目的 探讨bcl-2和P^53蛋白在鼻咽癌发生发展中的作用。方法 采用抗bcl-2和P^53蛋白的单克隆抗体,对25例鼻咽癌(Nasopharyngeal carcinoma,NPC)标本和16例正常鼻咽上皮(Nasopharyngeal epithelia,NE)标本进行免疫组化检测。结果 bcl-2和P^53蛋白的阳性表达率在鼻咽癌组织中分别为84.0%(21/25)和76.0%(19/25),与正常鼻咽上皮组相比差异有极显著性意义(P<0.005);鼻咽癌中有16例同时表达bcl-2和P^53蛋白。此外,bcl-2蛋白多见于分化差的鼻咽癌,而P^53蛋白则在相对分化较好的鼻咽癌中较多。结论 bcl-2原癌基因和P53抑癌基因在鼻咽癌的发病中可能有不同的意义,二种基因的协同作用可能与鼻咽癌持续生长有关。  相似文献

目的:探讨鼻咽癌组织中的环氧化酶2(COX-2)、潜伏膜蛋白-1(LMP1)的表达情况,同时分析了两者之间的关系。方法:53例鼻咽癌活检标本,8例鼻咽部慢性炎症常规石蜡切片标本作为对照组,用SP免疫组织化学法检测COX-2、LMP1的表达,采用sPsS12.0统计软件对实验数据进行统计。结果:在鼻咽癌组织中,COX-2阳性表达率为71.70%(38/5a);LMPI阳性表达率为66.04%(35/53)。COX2、LMP1的表达在颈部淋巴结转移组中均明显高于无颈部淋巴结转移组(P%O.05),但在年龄、性别、病理类型、临床分期的分组中均差异无统计学意义(均P〉0.05)。在鼻咽癌组织中,LMP1和COX-2的表达存在正相关(7—0.797,P〈0.01)。结论:COX-2、LMP1在鼻咽癌组织中均有表达。COX2、LMP1在鼻咽癌中的表达和鼻咽癌颈淋巴结转移存在显著相关性,但和年龄、性别、病理类型、临床分期无相关性。COX-2和LMP1在鼻咽癌组织中的表达呈正相关。LMP1可能通过多途径上调COX-2的表达,从而促进鼻咽癌颈部淋巴结转移。  相似文献

目的 探讨鼻咽癌中血管生长因子VEGF、bFGF的表达及其与颈淋巴结转移的关系。方法 采用免疫组化S-P法对35例鼻咽癌(其中15例同时取颈部转移癌组织),20例鼻咽粘膜不典型增生组织和10例慢性炎症组织中VEGF、bFGF的表达进行了检测。结果(1)35例鼻咽癌中17例(48.6%)VEGF表达阳性,明显高于非癌对照组(6.7%,P=0.000),有颈淋巴结转移鼻咽癌中VEGF的表达(73.3%)明显高于无颈淋巴结转移者(30%,P=0.018);晚期(Ⅲ、Ⅳ期,70.6%)明显高于早期(Ⅰ、Ⅱ期,27.8%,P=0.018):但咽癌原发灶中VEGF的表达与颈部转移灶中的表达无相关性(P>0.05);(2)35例鼻咽癌中有8例(22.9%)bFGF表达阳性,明显高于非癌对照组(3.3%,P=0.031)。但鼻咽癌中bFGF的表达与患者的年龄、性别、肿瘤的组织学类型和分期无关,也与肿瘤的N分期无关(P>0.05)。结论 (1)VEGF与鼻咽癌的生长及颈淋巴结转移有关,它可能可以作为预测鼻咽癌颈淋巴结转移的的指标;(2)bFGF可能与鼻咽癌的生长有关系,但与肿瘤的颈淋巴结转移可能无关。  相似文献

