血管内皮生长因子(VEGF)是内皮细胞特异性的促有丝分裂原,属分泌性的糖蛋白。有促进内皮细胞增生,增强血管通透性;加速新血管形成的作用。VEGF通过与其特异性受体结合发挥生物学作用。VEGF在生理和病理状态下新生血管形成中起重要作用。VEGF及其受体在缺血和/或缺 氧的 心肌细胞中表达升高。VEGF蛋白应用于心肌缺血可以通过促进缺血区新生血管形成,提高侧枝血流量而改善心功能。VEGF基因治疗心肌缺血具有广阔应用前景。  相似文献

In the last few years, there have been rapid advances in the application of angiogenic growth factor therapy to clinical settings. In particular, vascular endothelial growth factor and fibroblast growth factor were evaluated in a number of different trials that explored various forms of delivery (systemic vs local, gene vs protein) in various patient populations. Although all Phase I trials reported unqualified successes, the results of Phase II blinded trials were decidedly mixed, calling into question the entire concept of therapeutic angiogenesis. However, with continuing advances in the basic knowledge of new vessel growth in mature tissues and a better understanding of the various issues involved in patient selection, evaluation, and drug delivery, there is hope that growth factor therapy will take its place among other therapeutic modalities available to the clinician.  相似文献

The prevalence of diabetes is growing at epidemic rates in the USA. Diabetic retinopathy develops in a large proportion of patients and is a leading cause of blindness worldwide. Systemic management of diabetic retinopathy has included glycemic, hypertension, and lipid control. Local ophthalmic treatment in the form of focal/grid or panretinal laser photocoagulation has been shown to prevent vision loss in diabetic edema and proliferative diabetic retinopathy, respectively. The introduction of anti-vascular endothelial growth factor for diabetic macular edema and retinopathy has provided clinicians with improved clinical outcomes with potentially less damaging effects than laser.  相似文献

目的探讨血清血管内皮生长因子(VEGF)在POEMS综合征中的临床意义。方法采用ELISA方法检测POEMS综合征组(36例)、疾病对照组(多发性骨髓瘤29例、格林巴利综合征7例共36例)、正常对照组(36例)的血清VEGF浓度,进行组间比较,并与临床资料进行相关性分析。结果 POEMS综合征组血清VEGF浓度为(982.06±611.49)pg/ml,显著高于疾病对照组[(364.22±264.37)pg/ml]和正常对照组[(217.37±87.74 pg/ml)],P<0.001。VEGF诊断POEMS综合征曲线下面积为0.902,VEGF Cutoff值为360.62 pg/ml,敏感性为0.889,特异性为0.764。POEMS综合征组血清VEGF与肺动脉高压、外周血小板计数相关,与其他临床表现无显著相关性。结论血清VEGF浓度升高是POEMS综合征的一个重要特征,有助于POEMS综合征的诊断;血清VEGF浓度与肺动脉高压有一定相关性,可能是肺动脉高压发病机制之一。  相似文献

目的研究血管内皮生长因子对过氧化氢诱导的血管内皮细胞凋亡的影响,同时检测凋亡相关基因Bcl-2 mRNA与Fas mRNA表达的变化。方法体外培养人脐静脉内皮细胞,随机分成对照组、过氧化氢组和过氧化氢 血管内皮生长因子组,12 h后,采用原位末端标记法和流式细胞仪分别观察各组细胞的凋亡数和凋亡率,通过逆转录聚合酶链反应观察各组细胞中凋亡相关基因Bcl-2 mRNA与Fas mRNA表达的变化。结果过氧化氢组凋亡细胞数(27.83±2.14)明显高于对照组(2.50±1.05)(P<0.01)和过氧化氢 血管内皮生长因子组(13.00±2.10)(P<0.01)。过氧化氢组细胞凋亡率(14.17%±0.45%)明显高于对照组(1.55%±0.87%)(P<0.01)和过氧化氢 血管内皮生长因子组(5.69%±0.38%)(P<0.01)。与过氧化氢组比较,过氧化氢 血管内皮生长因子组Fas mRNA表达明显减少(40.67%±2.16%比94.50%±3.45%,P<0.01),而Bcl-2 mRNA表达明显增加(60.33%±1.75%比23.17%±1.17%,P<0.01)。结论血管内皮生长因子能拮抗过氧化氢诱导的内皮细胞凋亡,其抗凋亡的机制可能与上调Bcl-2 mRNA表达与下调Fas mRNA表达有关。  相似文献

血管内皮生长因子(VEGF)是作用最强的血管生成因子之一。研究证明,在脑缺血动物脑内注入外源性VEGF能促进血管生成,但同时在缺血半暗带内会发生“盗血”现象。另一方面,VEGF对中枢神经系统具有神经发生、直接神经营养和神经保护等多重保护作用。从分子水平了解这些神经保护机制,更有益于临床开发治疗脑缺血的措施。  相似文献

