AIM To certify the relationship between AFP mRNA and some pathological parameters of he-patocellular carcinoma (HCC).METHOD We detected the expression of AFP in mRNA level in tissue samples from 52 patients suffering from HCC by RT-PCR method.RESULTS The positive rate of AFP mRNA was 76.9% in the HCC tumor tissues, and 69.4% in the paratumor tissues from the HCC patients with severe cirrhosis. However, in HCC patients without cirrhosis, the positive rate reached 50% in tumor tissues, but no AFP mRNA expression was found in the related paratumor tissues.CONCLUSION The AFP protein was specially expressed by HCC cells and mutated hepatocytes. The AFP mRNA was positively related with cirrhosis, but no significant relationship was found between AFP mRNA and tumor size, capsule status and tumor metastasis.  相似文献

Summary. Blood alpha-fetoprotein messenger RNA (AFP mRNA) is thought to be a marker of hepatocellular carcinoma (HCC). Its value as a predictor of HCC in patients at risk is not known. A series of 201 patients with compensated cirrhosis (114 men, mean age 58 years) underwent surveillance with semi-annual ultrasound and serum alpha-fetoprotein measurements. Total RNA was extracted from peripheral blood mononuclear cells collected at different intervals and AFP mRNA was retrotranscribed and amplified by nested polymerase chain reaction. Ten patients with HCC and 30 blood donors were used as controls. Three patients with HCC, 39 with cirrhosis under surveillance and four blood donors circulated AFP mRNA (30, 20 and 13%, NS). During 50 months of surveillance, 27 patients with cirrhosis developed HCC: the tumour was detected more often in patients with higher than normal baseline serum AFP (≥7 IU/L) than in those with normal AFP levels (21% vs 9%, P  = 0.02). The incidence of HCC was the same in patients with and without AFP mRNA at baseline (15% vs 14%). In 53 patients, AFP mRNA was re-tested after 6–25 months of surveillance. HCC developed in two of 11 (18%) who were initially AFP mRNA positive and later became negative, in none of those who were initially negative and later became positive and in two of 39 (5%) who remained persistently negative. In conclusion, blood AFP mRNA is not a sensitive predictor of HCC in patients with compensated cirrhosis.  相似文献

[摘要]目的探讨肝癌细胞中Twist基因表达与WNT信号传导通路激活的关系。方法将0.05mol/L氯化锂加入人肝癌SMMC7721细胞中培养2h激活VeNT通路,然后分别培养6、12、24h,分别为A1、A2、A3组;将0.1mol/L氯化锂加入人肝癌SMMC7721细胞中培养2h激活WNT通路,然后分别培养6、12、24h,分别为B1、B2、B3组;另设空白对照组。用RT—PCR及荧光定量PCR法检测各组细胞的TwistmRNA。结果lit.PCR法测得TwistmRNA在A1、A2、A3组分别为0.38±0.05、0.60±0.08、0.69±0.06,B1、B2、B3组分别为0.56±O.05、0.59±0.06、0.69±0.04,以上各组细胞TwistmRNA与对照组的0.18±0.03相比,P均〈0.05。荧光定量法测得TwistmRNA在A1、A2、A3组分别为0.43±0.01、0.56±0.01、0.67±0.01,BI、B2、B3组分别为0.52±0.01、0.58±0.00、0.65±0.00,以上各组细胞TwistmRNA与对照组的0.21±0.00相比,P均〈0.05。结论肝癌细胞中Twist基因的表达程度与WNT通路的激活有关。  相似文献

Transplant surgeons have long dreamed of achieving a complete cure for hepatocellular carcinoma (HCC) by replacing the liver with a new graft. Although the early results of liver transplantation for HCC were disappointing, with 5-year survival less than 40%, improved results in patients who met the so-called Milan criteria rekindled the enthusiasm for the treatment of HCC with liver transplantation. Furthermore, the recent development of living-donor liver transplantation in adults has allowed timely grafting for HCC patients and tentative expansion of the criteria for transplant candidacy in patients with HCC — although such expansion is fraught with controversy. Identification of a noninvasive marker that could predict the biological behavior as well as the prognosis of HCC would indeed be a major breakthrough.  相似文献

目的 为探讨人外周血AFPm RNA 与肝癌转移之间的关系。方法 采用分子生物学反转录—巢式PCR 技术,检测了38 例不同肝病患者和9 例非肝病患者外周血中的AFPm RNA。结果 其检出结果分别为:对照组0/9 ;肝硬化组2/11 ;肝癌未发生转移组2/7 ;局部转移组6/12 ;远处转移组7/8 。其中良性组与恶性组之间、肝癌无远处转移与远处转移组之间有显著性差异( P< 0 .01 及 P< 0 .05) 。另外,肝癌患者中,直径> 5 cm 组外周血中AFPm RNA 阳性率明显高于直径< 5cm 组( P< 0 .05) 。肝癌患者血清AFP> 400ng/ml 组外周血中AFPm RNA 阳性率具有明显高于AFP< 400ng/ml组(P< 0 .01) 。肝功能分级与外周血中AFPm RNA 阳性率无关。结论 人外周血中AFPm RNA 的检测具有重要的临床指导意义,可用于预测肝癌远处转移,并为临床治疗提供实验依据  相似文献