目的:探讨抑癌基因PTEN(phosphatase and tensin homolog deleted in chromosome10),P^21wafl在鼻咽癌发病中的作用及其与EB病毒的关系。方法:分别应用免疫组化(SP)染色法检测鼻咽癌组,慢性鼻咽炎组,鼻咽部不典型增生或癌前状态组中PTEN、P21wafl的表达及分布情况。结果:慢性鼻咽炎组PTEN蛋白阳性表达率(20/25,80.0%)高于不典型增生或癌前状态组(8/11,72.7%)及鼻咽癌组(32/78.41.0%)。Ⅰ~Ⅱ期鼻咽癌PTEN蛋白阳性表达高于Ⅱ~Ⅳ期。慢性鼻咽炎组P^21wafl蛋白阳性表达率(6/25,24.0%)低于不典型增生或癌前状态组(4/11,36.4%)和鼻咽癌组(60/78,76.9%)。Ⅰ~Ⅱ期鼻咽癌P^21wafl蛋白阳性表达率低于Ⅲ~Ⅳ期,发生淋巴结转移鼻咽癌组P^21wafl阳性表达率高于无淋巴结转移组的阳性表达率。结论:PTEN的失活或降低与鼻咽癌发生及临床分期有关,是鼻咽癌发生发展的早期事件。P^21wafl蛋白表达增多与鼻咽癌发生及发展进程、临床分期、淋巴结转移有关,可能是鼻咽癌发生、发展过程中的晚期事件。  相似文献

目的探讨COX-2和VEGF表达活性与鼻咽低分化鳞癌颈淋巴结转移的相关性。方法56例鼻咽低分化鳞癌组织标本,免疫组化法检测其COX-2和VEGF蛋白表达活性。本组患者中,伴有颈淋巴结转移(N+)者44例,不伴颈淋巴结转移(N-)者12例。结果颈淋巴结转移组鼻咽原发灶组织标本COX-2和VEGF的表达阳性率分别为84.1%和70.5%,不伴颈淋巴结转移组相应指标的阳性率分别为50.0%和33.3%,组间差异具有统计学意义(P〈0.05)。原发灶组织COX-2和VEGF的活性表达相关性为r=0.372,P〈0.005。结论COX-2和VEGF表达活性与鼻咽低分化鳞癌颈淋巴结转移具有相关性。  相似文献

目的检测鼻咽癌中血管内皮生成因子c(VEGF-C)的表达及微淋巴管密度(microlymphatic vessel density,MLVD),探讨鼻咽癌淋巴管生成及淋巴转移之间的关系。方法采用免疫组化sP法检测58例鼻咽癌和20例鼻咽部炎性反应组织中VEGF—C的表达情况,并用淋巴管内皮细胞特异性抗体LYVE-1行免疫组化染色,计数肿瘤内MLVD,并结合临床病理特征进行分析。结果鼻咽癌组和炎性反应对照组VEGF.C的阳性表达率分别为84.5%(49/58)和15.0%(3/20),差异有统计学意义(x。:32.309,P〈0.01)。鼻咽癌组织MLVD为(28.6±6.2)个/视野,鼻咽炎性反应组为(10.5±3.0)个/视野,两组差异有统计学意义(t=12.491,P〈0.01)。鼻咽癌组织中,有淋巴转移组VEGF.C阳性表达率(87.8%)明显高于无淋巴转移组(76.5%);有淋巴转移组MLVD(30.2±6.4)个/视野高于无淋巴转移组(24.8±3.6)个/视野,经统计学分析,差异均有统计学意义(t=3.259,P〈0.01)。VEGF—C的表达与MLVD(t=3.512,P〈0.01)、淋巴转移(χ^2=7.715,P〈0.01)、临床分期(χ^2=4.250,P〈0.05),与病理分化程度无关(χ^2=0.000,P〉0.05)。VEGF-C的表达与MLVD(t=3.512,P〈0.01)、淋巴转移(χ^2=7.715,P〈0.01,r=0.712)、临床分期(χ^2=4.250,P〈0.05,r=0.481)相关,与病理分化程度无关(χ^2=0.000,P〉0.05)。结论在鼻咽癌组织中VEGF-C呈高表达,VEGF-C表达与鼻咽癌组织MLVD、淋巴转移、临床分期密切相关。VEGF-C可能参与了鼻咽癌发生、浸润和转移的过程。VEGF-C与肿瘤组织微淋巴管密切相关,在鼻咽癌发展过程中起重要作用,可能成为抗肿瘤治疗的潜在靶点。  相似文献

