香豆素(coumarins)是一类广泛存在于自然界中的天然化合物,具有广泛的药理作用,如抗肿瘤、抗人类免疫缺陷病毒(HIV)、抗氧化、抗菌、抗凝血、光敏等。同时已有研究证实,其药理作用与化学结构密切相关,因此可进一步研究开发更多的香豆素类化合物,从中寻找有效的先导化合物和活性基团,通过结构修饰和筛选,开发出高效低毒的临床药物。该文就近年来香豆素类化合物在肿瘤、艾滋病等治疗领域的研究作一综述。 相似文献

姜黄素类化合物(curcuminoids)是一类二芳基庚烷类化合物,主要存在于姜科、桦木科等植物中,以庚烷为母体,在1、7位有芳基取代,其中姜黄素、单去甲氧基姜黄素和双去甲氧基姜黄素最为常见。姜黄素类化合物具有良好的抗氧化、抗炎、抗肿瘤、降血糖、抗HIV、治疗心血管疾病及阿尔茨海默病等多种生物活性。由于其色泽稳定和毒性极低,也被广泛应用于食品添加剂、食品色素和医药领域中。1抗氧化的活性据研究,姜黄素类化合物具有较强的抗氧化活性,其抗氧化的活性是维生素E的10倍以上[1]。 相似文献

Drug development is a failure-prone endeavour, and more than 85% of drugs fail during clinical development, showcasing that current preclinical systems for compound selection are clearly inadequate. Liver toxicity remains a major reason for safety failures. Furthermore, all efforts to develop pharmacological therapies for a variety of chronic liver diseases, such as non-alcoholic steatohepatitis (NASH) and fibrosis, remain unsuccessful. Considering the time and expense of clinical trials, as well as the substantial burden on patients, new strategies are thus of paramount importance to increase clinical success rates. To this end, human liver spheroids are becoming increasingly utilized as they allow to preserve patient-specific phenotypes and functions for multiple weeks in culture. We here review the recent application of such systems for i) predictive and mechanistic analyses of drug hepatotoxicity, ii) the evaluation of hepatic disposition and metabolite formation of low clearance drugs and iii) the development of drugs for metabolic and infectious liver diseases, including NASH, fibrosis, malaria and viral hepatitis. We envision that with increasing dissemination, liver spheroids might become the new gold standard for such applications in translational pharmacology and toxicology. 相似文献

Coumarins have attracted intense interest in recent years because of their diverse pharmacological properties. Among these properties, their anticancer effect was most extensively examined. In this review, their broad range of effects on the tumours as shown by various in vitro and in vivo experiments as well as clinical investigations is discussed. Studies have indicated that coumarins elicit inhibitory effects on cell growth of various carcinoma cell lines and may be potential candidates for cancer therapy. These natural compounds have served as valuable leads for further design and synthesis of more active analogues. In view of the relative simplicity of the coumarin compounds and their mechanism of action, the coumarin pharmacophore may serve as an important model on which to develop new patterns in cancer chemotherapy. The aim of this review is to examine in detail the properties of the title compounds as anticancer agents. In view of their comparatively low toxicity, relative cheapness, presence in the diet and occurrence in various herbal remedies, it appears important to evaluate their anticancer potentialities. Moreover their synergistic activity in combination therapy with other well-known anticancer drugs could be the basis for the development of rational approaches to new forms of cancer chemotherapy. 相似文献

苍耳子为历代中医临床治疗鼻渊要药,有小毒,属传统记载的有毒中药。现代药理研究苍耳子具有抗菌、抗病毒、抗炎、镇痛、降血糖、抗过敏等作用。现代临床医学中苍耳子为鼻科常用药,尤其多用于治疗慢性鼻炎、慢性鼻窦炎。但临床中常因长期、过量、反复使用苍耳子以及其炮制不当或者未经炮制使用而发生中毒。毒理学研究提示其对多脏器均有损伤,主要对肝脏和肾脏的损害较为严重。查阅大量中外文献,综述苍耳子功效及毒性、化学成分和毒理方面的研究进展,并对其"效-毒"相关性问题进行探讨。 相似文献
