OBJECTIVE To investigate the effect of Helicobacter pylori (H pylon) infection on expression of hMSH2 and P53 and its possible carcinogenic mechanism. METHODS H pylori infection was detected using a rapid urease test and haematoxylin and eosin (H&E) staining. The hMSH2 and P53 proteins in gastric cancer (GC) tissue, its adjacent mucosa, gastritic mucosa and normal gastric mucosa were detected by the immunohistochemical SP method. RESULTS (1) The positive rate of hMSH2 expression in GC tissue (62.7%)was higher than those in non-cancer tissues (P〈0.001); the positive rate of hMSH2 expression in poorly differentiated adenocarcinoma (76.4%) was higher than that in other carcinomas (P〈0.05). The positive rate of P53 expression in GC tissue (51.9%) was higher than that in noncancer tissues (P〈0.001); the positive rate of P53 expression in well-differentiated adenocarcinoma (32.6 %) was lower than that in other carcinomas (P〈0.01). (2) The positive rate of hMSH2 expression in GC tissue with H pylori infection was lower than that without the infection (52.8% vs. 74.5%; P〈0.05). The positive rate of P53 expression in GC tissue with H pylori infection was higher than that without the infection (61.4% vs. 40.6%; P〈0.05).(3) The expression of hMSH2 and P53 in GC tissue correlated positively (r=0.457, P〈0.01). CONCLUSION High expression of hMSH2 and P53 as well as their interaction may be involved in gastric carcinogenesis; H pylori infection affecting expression of hMSH2 and P53 may be one of its carcinogenic mechanisms.  相似文献

Objective: To investigate the relationship between matrix metalloproteinase-2 (MMP-2), MMP-9 as well as microvessel density (MVD) and their clinicopathological features in gastric cancer. Methods: The expression of MMP-2, MMP-9 and MVD were detected by immunohistochemistry SP method on 65 cases of gastric cancer tissue and 32 adjacent gastric mucosa. Results: The positive expression of MMP-2, MMP-9 and MVD in cancer group were significantly higher than those of adjacent mucosa group (P〈0.01). The positive expression of MM P-2 and MMP-9 had closely correlation with clinical features of lymphatic metastasis, pathological type and stages (P〈0.05), and the high level of CD 34 had correlation with metastasis and stages (P〈0.01). The positive expression of MMP-2 and MMP-9 showed significant correlation with the level of MVD. Prognosis was mostly affected by lymphatic metastasis and stages of clinical features. The low cum survival rate was showed in the groups of positive expression of MMP-2, MMP-9 and high level of MVD. Conclusion: MMP-2, MMP-9 and MVD play an important role in metastases, invasion and prognosis of gastric cancer, which is a valuable maker to evaluate the prognosis.  相似文献

Objective: The aim of this study was to detect metastasis-associated in colon cancer-1 (MACC1) expression in Chinese gastric cancer and analyze the relationship between MACC1 expression and postoperative survival. Methods: The expression of MACC1 and c-MET protein in a sample of 128 gastric cancer tissues was detected by immunohistochemistry. A retrospective cohort study on the prognosis was carried out and data were collected from medical records. Results: The positive rate of MACC1 protein expression in gastric cancer was 47.66%, higher than that in adjacent noncancerous mucosa (P<0.001). MACC1 protein expression was not related to the clinicopathological variables involved. Kaplan-Meier analysis revealed that the survival of MACC1 positive group tended to be better than that of MACC1 negative group, particularly in patients with stage III carcinoma (P=0.032). Cox regression analysis revealed that MACC1 protein over-expression in gastric cancer tended to be a protective factor with hazard ratio of 0.621 (P=0.057). Immunohistochemical analysis showed that the positive rate of c-MET protein expression was much higher in cases with positive MACC1 expression in gastric cancer (P=0.002), but P53 expression was not associated with MACC1 expression. Conclusion: MACC1 over-expression implies better survival and may be an independent prognostic factor for gastric cancer in Chinese patients.  相似文献

