Histamine release (HR) after stimulation with anti-IgE, concanavalin A (ConA) and Formyl-Met-Leu-Phe (FMLP) from 97 cord blood samples was compared to results obtained in identically treated blood samples from adults.The maximal HR obtained with anti-IgE did not differ significantly from the values obtained in adult blood, although a ten times higher concentration of anti-IgE was required for maximum release.Passive sensitization with IgE-rich plasma caused a significant increase in maximal anti-IgE-induced HR in the majority of cord blood samples, and the dose-response curve was similar to that obtained in adult blood.Challenge with ConA and FMLP caused a HR similar to that seen in adult blood, but the close correlation between anti-IgE- and concanavalin A-induced HR seen in adult blood was absent in cord blood.  相似文献

Artificial blood     
Artificial blood is a product made to act as a substitute for red blood cells. While true blood serves many different functions, artificial blood is designed for the sole purpose of transporting oxygen and carbon dioxide throughout the body. Depending on the type of artificial blood, it can be produced in different ways using synthetic production, chemical isolation, or recombinant biochemical technology. Development of the first blood substitutes dates back to the early 1600s, and the search for the ideal blood substitute continues. Various manufacturers have products in clinical trials; however, no truly safe and effective artificial blood product is currently marketed. It is anticipated that when an artificial blood product is available, it will have annual sales of over $7.6 billion in the United States alone.  相似文献

Relative changes in local blood volume in 46 vascular regions of the body after moderate and severe blood loss are described. Moderate blood loss caused a redistribution of blood from the skin of the chest and hind limbs, most organs of the abdomen and pelvis, the muscular and bony tissues of the abdomen, pelvis, and limbs to the brain, heart, lungs, kidneys, stomach and to the muscles of the head and neck. After severe blood loss the changes were similar but the blood volume in the kidneys and stomach was reduced; a relative increase in the blood volume in the muscular and bony tissues of the thorax also was observed. Cord blood     
Screening of total IgE in 2814 cord blood samples was analysed by Phadebas IgE PRIST in 2 1-year birth cohorts (1983-1984 and 1985–1986) in Denmark (n =1189+ 1625). 48.6% of the sera contained less IgE than the detection limit 0.1 kU/1. Cord blood IgE values > 0.5 kU/1 were regarded as elevated. 13.2% of the sera contained at least 0.5 kU/1 of IgE, with a significant preponderance in boys. Geometric mean cord blood IgE was 0.13 kU/1 and 0.12 kU/1, respectively. Geometric mean cord blood IgE was significantly higher in boys. A significant seasonal variation with lowest IgE values in the autumn was found. No correlation between cord blood IgE and birth weight or gestational age was demonstrated. Only few newborns had cord blood igA values > 0.014 g/l, calculated as geometric mean cord blood IgA + 2 SD among children with no detectable cord blood IgE, indicating infrequent contamination with maternal blood.
Clinical aspects
Using a simple cord blood sampling method and a sensitive IgE analysis., Phadebas IgE P rist , reproducible results of cord blood IgE in 2 unselected series of newborns were obtained. Cord blood IgA determination is recommended in all infants with cord blood IgE ≥ 1.0 kU/1 in order to exclude contamination with maternal blood.  相似文献

