Rabbit antisera were raised to the malaria vector Anopheles tessellatus, head-thorax, abdomen, and midgut preparations. Reactivity of the antisera with An. tessellatus and the filariasis vector, Culex quinquefasciatus, were examined by enzyme-linked immunosorbent assay (ELISA) and Western blots. Although many antigens were shared between the two species of mosquitoes, tissue- and species-specific antigens also were identified.  相似文献

Larvicidal activity of methanolic plant extracts of Lantana cramera (P1) root and Anacardium occidentale (P2) leaf was investigated against the larvae of the three mosquito species (Culex quinquefasciatus, Anopheles stephensi, and Aedes aegypti reared in the laboratory), and the respective glutathione S-transferase (GST) activity was analyzed as an index of protection against the extracts. The LC50 (extract concentration that shows 50% mortality) values of P1 extract for An. stephensi, Ae. aegypti, and Cx. quinquefasciatus were 132.55, 27.82, and 11.68 ppm, respectively, whereas those of P2 extract were 56.81, 912, and 10.79 ppm, respectively. In general, in the untreated groups, the level of GST activity was significantly higher in Ae. aegypti in comparison with An. stephesi and Cx. quinquefasciatus. However, the enzyme activity failed to show any response when treated with either of the plant extracts in Ae. aegypti. However, an increase in the GST activity was recorded in extract-treated larvae of both An. stephensi and Cx. quinquefasciatus. The results of the current study suggest that both the plant extracts show species-specific mosquitocidal potential. Induction of GST activities in survived An. stephensi and Cx. quinquefasciatus larvae suggests the role of this enzyme in conferring resistance to the plant extracts.  相似文献

Seasonal and habitat influences on the egg-laying activity of four species of Culex were compared in south Florida using jar- and vat-type oviposition traps. Egg rafts of Culex nigripalpus Theobald, Cx. quinquefasciatus Say, and Cx. salinarius Coquillett were collected in every month of the year, but rafts of Cx. restuans Theobald were absent during spring and summer. Cx. nigripalpus was the dominant species in the summer and fall, and Cx. quinquefasciatus and Cx. salinarius peaked during the winter and spring. Cx. restuans oviposited most rafts in the fall (98.4%). Most Cx. nigripalpus egg rafts were laid in vats (88.5%), and most Cx. quinquefasciatus egg rafts were deposited in jars (58.7%). Fewer Cx. quinquefasciatus rafts were found in a wooded area (32.4%) with a dense undergrowth than in a more open area (67.6%), but Cx. nigripalpus showed no ovipositional preference between these two habitats. The number of rafts oviposited by all species was reduced during periods of rainfall.  相似文献

The egg hatchability, insemination, and longevity of Japanese Culex pipiens pallens Coquillett and Japanese Culex quinquefasciatus Say were compared at 25 and 30 degrees C. Egg hatchability was high in Cx. p. pallens at 25 degrees C, but it was very low at 30 degrees C because almost no females were inseminated at this temperature. In Cx. quinquefasciatus, the egg hatchability and insemination rates were very high, even at 30 degrees C. The longevity of adult females and males was generally shorter in Cx. p. pallens than in Cx. quinquefasciatus at both temperatures. Because high temperatures may restrict the spread of Cx. p. pallens, we suggest that even if this species spreads to Okinawa, the possibility of it becoming established is very low.  相似文献

The efficiency of laboratory colonies of mosquitoes such as Anopheles stephensi Liston, Aedes aegypti (L.) Liverpool strain, Ae. aegypti wild type, Aedes albopictus (Skuse), Culex tritaeniorhynchus Giles, Culex sitiens Wiedemann, and Armigeres subalbatus Coquillett in supporting the development of Wuchereria bancrofti (Cobbold) (Spirurida: Onchocercidae) microfilariae to infective larvae was investigated. The mosquitoes were fed on heparinized microfilaremic human blood by using a membrane-feeding unit with Parafilm as membrane. The rate of infection, parasite development, and parasite burden were compared with that in the known vector mosquito Culex quinquefasciatus Say. Cx. quinquefasciatus showed the highest percentage of infection, followed by Ae. aegypti Liverpool strain and An. stephensi. The rate of development of the parasite was more or less similar in all the three species, and infective larvae were found on day 13. When the larvae were harvested on day 17, Cx. quinquefasciatus yielded the highest numbers, followed by Ae. aegypti Liverpool strain and An. stephensi. The percentage of infection was low, and the development was slow in Cx. tritaeniorhynchus compared with the other susceptible species. The parasite developed to second-stage larvae only by day 22 and to infective larvae by day 28. When 2-wk-old Cx. tritaeniorhynchus were fed on microfilaremic blood, they could develop the parasite to infective larvae by day 13 postfeeding. All other species of mosquitoes tested were found to be refractory to parasite development. It is shown that Cx. quinquefasciatus is the most suitable mosquito host for the production of infective larvae. However, Ae. aegypti Liverpool strain, which is commonly used for Brugia malayi filarial parasite, also can be used for generation of W. bancrofti infective larvae to circumvent the problem of maintaining two mosquito species.  相似文献

