白细胞介素是由多种细胞产生并作用于多种细胞的一类细胞因子,最初是指由白细胞产生又在白细胞间起调节作用的细胞因子,故称之为“白介素”。现在人们对于白介素的分子结构和生物学功能已经基本明确,并将这些具有重要调节作用的细胞因子进行了系统分类和统一命名。目前已经得到承认的“白介素家族”成员有29个,分别命名为IL-1~29,功能复杂,互成网络,复杂重叠。  相似文献

白细胞介素-12(IL-12)家族是一类由异源二聚体分子组成的细胞因子,其成员有IL-12、IL-23、IL-27和IL-35。IL-12家族是桥连(bridge)天然免疫和适应性免疫反应的一类重要细胞因子,特别在经典的TH1、TH17和Treg细胞参与的适应性免疫过程中作用尤为突出。IL-12家族各成员通过共享部分亚基及受体,借助JAK-STAT信号通路共同参与适应性免疫反应,其调控网络极其复杂。在抵御微生物感染过程中,IL-12和IL-23主要表现出促炎作用,而IL-27和IL-35常表现出抑炎作用。本文就IL-12家族各成员在感染性疾病相关研究中的进展进行综述,为进一步揭示IL-12家族各成员在感染性疾病发生、发展中的作用提供参考。  相似文献

IL-23是最新发现的一种由P19及P40两条多肽链组成的异二聚体细胞因子,来源于活化的单核巨噬细胞、B细胞和树突状细胞(dendriticcell,DC),IL-23与IL-27同为IL-12家族成员。IL-23可促进CD4 T细胞的增殖并强化树突细胞抗原递呈能力,通过诱导T细胞和DC产生IFN-γ与IL-12,增强宿主针对肿瘤的免疫反应作用。  相似文献

白细胞介素22(IL-22)是2000年由Dumoutier等发现的-种新型细胞因子,与IL-10有23%的同源性,曾被命名为IL-TIF(Interleukin-10-related T cell—derived inducible factor)。鉴于与IL-10的结构相似,其受体是-个Ⅱ型细胞因子受体家族的新成员,以及其在白细胞内的产物和作用的确认,由Xie等将它重命名为人类细胞因子白细胞介素-22(Interleukin-22,IL-22)。目前对它的研究尚处于起始阶段。  相似文献

白细胞介素-23(IL-23)是造血细胞因子家族成员,它是由p19和p40两个亚基组成的活性复合体,IL-23受体由IL-12β1和IL-23R构成,IL-23的信号途径与白细胞介素-12(IL-12)有相同的Jak-Stat信号分子,IL-23的功能与IL-12也存在许多相同之处,但也有其独特的功能。IL-23可作用于活化的T细胞、记忆性T细胞和树突状细胞,产生γ干扰素(IFN-γ)和IL-12等细胞因子,在炎症性疾病、自身免疫性疾病的发病以及抗肿瘤和抗感染等方面发挥重要作用,有望成为新的免疫治疗因子。  相似文献

机体对感染的合理免疫反应应是保持一种平衡.既保证清除致病菌的同时,又使正常组织不被失控的炎症反应损伤,最近德国科研人员的一篇报道证实,白细胞介素-27(IL-27)在调节这一平衡中具有关键作用。采用盲肠结扎穿孔术(CLP)引起小鼠腹膜炎时,体内IL-27迅速增多。但IL-27EB13亚基缺陷的小鼠对CLP诱发腹膜炎的抵抗力明显强于野生型小鼠,而注射IL-27EB13亚基后这一优势受到抑制.EB13缺陷小鼠在CLP时白细胞聚集和“呼吸爆发”均明显增强,  相似文献