通过定量检测AFPmRNA在原发性肝癌(PHC)患者外周血中的表达,探讨AFPmRNA作为PHC患者早期诊断和微转移指标的可能性。建立荧光定量聚合酶链式反应(FQ-PCR),检测58例外周血AFPmRNA的表达量。其中,30例PHC患者中19例AFP≥20ng/m l,23例外周血中AFPmRNA有表达;肝癌患者外周血中AFPmRNA的基因拷贝数与肝癌患者有肝内外转移有明显相关,而肝炎及健康对照组外周血中AFPmRNA均未表达,肝硬化组有一例表达。应用荧光定量PCR检测PHC外周血中AFPmRNA表达情况,可以作为早期反映PHC患者外周血中是否存在肝癌细胞和发生微转移的早期灵敏指标。  相似文献

AIM:To study the relationship between N-ras gene mutation and p53 gene expression in the carcinogenesis and the development of human hepatocellular carcinomas (HCC).METHODS:The N-ras gene mutation and the p53 gene expression were analyzed in 29 cases of HCC by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP) and immunohistochemistry.RESULTS:Thirteen cases of HCCs were p53 positive (44.8%), which showed a rather high Cpercen-tage of p53 gene mutation in Guangxi. The aberrations at N-ras codon 2-37 were found in 79.31% of HCCs and 80.77% of adjacent non-tumorous liver tissues. More than 2 point mutations of N-ras gene were observed in 22 cases (75.86%). Twelve cases (41.37%) of HCCs showed both N-ras gene mutation and p53 gene expression.CONCLUSION:N-ras gene and p53 gene may be involved in the carcinogenesis and the development of HCC.That 38% of HCCs with N-ras gene mutation did not express p53 protein indicates that some other genes or factors may participate in the carcinogenesis and the development of HCC.  相似文献

目的寻找一种敏感的方法以检外外周血中的肝癌细胞.方法分离原发性肝癌患者以及肝硬化、急慢性肝炎、肝转移癌、肝脏良性肿瘤患者和健康志愿者的外周血单个核细胞,提取单个核细胞的总RAN,进行RT-PCR/Southern杂交检测AFPmRNA.结果原发性肝癌患者外周血AFP mRAN的检出率为41.4%,其检出率与肿瘤大小以及TNM分期有关,对照组均为阴性.结论肝癌患者外周血中的AFPmRNA可以作为肝癌细胞的血液微小转移的标志物.  相似文献

Background

In addition to the biochemical components secreted in bile, aquaporin (AQP) water channels exist in hepatocyte membranes to form conduits for water movement between the sinusoid and the bile canaliculus. The aim of the current study was to analyse AQP 9 expression and localization in human hepatocellular carcinoma (HCC) and non-tumourigenic liver (NTL) tissue from patients undergoing hepatic resection.

Methods

Archived tissue from 17 patients was sectioned and analysis performed using an antibody raised against AQP 9. Slides were blind-scored to determine AQP 9 distribution within HCC and NTL tissue.

Results

Aquaporin 9 was predominantly expressed in the membranes of hepatocytes and demonstrated zonal distribution relative to hepatic sinusoid structure in normal liver. In HCC arising in the absence of cirrhosis AQP 9 remained membrane-localized with zonal distribution in the majority of NTL. By contrast, AQP 9 expression was significantly decreased in the HCC mass vs. pair-matched NTL. In HCC in the presence of cirrhosis, NTL was characterized by extensive AQP 9 staining in the membrane in the absence of zonal distribution and AQP 9 staining in NTL was significantly greater than that observed in the tumour mass.

Conclusions

These data demonstrate that human HCC is characterized by altered AQP 9 expression and AQP 9 localization in the NTL mass is dependent on underlying liver pathology. Given the central role of AQPs in normal liver function and the potential role of AQPs during transformation and progression, these data may prove valuable in future diagnostic and/or therapeutic strategies.  相似文献

研究表皮生长因子 (EGF)在原发性肝癌患者中的含量变化及与甲胎蛋白的相关关系 ,以探讨表皮生长因子在原发性肝癌发生、发展过程中的作用和意义。采用放射免疫方法测定 30例原发性肝癌患者和 4 8例健康体检人群的血清表皮生长因子和甲胎蛋白 (AFP)。结果发现原发性肝癌组患者EGF含量明显低于健康对照组且有显著性差异 ;经相关分析血清EGF与AFP含量间无相关性关系 (r =0 16 4 3,P >0 0 5 )。表明EGF含量变化在肝癌早期诊断方面有一定的临床价值 ,同时与AFP无相关关系 ,也说明了两者在肝癌发生机制中的不同作用  相似文献
