Immunocytochemistry may be of valuable help in typing most malignant serous effusions. Various preparatory methods are described including centrifugation and smearing, cytocentrifugation, cell blocks and liquid based cytology. The value and usefulness of immunocytochemistry depends on the experience of each pathologist. The use of a panel of antibodies associating markers of carcinomas with markers of mesothelial cells is recommended.  相似文献

A continuing problem in cytology laboratories is the lack of adequate control material for immunocytochemical testing. Usual control procedures involve testing paraffin-embedded control materials along with the patient specimens. These control materials are fundamentally unlike cytologic preparations. We have developed a method to make control preparations for immunocytochemical analysis using cultured anaplastic cells with known antigenic features from commercial sources. Cell lines included melanoma, rhabdomyosarcoma, T-cell leukemia, and squamous-cell carcinoma. Modified Saccomano and acetone fixation coupled with the cytospin technique enabled good-quality preparations. Cell lines were tested with antibodies for HMB-45, actin, leukocyte common antigen (LCA), and cytokeratin, with avidin-biotin immunoperoxidase and diaminobenzidine (DAB) as chromogens. Our final preparations were easily interpretable with excellent morphologic preservation of cellular detail. Cultured cells provide a superior method for preparing almost unlimited numbers of control slides for immunocytochemistry for laboratories with access to a tissue culture facility. Diagn. In six cases of Whipple's disease the characteristic macrophages could be stained immunocytochemically using a first layer antiserum to group B streptococci. Other types of periodic acid Schiff positive macrophages were negative. The staining appears to be due to a rhamnose containing polysaccharide as shown by a blocking test. The method may be useful for confirming the diagnosis in rectal biopsy and peripheral nodes, especially if small intestinal findings are atypical.  相似文献

采用牛血清白蛋白(BSA)——肽抗原偶联技术,制备了免抗人防御素多克隆抗血清。并以此抗血清用免疫细胞化学染色法检查了一系列正常白细胞和白血病细胞株的免疫细胞化学反应。结果显示正常人中性粒细胞、急性髓性白血病细胞株KG1和HL-60细胞呈现强阳性反应。在正常人单个核白细胞、人红白血病K562细胞株及小鼠骨髓瘤SP2/0细胞株中均为阴性反应,甚至在健康家兔及豚鼠中性粒细胞中亦呈阴性反应。本实验提示防御紊可能作为粒系白细胞分化标志。  相似文献

Summary An immunocytochemical study of 30 retinoblastomas was carried out using antibodies to neuronal and glial markers. The tumours were found to react with antibodies to neuron-specific enolase (NSE), a marker for neuronal elements, and S-100 and glial fibrillary acidic protein (GFAP), both of which are proteins present in glia. Two distinct cell populations were found within the tumour: the first, composed of anaplastic tumour cells at various stages of differentiation, showed both NSE and S-100 immunoreactivity; the second cell type, which immunostained for S-100 and GFAP, resembled mature glial cells. The results of this study indicate that the retinoblastoma may arise from a pluripotential primitive cell partially retaining neuronal and glial characteristics.  相似文献