BACKGROUND: Statins reduce lipid levels, inflammation and cardiovascular events in patients with coronary artery disease; CKD patients show increased risk of cardiovascular and increased plasma levels of IL-6 and IL-8. AIM: To evaluate the in vitro effect of simvastatin (S) or fluvastatin (F) on the lipopolysaccharide (LPS) stimulated secretion of IL-6 and IL-8 from monocytes of chronic kidney disease patients (CKD) in K-DOQI stages 3-5. METHODS AND SUBJECTS: Monocytes enriched peripheral blood (PBMC) from 28 CKD (15 in K-DOQI stages 3-4, Group I, and 13 in K-DOQI stage 5 on hemodialysis, Group II) and 10 healthy subjects (HS), were isolated by Ficoll-gradient centrifugation. Cells were incubated with LPS 100 ng/ml or with LPS plus increasing doses of statins (from 10(-6) to 10(-8) M ) for 24 h. Surnatant IL-6 and IL-8 concentrations were determined by EIA. RESULTS: Basally the mean concentration of IL-6 and IL-8 was higher in patients than in HS and in Group II than in Group I (IL6: HS 285 +/- 77 pg/ml, Group I 365 +/- 178 pg/ml, Group II 520 +/- 139 pg/ml- IL8 HS 180 +/- 75 pg/ml, Group I 1722 +/- 582 pg/ml, Group II 4400 +/- 1935 pg/ml). After addition of LPS the mean concentration of IL-6 and IL-8 increased in all groups (IL6: HS 1740 +/- 178 pg/ml, Group I 3754 +/- 672 pg/ml, Group II 4800 +/- 967 pg/ml; IL8: HS 450+/-132 pg/ml, Group I 9700+/-2837 pg/ml, Group II 11608 +/- 2316 pg/ml). After the addition of LPS plus increasing doses of S or F from 10(-10) to 10(-6) M, a significantly lower cytokine concentration compared to the data after LPS alone was observed (IL6: HS 45%, Group I 75%, Group II 50%; IL8: HS 100%, Group I 65%, Group II 35%). CONCLUSIONS: These data confirm that cytokine release is increased in CKD patients and that is highest in the most severe patients. Furthermore they suggest that fluvastatin or simvastatin can be used in order to reduce the high cardiovascular risk.  相似文献

目的 探讨细胞因子TNF、IL-6与病毒性肝炎发生的相关性.方法 应用双抗体夹心ELISA方法检测患者血清中TNF、IL-6,并用全自动生化分析仪检测肝功能.结果 肝炎患者血清中 TNF、IL-6均明显高于对照组,P<0.01;而且TNF含量与ALT呈正相关,IL-6与球蛋白水平呈正相关.结论TNF、IL-6与病毒性肝炎发生的病理过程有关,可能参与肝细胞的损伤过程.  相似文献

目的探讨血清IL-12p70含量在强直性脊柱炎(AS)和健康个体间的差异及其临床意义。方法应用EHSA方法检测60例AS患者和30例健康对照血清IL-12p70水平;应用ESR自动化分析仪和特定蛋白分析仪分别测定红细胞沉降率(ESR)和血浆C反应蛋白(CRP);分析HLA-B27阳性和阴性的AS患者血清IL-12p70水平及与ESR、CRP的相关性。结果AS患者血清IL-12p70水平明显高于对照组(P〈0.01),且HLA-B27阳性和阴性AS患者均高于对照组(P〈0.01),两者比较,差异无显著性(P〉0.05)。在HLA-B27阳性AS患者中,血清IL-12p70水平与ESR及CRP均呈明显的相关性,但在HLA-B27阴性AS患者中则无显著相关性。结论AS患者血清IL-12p70水平明显上调,且这种上调与HLA-B27阳性与否无明显相关性。但AS患者血清IL-12p70水平与ESR及CRP是否呈显著相关竹却明显受到HLA-B27阳性与否的影响,进一步提示血清IL-12p70水平的改变可能与AS的发生和发展密切相关。  相似文献

目的:探讨白细胞介素6(IL-6)和白细胞介素18(IL-18)血清水平与类风湿性关节炎(RA)中医辨证分型的关系。方法:采用酶联免疫吸附试验(ELISA)双抗体夹心法检测RA患者和健康人的外周血IL-6和IL-18的血清水平,分析不同证型两指标之间及其与C反应蛋白、类风湿因子含量的相关性。结果:RA患者IL-6和IL-18血清水平与健康人比较差异具有显著性,活动期RA患者IL-6水平显著高于稳定期患者(P<0.05)。风寒湿阻型患者血清IL-6浓度明显高于痰瘀互结型与肝肾两虚型(P<0.05),而与风湿热郁型无明显差异(P>0.05);而风寒湿阻型患者血清IL-18浓度明显高于风湿热郁型与肝肾两虚型(P<0.05),而与痰瘀互结型无明显差异(P>0.05)。结论:IL-6血清水平与RA的活动性密切相关,且与其中医辨证分型存在一定的联系。  相似文献

The production and action of primary proinflammatory cytokines are strictly controlled by a series of circuits to avoid damage that they can cause if produced in excess. Interleukin-10 and interleukin-1 receptor antagonist contribute to the control of the magnitude of the inflammatory responses in vivo. Benzydamine, a non-steroidal anti-inflammatory drug that has been shown to have suppressive activity for the proinflammatory cytokines, tumor necrosis factor-alpha and interleukin-1beta, was investigated for its effects on interleukin-10 and interleukin-1ra production. The drug did not modify the production of interleukin-10 and interleukin-1ra by peripheral blood mononuclear cells stimulated with lipopolysaccharide, under conditions where tumor necrosis factor-alpha and interleukin-1beta were decreased. The antiinflammatory capacity of benzydamine might thus result from its ability to reduce the production of proinflammatory cytokines, without affecting antiinflammatory factors.  相似文献

