The acute toxicities of arsenate, arsenite, cadmium, chromate, copper, mercury, silver, vanadium, and zinc were determined, individually and in two environmentally relevant mixtures, to two life stages of chinook salmon in reconstituted fresh and brackish waters that stimulated potential conditions in the San Joaquin Valley, California. The relative individual toxicities of the elements varied over four orders of magnitude; from most toxic to least toxic, the rank order was cadmium greater than copper greater than mercury greater than zinc greater than vanadium greater than arsenite greater than arsenate greater than chromate (no definitive tests for silver). In general, young fish tested in fresh water were more sensitive to the individual elements and the two mixtures than were advanced fry tested in brackish water. A 13-element mixture simulating concentrations in the San Luis Drain had the same toxicity to fish as the same mixture plus selenate and selenite, thus demonstrating that the presence of selenium at its environmental ratio in the mixture contributed no toxicity in short-term tests. As judged by a comparison of the individual acute values for salmon to the expected environmental concentrations in the San Luis Drain, the margins of uncertainty for cadmium and copper in both waters, zinc in fresh water, and mercury in brackish water were less than 100, whereas for the two environmental mixtures the margins were less than 15 in both test waters. These low margins of uncertainty, especially for the mixtures, indicate a high potential for environmental hazard to chinook salmon in the reconstituted waters tested.  相似文献

A study was conducted to assess the potential for uptake of toxic chemicals by down-stream migrant salmon in an urban estuary. Juvenile chinook salmon (Oncorhynchus tshawytscha) were collected from the Duwamish Waterway (located in Seattle, Washington) and from the Nisqually River (a reference site). The mean concentrations of aromatic hydrocarbons and polychlorinated biphenyls (PCBs) in the stomach contents (food organisms) of salmon from the Duwamish Waterway were approximately 650 times and 4 times, respectively, higher than those in salmon from the Nisqually River. Similarly, the mean concentration of bile metabolites of aromatic compounds which fluoresce at benzo[a]pyrene wavelengths was 24 times higher in the urban salmon compared to the reference salmon, whereas the mean concentration of PCBs in liver of urban salmon was 3 times higher than that in reference salmon. The study clearly demonstrated that, during their residency in this urban estuary, juvenile chinook salmon bioaccumulate substantial levels of toxic chemicals. The possible effects of these chemical exposures on the health and survival of this species are not presently known.  相似文献

Structural changes of the intestinal mucosal cells of juvenile chinook salmon (Oncorhynchus tshawytscha) exposed in parallel experiments to a model mixture of petroleum hydrocarbons, chlorinated biphenyls, and the combined contaminants have been studied by light and electron microscopy. The mucosa was intact in the control and petroleum hydrocarbon-exposed fish; some exfoliation was observed in the group exposed to chlorinated biphenyls. The group fed the combined contaminants (petroleum and chlorinated biphenyls) had considerably increased sloughing indicating an interactive effect. The “goblet” or mucous cells appeared normal in all groups, but in the contaminant-exposed groups the columnar cells of the mucosa had distinct subcellular inclusions. The inclusions were not stained by carbohydrate-specific (PAS) or lipid-specific (Sudan black) reagents. Ultrastructurally, the inclusions that appeared after exposure to petroleum hydrocarbons were variable in size and contained a flocculent, finely granular material. In the petroleum hydrocarbon-exposed group the inclusions differed from both the chlorinated biphenyl and the combined-contaminant groups. The inclusions in the latter groups included large, irregularly shaped vesicles with relatively electron-transparent material and other vesicles with a range of electron densities. The cellular alterations were consistent within the exposed groups and presented a distinct change from normal morphology. Since the average gain in weight for all groups was similar, we could not conclude, however, that functional impairment from intestinal damage had occurred.  相似文献

We measured persistent organic pollutant (POP) concentrations in chinook salmon (Oncorhynchus tshawytscha) in order to characterize dietary exposure in the highly contaminated, salmon-eating northeastern Pacific resident killer whales. We estimate that 97 to 99% of polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), dichlorodiphenyltrichloroethane (DDT), and hexachlorocyclohexane (HCH) in returning adult chinook were acquired during their time at sea. Highest POP concentrations (including PCBs, PCDDs, PCDFs, and DDT) and lowest lipids were observed in the more southerly chinook sampled. While feeding by salmon as they enter some more POP-contaminated near-shore environments inevitably contribute to their contamination, relationships observed between POP patterns and both lipid content and delta13C also suggest a migration-related metabolism and loss of the less-chlorinated PCB congeners. This has implications for killer whales, with the more PCB-contaminated salmon stocks in the south partly explaining the 4.0 to 6.6 times higher estimated daily intake for sigmaPCBs in southern resident killer whales compared to northern residents. We hypothesize that the lower lipid content of southerly chinook stocks may cause southern resident killer whales to increase their salmon consumption by as much as 50%, which would further increase their exposure to POPs.  相似文献