目的:研究肿瘤转移相关基因Mtal(metastasis-associated gene)表达与喉癌淋巴结转移的关系。方法:应用RT-PCR技术,对30例喉癌组织、12例喉癌颈部淋巴结和10例正常喉部组织,进行了Mtal mRNA表达的检测。结果:有转移喉癌原发灶Mtal mRNA高表达率为100%(8/8),无转移为22.7%(5/22),P〈0.01;有癌转移淋巴结高表达率为87.5%(7/8),无癌转移为0(0/4),P〈0.05。30例喉癌有13例Mtal mRNA高表达,高表达率为43.3%。结论:Mtal mRNA表达与喉癌淋巴结转移呈相关关系。基因Mtal可成为判断喉癌转移有价值的指标。  相似文献

目的探讨FXYD6蛋白表达与鼻咽癌(NPC)患者临床病理特征的关系。方法采用免疫组织化学技术检测128例鼻咽癌组织中FXYD6蛋白的表达情况。结果 FXYD6蛋白在鼻咽癌组织中的表达率为84.3%(108/128),其蛋白在正常组中的阳性表达率为5.0%(4/80),两者差异有统计学意义(P〈0.01)。FXYD6蛋白在有颈部淋巴结转移组中的表达率为94.2%(65/69),高于无颈部淋巴结转移组72.8%(43/59),两组比较差异有统计学意义(P〈0.0 1)。FXYD 6蛋白的表达与NPC的病理组织学分型,临床分期无明显关系(P〉0.05)。结论 FXYD6蛋白高表达与NPC颈部淋巴结转移相关,提示FXYD6表达的上调可作为NPC患者预测淋巴结转移的参考指标。  相似文献

目的了解抑癌基因nm23-H1蛋白在鼻咽癌组织中的表达,探讨其与鼻咽癌发生发展及转移的关系。方法采用免疫组化法检测43例鼻咽癌患者鼻咽部组织中nm23-H1蛋白的表达,分析鼻咽癌患者不同临床分期及鼻咽癌是否伴有淋巴转移组织中nm23-H1蛋白表达的差异;同期选取17例慢性鼻咽炎患者鼻咽部组织作为对照。结果鼻咽癌组织中nm23-H1蛋白阳性表达率为53.4%,慢性鼻咽炎组织中nm-23H1阳性率为88.2%,两者比较有统计学意义(P〈0.05);鼻咽癌Ⅰ-Ⅱ期组织中阳性率为88.2%,Ⅲ-Ⅳ期组织中阳性率为30.8%,两者比较有统计学意义(P〈0.05);鼻咽癌伴有淋巴结转移组和不伴有淋巴转移组阳性率分别为38.5%、76.5%,两者比较有统计学意义(P〈0.05)。结论抑癌基因nm23-H1蛋白表达下调,可能与鼻咽癌发生发展、侵袭转移有关。  相似文献

目的探讨表皮钙黏素(E-cadherin,E-CD)基因在鼻咽癌组织中的蛋白表达及其意义。方法用免疫组化技术检测了38例鼻咽癌和43例鼻咽炎症黏膜组织中E-CD蛋白的表达。结果E-CD蛋白在鼻咽炎症黏膜组织中表达率(81.4%),明显高于鼻咽癌中的阳性表达率(55.3%),P<0.05。E-CD蛋白表达与鼻咽癌临床分期相关不明显,P>0.05。与肿瘤的浸润转移呈负相关,即有颈淋巴结转移的鼻咽癌中E-CD表达率(45.8%),明显低于无颈淋巴结转移的鼻咽癌中表达率(71.4%),P<0.05。结论E-CD蛋白表达程度与鼻咽癌浸润、转移有关,通过E-CD蛋白的检测可了解鼻咽癌生物学特性及判断其预后提供有价值的指标。  相似文献

