白细胞介素-23在类风湿关节炎滑膜中的表达及意义 总被引:11,自引:0,他引:11
目的白细胞介素(IL)-23由IL-23p19和IL-12p40亚单位组成,它在一些自身免疫疾病发生过程中起着重要的作用。研究分析类风湿关节炎(RA)患者关节滑膜组织中IL-23p19蛋白及其mRNA的表达,旨在探讨其在RA发病中的意义。方法应用反转录-聚合酶链反应(RT—PCR)和免疫组织化学方法检测RA患者关节滑膜组织中IL-23p19基因和蛋白的表达。并与骨关节炎(OA)患者和健康人作对照研究。结果IL-23p19mRNA和蛋白在RA患者关节滑膜组织中表达明显增高.而在OA患者中低表达,正常对照组中无表达。结论IL-23p19在RA关节滑膜组织中高表达.提示IL-23可能直接参与了RA的发病过程。 相似文献

目的:检测慢性心力衰竭患者(CHF)外周血白细胞介素(IL)-12和IL-18的水平并探讨其意义.方法:CHF患者72例,按照NYHA心功能分级,分为HF1(NYHAⅡ~Ⅲ级),HF2组(NYHA Ⅳ级).按照病因分类,分为缺血性心脏病(IHD)组(43例)和非缺血性心脏病(NIHD)组(29例).选取29例健康志愿者作为正常对照组(C组).采用双抗体夹心酶联免疫吸附法(ELISA)检测血清中IL-12和IL-18的水平,用彩色多普勒超声诊断仪测定左室舒张末期内径(LVEDd)和左室射血分数(LVEF).结果:HFI、HF2、IHD和NIHD组IL-12的水平显著升高,分别为(34.56±19.10),(35.12±15.18),(31.18±7.80),(32.73±11.54)ng/L,与C组(28.73±13.43)ng/L比较,均差异有统计学意义(P<0.05),IL-18的水平分别为(39.50±5.66),(46.76±12.66),(40.58±13.10),(42.53±6.16)ng/L,明显高于C组(31.82±5.79)ng/L,均差异有统计学意义(P<0.05或P<0.01),HF2组血清IL-18水平显著高于HF1组(P<0.05).与C组比较,HF1、HF2、IHD和NIHD组的LVEDd明显增大.LVEF显著降低.IL-12水平与LVEDd呈低度正相关(P<0.01),IL-18水平与LVEDd呈中度正相关(P<0.01),与LVEF呈中度负相关.结论:IL-12和IL-18在CHF患者血清中表达升高;IL-12的水平与患者基础病因和心功能均无明显相关性;IL-18的水平与CHF的基础病因无明显相关性,但与患者的CHF程度呈正相关. 相似文献

取类风湿关节炎(RA)患者的关节滑膜40份(RA组)及外伤后患者的关节滑膜40份(对照组),用胶原酶消化培养法培养至3-8代,用免疫组化法检测滑膜细胞IL-18表达,用酶联免疫吸附检测细胞培养上清液中IL-18水平。结果RA组滑膜细胞及上清液中IL-18分泌量明显高与对照组(P〈0.05)。认为IL-18高表达参与RA发病,可作为临床治疗的靶点之一。 相似文献

目的 研究类风湿关节炎(RA)患者血清、滑液中白细胞介素-18(IL-18)蛋白及滑膜组织IL-18mRNA表达水平,探讨其在RA致病中的作用。方法 应用双抗夹心酶免疫吸附(ELISA)法和细胞生物法分别测定RA患者血清、滑液中IL-28蛋白水平和生物活性,同时还检测NO、前列腺素E2的含量;有用半定量RT-PCR法检测膜组织IL-18m RNAG表达水平。以骨关节炎(OA)病人及因外伤截肢的正常人作对照。结果 RA患者血清、滑液中IL-18蛋白水平和生物活性均显著高于对照组,滑液中量及活化性比血清高;RA滑膜组织IL-18mRNA表达水平也明显高于对照组。结论 过度表达的IL-18参与了RA的致病过程,;选择性地抑制IL-18生物活性,将是RA治疗的新途径。 相似文献

系统性红斑狼疮(systemic lupus erythematosus,SLE)患者体内细胞因子反应非常复杂,Th1细胞因子和Th2细胞因子的表达均上调,在疾病的不同发展阶段可能有不同的细胞因子产生,细胞因子失衡在SLE的发病中起重要作用。白细胞介素(IL)-18是新发现的一种细胞因子,主要由活化的巨噬细胞产生,是IL-1家族的成员之一,与IL-12有相似的生物学功能。 相似文献