To determine the influence of wide variations in dietary levels of calcium, zinc and phytic acid (as sodium phytate) on growth and cataract incidence, juvenile chinook salmon held at 10-11 degrees C were fed daily to satiation for 105 d one of nine purified diets containing one of three levels (grams/kilogram) of calcium (averaged 4.8, 17.7, 50.2), zinc (averaged 0.05, 0.15, 0.39) and phytic acid (1.62, 6.46, 25.8). Diets were formulated to have a calcium-phosphorus ratio of close to unity when considering phosphorus sources other than sodium phytate. High dietary phytic acid concentration (25.8 g/kg) depressed chinook salmon growth, food and protein conversion [protein efficiency ratio (PER)] and thyroid function, increased mortality, promoted cataract formation (zinc at 0.05 g/kg) and induced anomalies in pyloric cecal structure. Calcium at 51 g/kg (or phosphorus) exacerbated the effects of high dietary phytate and low dietary zinc on cataract incidence. Moreover, high dietary levels of calcium (48-51 g/kg) coupled with phosphorus significantly impaired the growth and appetite of low phytic acid (1.62 g/kg) groups and led to nephrocalcinosis in low and high phytic acid groups. Plasma zinc levels were directly related to dietary zinc concentration and inversely related to dietary phytic acid level. Calcium (51 g/kg) and/or phosphorus reduced zinc bioavailability when the diet concurrently contained 0.05 g zinc and 25.8 g of phytic acid per kilogram. It is concluded that zinc is essential for normal eye development in juvenile chinook salmon. Further, zinc deficiency could not be induced in chinook salmon fed diets with high ratios of calcium (or phosphorus) to zinc alone. The Chinook salmon (Oncorhynchus tshawytscha) is endangered or threatened in several of its ranges. The uptake of metals by Chinook salmon eggs and how humic acid (HA) affects the uptake is a subject of interest. Humic acid (0, 0.001, 0.01, and 0.05g/l) reduces the uptake of the metal ions Hg(II), Cd(II), and Zn(II), (1.0 microM) by eggs. HA is more effective in reducing the uptake of Hg than that of Cd or Zn. At [HA] = 0.001 g/L Hg uptake is reduced by 44% compared to no HA, while Cd and Zn uptakes are slightly or not reduced. Once the metals are taken up by the eggs, Hg migrates more slowly from the chorion to the yolk than either Zn or Cd. In experiments in which the metal contents of the chorion and yolk were measured at up to 24 h and five days after uptake, the order of migration was Cd > Zn > Hg. This observation is important when discussing the effects of metals on biological processes in the yolk because when Hg is taken up by eggs, a smaller percentage reaches the yolk than does Cd and Zn.  相似文献

Double-crested cormorant (Phalacrocorax auritus) eggs at pipping and sibling 10-day-old chicks were collected from two colonies in Green Bay, WI, one colony in Lake Michigan, WI, and reference colonies in South Dakota and Minnesota. Egg contents and chicks were analyzed for organochlorine contaminants including polychlorinated biphenyl (PCB) congeners. Livers of embryos and chicks were assayed for hepatic microsomal ethoxyresorufin-O-dealkylase (EROD) activity. Eggshell thickness and the physical dimensions of embryo brains were measured. Concentrations of organochlorines, including p,p′-DDE (p,p′-dichlorodiphenyldichloroethylene), PCBs, and PCB congeners were generally an order of magnitude higher in eggs and chicks from Wisconsin than from reference locations. Total PCBs averaged 10–13 μg/g wet weight in eggs from three Wisconsin colonies compared to 0.9 μg/g PCBs from reference locations. Double-crested cormorant chicks accumulated on average 33–66 μg PCBs/day and 7–12 μg p,p′-DDE/day in the Wisconsin colonies compared to 0 μg PCBs/day and 1 μg p,p′-DDE/day in the reference colonies. At pipping, EROD activity in the livers of cormorant embryos was significantly higher in the Wisconsin colonies and significantly correlated with PCBs and the toxic equivalents (TEQs) of aryl hydrocarbon-active PCB congeners relative to 2,3,7,8-tetrachlorodibenzo-p-dioxin. However, in 10-day-old chicks EROD activity was not consistently different among colonies and was not correlated with PCBs or TEQs. A significant negative relationship between embryo brain asymmetry and the size of the egg suggested that physical constraint might be an important factor influencing the response of this bioindicator. Thinner eggshells in two colonies located near Door County, Wisconsin, suggested that historic p,p′-DDE residues associated with orchards are still an important source of p,p′-DDE in the local environment. Organochlorine contaminants (OCs) in fish were determined to evaluate the potential risk to humans consuming fish originating in Baiyangdian Lake, North China. Relatively low levels of PCBs, HCHs and DDTs were observed, with mean concentrations ranging from 0.28 to 3.28 ng/g, wet weight. Among various fish species tested, the highest burden of OCs was recorded in northern snakehead (7.39 ng/g, wet weight) and the lowest was in grass carp (2.04 ng/g, wet weight). The hazard ratios (HRs), based on noncancer risk were all less than 1.0, while the HRs based on cancer risk exceeded 1.0 only for PCBs based on the 90th percentile concentration.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are ubiquitous marine and freshwater sediment contaminants. Extensive data exist to confirm that PAHs are toxic to aquatic receptors. However, limited information is available regarding the bioavailability and genotoxicity of sediment PAHs to aquatic organisms. This study investigated an integrated biomonitoring approach using chemical analyses and biomarkers to characterize the bioavailability and genotoxicity of a complex PAH mixture in freshwater lake sediments associated with a former manufactured gas plant (MGP). Sediment PAH genotoxicity was assessed by flow cytometry (FCM), DNA adduct (32)P-postlabeling, and erythrocyte micronuclei in juvenile coho salmon (Oncorhynchus kisutch) caged in the water column. Significant PAH-induced genotoxicity was observed with FCM and (32)P-postlabeling, but not with erythrocyte micronuclei. Chromosome damage in peripheral blood and hepatic DNA adducts correlated with sediment, but not water column PAH concentrations. Total hepatic DNA adducts in salmon caged nearest the former MGP facility was 39+/-6.5 (RALx10(9)), while salmon caged in a reference lake had 28+/-2.3 total hepatic DNA adducts per 10(9) nucleotides. These results indicate that in situ biomonitoring using biomarkers and caged fish can be a sensitive indicator of genotoxic PAHs in sediments.  相似文献