Objective: To estimate the practical values of pepsinogen C (PGC) dynamic expression and the levels of serum pepsinogens in gastric cancer screening and diagnosis. Methods: 129 cases gastric mucosa biopsies and serum specimens were examined. The expression of PGC in stomach mucosa was detected by immunohistochemistry. The serum concentration of pepsinogen A (sPGA) and pepsinogen C (sPGC) were determined by ELISA. Results: The positive rate of PGC antigen expression decreased in superficial gastritis (100%), gastric ulcer or erosion (80.00%), atrophic gastritis (34.48%) and gastric cancer (11.43%) in sequence (P〈0.05). There was no statistics difference in concentration of sPGA and sPGC among the above 4 groups. The ratio of sPGA/sPGC decreased in superficial gastritis, gastric ulcer or erosion, atrophic gastritis and gastric cancer in sequence (P〈0.05). There was specific correlation between the expression of PGC in stomach mucosa and the levels of sPGA/sPGC ratio in serum (rs =0.297, P=0.001). Conclusion: Tissue expression of PGC has close relationship with different gastric diseases. The ratio of sPGA/sPGC is relative with the tissue expression of PGC antigen and may be a convenient and economic maker in screening and diagnosis of gastric cancer.  相似文献

Objective: To investigate the expression of fragile histidine triad (FHIT) protein in normal colorectal tissue, colorectal adenoma and colorectal cancer (CRC), and to study the relationships between the expression of FHIT protein and the clinical pathology, the apoptosis-associated protein (Bcl-2, Bax, Survivin), apoptosis in colorectal cancer. Methods: Tissue microarray (TMA) and immunohistochemistry SP were used to detect the expression of FHIT gene, Bcl-2, Bax and Survivin in 16 cases of the normal colorectal tissue, 16 cases of colorectal adenoma and 80 cases of the colorectal cancer. TUNEL was used to detect the apoptosis index (Al) in 80 cases of the colorectal cancer. Results: (1) The positive rates of FHIT gene expression in normal colorectal tissue, colorectal adenoma and adenocancer were 93.75%, 68.75% and 46.25% respectively. There were no significant differences in the relationships between the FHIT gene expression and histological types, the gender as well as the age (P〉0.05). There were significant relationships between FHIT gene expression and lymph node metastasis, histological grades, Duke's system as well as the 5-year survival rate after operation. (2) The positive rates of Bax, Bcl-2 and Survivin in colorectal adenocancer were 72.50%, 51.25%, 77.50% respectively. The expression of FHIT gene was positively correlated with that of Bcl-2, Bax and Survivin. (3) The mean AI in FHIT negative tumors was significantly lower than that in FHIT positive tumors (P〈0.01). Conclusion: FHIT gene may play a role in the oncogenesis and progression of colorectal cancer. The abnormal regulation of apoptosis may play an important role in the pathogenesis of colorectal cancer.  相似文献

Objective: To study the expression of CD44s mRNA in the occurrence, development and invasion of gastric carcinoma (GC). Methods: The expressions of CD44s mRNA in 66 cases of GC, 25 cases of superficial gastritis and 25 cases of atypical hyperplasia were examined by in situ hybridization (ISH). Results: There was no expression of CD44s mRNA in the group of superficial gastritis; the positive rate was 20%(5/25) in the group of atypical hyperplasia and 62.12%(41/66) in the group of gastric carcinoma. The positive rate in poor differentiation group was significantly higher than that in well differentiation group (P〈0.05), and the positive rate of lymph node metastasis group was significantly higher than that in negative lymph node metastasis group(P〈0.05). Conclusion: The expression of CD44s mRNA was related to cell differentiation degree and lymph node metastasis, the activation of CD44s gene was related to strong invasion of cancer cells and poor prognosis.  相似文献

