非心绞痛组、心绞痛组、AMI组bFGF、Fn分别为177.5±20.2μg/ml,350.2±26.4ug/ml,430.4±8.5tμg/ml;230.3±28.9μg/ml,240.2±27.4μg/ml,360.7±38.4μg/ml,与对照组比较有显著差异(P<0.05-0.01)。结论:bFGF水平:AMI组>心绞痛组>冠心病非心绞痛组。bFGF、Fn对了解冠心病的病情及预后有一定意义。  相似文献

目的研究碱性成纤维细胞生长因子(bFGF)在慢性间质性肾炎(CIN)患者肾组织中的表达及意义。方法2001-022002-12对中国医科大学附属第一医院的30例慢性间质性肾炎患者及5例正常人采用免疫组织化学SP法检测肾组织中bFGF的表达,并采用图像分析技术测定bFGF阳性表达部分的平均灰度值、平均光密度值及其与整个视野面积的比值,所得数据进行统计学比较。结果慢性间质性肾炎患者肾间质中bFGF表达明显增强,其平均灰度值、平均光密度值及阳性表达面积较正常对照组差异有显著性。结论bFGF能够促进慢性间质性肾炎患者肾间质成纤维细胞及单核细胞增生,从而导致肾间质纤维化。  相似文献

目的探讨碱性成纤维细胞生长因子(bFGF)在急性白血病(AL)中的表达及其临床意义。方法应用酶联免疫法(ELISA)检测AL初治及复发患者化疗前后血清bFGF的水平,并与正常对照组比较。结果47例AL初治患者血清bFGF水平为(60.57±25.62)pg/L,8例复发患者为(72.26±21.44)pg/L,均明显高于正常人对照组的(24.06±5.97)pg/L,差异有统计学意义(P<0.01);11例完全缓解(CR)患者血清bFGF水平(27.33±9.04)pg/L,较CR前明显下降(P<0.01),接近于正常人对照组的水平(P>0.05);急性淋巴细胞白血病(ALL)难治组血清bFGF的水平为(85.20±31.45)pg/L,明显高于非难治组的(60.03±22.97)pg/L,两者差异有统计学意义(P<0.05);但急性髓细胞白血病(AML)难治组血清bFGF水平(63.36±16.62)pg/L与非难治组的(55.02±22.84)pg/L差异无统计学意义(P>0.05)。结论血清bFGF的检测对了解AL的发生、发展及疗效观察和预后判断有一定临床价值。  相似文献

目的测定胃癌患者周围血碱性成纤维细胞生长因子(basic fibriblast growth factor,bFGF),探讨对胃癌诊断、预后的价值.方法采用定量夹心ELISA法测定胃癌患者周围血bFGF的含量.结果献血员(18例)、非炎性疾病(192例)、炎性疾病(54例)和胃癌组(82例)患者血清bFGF平均含量分别为91.8±15.4、95.0±21.2、122.9±21.4和146.8±29.7pg/ml.胃癌组bFGF平均含量显著高于献血员、非炎性疾病和炎性疾病组(P<0.05~0.01);献血员组bFGF平均含量与非炎性疾病组比较差别无显著性(P>0.05),但显著低于炎性疾病组(P<0.05);非炎性疾病组显著低于炎性疾病组(P<0.05);胃癌手术前bFGF含量明显高于手术后(P<0.05).bFGF含量与年龄、性别、胃癌类型无关.结论胃癌患者周围血中bFGF显著升高,对胃癌诊断和预后判断有一定的参考价值.  相似文献

目的观察碱性成纤维细胞生长因子(BFGF)在慢性乙型肝炎及肝癌组织中的表达和定位情况,探讨其在慢性肝病进展中的作用。方法对30例正常肝组织、肝癌组织及150例慢乙肝组织BFGF行免疫组化定量检测,比较其与肝脏纤维化程度及血清透明质酸(HA)、Ⅳ型胶原(CⅣ)的关系,并行统计学分析。结果肝组织BF-GF及血清HA、CⅣ随纤维化进展而表达增加,组间两两比较均具有显著性差异(P〈0.05);肝组织BFGF与血清HA、CⅣ呈正相关,r=0.869、0.793,(P〈0.01)。结论肝组织BFGF不仅可作为评价肝脏损伤严重程度的重要指标,并可提供针对其进行抗纤维化治疗及选择多靶点拮抗肿瘤血管生成的理论依据。  相似文献

