In hypokalaemic rats maintained on a potassium deficient diets for 10–50 days, the isolated Na-loaded and K-depleted (Na-rich) muscle fibers showed the membrane potential less than –115 mV in fresh muscles of normal rats in K+-free Krebs solution. Upon adding 5 mM K+ to the K+-free medium bathing the soleus muscles, the measured potentials of Na-rich muscles always exceeded the membrane potentials of fresh muscles in 5 mM K+. The hyperpolarization was dependent on the amount of intracellular Na+ concentration ([Na]i) accumulated during the potassium deficiency. The electrogenic Na-pump was activated by an increase of [Na]i of less than 5 mM. Further increases in [Na]i resulted in increases in membrane potential which appeared to approach a limit at [Na]i levels higher than 65 mM.  相似文献

Summary In vitro investigations have shown that the adsorption capacity of activated charcoal (Kohle-Compretten, Ultracarbon, E. Merck, Darmstadt, FRG) is just as high as that of Fuller's earth (Surrey powder, Laporte Industries Ltd., Luton, GB) or Bentonite BP W.B. (Steetley Minerals Ltd., Milton Keynes, GB). Fuller's earth (Fullererde) from another manufacturer has had very poor adsorption properties and is thus not suitable for the treatment of paraquat poisoning. Animal experiments have shown that the curative effect of activated charcoal given 0.5, 1, 2, and 3 h after ingestion of 200 and 300 mg paraquat/kg body weight is equally as good or even better than that of Fuller's earth or Bentonite BP W.B.. Activated charcoal is a substitute of equal value to these mineral soils.  相似文献

Using apparent fronto-parallel plane (AFP) monitoring techniques, the relative stability of the abathic plane, i.e. Euclidean visual space, was investigated in 16 volunteers with a median age of 23.5 years under 160 g/kg psilocybininduced ergotropic arousal. Handwriting area and pressure were also measured in the same subjects.Drug-induced contraction of nearby visual space was inferred from changes of AFP curvature and tilt, as well as from increased handwriting area at drug peak. The rising horizon (Rennert) in the drawings of schizophrenics is also considered a manifestation of the contraction of visual space and is described in terms of an arousal-dependent transformation of constancies. The projection of central nervous system activity as experience out there is also discussed as an arousal-dependent learned constancy.  相似文献

Summary Transitions in myosin patterns were examined in situ by immunofluorescence in two fast muscles of the developing chicken, the pectoralis and the posterior latissimus dorsi. Myosin isoforms were localized using stage-specific monoclonal antibodies against the heavy chain of pectoralis myosin. Two antibodies (12C5 and 10H10) recognize adult and late embryonic myosin. They reacted weakly with both the pectoralis and posterior latissimus dorsi at 10 days in ovo, but intensely at 18 days in ovo. Both muscles were completely unreactive with an adult-specific antibody (5C3), indicating that the staining with 12C5 and 10H10 at 18 days in ovo reflects embryonic myosin. Thus two different embryonic isoforms are expressed sequentially in each muscle. Both 12C5 and 10H10 reacted weakly again with these muscles after hatching. The reappearance of a strong positive response to both antibodies, at 28 days in the pectoralis and after 60 days in the posterior latissimus dorsi, correlated well with the first appearance of a response to the adult-specific antibody, 5C3, signalling the beginning of the adult pattern. Both muscles reacted strongly with an antibody (5B4) specific for neonatal myosin between 18 days in ovo and 60 days after hatching. In the pectoralis, embryonic was replaced by neonatal myosin in most fibres by 14 days after hatching; by 28 days, both adult and neonatal myosin were expressed in most fibres; and in the adult, neonatal myosin was replaced entirely by the adult isoform. In contrast, many fibres in the posterior latissimus dorsi still expressed both embryonic and neonatal myosins up to at least 60 days post-hatch, and the remaining fibres expressed the neonatal isoform; the neonatal isoform was present in some fibres even in the adult posterior latissimus dorsi. We have therefore demonstrated in situ four different heavy chain isoforms in two different fast muscles. Early embryonic, late embryonic, neonatal and eventually adult isoforms are expressed in each muscle and more than one isoform often coexists in the same fibre.  相似文献