Abnormal Wnt signaling and impaired cell–cell adhesion due to abnormal E-cadherin and β-catenin function have been implicated in many cancers, but have not been fully explored in nasopharyngeal carcinoma. The aim of this study was to analyze β-Catenin cellular location and E-cadherin expression levels in nasopharyngeal carcinoma. E-cadherin expression levels were also correlated with clinical data and underlying pathology. β-Catenin and E-cadherin expression were examined in 18 nasopharyngeal carcinoma and 7 non-tumoral inflammatory pharynx tissues using immunohistochemical methods. Patient clinical data were collected, and histological evaluation was performed by hematoxylin/eosin staining. β-catenin was detected in membrane and cytoplasm in all cases of nasopharyngeal carcinoma, regardless of histological type; in non-tumoral tissues, however, β-catenin was observed only in the membrane. As for E-cadherin expression levels, strong staining was observed in most non-tumoral tissues, but staining was only moderate in nasopharyngeal carcinoma tissues. E-cadherin expression was associated with β-catenin localization, study group, metastatic disease, and patient outcomes. Reduced levels of E-cadherin protein observed in nasopharyngeal carinoma may play an important role in invasion and metastasis. Cytoplasmic β-catenin in nasopharyngeal carcinoma may impair cell–cell adhesion, promoting invasive behavior and a metastatic tumor phenotype.  相似文献

目的:探讨上皮钙黏附蛋白(E-cadherin)、黏附分子CD44H、基质金属蛋白酶-3(MMP-3)、肿瘤转移抑制基因nm23H1和血管内皮生长因子(VEGF)基因的表达与鼻咽癌颈淋巴结转移及远处转移的关系。方法:应用免疫组织化学SP法检测62例鼻咽癌组织(鼻咽癌组)和40例正常鼻咽黏膜组织(对照组)中E-cadherin、CD44H、MMP-3、nm23H1和VEGF的表达。结果:在鼻咽癌有淋巴结转移组中,E-cadherin和nm23H1的表达减弱,CD44H和MMP-3的表达增强,与无淋巴结转移组的表达差异有统计学意义,VEGF在两组的表达差异无统计学意义;E-cadherin与CD44H、MMP-3的表达分别呈负相关关系,CD44H与MMP-3的表达呈正相关;在鼻咽癌远处转移组中,E-cadherin和nm23H1的表达减弱,CD44H、MMP-3和VEGF的表达均增强,与无远处转移组比较,差异均有统计学意义;CD44H与VEGF的表达呈正相关关系。结论:E-cadherin、CD44H、MMP-3和nm23H1与鼻咽癌颈淋巴结转移及远处转移的关系密切,VEGF仅与鼻咽癌颈淋巴结转移相关,它们是肿瘤转移的重要因素,可作为评估鼻咽癌转移的生物学指标。  相似文献

Adhesion molecules as prognostic factors in nasopharyngeal carcinoma.   总被引:3,自引:0,他引:3  
OBJECTIVE/HYPOTHESIS: To identify the significance of molecular markers in determining the risk of recurrence and distant metastases in nasopharyngeal carcinoma. STUDY DESIGN: In this retrospective case study, we evaluated archival nasopharyngeal carcinoma specimens for patterns of expression of E-cadherin, beta-catenin, c-erb-B2, and Ki-67, which have been demonstrated to be important in other tumors. METHODS: Fifty-four cases of nasopharyngeal carcinoma were identified, with a maximum follow-up of 13 years. The histopathological sections were stained using an automated immunohistochemical stainer (NexES, Ventana Medical Systems, Tucson, AZ) for E-cadherin (Zymed Laboratories [San Francisco, CA] and Transduction Laboratories [Lexington, KY] clones), beta-catenin (Zymed), c-erb-B2 (Ventana Medical Systems), and Ki-67 (Novocastra, Burlingame, CA). The numbers of positively staining cells were scored as follows: 0%, 1% to 33%, 34% to 66%, or greater than 67%. RESULTS: E-cadherin (Zymed) stained positively in only one case. The Transduction Laboratories clone demonstrated a spectrum of staining in all cases, from complete to disrupted to no identifiable membranous staining. The staining was consistently absent at the advancing tumor border, regardless of stage. The loss of beta-catenin expression did not correlate with that of E-cadherin or with clinical outcomes. No staining was identified for c-erb-B2. Ki-67 staining was variable and did not correlate with clinical outcomes. CONCLUSIONS: Altered expression or loss of E-cadherin, or both, may result in loss of function, particularly at the infiltrating edge, with resultant loss of cell polarity, cell migration, and eventual metastasis. The interpretation of E-cadherin staining depends on antibody source. In contrast to recent studies, beta-catenin expression is not altered and c-erb-B2 expression not identified, suggesting that these markers are not important in the prognosis of nasopharyngeal carcinoma.  相似文献