Culex quinquefasciatus and Mansonia annulifera are abundant in the rural areas of Kuttanadu, Kerala, India. Bloodmeal identification for individuals of these species collected in this region was determined by the agar-gel precipitin test. A total of 2,328 blood smears from Cx. quinquefasciatus (1,148) and Ma. annulifera (1,180) was tested. Results showed that Cx. quinquefasciatus and Ma. annulifera were highly anthropophilic and that human feeding accounted for 74 and 66% of the total bloodmeals tested. Feeding on cattle accounted for only 1.5 and 2.1% of Cx. quinquefasciatus and Ma. annulifera bloodmeals, respectively. This study showed the high anthropophilic feeding rates of Cx. quinquefasciatus and Ma. annulifera collected from an endemic belt of Malayan filariasis, where epidemiological studies revealed the coexistence of Bancroftian and Malayan filariasis.  相似文献

The succession of mosquito species and abiotic factors affecting their distribution and abundance in rice (Oryza spp.) fields was investigated over a 16-wk rice growing cycle covering the period between January and May 2006. Fifteen experimental rice plots were sampled for mosquito larvae and characterized based on rice height, number of tillers, floating vegetation cover, water depth, water temperature, turbidity, salinity, pH, dissolved oxygen, total dissolved solids, and conductivity. Microscopic identification of 3,025 larvae yielded nine mosquito species predominated by Anopheles arabiensis Patton (45.0%), Culex quinquefasciatus Say (35.8%), Anopheles pharoensis Theobald (9.0%) and Ficalbia splendens Theobald (7.1%). Other species, including Anopheles rufipes Gough, Anopheles coustani Laveran, Anonopheles maculipalpis Giles, Culex annulioris Theobald, and Culex poicilipes Theobald made up 3.1% of the total collection. Anopheles gambiae s.l., Cx. quinquefasciatus, and An. pharoensis occurred throughout the cycle, but they were more abundant up to 4 wk posttransplanting with peaks after fertilizer application. As rice plants became established, three groups of mosquitoes were recognized: the first groups included An. rufipes, Fl. splendens, and Cx. annulioris, which occurred throughout much of the second half of the rice cycle, whereas the second group included Cx. poicilipes, which was found in the middle of the rice cycle. An. coustani and An. maculipalpis formed the third group occurring toward the end of the cycle. Dissolved oxygen, number of tillers, and rice height were negatively associated with the abundance ofAn. arabiensis and Cx. quinquefasciatus larvae. In addition, Cx. quinquefasciatus also was associated with water depth (-ve) and turbidity (+ve). Abundance of An. pharoensis larvae was significantly associated with water temperature (+ve), the number of tillers (-ve), and rice height (-ve), whereas Fl. splendens was significantly associated with the number of tillers (+ve). The results demonstrate a complex nature of the interactions between some of the factors in the ecosystem and mosquito species abundance and calls for time-dependent and species-specific mosquito control operations.  相似文献