Summary Tension responses of rat ventricular trabeculae subjected to successive treatment with EGTA and Triton X-100 are described in order to investigate the effects of chemical skinning techniques. In some preparations the alkaloid saponin was also used before Triton. Ultrastructural evidence is cited that the EGTA-treatment fails to render cells hyperpermeable, i.e. freely permeable to small ions, whereas both saponin and Triton do so. In this paper we show that contractile responses like those described previously for the EGTA-treated tissue can be obtained. However, more detailed examination shows that such behaviour is quantitatively distinct from that of conventionally skinned fibres in a way that is incompatible with the notion of hyperpermeability. The Ca-sensitivity after treatment with either EGTA, saponin or Triton is identical in our hands. However, this is not explained by free access of Ca (and EGTA) to the intracellular space in the EGTA-treated preparation: contractures develop with very different time courses, being fastest after Triton and only marginally slower when first exposed to saponin but a factor of five times slower after EGTA-treatment alone. This applies to contractures evoked direct from Ca2+ concentration 10–9 m to the test Ca2+ concentration at constant total buffer concentration.EGTA-treated fibres develop tension when ATP or creatine phosphate (CrP) are removed from the bath. However, responses to ADP and to CrP changes persist with millimolar levels of ATP present, quite unlike the Triton-skinned muscle. Exposure to each of a variety of solutions for 24h produce preparations showing similar behaviour: whatever the explanation for the EGTA-skinning phenomenon it is not dependent upon low bathing Ca2+ concentration. On the basis of the functional characteristics described here, and the structural results cited, we conclude that the cell membrane continues to function as a selective permeability barrier after EGTA-treatment: this treatment does not produce a model of a selectively skinned cardiac cell.  相似文献

Proteasomes catalyse the degradation of proteins responsible for the regulation of mitosis enabling the cell to complete cell division. We have studied the effect of an inhibitor of the chymotrypsin-like activity of the proteasome on the trilaminar structure of the kinetochore in HeLa cells. Whereas a role for the proteasome in the degeneration of the kinetochore was predicted, we found instead that the inhibitor strongly retarded kinetochore development. We observed different developmental stages of the kinetochore from the fibrous ball of a prekinetochore to the mature kinetochore in one cell. The data presented here support the proposition that proteasomes are involved in kinetochore formation.accepted for publication by H. C. Summary In an electron-microscopical study the occurrence and ultrastructural features of electron-dense dark variants of pinealocytes were evaluated in the gerbil pineal gland. A few dark pinealocytes, which tended to form small clusters of contiguous cells, could consistently be detected in pineals fixed and embedded by various procedures. Apart from the different degree of electron density, the only conspicuous difference between dark and electron-lucent light pinealocytes concerned their compartment of synaptic-like microvesicles. Thus, both variants of pinealocytes contained abundant clear microvesicles of variable size which accumulated in dilated process terminals. However, the vesicles within the process endings of dark pinealocytes showed an unusually dense arrangement throughout the cytoplasm. As was demonstrated by immunogold staining, the accumulations of vesicles in the dark terminals contained synaptophysin, a major synaptic vesicle-associated protein. This protein is present in small clear vesicles with putative secretory functions in a wide variety of neuroendocrine cells and has previously been shown to be a common constituent of microvesicles in mammalian pinealocytes. Since gerbil pinealocytes displayed distinct gradations of electron density, their ultrastructural heterogeneity may be the expression of different states of secretory activity of one pinealocyte cell type. On the other hand, differences in the content of synaptic-like microvesicles in the process terminals of light and dark cells could also indicate a principal functional heterogeneity of the microvesicular compartment among pinealocytes, pointing to the existence of different types of pinealocytes.  相似文献