目的碱性成纤维生长因子(bFGF)作为非小细胞肺癌(NSCLC)重要的血管新生因子,参与疾病的发生和发展。本研究检测bFGF在NSCLC患者血清中的表达,探讨bFGF在NSCLC患者疾病分期、治疗反应和预后中的价值。方法采用ELISA方法检测66例晚期NSCLC患者和15例健康体检者血清bFGF的表达水平,并和临床指标进行比较分析。结果 NSCLC患者血清bFGF的表达水平显著高于正常对照组(3.2vs 20.1 pg/ml,P0.001);在治疗结束后bFGF水平逐步下降(20.1 vs 10.6 pg/ml,P0.05);生存分析发现NSCLC患者治疗前血清bFGF水平和患者无疾病进展生存显著相关,血清bFGF20 pg/ml患者的PFS显著短于bFGF水平较低患者(5.6月vs 9.6月,P0.001)。结论 NSCLC患者血清bFGF水平和疾病发生发展、疾病分期和疾病生存有关,可以作为NSCLC血管新生和预后的新指标。  相似文献

Previous studies suggest that members of the fibroblast growth factor (FGF) family are mitogenic to mammary epithelium. In order to determine expression of acidic and basic FGF (aFGF and bFGF) during mammary development, mice were euthanized as virgins, early pregnant, mid-pregnant, late-pregnant, or during early lactation. Mammary expression of both aFGF and bFGF mRNA increased through pregnancy. Acidic FGF mRNA continued to increase during early lactation, but basic FGF message level decreased drastically during early lactation. Western blots probed with anti-aFGF showed four immunoreactive bands approx 30, 48, 52, and 55-kDa in size. The 30-, 48-, and 55-kDa bands for aFGF were expressed at low levels during virgin and early pregnant stages but were more prominent during the later stages. The 52-kDa band was high during the virgin and early pregnant stages and low in mid-pregnancy through early lactation. Blots probed with anti-bFGF showed two bands approx 30 and 50 kDa in size. Both bands increased through early-pregnancy, but during late-pregnancy there was a decrease in immunoreactive protein levels, which remained low during early lactation. Experiments to determine where FGF mRNAs are produced in the mammary gland suggest that both FGFs may be produced in the stroma, leading to the hypothesis that aFGF and bFGF are stromally produced growth factors and probably act on the epithelial component of the gland in a paracrine fashion.  相似文献

目的利用生长因子通过诱导血管生成治疗缺血性心脏病,但生长因子的输入方式还存在一些争议。本研究旨在探讨心肌内注射碱性成纤维生长因子(bFGF)对急性心肌梗死(AM I)血管生成和心肌功能的作用。方法AM I犬24只,随机分成对照组(M I区注射生理盐水15 m l)和实验组(M I区注射50 mg bFGF与生理盐水的混合液15 m l);每组观察4个不同的时间点(术后0、2、9、16 d)。各组动物分别在处死前应用敏感编码技术(sensitivity en-coded techn ique,SENSE)行磁共振电影成像(c ine m agnetic resonance im aging,c ine-MR I);电镜直接观察新生血管生长情况;免疫组织化学方法检测各组微血管数量;以左心室射血分数(LVEF)评价心脏功能的改变。结果实验组LVEF自9 d明显增加[9 d:对照组(24±3)%、实验组(46±6)%;16 d:对照组(31±5)%、实验组(53±5)%];除0d外各个时间点的微血管数量实验组均比对照组明显增多[2 d:对照组(92±12)支/视野、实验组(147±12)支/视野;9 d:对照组(125±12)支/视野、实验组(183±14)支/视野;16 d:对照组(125±14)支/视野、实验组(224±20)支/视野]。结论心肌内注射bFGF具有促进M I区域毛细血管形成及改善左心室功能的作用。  相似文献

Summary The release of growth factors from ischaemic retina has been hypothesized as the central stimulus for retinal neovascularization in proliferative diabetic retinopathy. Two of the growth factors implicated are insulin-like growth factor-I and basic fibroblast growth factor. We examined the effect of insulin-like growth factor-I on in vivo neovascularization using the established angiogenic model of the rabbit cornea (n=30), and also compared the effects of insulin-like growth factor-I and basic fibroblast growth factor using two new in vivo systems. Either supraphysiologic concentrations of each growth factor (600 g) were injected intravitreally into pigmented rabbits (n=21) or porous polyfluorotetraethylene chambers filled with an emulsion containing collagen and growth factor (500 ng) were placed on the retina surface (n=8). Our results demonstrate that when insulin-like growth factor-I was implanted together with a slow release carrier into the pocket of the normally avascular cornea, insulin-like growth factor-I (10 g/pellet) induced angiogenesis in all rabbits. This degree of angiogenesis was comparable to that previously shown for basic fibroblast growth factor. For the intravitreal studies, the fibrotic component was greater in the basic fibroblast growth factor injected eyes, whereas the vascular component was accentuated in the eyes injected with insulin-like growth factor-I. Light and electron microscopy demonstrated areas of vascular proliferation in both groups. Porous polyfluorotetraethylene chamber studies with insulin-like growth factor-I and basic fibroblast growth factor demonstrated vascular proliferation in the vicinity of the chamber similar to the intravitreal injected eyes, but to a lesser degree than the injected eyes. Our experiments overall support the angiogenic potential of both insulin-like growth factor-I and basic fibroblast growth factor and support distinct but complimentary roles for each growth factor in the pathogenesis of proliferative diabetic retinopathy.  相似文献