OBJECTIVE To evaluate the REGγ expression in gastric cancer tissue and gastric cancer cell lines of various differentiation levels and its clinical significance. METHODS Immunohistochemistry was used to detect the expression of REGγ protein in 70 specimens of gastric cancer and 30 specimens of normal gastric mucosa. The relationship between the expression of REGγ protein and the biological behaviors of gastric cancer was analyzed. RT-PCR and Western blot were used to detect the mRNA level and the protein expression of REGγ in normal gastric cell line GES-1, well differentiated gastric cancer cell line MKN-28, moderately differentiated gastric cancer cell line SGC-7901 and poorly differentiated gastric cancer cell line BGC-823. RESULTS The expression rate of REGγ protein in gastric cancer tissue (52/70, 74.29%) was significantly higher than that in normal gastric tissue (12/30, 40%) (P 〈 0.01). The expression rate of REGγ was correlated with tumor size (P 〈 0.01), lymph node metastasis (P 〈 0.05), differentiation degree (P 〈 0.01), infiltration depth (P 〈 0.01) and distant metastasis (P 〈 0.05). RT-PCR analysis showed that the expression of REGγ mRNA was 0.459 ± 0.079 in the normal gastric mucosa cell ling 0.588 ±0.118 in the well differentiated gastric cancer cell line, 0.715±0.066 in the moderately differentiated gastric cancer cell line, and 0.873 ± 0.099 in the poorly differentiated gastric cancer cell line, showing a negative correlation between REGγ mRNA expression and differentiation level (P 〈 0.05). Western blot analysis showed that the expression of REGγ protein was 0.712±0.065 in the normal gastric mucosa cell line, 1.176±0.185 in the well differentiated gastric cancer cell line, 1.533 ± 0.127 in the moderately differentiated gastric cancer cell line, and 2.061± 0.398 in the poorly differentiated gastric cancer cell line, showing a negative correlation between REGγ protein expression and differentiation level (P 〈 0.05). CONCLUSION REGγ is expressed in gastric cancer tissue and normal gastric tissue. In gastric cancer tissues, REGγ expression is positively correlated with the tumor size, lymph node metastasis, differentiation degree, infiltration depth and distant metastasis. Detecting the expression of REGγ mRNA and protein is helpful for early diagnosis and predicting prognosis of gastric cancer.  相似文献

Objective: Human carbonic anhydrases II (CAII) gene plays an important role in different cancer. However, its relevance to gastric cancer (GC) remains unclear. In the present study, we aimed to investigate the expression of CAII in GC and explore its correlation with some clinicopathologic characteristics of GC. Methods: The expression of CAII in 20 specimens of normal gastric mucosa, 38 specimens of intraepithelial neoplasia and 112 specimens of gastric carcinoma were detected by immunohistochemical techniques. Survival in GC with CAII expression was studied. Results: The positive rate of CAII protein in normal gastric mucosa was significantly higher than that in intraepithelial neoplasia and gastric carcinoma (100% vs. 63.16% and 28.57%, P<0.001). The positive rate of CAII protein was significantly higher in gastric carcinoma at early stages than that at advanced stages (70.0% vs. 19.57%, P<0.001). The positive rate of CAII protein was significantly lower in gastric carcinoma with lymph node metastases than that without lymph node metastases (10.81% vs. 37.33%, P<0.05). Furthermore, the positive rate of CAII protein was significantly lower in poorly-differentiated gastric carcinoma than in moderately- or well-differentiated gastric carcinoma (15.94% vs. 31.03% or 60.00%, P<0.05). Moreover, CAII expression was not related with sex, age and tumor size. The patients with CAII-positive tumors showed a better survival rate than those with CAII-negative tumors (P=0.024, log-rank test). Conclusion: CAII expression was related with stages and lymph node metastases in gastric carcinoma. The reduction of CAII expression in GC might promote tumor cell motility and contribute to tumor growth and metastasis.  相似文献

Objective: To analyse the expression of telomerase and apoptosis related protein, and explore the possible mechanism of breast cancer development. Methods: Immunohistochemistry method (SP) was used to detect the expression of hTERT, p53 and bcl-2 in the tissues of 48 cases of human breast cancer and 42 cases of benign lesions in breast. Results: The positive rates of expression of hTERT, p53 and bcl-2 in breast cancer were 87.50%, 56.25% and 54.17%, respectively. Compared with the groups of adjacent noncancerous and benign lesions, there was a significant difference among three types of tissues (P 〈 0.05). The positive rates of expression of p53 and bcl-2 in the group with positive expression of hTERT were 64.28% and 61.90%, respectively, and their difference was significant compared with the negative group (P 〈 0.05). Conclusion: There is a correlation between the activation of telomerases and p53 gene mutation in the development of breast cancer, and they are perhaps relation to the down regulation of bcl-2.  相似文献