Normal human subjects, sitting in a chair, were required to maintain stable elbow flexion against loads of 0.5 kg or 1.0 kg. Unloading was affected either passively by the experimenter, or actively with the subject's own contralateral arm. Elbow angle, force exerted by the load, and electromyographic activity (EMG) of biceps and triceps muscles of both arms were recorded and averaged. Passive unloading was followed by a reduction of biceps EMG activity, starting 50–80 ms after weight lift, and by an upward deflection of the forearm. With active unloading, however, a reduction of the biceps EMG activity slightly preceded the onset of unloading (0–30 ms). This reduction of the actively unloaded arm occurred at about the same time as the activity of the contralateral unloading arm. In this experiment, the unloaded forearm maintained an almost stable position. Thus, the anticipatory adjustment of elbow posture, observed when unloading was performed by the subject, appears to optimize limb stability during the mechanical perturbation.  相似文献

The exposure of amphibian muscle to osmotic shock through the introduction and subsequent withdrawal of extracellular glycerol causes vacuolation in the transverse tubules. Such manoeuvres can also electrically isolate the transverse tubules from the surface (detubulation), particularly if followed by exposures to high extracellular [Ca2+] and/or gradual cooling. This study explored factors influencing vacuolation in Rana temporaria sartorius muscle. Vacuole formation was detected using phase contrast microscopy and through the trapping or otherwise of lissamine rhodamine dye fluorescence within such vacuoles. The preparations were also examined using electron microscopy, for penetration into the transverse tubules and tubular vacuoles of extracellular horseradish peroxidase introduced following the osmotic procedures. These comparisons distinguished for the first time two types of vacuole, open and closed, whose lumina were respectively continuous with or detached from the remaining extracellular space. The vacuoles formed close to and between the Z-lines, but subsequently elongated along the longitudinal axis of the muscle fibres. This suggested an involvement of tubular membrane material; the latter appeared particularly concentrated around such Z-lines in the electron-micrograph stereopairs of thick longitudinal sections. Open vacuoles formed following osmotic shock produced by extracellular glycerol withdrawal from a glycerol-loaded fibre at a stage when one would expect a net water entry to the intracellular space. This suggests that vacuole formation requires active fluid transport into the tubular lumina in response to fibre swelling. Closed vacuoles only formed when the muscle was subsequently exposed to high extracellular [Ca2+] and/or gradual cooling following the initial osmotic shock. Their densities were similar to those shown by open vacuoles in preparations not so treated, suggesting that both vacuole types resulted from a single process initiated by glycerol withdrawal. However, vacuole closure took place well after formation of open vacuoles, over 25 min after glycerol withdrawal. Its time course closely paralleled the development of detubulation reported recently. It was irreversible, in contrast to the reversibility of open vacuole formation. These findings identify electrophysiological detubulation of striated muscle with closure of initially open vacuoles. The reversible formation of open vacuoles is compatible with some normal membrane responses to some physiological stresses such as fatigue, whereas irreversible formation of closed vacuoles might only be expected in pathological situations as in dystrophic muscle.This revised version was published online in September 2005 with corrections to the Cover Date.  相似文献

Summary Mechanisms of resistance to beet necrotic yellow vein virus (BNYVV) were studied by comparing the multiplication and distribution of BNYVV in root tissue of some beet accessions. Seedlings were infected either by soil containing resting spores ofPolymyxa betae with BNYVV, or by a viruliferous zoospore suspension. With both inoculation methods high virus concentrations were obtained in rootlets of the susceptible cultivar Regina. Using infested soil, low virus concentrations were found in the partially resistant cultivar Rima and in the resistant accessions Holly and WB42. When a zoospore suspension was used, similar virus concentrations occurred in Rima and Holly as in Regina, while a low virus concentration was found in WB42. By in situ localisation studies, using immunogold-silver labelling, virus was detected in Regina after infection by soil or a zoospore suspension, but it could only be detected in the resistant accessions after infection by a zoospore suspension. In rootlets of Regina, Rima and Holly, virus was found in the epidermis, cortex parenchyma, endodermis, and interstitial parenchyma, but in general not inside the vascular tissue. In WB42 the virus, occurring in small aggregates, seemed to be restricted to the epidermis and some cortex parenchyma cells. Comparing both the multiplication and distribution of BNYVV in rootlets of the accessions studied, it is concluded that the virus resistance mechanism in Rima and Holly is different from that in WB42.  相似文献