目的:研究胃癌患者血清和组织中VEGF,bFGF的表达与胃癌临床特征之间的关系,研究二者的相关性及组织和血清之间的相关性,探讨VEGF,bFGF在胃癌的发生、发展、侵袭和转移中的作用方法:应用酶联免疫技术(ABC-ELISA方法)检测73例胃癌患者术前血清和20例健康体检者血清中的VEGF,bFGF的表达水平,同时应用免疫组织化学染色方法检测癌组织和癌旁组织中VEGF,bFGF的表达.结果:胃癌患者术前血清VEGF,bFGF表达水平均明显高于健康体检者(YEGF:101.8±53.3 ng/L vs 16.1±22.5ng/L,P<0.05;bFGF:152.9±42.7ng/L vs 25.0±11.4ng/L,P<0.05).胃癌患者术前血清VEGF,bFGF的表达水平均随胃癌的浸润深度、TNM分期、淋巴结转移、远处转移而增高(P<0.05),而与年龄、性别及病理类型无关.胃癌组织VEGF的阳性表达率为71.2%,癌旁组织中VEGF均未见阳性表达,二者之间有显著性差异(X~2=32.1,P<0.05);胃癌组织中bFGF的阳性表达率为63.0%、癌旁组织中bFGF阳性表达率为(10%),二者之间亦有显著性差异(X~2=17.7,P<0.05).胃癌患者组织VEGF,bFGF的表达水平均与胃癌的浸润深度、TNM分期、淋巴结转移、远处转移有关(P<0.05),而与年龄、性别及病理类型无关.胃癌患者血清VEGF的表达水平与血清bFGF的表达水平呈明显正相关(r=0.439,P<0.01),胃癌患者组织VEGF的表达水平与组织bFGF的表达水平呈明显正相关(r=0.391,P<0.01);胃癌患者术前血清VEGF的表达水平与组织VEGF的表达呈正相关(r=0.346,P<0.01),术前血清bFGF的表达水平与组织bFGF的表达呈正相关(r=0.304,P<0.01),均有显著性差异.结论:VEGF,bFGF在胃癌的发生、发展、转移及预后起着重要的作用,有望成为胃癌术前诊断、术后随访、复发转移监测、评价抗血管生成药物疗效和化疗效果判定的新的肿瘤标志物.  相似文献

目的 观察重组人碱性成纤维细胞生长因子 (rhb FGF)对缺血心肌血管结构的作用。方法 建立兔急性心肌梗死模型 ,将 rhb FGF直接四点注射入兔缺血心肌内 ,通过病理切片图像分析观察缺血心肌血管管壁及管腔变化情况。结果  rhb FGF组与生理盐水组平均管壁厚度分别为 6周 :(2 0 .70± 9.94 ) μm,(18.88± 9.6 5 ) μm,P>0 .0 5 ;12周 :(2 9.87± 12 .96 )μm,(18.13± 11.33)μm,P<0 .0 1;管壁管腔比值分别为 6周 :0 .2 5± 0 .12 ,0 .2 4± 0 .0 2 ,P>0 .0 5 ;12周 :0 .33± 0 .14 ,0 .2 4± 0 .0 9,P >0 .0 5。结论  rhb FGF促缺血心肌血管新生到一定时期 (可能小于 12周 ) ,血管数量不再增长 ,只是血管管腔增大 ,管壁增厚 ,发生血管重构。  相似文献

Based on previous results showing developmental regulation of aFGF and bFGF, we evaluated the effect of various hormones on aFGF and bFGF mRNA levels, in mammary gland. Northern blots indicated that estrogen alone increased aFGF but had no effect on bFGF messenger RNA level. Progesterone alone increased aFGF and bFGF mRNA levels. Estrogen and progesterone together increased aFGF mRNA level in mammary gland, but the increase was no greater than that caused by either hormone alone. However, the combination of estrogen and progesterone had no effect on bFGF message level. PRL or GH, when administered with estrogen and progesterone, increased aFGF, but did not have any effect on bFGF message level. However, when PRL and GH were administered together with estrogen and progesterone, they increased bFGF messenger RNA level. Ovarian steroid withdrawal increased aFGF, but did not have any effect on bFGF mRNA accumulation. PRL alone (in the absence of estrogen and progesterone) decreased aFGF, whereas it had no effect on bFGF message level. Hydrocortisone alone decreased aFGF, but increased bFGF mRNA level. However, PRL and hydrocortisone increased aFGF, but did not have any significant effect on bFGF message level. In the overall model, during growth of the mammary gland, ovarian steroids cause an increase in aFGF mRNA. During lactogenesis, ovarian steroid withdrawal causes an increase in aFGF messenger RNA levels. Lactogenic hormones together cause a further increase in aFGF message levels.  相似文献