目的研究转录因子Snail、Slug和E-cadherin在鼻咽癌组织中的表达,并探讨其临床意义。方法采用实时定量PCR和免疫组化法检测65例鼻咽癌样本和18例鼻咽炎性组织中Snail、Slug、E-cadherin基因的mRNA和蛋白表达情况,分析三者表达和鼻咽癌临床分期及转移的关系。结果 随着鼻咽癌分期的增高,Slug基因的表达逐渐增加(I期0.54±0.20,Ⅱ期0.85±0.28,Ⅲ期0.89±0.21,Ⅳ期1.03±0.18),E-cadherin的表达呈下降趋势(I期58.54±20.13,Ⅱ期45.12±13.56,Ⅲ期24.68±10.08,Ⅳ期14.37±9.16),而Snail的表达无显著变化(I期0.57±0.19,Ⅱ期0.70±0.22,Ⅲ期0.75±0.24,Ⅳ期0.66±0.31)。E-cadherin的表达与鼻咽癌的淋巴结转移和远端转移有相关性(P〈0.05),Snail、Slug的表达与淋巴结转移及远端转移无统计学相关性(P〉0.05)。结论鼻咽癌中存在Slug和E-cadherin两个基因的表达失调,提示两者参与了鼻咽癌的发病过程。  相似文献

OBJECTIVE: The present study was aimed to determine whether p16/MTS1, nm23-H1, E-cadherin, and CD44 proteins were expressed in nasopharyngeal carcinoma (NPC) and whether those expressions were pathologically significant in the progress of NPC. METHOD: We examined non-cancerous nasopharyngeal mucosa (20 cases) and NPC (80 cases) using immunohistochemistry with six different types of monoclonal antibodies against p16, nm23-H1, E-cadherin, CD44H, CD44v3, and CD44v6 proteins. RESULTS: The results showed that 1) the rates of positive p16 protein expression and of preserved E-cadherin protein expression in NPC were significantly lower than those in non-cancerous tissue (P <.01); 2) no significant difference in the rate of positive expression of nm23-H1, CD44H, CD44v3, and CD44v6 proteins were observed between non-cancerous nasopharyngeal mucosa and NPC; 3) no significant difference in the expression of those proteins were found by respective correlation analyses of sex, stage, and size of primary tumor in NPC; and 4) no significant difference in the rates of positive expression of CD44H, CD44v3, and CD44v6 proteins were observed in NPC between with and without lymph node metastasis, indicating that those gene products did not correlate with lymph node metastasis in NPC. However, there were inverse correlations between the expression of p16, nm23-H1, or E-cadherin protein and lymph node metastasis (P <.05), indicating that the expression of p16, nm23-H1, and E-cadherin gene were related to the carcinogenesis and tumor progression of NPC. CONCLUSION: Detecting the expressions of those gene products may provide clinically valuable information for therapeutic strategy and for predicting the prognosis of patients with NPC.  相似文献

目的:F-box蛋白家族中的一员Skp2参与细胞周期抑制蛋白P27kip1泛素化降解,研究发现Skp2在乳腺癌、胃癌、前列腺癌等多种恶性肿瘤中表达增加。本研究旨在探讨Skp2与P27kip1在鼻咽癌组织中的表达及与鼻咽癌多项临床病理特征的关系,并探讨两者的相关性。方法:鼻咽癌组39例,慢性鼻咽炎组24例。标本均经石蜡包埋,HE染色确诊。Skp2和P27kip1蛋白的检测采用免疫组化染色(PV-9000二步法)。结果:(1) Skp2在鼻咽癌组织中的阳性表达率为71.79%,明显高于其在慢性鼻咽炎组织中的阳性表达率(41.67%),差异有统计学意义(P<0.05);P27kip1在鼻咽癌组织中的阳性表达率为35.90%,明显低于其在慢性鼻咽炎组织中的阳性表达率(79.17%),差异有统计学意义(P<0.01);(2) Skp2、P27kip1的蛋白表达与鼻咽癌的临床分期、颅神经侵犯有关(P<0.05),与年龄、性别、分化程度、颈淋巴结转移无关(P>0.05);(3) Skp2、P27kip1在鼻咽癌组织中的表达呈负相关(rs=-0.373,P=0.019)。结论:鼻咽癌中Skp2蛋白表达与靶蛋白P27kip1蛋白降解有关,Skp2、P27kip1可能与鼻咽癌的发生发展有关,联合检测Skp2、P27kip1蛋白表达可作为判断鼻咽癌生物学行为的指标之一。  相似文献

