Abstract

Angiogenic imbalance contributes to the development of preeclampsia. We evaluated the protein expression of the proangiogenic placental growth factor (PlGF) and transforming growth factor beta 1 (TGF-β1) compared with the anti-angiogenic soluble fms-like tyrosine kinase receptor (sFlt1) and soluble endoglin (sEng) in HIV-infected normotensive and pre-eclamptic pregnancies.Blood was obtained from 110 pregnant women, enrolled in four groups, namely, HIV-negative normotensives (27); HIV-positive normotensives (31); HIV-negative pre-eclamptics (27) and HIV-positive pre-eclamptics (25), and was used to measure PlGF, TGF-β1, sFlt1 and sEng levels.Increased sFlt1 and sEng levels were associated with the pre-eclamptics (HIV negative and positive) compared with their counterparts. Decreased PlGF levels were observed between the HIV-negative pre-eclamptics versus HIV-negative normotensives, but levels differed significantly (p = 0.02) among the normotensives (HIV negative and positive). TGF-β1 remained unchanged across all groups. Higher sEng/TGF-β1 ratios were associated with the pre-eclamptics (HIV negative and positive) compared with their counterparts. This study demonstrated increased sFlt1 and sEng levels in pre-eclamptic compared with normotensive pregnancies, irrespective of the HIV status.  相似文献

随着葡萄糖浓度的增加,人肾小球系膜细胞转化生长因子β1(TGF-β1)、纤连蛋白(FN)和Ⅳ型胶原(ColⅣ)的表达显著增加(P<0.01).在30 mmol/L的高糖条件下,40~160 nmol/L的色素上皮衍生因子(PEDF)显著抑制TGF-β1表达,呈剂量依赖性(P<0.01);5~40 nmol/L的PEDF也剂量依赖性地抑制FN和ColⅣ的表达(P<0.01),提示PEDF可能通过其抗纤维化的作用延缓糖尿病肾病的进程.
Abstract:
The expressions of transforming growth factor β1 (TGF-β1), fibronectin (FN), and collagen Ⅳ in human mesangial cells were augmented with increased concentrations of glucose.Pigment epithelium-derived factor (PEDF) at concentrations of 40-160 nmol/L significantly inhibited TGF-β1 expression in a dose-dependent manner(P<0.01).PEDF at concentrations of 5-40 nmol/L significantly decreased FN and collagen Ⅳ expressions in a dose-dependent manner(P<0.01) ,suggesting that PEDF may play a salutary role in diabetic nephropathy by its antifibrogenic activity.  相似文献

The pathogenesis and aetiology of pre-eclampsia (PE) is still unclear. We investigated the role of angiogenic, antiangiogenic and vasoactive factors in black South African women with early- and late-onset PE. Serum soluble fms-like tyrosine kinase 1 (sFlt-1), soluble vascular endothelial growth factor (VEGF) and placental growth factor (PlGF) levels were determined using the ELISA technique, and placental mRNA expression levels of sFlt-1, VEGF, PlGF and AT1 receptors were determined using real-time PCR. Serum sFlt-1 levels were significantly elevated and PlGF significantly reduced in early-onset PE compared to the normotensive group. Placental VEGF mRNA expression levels were significantly reduced in the late-onset preeclamptic group compared with the normotensives. The placental mRNA expression of AT1 receptor in the late-onset pre-eclamptic group was relatively raised compared to the normotensives, suggesting hypersensitivity to pressor agents. We believe that the excess of serum sFlt-1 and reduced VEGF and PlGF levels favour an anti-angiogenic state and endothelial dysfunction leading to PE, and that the aetiology and pathogenesis of early- and late-onset PE differ.  相似文献