We have used caged-ATP to investigate the kinetic behavior of KATP channels in ventricular cells from rat heart. In whole cells, loaded with caged-ATP, an increase of intracellular [ATP] following a UV light flash produced a decrease of KATP channel current that was too slow ( 300 ms) to be explained by the expected timecourse of ATP release ( 3 ms) and the time-course of channel blockade by ATP ( 20 ms). In isolated membrane patches, caged-ATP itself caused partial blockade of KATP channels. Under these conditions, photorelease of ATP caused channel activity to decline further. The results suggest that caged-ATP can bind to the KATP channel but, on binding, decreases the open probability to a lesser extent than does ATP. Additionally, the observations indicate that for photolytically-generated ATP to bind to the channel, caged-ATP must first unbind (slowly) from the channel. We conclude that caged-ATP is not fully caged with respect to its allosteric action on the KATP channel.  相似文献

Summary The effects of saphenous nerve stimulation on discharge activity of supraoptic neurosecretory (NS) cells were studied in anesthetized rats. Of 112 supraoptic neurosecretory cells, 62 exhibited a phasic discharge pattern. The nerve stimulation transiently excited 46 of these 62 phasic units, as well as 35 of the 50 remaining non-phasic units. No appreciable blood pressure change was noted using PSTHs with 1-ms resolution. Though the nerve stimulation also evoked a flexor reflex of the ipsilateral hind limb, blockage of the hind limb movement with gallamine did not alter the amplitude of the supraoptic cell excitation. The threshold of the nerve stimulation was higher for the excitation than for the flexor reflex. Effects of hypovolemic and hyperosmotic stimuli on discharge activity of phasic cells during saphenous nerve stimulation were studied to find a possible interaction between these stimuli. Hemorrhage potentiated the transient excitation evoked by the nerve stimulation in all of the 8 phasic cells tested, while no such effect was seen after an injection of hypertonic sodium chloride solution in the 7 phasic cells tested. These electrophysiological data suggest that hypovolemic and noxious stimuli potentiate VP secretion in a synergistic manner but that hyperosmotic and noxious stimuli do not.Supported by grants from the Ministry of Education, Science and Culture, Japan  相似文献

This study focuses on recent improvement in epithelial monolayer cultures originating from whole extirpated Botryllus schlosseri (Urochordata) buds. Buds (n = 2,000) were taken at different (A to D) blastogenic stages. We tested the suitability of 35 combinations of various substrates and media on attachment, cell spread, epithelial growth frequencies and on monolayer lifespans. Under favorable conditions, cultured buds at blastogenic stages B to D (but not stage A) started to attach to the substrates following a 3-day transient period that leads to formation of spheres and attached monolayers. Substrate type is important for the attachment and the development of monolayers. Under various culture conditions, some of stages B and C buds develop (3–20 days) one or more large (1 mm diameter) spheres. Stage D buds develop monolayers (up to 20% of buds) without going through a sphere phase. Neither spheres nor attached monolayers of epithelium were observed in stage A bud cultures. Spheres grew at a rate of 60 m in diameter per day using specific medium types and did not attach unless the appropriate substrate was present. When attached, epithelial monolayers expanded at a rate of 200 m in diameter per day, for 3–15 days, and subsequently detached and died. Sixteen types of media were tested. Medium and substrate combinations were found to determine epithelial lifespan. These results revealed significant improvements in the culture of epithelial monolayers from Botryllus palleal buds. However, an early senescence of the developed epithelial sheets (up to two weeks from onset of appearance) may indicate an internal ageing clock that should be taken into consideration in future approaches.  相似文献