目的探讨白细胞介素-2(IL-2)联合白细胞介素-12(IL-12)对人脐血单个核细胞(CBMC)抗肿瘤作用的影响。方法采用噻唑蓝还原法和半固体培养方法研究IL-2联合IL-12激活的CBMC对白血病细胞株K562和HL60的抗肿瘤活性及其造血祖细胞活性。结果①10U/mlIL-12激活的CBMC对K562及HL60细胞杀伤率分别为(35.12±3.46)%和(45.60±4.32)%;500U/mlIL-2激活的CBMC对K562及HL60细胞的杀伤率分别为(37.23±4.82)%和(23.69±4.86)%;IL-2、IL-12联合可产生明显的协同作用,对K562及HL60细胞的杀伤率分别达(68.60±4.68)%和(67.69±4.84)%。②CBMC经IL-2和IL-12刺激72h,可产生更多的肿瘤坏死因子-α、白细胞介素-6、γ-干扰素。③IL-2、IL-12单独或联合应用激活CBMC产生抗肿瘤活性的同时对CBMC原有的造血祖细胞活性无明显影响。结论IL-2联合IL-12可提高CBMC的抗肿瘤活性,为临床应用CBMC治疗白血病提供了实验依据。  相似文献

Alcohol abuse is a major cause of liver cirrhosis as well as chronic liver disease. The aim of the present study was to investigate the possible correlation, between liver dysfunction biological markers and vitamin B12, with interleukin-6, in the serum of alcohol-dependent individuals without liver disease (AWLD). In a sample of 43 alcohol abusing/dependent subjects (33 males and 10 females) treated on an inpatient basis according to a standard detoxification protocol, the serum activities of the hepatic enzymes (ASAT, ALAT, gamma-GT), as well as the concentration of B12 and IL-6, were determined on admission. A strong positive correlation has been observed between IL-6 and B12, ASAT, ALAT, and gamma-GT at the beginning of the detoxification period. The results confirmed that in alcohol-dependent individuals, the median serum concentration of IL-6, before the beginning of the treatment, had a significant positive correlation with the liver dysfunction biological markers and B12. In conclusion, IL-6 might be used as an additional diagnostic marker for the degree of liver dysfunction in alcohol dependent individuals.  相似文献

The recent discoveries of the two interleukin (IL)-12-related cytokines, IL-23 and IL-27, reveal that the regulation of cellular immunity is more complex than originally thought. Until these discoveries, the role of IL-12 in modulating cellular immune responses had been overestimated due to the belief that the IL-12 p40 subunit was unique to IL-12. However, because p40 is shared between IL-12 and IL-23, it would be expected that p40-/- mice are doubly deficient in IL-12 and IL-23. Indeed, the essential role previously attributed to IL-12 in experimental autoimmune encephalitis during studies of p40-/- mice has been shown to be due to IL-23 instead. The newest addition to the IL-12 cytokine family, IL-27, has unique features as well. Its specific action on naive CD4+ T cells in both mice and humans appears to distinguish it from the other IL-12 family members. Although related, the IL-12 family of cytokines and their receptors have distinct patterns of expression and unique effects on developing immune responses. This review summarises much of the pertinent literature on the IL-12 cytokine family and provides predictions regarding their potential therapeutic roles based on what has been learned about their functions in vitro and in vivo in gene-deficient mice.  相似文献

目的探讨强直性脊柱炎(AS)患者HLA-B27和细胞因子等项目联合检测的临床价值。方法应用流式细胞术(FCM)对自愿献血者356例和疑似AS患者347例进行HLA-B27检测,筛选HLA-B27阳性对照组30例及确诊AS患者92例,对比ELISA法检测28例HLA-B27( )AS患者治疗前后IL-1、IL-6、IFN-γ和SIL-2R等项目的变化。结果自愿献血者HLA-B27抗原阳性率8.7%(31/356),AS患者HLA-B27阳性率92.4%(85/92),28例HLA-B27( )AS患者治疗前C反应蛋白(CRP)及血沉(ESR)显著升高,IL-1、IL-6、IFN-γ和SIL-2R检测值分别为263.1±32.5、285.4±32.2、98.5±16.2(pg/ml)和368.2±152.1(μg/ml),与对照组的差异有显著性(P<0.01),且患者治疗后IL-6水平仍高于正常。结论AS发病与HLA-B27的协同作用有关,IL-1、IL-6、IFN-γ诸因子组成网络共同参与AS的发病,CRP和ESR可作为判断患者处于活动期的指标。  相似文献