目的 观察色素上皮衍生因子(PEDF)、血小板反应蛋白-1(TSP-1)在糖尿病大鼠肾脏的表达变化,探讨促血管生成素-1(Ang-1)对上述因子的影响. 方法 将雄性SD大鼠分正常对照(NC)组、DN组、空载处理(BV)组、Ang-1处理(AV)组.采用STZ腹腔注射诱导大鼠DN模型.成模8周后尾静脉注射Ang-1腺病毒载体.多时点检测24 hUAlb和肾组织PEDF、TSP-1蛋白及mRNA表达水平.结果 DN、BV、AV组24 hUAlb均升高,其中AV组于20周后降低(P<0.05).DN、BV、AV组肾组织PEDF蛋白及mRNA表达下调,TSP-1表达上调(P<0.05),其中AV组12周后肾组织PEDF和TSP-1mRNA及蛋白表达变化较DN、BV组改善明显(P<0.05). 结论 糖尿病大鼠肾脏PEDF、TSP-1异常表达参与DN发生发展,给予Ang-1可改善糖尿病大鼠肾脏PEDF、TSP-1异常表达.  相似文献

目的研究可溶性血管内皮生长因子受体1(sVEGFR-1)和血管内皮生长因子(VEGF)在子痫前期及子痫患者血清中的浓度变化,探讨sVEGFR-1和VEGF在妊娠期高血压(子痫前期及子痫)中的作用。方法采用酶联免疫吸附试验(ELISA)分别测定轻度子痫前期(n=9)、重度子痫前期(n=10)及子痫(n=5)患者(子痫组,总共n=24)与正常妊娠妇女(对照组,n=18)外周血清中sVEGFR-1和VEGF的浓度。结果(1)子痫组各组外周血清中sVEGFR-1水平[子痫前期轻度:(58±7)mg/L、子痫前期重度:(82±15)mg/L、子痫:(96±13)mg/L]明显高于对照组[(11±3)mg/L,P<0·01],子痫前期重度和子痫组高于子痫前期轻度组(P<0·05);(2)子痫组各组外周血血清中VEGF水平[子痫前期轻度:(0·37±0·04)mg/L、子痫前期重度:(0·24±0·04)mg/L、子痫:(0·16±0·01)mg/L]明显低于对照组[(0·43±0·04)mg/L,P<0·05~0·01],子痫前期重度和子痫组低于子痫前期轻度组(P<0·05~0·01);(3)正常妊娠妇女外周血血清中sVEGFR-1水平与妊娠周数成正相关(r=0·508,P<0·05),子痫前期及子痫组血清中sVEGFR-1水平与妊娠周数无关。结论外周血中升高的sVEG-FR-1和降低的VEGF可能与妊娠期高血压(子痫前期及子痫)的发病有关,并参与了子痫前期及子痫的病理生理过程。  相似文献