目的探讨白细胞介素-1受体(IL-1R)家族中的3个成员IL-1RⅠ、IL-1RⅡ和IL-1R协同蛋白(IL-1RAcP)在骨关节炎(OA)滑膜中的表达和生物学意义。方法采用免疫组织化学和反转录聚合酶链反应(RT-PCR)方法检测了不同病程的107例OA患者膝关节滑膜组织中三者的表达情况。结果免疫组织化学显示IL-1RⅠ和IL-1RAcP在OA滑膜中强阳性表达,IL-1RⅡ在OA滑膜中阳性表达,在对照组中均呈阴性或弱阳性表达。RT-PCR显示IL-1RⅠ、IL-1RⅡ、IL-1RAcP在Ⅰ度OA滑膜中的表达均显著高于正常滑膜(P<0.05),且IL-1RⅠ的表达显著高于IL-1RⅡ(P<0.05),而与IL-1RAcP的差异无统计学意义(P>0.05)。但在Ⅱ度和Ⅲ度OA滑膜中,IL-1RⅠ、IL-1RAcP的表达与在正常滑膜中的表达差异无统计学意义(P>0.05),IL-1RⅡ在Ⅲ度OA滑膜中的表达显著高于正常组(P<0.05)。结论IL-1RⅠ、IL-1RⅡ、IL-1RAcP在OA的发病机制中起着重要的作用,尤以IL-1RⅠ和IL-1RAcP为主,但其表达只在OA早期短期增加,与OA病情的发展可能无关。 相似文献

目的:观察白细胞介素-12(IL-12)及白细胞介素-18(IL-18)能否导致脾虚,研究脾虚与IL-12及IL-18的关系。方法:60只小鼠随机分为脾虚组、IL组和对照组。采用利血平制作脾虚动物模型组,IL组每日腹腔注射IL-12、IL-18。采用免疫分析技术检测3组小鼠血浆IL-12、IL-18的含量。结果:IL-12及IL-18引起明显脾虚证的表现,与利血平制作脾虚动物模型组相似。脾虚组和IL组血浆IL-12、IL-18的含量明显升高。结论:IL-12、IL-18可导致脾虚,血浆IL-12、IL-18的含量可作为脾虚证的实验室参考指标。 相似文献

目的 探讨类风湿关节炎 (RA)外周血白细胞介素 18(IL 18)的表达及其与Th1/Th2平衡和疾病活动的关系。方法 以 β actin为内参照 ,用半定量反转录多聚酶链反应 (RT PCR)法测定 16例RA患者及 15名正常对照外周血单个核细胞 (PBMC)中IL 18、IL 4、IFN γmRNA水平 ,观察IL 18与IL 4、IFN γ及疾病活动指标红细胞沉降率 (ESR)、C反应蛋白 (CRP)的相关性。结果 ①RA患者PBMC中IL 18mRNA表达与正常组相比增高非常显著 (1 82± 0 39比 0 45± 0 30 ,P <0 0 0 1)。②RA组IFN γ、IL 4均显著高于对照组 ,但IL 4/IFN γ显著低于对照组。③相关分析显示 :RA组IL 18mRNA与IFN γ、ESR显著正相关 (16例 ,r分别为 0 836 ,0 75 3,P均 <0 0 5 )。结论 RA患者外周血IL 18mRNA表达水平显著高于正常对照组 ,且与RA患者外周血IFN γ的产生及疾病活动性相关。 相似文献

目的:研究检测高血压病患者血清白细胞介素-18(IL-18)和C反应蛋白(CRP)的临床意义。方法:选择168例高血压患者和58例健康体检者,以酶联免疫吸附试验(ELISA)法和免疫比浊法检测其血清IL-18和CRP水平,分析其与高血压的相关性和临床意义。结果:与正常对照组比较,高血压病(1~3级)各组血清IL-18[(120.7±76.4)ng/L:(224.8±80.7)ng/L:(478.9±184.1)ng/L:(669.1±210.3)ng/L]、CRP[(2.4±1.6)mg/L:(5.9±3.8)mg/L:(24.1±12.6)mg/L:(41.6±16.5)mg/L]水平均显著升高(P均0.01);且随高血压分级水平升高,血清IL-18及CRP水平均逐渐升高(P0.01)。结论:血清白细胞介素-18、C反应蛋白水平与高血压的发病及分级水平有关,其测定很有意义。 相似文献