目的:染色质重塑因子1(chromatin remodeling factor 1,Rsf-1)可作为肿瘤靶向治疗的靶点,因此探讨其在胃癌(gastric cancer)及癌前病变(precancerous lesions)组织中的表达及临床意义。方法:采用免疫组化SP法检测30例萎缩性胃炎、30例肠上皮化生、22例不典型增生Rsf-1蛋白表达水平。并检测64例胃癌与配对癌旁组织(手术切缘距离肿瘤>5 cm)中Rsf-1和p53蛋白表达水平并分别分析与胃癌临床病理参数间的关系,同时分析Rsf-1与p53在胃癌中表达的相关性。结果:Rsf-1在胃癌、肠上皮化生、不典型增生组织中的阳性表达率均高于癌旁组织(P<0.05)。Rsf-1在胃癌中表达与淋巴结转移有关(P<0.05)。p53在胃癌中的阳性表达率高于癌旁组织,其表达与淋巴结转移有关(P<0.05)。p53和Rsf-1在胃癌中的表达存在正相关性(r=0.38,P<0.05),两者在胃癌中共阳性表达与淋巴结转移有关(P<0.05)。结论:Rsf-1可能参与了胃癌病变的发生及发展,对胃癌早期筛查具有重要指导意义。Rsf-1和p53两者共阳性表达对于预测胃癌淋巴结转移可能具有一定的参考价值。  相似文献

背景与目的:新辅助治疗越来越受到关注和重视,有关术前区域动脉灌注化疗联合短期放疗对直肠癌Survivin表达有何影响仍不清楚。本研究旨在探讨术前区域动脉灌注化疗或放疗对Survivin表达影响及其与P53、Bax表达之间的关系。方法:60例低位直肠癌患者随机分为术前区域动脉灌注化疗联合放疗组19例(简称放化疗组)、术前短期放疗组15例(简称放疗组)和单纯手术组26例(简称对照组)。术前区域动脉灌注化疗采用Seldinger技术,术前短期放疗采用前后双野照射,放疗剂量每天每野2Gy,共5天20Gy,术后追加放疗30Gy,对照组术前不做放化疗,术后放疗50Gy。应用免疫组织化学方法检测新辅助治疗前后Survivin、P53和Bax表达。结果:新辅助治疗后,放化疗组、放疗组与对照组Survivin、P53、Bax阳性率无显著差异(P>0.05)。放化疗组Survivin阳性率由治疗前63.16%(12/19)降低至治疗后的26.32%(5/19),P53高表达率由治疗前57.89%(11/19)下降至26.32%(5/19);放疗组Survivin高表达率由治疗前53.33%(8/15)降低至治疗后的13.33%(2/15)(P<0.05),P53阳性率治疗前后分别为73.33%(11/15)、60.00%(9/15),无显著变化(P>0.05)。放化疗组、放疗组Bax阳性率分别由治疗前42.11%(8/19)、46.67%(7/15)增加到治疗后的63.16%(12/19)、73.33%(11/15),有升高趋势(P>0.05);新辅助治疗后(包括放化疗组和放疗组),Survivin表达“减少”组平均生存期(50.05个月)高于表达“增加”组(42.61个月)(P<0.01);Survivin表达变化与P53、Bax表达变化无相关性(P<0.05)。结论:新辅助治疗可以降低P53和Survivin表达,可能上调Bax表达,促进癌细胞凋亡,对低位直肠癌具较好的近期疗效。  相似文献