The effectiveness of traps baited with (5R,6S)-6-acetoxy-5-hexadecanolide (the synthetic oviposition pheromone) and grass infusions in sampling a population of gravid Culex quinquefasciatus Say was conducted in Muheza, Northeast Tanzania. A counterflow geometry (CFG) trap baited with pheromone and set outdoors, adjacent to a pit latrine building, collected more gravid Cx. quinquefasciatus than a CDC trap baited with pheromone and operated without light. Inside pit latrine buildings, significantly more gravid Cx. quinquefasciatus were collected in a CFG trap-baited with pheromone or grass infusion than in traps baited with tap water. CFG traps baited with either grass infusion or pheromone and set outdoors, away from known breeding sites, caught significantly more gravid Cx. quinquefasciatus than traps baited with tap water. CFG traps baited with pheromone + grass infusion caught significantly more gravid Cx. quinquefasciatus than CFG traps baited with either grass infusion or pheromone. In both cases, the proportion of gravid mosquitoes increased as traps were moved away from a natural emergence site. More gravid Cx. quinquefasciatus were collected in a pheromone-baited CFG trap than were egg rafts deposited in a jar with pheromone-treated water. It is concluded that CFG traps baited with oviposition attractants can be used effectively to sample gravid Cx. quinquefasciatus.  相似文献

We studied the toxicity and delayed effects of a mosquitocidal toxin (Mtx1) and a binary toxin (Bin) produced in Escherchia coli E-TH21 and Bacillus thuringiensis B-CW1, respectively, on Culex quinquefasciatus (Diptera: Culicidae). Bioassay results showed that both E-TH21 powder and B-CW1 sporulated culture were highly toxic against susceptible Cx. quinquefasciatus, with LC50 values of 0.65 and 1.70 mg/liter against third and fourth instars at 48 h, respectively. After initial 48-h exposure of larvae to different concentrations of Mtx1 and Bin, significant continued mortality could be observed in larval, pupal, and emergence stages of Cx. quinquefasciatus. Importantly, the Mtx1 could induce higher cumulative larval and preadult mortalities than Bin toxin on the target mosquito. This finding is important for understanding the mode of action of Mtx1 and Bin toxins and for developing a new bioassay procedure for evaluation of toxicity of Bacillus sphaericus Neide, some strains of which produce Mtx1 and Bin, in the laboratory and field.  相似文献

本文针对尖音库蚊复合组,选择了4个分类性状,包括雄蚊阳茎DV/D的比值、四龄幼虫呼吸管指数、四龄幼虫呼吸管1-S第1对毛的分枝数和第2对毛的分枝数,应用最小距离法对我国22个地理株及尖音库蚊、淡色库蚊和致倦库蚊的实验室杂交后代分别进行聚类分析,发现尖音库蚊乌鲁木齐株和骚扰库蚊北京株与其它株距离最远,在剩余的20株蚊虫中,除思茅株外,聚为三类,分别代表分布在我国的淡色库蚊、致倦库蚊、以及淡色库蚊和致倦库蚊的中间型。在杂交后代的聚类中发现,尖音库蚊和致倦库蚊的杂交后代与淡色库蚊没有聚在一类。  相似文献

本文针对我国重要的媒介蚊虫淡色库蚊和致倦库蚊,进行了3个地理株的实验室内杂交研究。结果发现淡色库蚊北京株与淡色库蚊南京株杂交,以及淡色库蚊南京株与致倦库蚊广州株杂交,都可以产生具有能育性的杂种子一代并且也得到了杂种子二代,但是杂种子二代成成蚊率很低,介于0%—5.54%之间,提示这可能是尖音库蚊复合组蚊虫的分化在遗传学上的反映  相似文献

The spatial and temporal distribution of 28 species of female mosquitoes resting in natural (swamp, woods, and swamp-woods ecotone) and human-made (beneath bridges) habitats in blackwater stream-associated ecosystems in Duplin County, N.C., was determined by sampling with a D-Vac vacuum aspirator during 1984 and 1985. Two types of resting distributions were evident. In type one, species including Aedes atlanticus Dyar & Knab, Ae. canadensis (Theobald), Ae. triseriatus (Say), and Psorophora ferox (von Humboldt) rested predominantly on vegetation and were not collected beneath bridges. In type two, species including Anopheles punctipennis (Say), Culex erraticus (Dyar & Knab), Cx. peccator Dyar & Knab, Cx. pipiens L. and Cx. quinquefasciatus Say and their hybrids, Cx. restuans Theobald, Cx. territans Walker, and Uranotaenia sapphirina (Osten Sacken) rested on vegetation, in natural shelters in swamp habitats, and beneath bridges. Differences in the proportion of gravid mosquitoes among habitats were evident for Ae. canadensis, An. punctipennis, Cx. pipiens and Cx. quinquefasciatus and their hybrids, Cx. restuans, Cx. territans, Ps. ferox, and Ur. sapphirina, indicating that gonotrophic condition may influence resting site selection.  相似文献