AIM: To explore the effect of intratumoral expressions of interleukin-12 (IL-12) and interleukin-18 (IL-18) on clinical features, angiogenesis and prognosis of gastric carcinoma. METHODS: The expressions of IL-12 and IL-18 from 50 samples of gastric cancer tissue were analyzed by immunohistochemistry, and microvessel density (MVD) was determined with microscopic imaging analysis system. RESULTS: The positive expression rates of IL-12 and IL-18 were 44% (22/50) and 26% (13/50), respectively. IL-12 was significantly associated with pathologic differentiation, depth of invasion, lymph node metastasis, distant metastasis, and TNM stage, and IL-18 was closely related to distant metastasis. Intratumoral IL-12 and IL-18 expressions were not statistically related to MVD scoring. IL-12-positive patients survived significantly longer than those with IL-12-negative tumors, but there was no significant difference between IL-18-positive patients and IL-18-negative ones. The multivariate analysis with Cox proportional hazard model revealed IL-12, MVD and T stage were independent prognostic factors. CONCLUSION: The positive expressions of IL-12 and IL-18 can play an important role in progression and metastasis of gastric cancer, and IL-12 might be an independent factor of poor prognosis in gastric carcinoma. 相似文献

目的 探讨白细胞介素-18(IL-18)的表达与慢性HBV感染后不同疾病转归患者的相关性及其临床意义。方法 收集230例慢性HBV感染后不同疾病患者肝脏组织和临床资料,采用半定量SP免疫组织化学法检测HBV感染者肝组织中IL-18的表达情况,并与患者的临床生物化学指标、肝脏炎症分级和纤维化分期进行比较,分析肝脏组织中IL-18表达与上述指标的相关性。结果 IL-18在肝脏组织中以肝细胞细胞质表达为主,半定量分析结果显示,在健康者、CHB、肝硬化、HBV相关性肝癌患者肝组织中的IL-18表达,差异均有统计学意义(P〈0.05);IL-18在HBV感染者肝组织中的表达与ALT水平、肝脏炎症及纤维化程度均呈正相关(r=0.184、0.169、0.085,P=0.015、0.021、0.032)。结论IL-18表达强弱与HBV感染的免疫发病机制有关,IL-18参与了HBV感染的慢性化和疾病的转归。 相似文献

目的观察冠心病急性心肌梗死(AMI)患者溶栓治疗前后血浆心肌营养素-1(CT-1)和血清白介素-18(IL-18)的变化及意义。方法溶栓组于溶栓即刻,溶栓后2、4、6、8、10、12h测定血中CT-1和IL-18浓度,未溶栓组于症状出现后2、4、6、8、10、12、14h测定血中CT-1和IL-18浓度;同时测定血清cTnI浓度。结果AMI患者IL-18浓度明显高于对照组,升高时间早于cTnI升高的时间(P〈0.01);AMI患者CT—1水平明显高于对照组,升高时间早于cTnI及IL-18升高的时间(P〈0.01)。与cTnI相比,CT-1和IL-18检测的灵敏度和特异性均较高,尤其是发病早期患者血中尚不能检测到cTnI,而CT-1和IL-18水平即明显升高。②溶栓后4、6、8h,再通组CT—1和IL-18浓度显著高于未再通组(P〈0.01),未再通组CT-1和IL-18浓度与未溶栓组相比差异无统计学意义。结论@AMI患者血清中很早即可测到较高浓度的cT-1和IL—18,且水平明显高于对照组,时间早于cTnI升高的时间,故认为CT-1和IL-18可作为AMI早期诊断的参考指标之一。②溶栓后4、6、8h,溶栓再通组可检测到明显升高的CT-1和IL-18,故认为CT-1和IL-18可以作为判定AMI溶栓治疗是否再通的指标之一。 相似文献

Objectives: To investigate cross-sectional associations between serum levels of resistin and interleukin-17 (IL-17) and cartilage defects and bone marrow lesions (BMLs) in patients with knee symptomatic osteoarthritis (OA).Methods: One hundred and ninety-four consecutively-selected patients with knee symptomatic OA (mean 55.4 years, range 34–74, 87% females) were included in Anhui Osteoarthritis (AHOA) Study. Knee cartilage defects and BMLs were determined at different sites using T2-weighted fat-suppressed fast spin echo MRI. Serum resistin, IL-17, and high-sensitivity C-reactive protein (hs-CRP) levels were measured using ELISA.Results: In multivariable analyses, serum resistin was positively associated with cartilage defects at lateral femoral, lateral tibial, and medial tibial (all p?0.05) sites. The significant associations were also present with BMLs at lateral femoral and tibial sites (ORs: 1.13–1.19, both p?0.05). In patients with the highest quartile of hs-CRP (>2.45?pg/ml), IL-17 was positively and significantly associated with cartilage defect score at nearly all sites (ORs: 1.33–1.44, all p?0.05), and BMLs at lateral and medial femoral sites (ORs: 1.26–1.51, both p?0.05).Conclusions: Serum levels of resistin were positively and independently associated with cartilage defects and BMLs in patients with knee OA. Serum IL-17 was significantly associated with cartilage defects and BMLs in patients with an increased inflammatory status. These suggest that metabolic and inflammatory mechanisms may have a role to play in knee OA. 相似文献