目的观察冬虫夏草对高糖诱导的大鼠肾小球系膜细胞色素上皮衍生因子(PEDF)和血管内皮生长因子(VEGF)表达的影响,探讨其肾脏保护的作用机制。方法35只8周龄清洁级雄性sD大鼠,体重260~280g,根据体重按随机数字表法分为正常血清组(n=18)、低剂量虫草提取液喂养组(5g·kg^-1·d^-1,n=8)和高剂量虫草提取液喂养组(10g·kg^-1·d^-1,n=9),采用生理盐水及不同剂量虫草提取液灌胃8d后,分离静脉血提取对照血清及不同浓度的冬虫夏草含中药血清。体外培养HBZY-1大鼠肾小球系膜细胞,之后分别加入正常血糖正常血清[正常血糖组(NC组):5.6mmol/L葡萄糖+10%胎牛血清(FBS)+1%正常大鼠血清]、高血糖正常血清[高血糖组(HG组):30.0mmol/L葡萄糖+10%FBS+1%正常大鼠血清]、低剂量虫草含药血清[低药组(LD):30.0mmol/L葡萄糖+10%FBS+1%低剂量虫草含药血清]和高剂量虫草含药血清[高药组(HD):30.0mmol/L葡萄糖+10%FBS+1%高剂量虫草含药血清]进行培养,采用逆转录.聚合酶链反应(RT—PCR)测定各组细胞PEDF及VEGFmRNA的表达,酶联免疫吸附试验(ELISA)测定各组细胞上清液中PEDF及VEGF的水平。组间资料比较采用单因素方差分析,样本间资料比较采用t检验。结果与NC组相比,HG组PEDFmRNA表达明显下降(24h:0.375±0.011比0.507±0.008,t=16.828,P〈0.0l;72h:0.376±0.014比0.515±0.010,t=14.034,P〈0.01),VEGFmRNA表达明显上升(24h:1.005±0.011比0.864±0.012,t=14.963,P〈0.01;72h:1.168±0.012比0.873±0.011,t=31.417,P〈0.01);LD组和HD组PEDFmRNA表达较HG组显著上调(24h:0.505±0.018、0.639±0.012比0.375±0.011,F=354.719,P〈0.01;72h:0.612±0.023、0.700±0.019比0.376±0.014,F=97.82,P〈0.01),VEGFmRNA表达明显下降(24h:0.838±0.014、0.767±0.017比1.005±0.011.F=79.55,P〈0.01:72h:0.923±0.016、0.784±0.012比1.168±0.012,F=244.28,P〈0.01),且HD组疗效更明显。经对照血清及CS血清培养24、72h后,与NC组相比,HG组PDEF水平明显降低[24h:(267±8)比(303±10)ng/L,t=6.493,P〈0.01;72h:(265±27)比(338±42)ng/L,t=3.259,P〈0.01],VEGF明显升高[24h:(128±3)比(113±8)ng/L,t=3.737,P〈0.01;72h:(144±7)比(125±7)ng/L,t=4.215,P〈0.01;与HG组相比,LD组和HD组PEDF明显升高[24h:(297±12)、(296±26)比(267±8)ng/E,F=5.427,P〈0.01;72h:(323±20)、(332±33)比(265±27)ng/L,F=5.690,P〈0.01],VEGF明显降低[24h:(106±6)、(109±11)比(128±3)ng/L,F=5.738,P〈0.01;72h:(124±9)、(125±7)比(144±7)ng/L,F=7.878,P〈0.01]。结论冬虫夏草可能通过降低肾小球系膜细胞VEGF的表达,升高PEDF的表达,对肾脏起到保护作用。  相似文献

目的 观察外源性牛初乳短链胰岛素样生长因子 - 1(BC- t IGF- 1)对糖尿病生长激素 GH/ IGF- 1轴的影响。方法 链脲佐菌素 (STZ) 5 5 mg/ kg空腹腹腔注射 (IP)制备糖尿病大鼠模型 ,随机分为糖尿病对照组 (DM- C,生理盐水0 .1ml/ d IP)和糖尿病治疗组 (DM- BC,BC- t IGF- 12 5 0 ng/ kg· d- 1 IP)。并设正常对照组 (NC) ,每组 n=5。治疗前测定尾尖血血糖 (BG0 ) ,治疗 6周后测定尾尖血血糖 (BG)、血清果糖胺 (FMN)、甘油三酯 (TG)、胆固醇 (CHO)、GH、IGF- 1和肝脏 IGF- 1含量。结果 与 NC相比 ,DM- C血清 FMN、TG明显升高同时血清 GH升高而血清和肝脏 IGF- 1降低 ;与DM- C相比 ,DM- BC血清 FMN、TG明显降低同时血清 GH降低而肝脏 IGF- 1显著升高。结论 外源性 BC- t IGF- 1治疗改善糖尿病大鼠代谢同时改善了糖尿病时的 GH/ IGF- 1轴的紊乱  相似文献