Aedes aegypti (L.), Culex sitiens Weidemann, Culex annulirostris Skuse, and Culex quinquefasciatus Say mosquitoes colonized at the Queensland Institute of Medical Research, Brisbane Australia, were fed on blood containing Barmah Forest virus (BF). Only Cx. annulirostris was susceptible to infection, with a median cell culture infectious dose (CCID50) of 10(3.36) per mosquito. Ae. aegypti and Cx. quinquefasciatus were infected experimentally, but at rates of < 9%. Cx. sitiens did not become infected. Infection rates for Cx. annulirostris fed 10(3.5) CCID50 of virus per mosquito, varied from 9 to 50% between 2 and 13 d after infection. Virus transmission to suckling mice by Cx. annulirostris occurred from 2 d after infection. Transmission of BF virus by Cx. annulirostris was 10% at 2 d after infection and did not exceed 8% thereafter. Although Cx. annulirostris may be infected and is able to transmit BF virus to suckling mice, it is nonetheless a relatively inefficient vector of the virus.  相似文献

Mosquito sugar-feeding and host-seeking rhythms were recorded in the laboratory using remote sensors. In all five species studied, the two rhythms were nearly synchronous. In Anopheles quadrimaculatus Say, Culex quinquefasciatus Say, and Aedes triseriatus (Say), host-seeking activity increased slightly earlier than sugar feeding in the evening. The evening sugar-feeding period was longer than the evening host-seeking period in Aedes aegypti (L.), but the two periods were of similar duration in Aedes albopictus (Skuse). In the morning, the only clear difference between the two behaviors occurred in Cx. quinquefasciatus, which had a host-seeking peak but lacked a sugar-feeding peak. When exposed concurrently to both host and sugar stimuli, sugar feeding practically ceased in Ae. albopictus and Ae. aegypti. In the presence of host stimuli, the sugar-feeding rhythms of An. quadrimaculatus, Cx. quinquefasciatus, and Ae. triseriatus differed from those obtained in the absence of host stimuli by having an earlier onset of evening activity, a relatively smaller evening activity peak, and a more irregular activity pattern during early scotophase, respectively. We concluded that inseminated, nulliparous females have nonspecific appetitive feeding periods, and that the order in which sugar and blood are sought largely may be a function of factors other than differences in diel rhythms.  相似文献

Groups of wild-caught Culex quinquefaciatus Say, previously tested for the presence of hepatitis B surface antigen (HBsAg), were tested for the presence of hepatitis B e antigen (HBeAg). This antigen was detected at low levels in blood-fed, half-gravid, and gravid groups. A colony of Cx. quinquefasciatus was established in the laboratory and tested for the persistence of HBsAg and HBeAg. Five days after feeding on blood infected with HBsAg and HBeAg, 9 of 20 (45%) pools of Cx. quinquefasciatus were HBeAg-positive and 5 of 20 (25%) pools were HBeAg-positive; low levels of HBsAg and HBeAg were still detectable 28 d after the infective meal in 2 of 20 (10%) and 1 of 20 (5%) pools, respectively. A crude protease extract was prepared from colony mosquitoes, and the effect of this extract on HBsAg and HBeAg present in human serum was tested in vitro. After 20 h, tests for both antigens were still strongly positive. Low levels of HBsAg were detected in ovaries 7 d after infection. Salivary glands were HBsAg- and HBeAg-negative.  相似文献

Field studies were conducted in a rice, Oryza sativa L., agroecosystem in Mwea Kenya to compare the efficiency of CO2-baited Centers for Disease Control (CDC) light traps against nonbaited CDC light traps and gravid traps against oviposition traps in outdoor collection of Culex quinquefasciatus Say (Diptera: Culicidae) and other mosquitoes. Collectively, 21 mosquito species from the genera Culex, Anopheles, Mansonia, Ficalbia, and Aedes were captured during the 10-wk study period. Cx. quinquefasciatus was the predominant species in all trap types with proportions ranging from 57% in the nonbaited CDC light traps to 95% in the gravid traps. Significantly higher numbers of Cx. quinquefasciatus and Culex annulioris Theobald were collected in the CO2-baited CDC light traps than in the nonbaited CDC light traps, but the numbers of other mosquito species, including malaria vectors Anopheles arabiensis Patton and Anopheles funestus Giles did not differ significantly between the two trap types. More Cx. quinquefasciatus females were collected in grass infusion-baited gravid traps than egg rafts of this species in oviposition traps containing the same infusion. Although most mosquitoes captured in CO,-baited and nonbaited CDC light traps were unfed, most of those collected in gravid traps were gravid. From these findings, it is concluded that at least in the rice-growing area of Mwea Kenya, CO2-baited CDC light traps in conjunction with gravid traps can be used in monitoring of Cx. quinquefasciatus both for control and disease surveillance.  相似文献