目的 本实验以ConcanavalinA(ConA)诱发小鼠急性肝损伤模型,检测白细胞介素-18(Interleukin-18,IL-18)及一氧化氮(Nitric oxide,NO)血清浓度变化。并探讨此模型中IL-l8和NO的关系、意义及作用机制。方法 用ABC-ELISA法及硝酸还原酶法分别检测了20只正常小鼠(对照组)及20只(实验组)小鼠6h血清中IL-l8及NO的浓度。结果 实验组小鼠血中IL-l8及:NO较对照组均明显增高,差异有显著性。经直线相关分析,IL-18与NO呈显著相关性。结论 IL-18在肝损伤中起关键作用,NO对肝损伤亦有重要作用,NO浓度随IL-l8表达水平的变化而变化,两者对肝损伤既有独立又有协同作用。IL-l8的浓度高低可用于判断肝损伤的严重程度及预后,IL-18使血清中NO水平升高是其致肝细胞损伤的机理之一。 相似文献

AIM To determine whether serum interleukin-18 (IL-18)levels correlated with clinicopathologic features and prognosis in patients with hepatocellular carcinoma (HCC).METHODS Serum IL-18, IL-6 and IL-12 levels were measured by enzyme-linked immunosorbent assay (ELISA) from 70 patients with HCC and 10 healthy controls.RESULTS Serum IL-18, IL-6 and IL-12 levels of patients with HCC were significantly higher that those of the controls. The levels of IL-18 correlated significantly with the presence of venous invasion and advanced tumor stages classified by Okuda's criteria. Patients with high serum IL-18 levels (≥ 105 pg/mL) had a poorer survival than those with low serum IL-18 levels (< 105 pg/mL)(4 and 11 mo, respectively, P = 0.015). Multivariate analyses showed that serum IL-18 level, but not IL-6 and IL-12 levels, was a significant and independent prognostic factor of survival.CONCLUSION These findings demonstrate that serum IL-8 may a useful biological marker of tumor invasiveness and an independent prognostic factor of survival for patien ts with HCC. Thus, the detailed mechanisms of IL-18 involving in tumor progression should be further investigated. 相似文献

目的:检测克罗恩病(CD)和溃疡性结肠炎(UC)肠黏膜组织中IL-27 p28 mRNATL其蛋白、IL-27受体mRNA的表达,探讨其在CD和UC中的发病意义.方法:应用反转录-聚合酶链反应(RT-PCR)和蛋白质免疫印迹(Western blot)方法检测炎症性肠病患者炎症肠黏膜组织中IL-27 p28基因及蛋白、IL-27受体基因的表达,并与健康者作对照.结果:IL-27 p28 mRNA在CD患者中的阳性率和灰度分析表达较UC患者明显增高(X2=6.64,P<0.05;t=11.01,P<0.01),IL-27受体阳性率和灰度分析在CD患者较UC患者和健康对照者明显增高(阳性率:X2=10.91,P<0.016,X2=18.84,P<0.016).IL-27蛋白阳性率和灰度分析在CD患者中表达明显高于UC患者(X2=5.24,P<0.05;t=3.37,P<0.05),并且IL-27mRNA的表达与蛋白质表达密切相关.结论:IL-27 p28及其受体的上调,可能有助于CD患者炎症发展过程. 相似文献

Aim of the workTo further investigate the possible role of IL-18 in the pathogenesis of systemic lupus erythematosus (SLE) and development of lupus nephritis (LN), and to explore its relationship with pathological classes of LN, degree of acute renal activity and chronic damage.Patients and methodsForty-one SLE patients with LN, thirty-one lupus non-nephritis patients and fifteen age and sex matched healthy controls were enrolled in this study. SLE patients were subjected to disease activity assessment by SLEDAI, renal disease activity assessment by the Systemic Lupus International Collaborating Clinics (SLICC) Renal Activity Score, laboratory investigations including measurement of serum interleukin-18 using Enzyme Linked Immunosorbent Assay. Renal biopsy was obtained from LN patients and pathological classification was made according to World Health Organization (WHO) criteria. Analysis of activity and chronicity indices was done on these biopsy specimens.ResultsSerum levels of IL-18 were significantly higher in patients with LN than lupus non-nephritis patients and healthy controls (p < 0.001). There were significant correlations between IL-18 and SLEDAI (p = 0.002), proteinuria (p = 0.027), renal activity score (p = 0.003) and activity index (p = 0.039) in patients with LN. There was no significant difference in the serum levels of IL-18 between WHO classes of LN.ConclusionIL-18 appears to have a pathogenic role in the development of SLE and plays a crucial role in triggering inflammation in LN. Serum IL-18 levels could be a useful biomarker to assess the activity of renal disease in SLE. 相似文献