背景:随着生物医学工程学和生物材料学的发展,人工血管的研究得到广泛的应用,人工血管材料得到不断更新,如何提高血管的通畅性和人工血管材料的相容性是在近年来人工血管研究的重点。 目的:利用CNKI数据库文献检索和深度分析功能,对于人工血管材料学研究的文献资料趋势进行多层次探讨分析。 设计:文献计量学分析。 资料提取:以电子检索方式对CNKI数据库2002-01/2011-12有关人工血管材料学研究的文献进行分析,采用检索词为“人工血管(Artificial blood vessels);生物材料(Biomaterials)”,对检索的相关文献运用数据库中自带的分析功能和Excel软件绘制图表的功能进行分析,通过文字和图表的形式将统计和计量数据分析,描述其分布特征。 入选标准:纳入标准:①与人工血管材料相关的基础研究论文。②与人工血管材料临床应用相关的论文。排除标准:①与文章目的无关的文献。②重复研究的文献。③刊社信息。④未发表的文章。⑤需电话追踪和手工检索逐一分析的文章。⑥年鉴。 主要数据判定指标:以CNKI数据库学术期刊文献出版时间、文献数量、学科类别、研究机构、来源期刊、文献被引频次、文献下载频次、关联文献、作者分布、基金资助情况和主要关键词进行相关分析。并对CNKI数据库中博士学位论文、优秀硕士学位论文、专利技术和重要会议论文进行分析。 结果:在CNKI数据库学术期刊收录2002/2011的文献中,共检索到122篇与人工血管材料学研究相关的文献。文献数量产出趋于上升趋势,2008年文献产出数量最多共17篇;《中国组织工程研究与临床康复》杂志发表文献量最多为27篇,占全部文献的22.1%;人工血管材料学研究基金资助以国家自然科学基金项目为主,共发表文献30篇;人工血管材料学研究以聚氨酯的研究为主,突出材料学生物相容性和血液相容性的研究。 结论:通过文献计量学方法对CNKI数据库学术期刊关于人工血管材料的文献进行分析,可为中国从事人工血管材料学基础研究和临床实施的医务工作者进一步确定科研思路提供有价值的参考。  相似文献

The thermally stimulated discharge (TSD) method, intended for the analysis of charged dielectrics, was used for human blood research. Above-room-temperature TSD spectra of blood consist of three peaks. There are indications that the low-temperature peak (40-50 degrees C) corresponds to the thermally stimulated destruction of hydrate shells surrounding blood components while the mid-temperature peak (70-90 degrees C) is related to thermal denaturation of blood proteins. The intensive high-temperature peak (105-120 degrees C) is observed when a phase transition of blood, accompanied by the formation of a firm dry film of blood, occurs. The position of the high-temperature peak depends on the blood group. Data is discussed which suggests that the spontaneous "quasi-electret effect" of blood relates to the character of the biochemical processes taking place in human organisms. The TSD method might be used as simple and informative means of diagnostics in cooperation with medical and physical investigations.  相似文献

目的 为准确模拟血流,研究红细胞变形性对血液流动的影响。方法 基于血液流变特性和红细胞力学特性分析,对现有血液两相流流动模型进行改进,改进模型中考虑了易变形红细胞受剪切流场或血管壁面作用而产生的非惯性升力的影响。利用改进模型对多个不同直径血管内的血液流动进行模拟。结果 由红细胞所受非惯性升力导致的径向运动对血管内红细胞体积分数、运动速度分布有明显影响;当血管直径为0.1~3.0 mm时,用改进模型得到的血液相对黏度的模拟值与测量值接近。结论 非惯性升力是血流呈现Fahraeus-Lindqvist效应的主要原因之一。考虑非惯性升力的改进模型可以准确模拟血液流动,为循环系统诊疗机制和细胞分选等过程的模拟提供更为准确的方法。  相似文献

IntroductionBlood is a valuable as well as a perishable commodity that has a limited lifespan. So, keeping it in a blood bank requires careful attention and management. In this study, a management model has been designed for the blood bank that can cover all the requests sent through the hospital subunits in such a way that it has the least waste.Materials and MethodsFirst, by examining the blood inventory system of a hospital, the number of inventories per month and the distribution of blood products in subunits of the hospital, and the amount of blood wastage every six months were estimated. Then, using ARENA simulation, all parts of the blood bank, including the request section, emergency blood section, donated blood section, and other sections were simulated. Finally, 10 scenarios were extracted by placing various parameters and the best scenarios were discussed and concluded.Results and discussionThe scenarios for the four blood groups included A, B, AB, and O were applied for six months in one year. The results showed that if the amount of blood injected into the blood bank is appropriate and at the right time, the amount of wastage can be reduced by up to 80% for most of the blood groups.ConclusionThe results of this study showed that if the system of receiving blood products built upon a precise management model is predetermined and placed with appropriate parameters, the amount of wastage will be reduced to the minimum possible number.  相似文献