目的:分析CDGSH铁硫结构域2(CDGSH iron sulfur domain 2,CISD2)在胃腺癌组织中的表达及其与血管内皮生长因子(vascular endothelial growth factor,VEGF)和肿瘤微血管密度(microvessel density,MVD)的相关性及临床意义。方法:收集胃腺癌组织(胃癌组)及配对的癌旁组织(癌旁组)各357例,免疫组化法检测CISD2、VEGF的表达,CD34标记血管内皮细胞并计算MVD值。分析CISD2、VEGF的表达及其与临床病理参数的相关性。分析MVD、CISD2和VEGF的两两相关性。结果:胃癌组中,CISD2和VEGF的表达均高于癌旁组(70.87%比25.77%,61.34%比19.89%,均P<0.001)。CISD2、VEGF的表达与胃腺癌的浸润深度、分化程度、淋巴结转移、脉管及神经侵犯、肿瘤的TNM分期均相关(均P<0.05)。胃癌组中,CISD2阳性者的MVD值高于CISD2阴性者(35.67±6.06比31.98±4.87,P<0.001),VEGF阳性者的MVD值高于VEGF阴性者(36.64±6.08比31.38±4.54,P<0.001);癌旁组中亦然。胃癌组中,CISD2与VEGF的表达呈正相关(r=0.706,P<0.001)。结论:CISD2在胃腺癌中高表达,且与组织学分化、浸润、转移及高微血管密度等相关。CISD2可能与VEGF共同参与胃腺癌组织中微血管的形成。  相似文献

目的 探讨胃腺癌17号染色体倍性改变及TP53和拓扑异构酶Ⅱα(TOP Ⅱα)蛋白的表达在胃癌病理诊断中的意义.方法 选取胃癌中异倍体出现频率较高的17号染色体着丝粒探针,应用荧光原位杂交技术对99例胃腺癌、18例癌旁组织及5例远端胃黏膜进行分析.复阅相应肿瘤患者癌和癌旁组织病理切片,构建组织微阵列,对位于17号染色体上的TP53和TOP Ⅱα蛋白进行免疫组织化学检测和分析.应用SPSS 11.5软件进行统计.结果 胃癌和癌旁组织的17号染色体异倍体发生频率及TP53和TOP Ⅱα蛋白表达率分别为58.6%、45.5%、84.7%和0、12.1%、14.1%,差异有统计学意义(P=0.000).在58例检测到17号染色体异倍体的胃癌组织中,TP53表达阳性26例,TOP Ⅱα表达阳性49例.TP53在伴有不典型增生的癌旁黏膜中的表达率高于无不典型增生的癌旁黏膜(P=0.009).高、中分化腺癌的17号染色体异倍体发生率(69.8%)显著高于低分化腺癌(50.0%,P=0.048).贲门部胃腺癌的17号染色体异倍体发生率高于幽门部(P<0.05).肿瘤≥5 cm患者的TP53表达高于肿瘤<5 cm者(P=0.034).TOP Ⅱα蛋白在淋巴结转移数目≤6枚患者和pTNM分期早的患者中表达较高(P=0.010,P=0.003).17号染色体异倍体及TP53和TOP Ⅱα蛋白表达之间无相关性.结论 17号染色体异倍体及TP53、TOPⅡα蛋白表达的检测对胃腺癌辅助诊断及预后判断具有一定意义.  相似文献

BACKGROUND AND OBJECTIVES: Cyclooxygenase-2 (COX-2) expression may contribute to the synthesis of prostanoids, which have been related to carcinogenesis and tumor progression. It is well known that the gastric remnant is at greater risk of the development of gastric cancer than is the whole stomach; incidence rates for gastric cardia adenocarcinoma are rising in the United States and Europe. Our objective was to determine the involvement of COX-2 in primary and remnant gastric cancer tissues as well as in adjacent noncancerous mucosa. METHODS: We investigated the expression of COX-2 in 91 human gastric cancer tissue and adjacent noncancerous mucosa samples (40 remnant gastric cancer, 37 gastric cardia cancer, and 14 gastric corpus and antrum cancer), using immunohistochemistry. In addition, p53 expression, Helicobacter pylori infection, and vascular endothelial growth factor in the tissues were evaluated by immunohistochemical staining and compared with COX-2 expression. RESULTS: There were no significant differences in clinicopathological data in the gastric cancer tissues. There was a significant relation between the expression of COX-2 and p53 in gastric cancer tissues (P = 0.0048). However, vascular endothelial growth factor expression and Helicobacter pylori infection showed no correlation with the expression of COX-2. In the case of adjacent noncancerous mucosa, the positive rate of COX-2 expression was significantly higher in the remnant gastric cancers (75.0%) than in the primary gastric cancers (25.5%) (P < 0.0001). CONCLUSIONS: This information may help in the analysis of the carcinogenesis of gastric cancer; there is also a possibility that the COX-2 selective inhibitor to the remnant gastric cancer has a chemopreventive effect.  相似文献