目的:观察表皮生长因子受体(EGFR)在大鼠糖尿病性心肌病(DCM)纤维化中的作用。方法:将雄性Wistar大鼠25只随机分为正常对照组(n=10)和DCM组(建立的DCM大鼠模型,n=15),比较两组各指标的差异显著性。结果:造模2周后开始,与正常对照组相比,DCM组体重显著降低(P<0.05),空腹血糖水平明显升高(P<0.01);8周时超声心动图显示,与正常对照组相比,DCM组左室收缩末期内径(LVESd)[(3.23±0.19)mm比(3.65±0.26)mm]明显增加,左室舒张末期内径(LVEDd)[(7.07±0.43)mm比(6.21±0.25)mm]明显减小,左室射血分数(LVEF)值[(82.71±2.18)%比(72.5±3.02)%]明显降低(P均<0.05)。HE染色可见心肌细胞增生肥大、排列紊乱;Masson染色可见心肌纤维细胞增生明显,心肌胶原容积分数显著增大;Western blot方法检测EGFR表达水平显著高于正常对照组[光密度值(4.18±0.54)比(1.53±0.12)],P均<0.01。结论:表皮生长因子受体可加速糖尿病性心肌病大鼠心肌纤维化。  相似文献

Aims/hypothesis Our aim was to study whether pre-eclampsia and pregnancy-induced hypertension are predictors of diabetic nephropathy in type 1 diabetic women. Materials and methods A total of 203 type 1 diabetic women, who were pregnant between 1988 and 1996 and followed at the Department of Obstetrics and Gynaecology in Helsinki, were re-assessed after an average of 11 years within the nationwide, multi-centre Finnish Diabetic Nephropathy Study. Diabetic nephropathy was defined as microalbuminuria, macroalbuminuria or end-stage renal disease. Results Patients with prior pre-eclampsia had diabetic nephropathy more often than patients with a normotensive pregnancy (diabetic nephropathy vs normal albumin excretion rate: 41.9% vs 8.9%; p<0.001), whereas patients with a history of pregnancy-induced hypertension did not (10.3% vs 8.9%; p=0.81). CHD was more prevalent in patients with a history of pre-eclampsia than in patients with a normotensive pregnancy (12.2% vs. 2.2%; p=0.03). Pre-eclampsia (odds ratio [OR] 7.7, 95% CI 1.6-36.1; p=0.01) and HbA1c (OR 2.0, 95% CI 1.1-3.8; p<0.05) were associated with incident diabetic nephropathy even when adjusted for follow-up time, BMI, smoking, diabetes duration and age. Conclusions/interpretation These data suggest that a history of pre-eclamptic pregnancy but not pregnancy-induced hypertension is associated with an elevated risk of diabetic nephropathy. 目的 观察人工发酵虫草菌丝体干粉对糖尿病大鼠肾组织中色素上皮衍生因子(PEDF)、基质金属蛋白酶-2(MMP-2)和转化生长因子-β1(TGF-β1)表达的影响.方法 依照随机区组设计将45只体质量为180~220 g的8周龄雄性SD大鼠随机分为3组:正常对照组(A组,n=15)、糖尿病对照组(B组,n=15)、人工发酵虫草菌丝体干粉干预组(C组,n=15).B组和C组大鼠腹腔注射55 μg/g链脲佐菌素制作糖尿病动物模型.造模成功后第3天,C组给予4 μg·g-1·d-1人工发酵虫草菌丝体干粉定时灌胃,A组、B组以等体积生理盐水灌胃.实验12周末,观察比较3组肾脏指数(KI)、血肌酐(Scr)、尿素氮(BUN)、24 h尿量、24 h尿白蛋白排泄量(UAE)、总胆固醇(TC)、甘油三酯(TG).免疫组化方法分析比较3组PEDF、MMP-2和TGF-β1在肾组织中的表达,实时定量-聚合酶链反应(RT-PCR)分析比较3组PEDF mRNA的表达情况.两组间比较采用t检验,多组间比较采用方差分析.结果 B、C组大鼠KI、BUN、SCr、UAE、TC、TG均高于A组(均P〈0.01),C组均较B组降低(均P〈0.01).免疫组化显示,B组和C组肾脏PEDF(1.53±0.12、2.60±0.15)、MMP-2(1.8±0.5、3.3±0.4)表达均低于A组(3.96±0.14、4.3±0.6)(均P〈0.01),TGF-β1(4.60±0.15、2.60±0.09)表达均高于A组(1.57±0.09)(均P〈0.01),而C组PEDF、MMP-2表达高于B组(P〈0.01),TGF-β1表达低于B组(P〈0.01).RT-PCR分析显示B、C组肾脏组织中PEDF mRNA表达量为0.3±0.1和0.8±0.2,显著低于A组(1.2±0.3)(均P〈0.01),而C组高于B组(P〈0.01).结论 肾脏组织中PEDF、MMP-2和TGF-β1表达的改变可能参与了糖尿病肾病的发生.人工发酵虫草菌丝体干粉可改善肾功能、调节血脂,还可通过调节PEDF、MMP-2及TGF-β1在肾脏的表达发挥肾脏保护作用.  相似文献