Summary The vestibular signal of head motion in space must be complemented by a neck signal of the trunk-to-head excursion in order to provide the individual with information on trunk motion in space. This consideration led us to study psychophysically the role of vestibular-neck interaction for human self-motion perception. Subjects (Ss) were presented with passive horizontal rotations of their trunk and/or head (sinusoidal rotations, f=0.025 –0.4 Hz) in the dark for vestibular and neck stimulation, as well as for combinations of both. Ss' perception was evaluated in terms of gain (veridical perception of stimulus magnitude, G=1), phase, and detection threshold. (1) Perception of trunk rotation in space. During vestibular stimulation (whole-body rotation) and neck stimulation (trunk rotation with the head kept stationary) the frequency-transfer characteristics underlying this perception were very similar. The gain fell short; it was only about 0.7 at 0.4 and 0.2 Hz stimulus frequency and was further attenuated with decreasing frequency. In contrast, the phase was close to that of actual trunk position. The gain attenuation was found to be a function of the peak angular velocity of the stimulus, a fact, which we related to a velocity threshold of the order of 1 deg/s. During the various vestibular-neck combinations used, Ss' perception was again erroneous, reflecting essentially the sum of its two non-ideal constituents. However, there was one noticeable exception; during the combination head rotation on stationary trunk, Ss veridically perceived their trunk as stationary (compatible with the notion that the sum yielded zero). (2) Perception of head rotation in space. During vestibular stimulation, Ss' estimates showed the same non-ideal gain-vs.-frequency characteristics as described above for the trunk. Neck stimulation induced an illusion as if the head had been rotated in space. This neck contribution was such that, when it was combined with its vestibular counterpart during head rotation on stationary trunk, the perception became almost veridical. On closer inspection, however, this neck contribution was found to reflect the sum of two components; one was the non-ideal neck signal contributing to the perception of trunk in space, the other was an almost ideal neck signal of head-on-trunk rotation. (3) The results could be described by a simple model. In this model, the erroneous vestibular signal head in space is primarily used to create an internal representation of trunk in space. To this end, it is combined with the closely matching neck signal of trunk to head. The perception of head rotation in space is achieved by summing this trunk in space signal with the almost ideal head on trunk signal, again of nuchal origin. These seeming complex interactions have two implications: (i) the head is referred to trunk coordinates, whereas the trunk is referred to space coordinates; (ii) there is at least one condition in the dark where orientation is correct (despite an erroneous vestibular signal), i.e., during head rotation on stationary trunk.Supported by Deutsche Forschungsgemeinschaft, SFB 325  相似文献

Summary Event-related potentials were recorded when a subject evaluated the outcome of a simple TV game as successful/unsuccessful, where the goal was specified randomly as one of two areas on the screen. The experimental metastasis patterns of low metastatic Lewis lung tumor (LLT) and liver selected high metastatic LLT-HH were studied following their arterial dissemination. In previous reports it was shown that both tumor lines develop metastases only in the first encountered organ. Here the liver preference of the liver selected cell line is demonstrated. The model of two LLT cell lines can provide experimental evidence for both the mechanical and seed and soil theories of metastasis formation, depending on the site of tumor cell injection.  相似文献