It is necessary to develop methods for accurate monitoring of cell-free hemoglobin in circulation. Routine monitoring of circulating cell-free hemoglobin will be useful for evaluating the efficacy of blood substitute administration andfor determining the clearance rates of the blood substitute from circulation. In addition, discriminating between cell-free hemoglobin and cell-associated hemoglobin will enable accurate determination of RBC indices, mean cell hemoglobin and mean corpuscular hemoglobin concentration, in individuals receiving hemoglobin-based blood substitutes. As colorimetric methods used by hematology analyzers to quantitate the hemoglobin value of a blood sample cannot distinguish between cell-associated and cell-free hemoglobin, it is currently not feasible to quantitate the levels of hemoglobin substitutes in circulation. The advent of a technology that measures volume and hemoglobin concentration of individual RBCs provides an alternative strategy for quantitating the cell-associated hemoglobin in a blood sample. We document that the combined use of cell-based and colorimetric hemoglobin measurements provides accurate discrimination between cell-associated and cell-free hemoglobin over a wide range of hemoglobin levels. This strategy should enable rapid and accurate monitoring of the levels of cell-free hemoglobin substitutes in the circulation of recipients of these blood substitutes.  相似文献

BackgroundTo accurately identify ABO blood typing in pre-transfusion testing is very important to ensure blood transfusion safely, which is a major responsibility of blood station.MethodsEighty-one blood donors samples with ABO blood group typing discrepancy was collected among 61952 donor samples in our blood station from January 2019 to July 2020. Blood group serological method was used to detect ABO blood group. DNA Sequencing was used to determine the genotype. The antibody screening test detects antibodies other than ABO.ResultsIn total, 61,952 donor samples were analysed for ABO typing discrepancies. The incidence among blood donors was 0.13% (81/61952). The most common reason of ABO typing discrepancies was due to specific antibody or non-specific agglutination (54.32%, 44/81), mainly anti-M antibody, cold autoantibody, anti-D antibody, anti-N antibody and anti-Lea antibody. The major cause of forward typing discrepancies among blood donors was ABO subgroups (25.93%, 21/81), including 10 cases of A subtype (1 case of A2, 2 cases of A3, 2 cases of Ax, 3 cases of AxB, 1 case of Ael, 1 case of Ahm), 6 cases of B subtype (2 cases of B3, 1 case of Bel, 3 cases of AB3), 2 cases of B subtype (A), 1 case of cisAB, and 2 cases of acquired B. The serum antibody was weakened in 16 cases (19.75%).ConclusionsThe blood types should be correctly identified by combining serology with gene sequencing to ensure the safety of clinical blood transfusion, when the forward and reverse typing discrepancies among the blood donors.  相似文献

Summary In experiments on dogs a study was made of the role played by the lymphatic system in the acceleration of blood coagulation after acute blood loss. After blood letting the clotting time decreased in all the experiments except one. As distinct from blood, the rate of lymph coagulation remained unchanged. Disturbed lymph circulation, caused by cannulzation of the chief lymph collectors, did not exclude acceleration of blood coagulation after blood letting. Thus, acceleration of blood coagulation following blood loss was not caused by the penetration of factors stimulating hemocoagulation through the lymph. The use of a diabetic blood glucose meter for radiochromic film dosimetry in blood irradiation using x-ray beams on a medical linear accelerator has been investigated. The glucose meter provides optical density analysis in the visible and infrared region using a reflectance measurement technique. By comparing the 'blood sugar' level output with standard calibration gafchromic films a calibration curve is produced for quantitative analysis. Results show that a reproducible dose to meter output curve can be fitted using a second order polynomial function and that blood irradiation doses in vitro were measured to within 7.9% mean error (as compared to ionization chamber results) using the blood glucose meter. This level of accuracy falls below that measured with a standard densitometer (4.3%); however, results show that the blood glucose meter, which would be available in any haematology department, produces an adequate measure of gafchromic film optical density for blood irradiation dosimetry.  相似文献