为了观察灭蚊真菌贵阳腐霉感染致倦库蚊幼虫后的组织形态学变化,探讨贵阳腐霉对致倦库蚊幼虫的致死机理,采用石蜡切片、 H. E.染色和比较组织学方法,以未经贵阳腐霉作用的致倦库蚊幼虫为对照,连续观察用贵阳腐霉感染24~72 h致倦库蚊幼虫的组织学变化。结果发现被贵阳腐霉感染的致倦库蚊幼虫中肠上皮细胞出现细胞间隙增大,随着感染时间的延长,细胞间隙进一步增大并出现空泡化、质膜破损、细胞裂解一系列病理变化。结果表明贵阳腐霉可导致致倦库蚊幼虫中肠上皮细胞的破坏,推测这可能是导致其死亡原因之一。  相似文献

本文报告了我国尖音库蚊复合组尖音库蚊、淡色库蚊和致倦库蚊表皮碳氢化合物的气相色谱分析,并与本复合组分布于日本的骚扰库蚊以及亲缘种迷走库蚊进行了比较。结果表明,尖音库蚊、淡色库蚊、致倦库蚊和骚扰库蚊表皮碳氢化合物的主要组分相同,含量存在着某些差异,与迷走库蚊相比则有较大的差异,而在一定程度上表现出复合组的特征。  相似文献

Starch gel electrophoretic zymograms were used for the taxonomic separation of mosquitoes of the genus Culex, possible vectors of encephalitis in Argentina. Catalase, hexokinase, malic enzyme, isocitrate and malate dehydrogenases, and leucine aminopeptidase were species markers that allowed the clear identification of individual Culex apicinus Philippi, Cx. bidens Dyar, Cx. brethesi Dyar, Cx. declarator Dyar and Knab, Cx. dolosus Arribalzaga, Cx. maxi Dyar, Cx. quinquefasciatus Say, Cx. saltanensis Dyar, and Cx. tatoi Casal & García. Intraspecific polymorphism was revealed but did not preclude species separation. On the basis of allele frequencies, an index of genetic distance between species was calculated. Genetic affinities determined by electrophoretic data agreed with taxonomic affinities ascertained by conventional systematics. Thus, electrophoretic zymograms proved to be a powerful tool for the study of the genus Culex in Argentina.  相似文献

Attraction of Culex quinquefasciatus Say, Culex tarsalis Coquillett, Culex nigripalpus Theobald, and Aedes aegypti (L.) to avian and other host odors was investigated in a dual-port olfactometer. Although attraction to a human arm was high for Ae. aegypti (>80%) and low for all Culex spp. (<25%), all species responded similarly to a chicken (55.3-73.6%). Responses of Ae. aegypti, Cx. quinquefasciatus, and Cx. nigripalpus to feathers were low (<20%) but greater than to controls. There was no difference in attraction of Cx. tarsalis to feathers or controls. Responses to CO2 (5 ml/min) were low for all species (<15%) except Cx. tarsalis, which were moderate (24.5%). When feathers were combined with CO2, the resulting attraction was additive or lower than responses to feather and CO2 alone for all species except for Cx. tarsalis, which had responses that were three-fold greater than expected if responses were additive. The CO2-feather treatments were less attractive than a chicken for all species. When olfactometer assays were extended from 3 to 20 min, responses by Ae. aegypti significantly increased to a chicken and CO2 and attraction of Cx. quinquefasciatus significantly greater to chickens, CO2, and feathers. None of the volatile compounds previously identified from feathers or uropygial glands tested were attractive. Both feather-rubbed cotton balls and hexane extracts of feathers were attractive and as attractive as feathers; however, ether extracts were not attractive. Feathers clearly contribute to the attraction of host-seeking Culex spp., and future studies will focus on identification of the attractant compounds.  相似文献