Summary The experiments reported in this paper were carried out to determine the effect moving background patterns have on the response characteristics of directionally specific neurons in the pigeon optic tectum. First, care was taken to select the optimal single stimulus for each cell, then large textured patterns were added to the test stimulus and moved either in-phase or anti-phase. Altogether 214 cells were studied in 77 white Carneaux pigeons and it was found that all cells below a depth of 400 microns were inhibited by backgrounds moved in-phase with the optimal test stimulus, while few cells above this level were affected in any way by backgrounds. All directions of background motion containing an in-phase vector resulted in rather profound inhibition of the directional response while directions with an anti-phase vector produced less inhibition and sometimes were even facilitated by direct anti-phase. The velocity tuning curves obtained with an optimal single test stimulus and by anti-phase movement of backgrounds were essentially similar.In-phase inhibition can also be produced by a second spot stimulus located some distance from the test stimulus. This latter effect was used to map the outer margins of the inhibitory receptive fields of deep tectal neurons displaying these effects and it was found they were extremely large, often in excess of 100 ° in diameter. When masks were used to prevent the moving background from stimulating the excitatory receptive field, anti-phase movement always produced facilitation. This suggests a double opponent-process directionally specific receptive field organization.These neurons seem well suited to respond to local (object) motion and to ignore translation of the visual image arising from body, head and eye movements.Supported by a grant No. A0353 from the Natural Sciences and Engineering Research Council of Canada to B.J. Summary The sizes of adrenergic vesicles and their cores, as made visible by an acrylic aldehyde in sodium dichromate fixative, have been measured in electron micrographs of the sympathetic nerves amongst the frog's ventricular muscle. The animals were either normal or previously treated with drugs expected to affect the catecholamine content of the heart. The sympathetic nerves contain two overlapping populations of vesicles. A graphical method was used to separate these and determine the mean diameter of each population. The distribution of vesicles between the large and small populations is variable in normal animals. No changes could be detected in the experimental animals. The mean size of the large vesicles is variable in normal animals. No changes could be detected in the experimental animals. Four injections of 5-hydroxydopamine caused a 5.8% increase in the diameter of the small vesicles. No other treatment produced significant changes in vesicle size. Four injections of 5-hydroxydopamine caused a 50% increase in the diameter of the cores of the small vesicles. Two injections of reserpine caused a 20% reduction in the diameter of the visible cores in the small vesicles, and 34% of the vesicles lost their cores entirely. One injection of 6-hydroxydopamine or ten injections of -methyl-tyrosine caused small reductions in small core diameter. It is postulated that core formation in adrenergic nerves under these conditions is not solely dependent on their catecholamine content, but on this and another factor which may be part of the storage complex.  相似文献

Summary Electron microscope images of rotary shadowed myosin heads and subfragment-1 (S1) have been computationally aligned and averaged using correlation methods. Average images show reproducible detail within the pear-shaped envelope of the head; the major features are invariant in S1 and in intact heads, in two mirror-related views of the head, and in the presence and absence of ATP. The averages support the view that the head contains two main structural domains separated by a cleft, and that the region of the neck close to the head-rod junction is flexible. They also reveal the inadequacy of the conventional method of correcting the measured dimensions of shadowed particles for the supposed thickness of the metal coat.  相似文献

Summary Cells of the PC-12 rat pheochromocytoma cell line respond to nerve growth factor (NGF) by sprouting neurites and biochemically differentiating into sympathetic ganglion-like cells. NGF-stimulated (differentiated) and unstimulated (undifferentiated) cells were studied by cytochemical techniques for the localization of the enzymes acid phosphatase (ACPase) and thiamine pyrophosphatase (TPPase), and by a morphometric analysis of the distribution of endocytosed wheat-germ agglutinin labelled with horseradish peroxidase (WGA-HRP). Both cytochemical stains showed the enzymes to be distributed in lysosomes and certain cisternae of the Golgi apparatus in both NGF stimulated and unstimulated cells. ACPase was not confined to GERL (Golgi-endoplasmic reticulum-lysosome) as in certain other cells. The morphometric studies demonstrated that the reaction product of the internalized WGA-HRP occupied 4.7% of the cytoplasmic area in unstimulated cells and 4.5% in NGF-stimulated ones. Despite this similarity, the distribution of the WGA-HRP among the studied intracellular compartments in these two cell groups varied. In the NGF-stimulated cells 3.3% of the WGA-HRP reaction product was found in the innermost Golgi cisterna(e) while in unstimulated cells only 0.3% was seen in this compartment. Similarly, 4.3% of the WGA-HRP stain was found in small vesicles at the trans aspect of the Golgi apparatus in stimulated cells, when only 0.3% of the stain occupied this compartment in undifferentiated cells. The morphometric analysis also revealed that when the PC-12 cells were stimulated with NGF, the Golgi apparatus increased in area by approximately 70%. These findings are consistent with the hypothesis that NGF induced differentiation of PC-12 cells is coupled with enhanced endocytosis of WGA and probably of its receptor to the innermost Golgi cisterna(e) and the closely associated vesicles.  相似文献