Objective : The expression of vascular endothelial growth factor (VEGF), a glycoprotein that selectively promotes proliferation of endothelial cells, has been associated with cancer development. The aim of the present study was to determine whether serum levels of VEGF correlate with disease progression in patients with colorectal cancer. Methods : VEGF levels were measured by a highly sensitive enzyme-linked immunosorbent assay in sera from 67 patients with colorectal cancer, 14 patients with colorectal adenomas, and 72 healthy volunteers, and in tissue homogenates from 10 patients with colorectal cancer. Results : Serum VEGF levels were significantly higher in patients with colorectal cancer than in patients with colorectal adenomas or in normal controls (   p < 0.01  ). In patients with colorectal cancer, serum VEGF levels were significantly associated with Dukes stage (   p < 0.01  ) and with carcinoembryonic antigen levels (   r = 0.725  ,   p < 0.001  ). Patients with hepatic and/or lymph node metastasis had higher serum VEGF levels than those without. Surgical resection of the colorectal tumor led to a decrease in serum VEGF levels whether or not metastasis was present (   p < 0.05  ). The tumor-bearing tissue contained significantly more VEGF than normal-appearing mucosa (   p < 0.05  ). Conclusions : VEGF is involved in the development of colorectal cancer. Measurement of VEGF in the serum may be a useful noninvasive clinical marker for evaluating the disease status.  相似文献

Background: Vascular endothelial growth factor (VEGF) has a key role in angiogene-sis and in transplantation. The level of VEGF is related to the differences in the DNA sequence of its promoter region. Objectives: In this study, the association between the combination of VEGF –1154 G and –2578 C alleles and VEGF production by LPS-stimulated PBMCs was investigated. In addition; the relationship between VEGF poly-morphisms and the influence of TNF-α and IL-4 on VEGF production was studied. Methods: VEGF –1154 G/A and –2578 C/A were detected using ARMS-PCR. To de-termine the impact of combinations of these two polymorphisms on VEGF production; PBMCs were stimulated by LPS and VEGF production was measured by ELISA. Re-sults: The combinations of –1154 GG/-2578 CC and –1154 GG/-2578 CA were signifi-cantly associated with higher VEGF production (p<0.0001). Production of VEGF was significantly influenced by TNF-α in individuals who had certain VEGF genotype com-binations. Although VEGF production was dramatically suppressed by IL-4, it was not dependent on VEGF genotype. Conclusions: Since TNF-α has influence on the graft outcome, to avoid allocation of grafts from high TNF-α producer donors to recipients, it might be useful to predict and minimize graft rejection by having prior knowledge of TNF-α and also VEGF genotypes especially -1154 G/A and -2578 C/A VEGF.  相似文献

The clinical significance of circulatingvascular endothelial growth factor (VEGF) in patientswith various liver diseases was investigated. Twenty-onepatients with acute hepatitis (AH), 40 with chronic hepatitis (CH), 34 with cirrhosis (LC), 16 withfulminant hepatitis (FH), 10 with primary biliarycirrhosis (PBC), 12 with autoimmune hepatitis (AIH), and120 healthy individuals were included. Serum VEGF levels were measured by a chemiluminescenceenzyme-linked immunosorbent assay. The mean values ofserum VEGF levels in the patients with AH, CH, LC, FH,AIH, PBC, and control were 172.7, 58.0, 44.1, 37.3, 49.7, 74.9, and 65.0 pg/ml, respectively. Thepatients with AH had a level of serum VEGF significantlyhigher than that of the control group (P < 0.001).The serum VEGF levels in survivors of FH weresignificantly increased, but not in the nonsurvivors in therecovery phase compared with the levels on admission (P< 0.05). In the LC patients, serum VEGF levels weresignificantly lower than those of the control group (P < 0.05). These findings suggest thatserum VEGF level may be associated with hepatocyteregeneration grade.  相似文献

目的分析评价血管内皮生长因子在糖尿病咽部并发症中的价值作用。方法将该院2017年2月—2018年2月收治的100例糖尿病眼部病变患者分成两组,每组50例;对照组患者采取曲安奈德注射液治疗,观察组患者采取血管内皮生长因子药物中的康柏西普玻璃体腔注药治疗,比较两组临床治疗效果。结果在眼部并发症总发生率方面,观察组比对照组明显更低。在最佳矫正视力下降率、最佳矫正视力不变率方面,观察组明显低于对照组。在最佳视力矫正提高率方面,观察组明显高于对照组。结论针对糖尿病眼部病变患者,采取血管内皮生长因子治疗具备显著的疗效,可预防眼部并发症的发生;值得采纳及应用。  相似文献