目的探讨凋亡抑制基因survivin在喉鳞癌中的表达及其与Bcl-2、Bax基因表达的相关性。方法应用免疫组织化学链酶卵白素-生物素复合体(strept avidin-biotin complex,SABC)方法检测45例喉鳞癌及10例正常喉黏膜组织中survivin,Bcl-2及Bax基因的表达。结果喉鳞癌组织中,Survivin的表达率为80%,显著高于正常喉黏膜组织的表达率20%(P均<0.05)。Survivin蛋白表达与喉鳞癌的TNM分期及淋巴结转移有关(P均<0.05)。Survivin表达与Bcl-2蛋白表达呈正相关(P<0.05),而与Bax蛋白表达呈负相关(P<0.05)。结论Survivin在喉鳞癌的发生发展中起重要作用,可能成为喉鳞癌基因治疗的新靶点;survivin基因可能与凋亡相关基因Bcl-2、Bax在喉鳞癌的发展中分别起协同和拮抗作用。  相似文献

Objective:To investigate microvessel density (MVD) and expression of Survivin in non-small cell lung cancer (NSCLC),and to explore their correlations to clinicopathologic features of NSCLC.Methods:66 specimens of tumor adjacent tissues,and 20 specimens of benign lesions were detected by SP immunohistochemistry;their interrelations and correlations to clinicopathologic features of NSCLC were analyzed.Results:Positive rate of Survivin in NSCLC was significantly higher than that in adjacent tissues and benign lesions (54.5% vs.30%,P<0.05),its positive rate was 63.6% in lung adenocercinoma tissues,and its positive rate was 54.5% in lung SCC,there was no significant difference between them.Its expressions were respectively 41.2%,87.5% in well-differentiated and poorly differentiated lung SCC;its expressions were respectively 47.1%,81.2% in well-differentiated and poorly differentiated lung adenocarcinoma;the discrepancy had statistical significance.Its expression was 74.1% in lymphatic metastasis group,which was obviously higher than that in no lymphatic metastasis group,which was 48.7%.The rate of MVD in Survivin positive group was obviously higher than that of Survivin negative group.Conclusion:The expression of Survivin is connected with the degree of differentiation on lung cancer and lymphatic metastasis,which indicates Survivin can be as an index for the judgment of condition and the evaluation of prognosis,the expression of Survivin is direct correlation with that of MVD.It is evident that Survivin functions the promotion of tumor angiogenesis.  相似文献

目的 探讨甲状腺癌中B细胞淋巴瘤-2相关X蛋白(Bax)、生存素(Survivin)及Livin的表达及意义.方法 采用免疫组化法检测82例甲状腺癌组织及相应癌旁正常组织中Bax、Survivin及Livin的阳性表达,分析三者与临床病理参数的关系.结果 甲状腺癌组织中Bax的阳性表达率明显低于癌旁正常组织(P﹤0.001),Survivin及Livin的阳性表达率明显高于癌旁正常组织(P﹤0.001).Ⅲ~Ⅳ期甲状腺癌患者的Bax阳性表达率明显低于Ⅰ~Ⅱ期患者,有淋巴结转移患者的Bax阳性表达率明显低于无淋巴结转移患者(P﹤0.01).Ⅲ~Ⅳ期甲状腺癌患者的Survivin及Livin阳性表达率明显高于Ⅰ~Ⅱ期患者,有淋巴结转移患者的Survivin及Livin阳性表达率明显高于无淋巴结转移患者(P﹤0.01).甲状腺癌组织中Bax与Survivin的表达呈负相关(r=-0.435,P﹤0.01),Bax与Livin的表达也呈负相关(r=-0.551,P﹤0.01),Survivin与Livin的表达呈正相关(r=0.738,P﹤0.01).结论Bax在甲状腺癌组织中低表达,Survivin及Livin在甲状腺癌组织中高表达,并与TNM分期及淋巴结转移密切相关,三者联合检测可能成为甲状腺癌预后的参考指标.  相似文献