目的:研究鼻咽癌中血管生成拟态(vasculogenic mimicry,VM)和上皮-间质转化(epithelial-mesenchymal transition,EMT)相关标记物的表达及与淋巴结转移的关系。方法免疫组化检测82例鼻咽癌和20例鼻咽慢性炎组织E-cadherin、Twist和Vimenitn表达,CD31联合PAS染色检测VM。结果 VM在鼻咽癌组织的阳性率为28.05%,与淋巴结转移具有正相关关系(χ2=6.927, P=0.008)。27例出现E-cadheirn表达减弱甚至缺失,23例和35例出现Vimentin和Twist阳性表达,E-cadherin表达与淋巴结转移具有密切负相关关系(χ2=9.694,P=0.002), Vimentin和Twist表达均与淋巴结转移具有密切正相关关系(χ2=6.411,P=0.011;χ2=14.576,P=0.000),VM和EMT标志物具有密切相关性(P均<0.000)。结论鼻咽癌中存在VM和EMT,存在VM和发生EMT的鼻咽癌易发生淋巴结转移,VM和EMT具有密切相关性,两者对鼻咽癌转移具有一定的预测价值。  相似文献

OBJECTIVE: To explore the value of E-cadherin and focal adhesion kinase (FAK) expression in the prediction of cervical lymph node metastases in squamous cell carcinoma of the supraglottic larynx. DESIGN: Immunohistochemical analysis of retrospectively selected cases. Patients The study population was composed of 95 previously untreated men with squamous cell carcinoma of the supraglottic larynx. Intervention All the patients underwent surgical resection of the tumor and bilateral neck dissection. MAIN OUTCOME MEASURES: E-cadherin and FAK expression in relation to nodal metastases. RESULTS: Decreased E-cadherin expression was correlated with the presence of nodal metastases (P = .006). The combination of E-cadherin and FAK expression resulted in a superior accuracy in assessing nodal metastasis (P = .001). Histological grade was also associated with nodal metastases (P = .005). Multivariate analysis confirmed that these parameters were independent predictors of nodal metastases. In addition, the cases with decreased E-cadherin and increased FAK expression presented a significantly reduced disease-specific survival (P = .005). CONCLUSION: The combination of the expression of E-cadherin and FAK could increase our ability to identify patients with clinically negative lymph nodes who are at considerable risk for occult metastases.  相似文献

目的:研究COX-2、PCNA及p53的基因产物在鼻咽癌组织中的表达及与其生物学行为和生存率之间的关系。方法:采用免疫组化SP法检测60例鼻咽癌及30例正常鼻咽组织中COX-2、PCNA 及p53的蛋白表达水平。结果:(1) 鼻咽癌组织COX-2、PCNA 及p53阳性表达率分别为90%、75%和68% ,明显高于正常鼻咽组织;(2) 鼻咽癌组织COX-2、PCNA及p53的表达与临床分期、分化程度和淋巴结转移相关(P<0.05);(3) COX-2、PCNA及p53的表达密切相关;(4) 鼻咽癌组织COX-2、PCNA 阳性表达和生存率呈负相关,是鼻咽癌复发的显著预后因素。结论:鼻咽癌组织中COX-2、PCNA表达水平明显上调,与鼻咽癌的发生发展及预后不良有关,免疫组化检测COX-2、PCNA表达可用以判断鼻咽癌患者的预后。  相似文献