通过转染表达人全长脂联素基因的重组腺病毒载体来提高2型糖尿病大鼠血清脂联素水平.结果表明脂联素干预组血清可溶性细胞间黏附分子1(sICAM-1)和C反应蛋白(CRP)水平较糖尿病组明显下降[(18.73±4.66对23.60±4.25)mg/L和(14.91±1.79对19.09±2.95)ng/ml,均P<0.01],提示提高循环脂联素水平可改善糖尿病炎症反应状态.  相似文献

AIMS: The aim of this study was to examine the prevalence of pre-eclampsia in Southern, Eastern, and Northern Finland, and the relationship between history of pre-eclampsia and maternal coronary artery disease (CAD) risk factors. METHODS AND RESULTS: Women aged 25-64 years, who participated in a cross-sectional population survey and had been pregnant (n=3650), were studied. The proportion of women who had ever had pre-eclampsia was lower in Southern (7.9%) compared with Northern Finland (13.9%) (P=0.001), but did not differ from Eastern Finland (11.1%). In the logistic regression model, the age-adjusted prevalence of pre-eclampsia was 1.92-fold in Northern (95% CI: 1.46-2.53, P<0.001) and 1.47-fold in Eastern Finland (95% CI: 1.11-1.96, P=0.008) compared with Southern Finland. The odds ratios (ORs) were 1.70 (95% CI: 1.21-2.38, P=0.002) and 1.16 (95% CI: 0.82-1.64, P=0.40), respectively, when adjusted for age at first birth, current age, parity, body mass index (BMI), increased blood cholesterol, hypertension, diabetes/impaired glucose tolerance, CAD, and mother's myocardial infarction. History of pre-eclampsia was associated with increased blood cholesterol, higher current BMI and blood pressure, and higher current prevalence of hypertension, diabetes/impaired glucose tolerance. CONCLUSIONS: Pre-eclampsia is most prevalent in the Northern part of Finland and could only be partly explained by higher prevalence of CAD risk factors.  相似文献

AimTo discuss available information on the opportunity for pregnant women affected by diabetes/obesity to receive COVID-19 vaccine.Data synthesisPregnant women with SARS-CoV-2 (COVID-19) infection are at high risk for severe acute respiratory syndrome and adverse outcomes. Pregnant women with severe COVID-19 present increased rates of preterm delivery (<37 gestational weeks), cesarean delivery and neonatal admissions to the intensive care unit. Comorbidity such as diabetes (pregestational or gestational) or obesity further increased maternal and fetal complications. It is known that diabetic or obese patients with COVID-19 present an unfavorable course and a worse prognosis, with a direct association between worse outcome and suboptimal glycol-metabolic control or body mass index (BMI) levels. Critical COVID-19 infection prevention is important for both mother and fetus. Vaccination during pregnancy is a common practice. Vaccines against COVID-19 are distributed across the world with some population considered to have a priority. Since pregnant women are excluded from clinical trials very little information are available on safety and efficacy of COVD-19 vaccines during pregnancy. However, it is well known the concept of passive immunization of the newborn obtained with transplacental passage of protective antibodies into the fetal/neonatal circulation after maternal infection or vaccination. Moreover, it has been reported that COVID-19 vaccine-induced IgG pass to the neonates through breastmilk. Therefore, maternal vaccination can protect mother, fetus and baby.ConclusionsAfter an individual risk/benefit evaluation pregnant and lactating women should be counselled to receive COVID-19 vaccines.  相似文献

