目的: 观察丹参酮ⅡA磺酸钠(sodium tanshinone ⅡA sulfonate,STS)对低氧诱导的大鼠肺动脉平滑肌细胞(pulmonary arterial smooth muscle cells,PASMC)增殖的影响及其潜在的作用机制。方法: 将大鼠PASMC分为常氧组、常氧+STS组(5、10、20 ng/mL)、低氧组(3%O2)、低氧+STS组(5、10、20 ng/mL),培养60 h后,采用CCK-8法测定PASMC的增殖率;另将大鼠PASMC分为常氧组、低氧组、低氧+STS组(10 ng/mL),qRT-PCR测定mTOR、eIF2α、c-myc mRNA的相对表达量;再将PASMC分为常氧组、低氧组、低氧+STS组(10 ng/mL)、低氧+雷帕霉素组(20 nmol/L),蛋白质印迹法检测mTOR、p-mTOR、eIF2α、p-eIF2α、c myc蛋白的表达量。结果: 与常氧组比较,低氧组细胞增殖率明显升高(P<0.05);与低氧组比较,低氧+STS组细胞增殖率明显降低(P<0.05)。qRT-PCR显示低氧组mTOR、eIF2α和c-myc mRNA相对表达量较常氧组明显升高(P均<0.05),低氧+STS组3种mRNA相对表达量较低氧组降低(P均<0.05)。蛋白质印迹显示,与常氧组比较,低氧组mTOR、p-mTOR、eIF2α、p-eIF2α、c-myc蛋白表达均显著升高(P均<0.05);与低氧组比较,低氧+STS组和低氧+雷帕霉素组的mTOR、p-mTOR、eIF2α、p-eIF2α、c-myc蛋白表达均升高(P均<0.05);而低氧+雷帕霉素组与低氧+STS组无明显差异(P>0.05)。 结论: STS可抑制低氧诱导的大鼠PASMC增殖,可能通过抑制mTOR/eIF2α信号通路而发挥作用。  相似文献

目的:研究低氧对复合培养的平滑肌细胞中Janus激酶(JAK)基因表达的作用。方法:鼠肺微血管内皮细胞与肺动脉平滑肌细胞复合培养,采用半定量RT-PCR技术检测常氧组、低氧2、6、12h组的JAK1、JAK2和JAK3基因的表达水平,并对PCR产物进行测序。结果:低氧复合培养2h组肺动脉平滑肌细胞中JAK1、JAK2、JAK3 mRNA表达水平升高,在低氧6h组达最高,与正常组比较均有明显差别。低氧12h组的表达量低于6h组,但高于正常组。RT-PCR产物直接测序证实扩增产物片断与Genebank中相应序列相同。结论:低氧增强复合培养的肺动脉平滑肌细胞中JAK基因的表达,而JAK基因在低氧所致的肺动脉收缩和低氧肺动脉高压的信号转导中可能发挥重要作用。  相似文献

观察内皮素-1(ET-1)和一氧化氮(NO)在缺氧性肺血管结构重组中的作用,发现ET-1可促进肺动脉平滑肌细胞(PASMC)DNA合成,其促进作用呈剂量依赖性;NO供应剂SNP则起抑制作用,NO的抑制增殖作用主要由cGMP介导。在此基础上观察到缺氧可促进PASMC对ET-1的增殖反应,同时可抑制PASMC胞浆内可溶性鸟苷酸环化酶活性而降低PASMC对NO等舒血管药物的反应性。提示ET-1和NO及缺氧在缺氧性肺血管结构重组中具有重要的作用。  相似文献

Summary In order to study the effect ofErigeron Breviscapus (EB) on proliferation of pulmonary artery smooth muscle cells (PASMC) in hypoxic porcines, immunohistochemical and MTT methods were employed to measure the proliferation of PASMC. It was found that the proliferation of PASMC in porcines was obvious, and the expression of proliferating cell nuclear antigen (PCNA) was significantly high within 48 h after exposure to hypoxia. The EB could inhibit the proliferation and the expression of PCNA in PASMC under hypoxia, but it had no effect on the proliferation and expression of PCNA in PASMC under normal condition. The EB could inhibit the proliferation and the expression of PCNA in PASMC induced by phorbol 12-myristate 13-acetate (PMA), an agonist of PKC in normal and hypoxic conditions. It was concluded that the hypoxia could enhance the proliferation and expression of PCNA in PASMC. The EB can inhibit the proliferation and expression of PCNA in PASMC under hypoxia through PKC-signal way. The EB may be used in treating the pulmonary hypertension by inhibiting the proliferation of PASMC and the pulmonary vascular remodeling. 目的 探讨血栓素A2(TXA2)合成酶抑制剂奥扎格雷钠对低氧肺动脉平滑肌细胞(PASMC)增殖及凋亡的影响.方法 采用四甲基偶氮唑盐微量比色法、免疫组化法及原位末端脱氧核苷酸转移酶标记法观察TXA2激动剂U46619及TXA2合成酶抑制剂奥扎格雷钠对PASMC增殖及凋亡和对反映细胞增殖状态的重要标记物Ki67表达的影响.结果 U46619对常氧和低氧PASMC的增殖和低氧PASMC中Ki67的表达均有明显促进作用,并可明显抑制PASMC的凋亡;奥扎格雷钠对常氧PASMC的增殖无明显影响,但对低氧PASMC的增殖和Ki67的表达均有明显抑制作用,可促进低氧PASMC的凋亡;奥扎格雷钠可明显拮抗U46619在常氧和低氧条件下对PASMC的促增殖和Ki67表达的作用,以及U46619抑制低氧PASMC的凋亡的作用.结论 奥扎格雷钠有可能通过抑制低氧PASMC的增殖及血管重塑、促进凋亡,从而对低氧性肺动脉高压起治疗作用.
Abstract:
Objective To study the effect of Thromboxane A2 (TXA2) inhibitor on the proliferation of hypoxic pulmonary artery smooth muscle cells (PASMC) of porcines. Methods The methods of 3(4,5-dimethylthiazol-2-yl) -2,5-diphenyl tetrazolium bromide ( MTT), immunohistochemistry of Ki67 and TdTmediated dUTP nick end labeling(TUNEL) were employed to measure the proliferation, inhibition rate and apoptosis of PASMC. Results U46619, a thromboxane A2 agonist, could promote the proliferation and expression of Ki67 in PASMC under hypoxic and normal conditions. It could also inhibit the apoptosis of PASMC. Ozagrel, a thromboxane A2 inhibiter, inhibited the proliferation and the expression of Ki67 in PASMC under hypoxia. The inhibition rate was 71.4%, but it had no effect on the proliferation and expression of Ki67 in PASMC under normal conditions. It promoted the apoptosis of PASMC. Ozagrel could inhibit the action of U46619 in promoting the proliferation and expression of Ki67 in PASMC and inhibit the apoptosis of PASMC. Conclusion Ozagrel may be used in treating the pulmonary hypertension by inhibiting the proliferation of PASMC, pulmonary vascular remodeling, and promote the apoptosis of PASMC.  相似文献