目的预测患者某一具体HLA表型在脐带血库查询到相合脐带血的可能性.方法以脐带血库单体型频率和脐带血库容量为基础计算出某一具体表型的理论值与实际查询结果相比较.结果6个等位基因相合脐带血的理论值最低为0.128,最高为2.808,平均为1.029,5个等位基因相合脐带血的理论值最低为0.004,最高为0.427,平均为0.067,4个等位基因相合脐带血的理论值最低为0.0002,最高为0.045,平均为0.006 5.结论查询到与患者6个等位基因相同脐带血的理论值与实际查询结果相比较,准确率能达到80%以上.尤其是划定某一表型是否能找到6个等位基因相合的脐带血时可做出较为准确的判断.但对预测找到5个或4个等位基因相合的脐带血区别能力较差.  相似文献

The direction of pituitary blood flow, the amount of pituitary blood flow, its regional control, and the role of the median eminence microcirculation are the subjects of this review. Present concepts of pituitary blood flow are focused almost entirely on its direction and arouse from studies of pituitary vascular anatomy performed almost 50 years ago. The development of new anatomic techniques has led to a reappraisal of pituitary angioarchitecture, stimulated physiological studies to clarify the pattern of blood flow within the entire gland, and led to a reappraisal of accepted concepts of directional pituitary blood flow. The availability of techniques to accurately measure organ blood flow has permitted study of pituitary blood flow; and, when combined with knowledge of pituitary anatomy, the application of these techniques promises to provide a means to develop insight into control of the mechanisms by which chemical messengers are delivered to the pituitary to control its function. New anatomic techniques promise to develop new understanding of the three-dimensional arrangement of median eminence microvasculature and yield new concepts of blood flow regulation within the median eminence that can be tested by physiological means.  相似文献

背景:内皮祖细胞可从外周血与脐血中获取,是修复各种疾病所致损伤的血管内皮细胞不可或缺的细胞来源。 目的:比较人外周血与脐血来源的内皮祖细胞经体外培养后生物学特性的差异。 方法:通过密度梯度离心法和6%羟乙基淀粉结合密度梯度离心法分别分离人外周血与脐血中的单个核细胞,分别设为脐血源组和外周血源组,计数各组单个核细胞数量,按1.0×106/cm2接种于大鼠尾胶包被的培养皿中,用内皮细胞培养基进行诱导,共培养7 d。 结果与结论:外周血与脐血体外培养分离出内皮祖细胞具有类似的形态学特征。光学显微镜下观察,随着培养天数的增加,大多数细胞由早期的贴壁圆形转变为梭形。外周血源组内皮祖细胞有细胞集落形成,脐血源组可见梭形细胞自行排列生长为典型的线样结构。锥虫蓝染色及绘制细胞生长曲线后发现,外周血源组单个核细胞及内皮祖细胞数量、内皮祖细胞活率及增殖能力均低于脐血源组(P < 0.05)。外周血源组和脐血源组内皮祖细胞在接种后第3天增殖速度达到峰值,在随后的培养中细胞增殖呈衰减态。流式细胞仪及免疫荧光染色检测结果显示,外周血源组和脐血源组内皮祖细胞均可表达具有内皮祖细胞表型的CD133、CD34和血管内皮细胞因子受体2表面标志物,两组既摄取Dil标记乙酰化低密度脂蛋白,也能标记体外内皮祖细胞的标志物荆豆凝集素Ⅰ。结果证实,脐血来源的内皮祖细胞与外周血来源的内皮祖细胞生物学特性相近,脐血来源的内皮祖细胞增殖能力更强。  相似文献

Summary Experiments were performed on various species of monkeys. It was established there was a decreased number of erythrocytes, leucocytes (of the total number, as well as of their separate forms) and hemoglobin in the blood taken from the vein in comparison with the blood taken from capillaries of the ear lobe. The color index (which reflects the degree of saturation of a single erythrocyte by hemoglobin) and the ratio of the lymphocytes and neutrophils are the same in both specimens of the blood. The difference between the total number of leucocytes in the ear capillary and in the vein is greatest during the first few days when the animal is not yet used to the conditions in which the experiment is performed. With a decline of the reaction to the environmental conditions, the difference between the total number of leucocytes taken from the ear capillary and the vein becomes less pronounced due to the decrease of leucocytes in the blood taken from the ear. This demonstrates that uneven distribution of the cellular elements and hemoglobin in various vessels of the organism is the result of redistribution of the blood.Presented by Active Member Acad.