ObjectiveThe purpose of the present study was to investigate the fear of hypoglycemia (FoH) and its predictors among diabetic pregnant women.Study designCross-sectional conducted between January to August 2022.MethodsIn the present study, 250 diabetic pregnant women from Qazvin province participated. Demographic and fertility characteristics, FoH, adherence to treatment, self-efficacy, anxiety and depression were assessed. Data were analyzed using univariable and multivariable linear regression models.ResultsThe participants’ mean age was 31.02 years (SD=4.72). The FoH mean score was 32.88 (out of 72). Based on the multivariable linear regression model, having a history of hypoglycemia (β = 0.44, p < 0.001), lower education (β = 0.17, p = 0.001), being treated with insulin (β = 0.22, p < 0.001), being treated with both insulin and diet (β = 0.16 p = 0.003), being of younger age (β = ?0.13, p = 0.008), and depression (β = 0.16, p = 0.002) were independent predictors of FoH among pregnant women.ConclusionDiabetic pregnant women experience FoH, particularly those with a history of hypoglycemia. Therefore, providing education and counseling concerning hypoglycemia, complications, and necessary measures for this group of diabetic pregnant women are needed along with those who are younger, less educated, and have comorbid mental health conditions.  相似文献

Objective:To investigate the expression of hypoxia inducible iaclor-1α(HIF-1α)and vascular endothelial growth factor(VECF)in diabelic retinopathy(DR)rats and its effect on the DR occurrence and development.Methods:A total of 120 SD rats were randomly divided into trial group and control group with 60 in each.STZ.i.p.was used in the trial group to establish the DM model,citrate buffer salt of same amount was used up.to the control group.1,3 and 6 months after injection,respective 20 rats were sacrificed in each group to observe expression of HIF-1αand VEGF in the rat retina tissue at different lime points.Results:Expression of HIF-1αand VEGF were negative in the control group;expression of HIF-1αand VKGF protein in retinal tissue were weak after 1 month of DR mold formation.It showed progressive enhancement along with the progression in different organizations,differences between groups were significant(P0.05).Conclusions:Expressions of HIF-1αand VF.GF were;correlated with disease progression in early diabelic relinopathy.Retinal oxygen can induce over-expression of HIF-1αand VEGF.It shows that HIF-1αand VEGF play an important role in the pathogenesis of DR.  相似文献

目的 探讨血红素氧合酶-1(HO-1)及抗血管生长因子——Avastin与DR的相关性. 方法 选取T2DM无视网膜病变组(NDR) 55例,背景期DR组(BDR) 53例,增殖期DR组(PDR) 56例,老年单纯白内障对照组(Con) 50例.采用ELISA对血液及房水的HO-1、Avastin进行检测. 结果 与Con组比较,BDR组和PDR组血液及房水HO-1[血:(17.47±5.75)、(19.25±6.95) vs(15.32±3.56)ng/ml;房水:(0.77±0.45)、(0.89±0.36)vs(0.52±0.47) ng/ml]、Avastin[血:(5.69±4.11)、(7.32±3.45)vs(1.49±0.86) ng/ml;房水:(32.58±18.74)、(55.12±32.17)vs(7.85±6.73) pg/ml]均升高(P<0.01),且随DR严重程度升高更明显. 结论 HO-1、Avastin与DR相关,可能参与了DR的发生发展过程.  相似文献
