Since the new century,stem cell research has become a hot field in medicine and life science.Scientists in Harvard Plans Stem Cell Center in 2004 have dreamed that one day they can lead stem cells to grow into tissues and even organs in the lab,which can be used to replace the damaged tissues or organs in severe injuries or applied in diseases at the end stage(incurable diseases)in order to prolong the life time and improve the life quality of patients.But this is a long way full of difficulties! Stem cells are defined by their capacity of self-renewal and multilineage differentiation, which make them uniquely situated to treat a broad spectrum of human diseases. Based on a series of remarkable studies in several fields of regenerative medicine, their application is not too far from the clinical practice. Full-thickness burns and severe traumas can injure skin and its appendages, which protect animals from water loss. temperature change, radiation, trauma and infection. In adults, the normal outcome of repair of massive full-thickness burns is fibrosis and scarring without any appendages, such as hair follicles, sweat and sebaceous glands. Perfect skin regeneration has been considered im-possible due to the limited regenerative capacity of epider- mal keratinocytes, which are generally thought to be the key source of the epidermis and skin appendages. Currently, researches on stem cells, such as epidermal stem cells, dermal stem cells, mesenchymal stem cells from bone marrow, and embryonic stem cells, bring promise to functional repair of skin after severe burn injury, namely, complete regeneration of skin and its appendages. Low back pain(LBP),as a leading cause of disability,is a common musculoskeletal disorder that results in major social and economic burdens.Recent research has identified inflammation and related signaling pathways as important factors in the onset and progression of disc degeneration,a significant contributor to LBP.Inflammatory mediators also play an indispensable role in discogenic LBP.The suppression of LBP is a primary goal of clinical practice but has not received enough attention in disc research studies.Here,an overview of the advances in inflammation-related pain in disc degeneration is provided,with a discussion on the role of inflammation in IVD degeneration and pain induction.Puncture models,mechanical models,and spontaneous models as the main animal models to study painful disc degeneration are discussed,and the underlying signaling pathways are summarized.Furthermore,potential drug candidates,either under laboratory investigation or undergoing clinical trials,to suppress discogenic LBP by eliminating inflammation are explored.We hope to attract more research interest to address inflammation and pain in IDD and contribute to promoting more translational research.  相似文献

Orthopaedics in China originated in traditional Chinese medicine and Western medicine. In the first half of twentieth century, Dr. Meng Chimao(Beijing), New Way Sung(Shanghai), Fang Hsienchi, and others contributed a great deal to the development of modern orthopaedics in China. The orthopaedic group was organized under the auspecies of Chinese Medical Association in 1937. The Chinese Medical Association Orthopaedic Society was established in 1980. The main achievements of Orthopaedics in China include replantation of severed limb or fingers, toe transplantation for thumb and finger reconstruction, nerve transfer for brachial plexus avulsion, research and manufacturing of domestic prostheses and the techniques of artifical joint replacement, surgical treatment of severe scoliosis and classification of idiopathic scoliosis, operative treatment of bone tumor, and basic research on giant cell tumor of bone.  相似文献

This paper aims to overcome slacklining's limited formulated explanatory models. Slacklining is an activity with increasing recreational use, but also has progressive adoption into prehabilitation and rehabilitation. Slacklining is achieved through self-learned strategies that optimize energy expenditure without conceding dynamic stability, during the neuromechanical action of balance retention on a tightened band. Evolved from rope-walking or ‘Funambulus', slacklining has an extensive history, yet limited and only recent published research, particularly for clinical interventions and in-depth hypothesized multi-dimensional models describing the neuromechanical control strategies. These ‘knowledge-gaps' can be overcome by providing an, explanatory model, that evolves and progresses existing standards, and explains the broader circumstances of slacklining's use. This model details the individual's capacity to employ control strategies that achieve stability, functional movement and progressive technical ability. The model considers contributing entities derived from: Self-learned control of movement patterns; subjected to classical mechanical forces governed by Newton's physical laws; influenced by biopsychosocial health factors; and within time's multi-faceted perspectives, including as a quantified unit and as a spatial and cortical experience. Consequently, specific patient and situational uses may be initiated within the framework of evidence based medicine that ensures a multi-tiered context of slacklining applications in movement, balance and stability. Further research is required to investigate and mathematically define this proposed model and potentially enable an improved understanding of human functional movement. This will include its application in other diverse constructed and mechanical applications in varied environments, automation levels, robotics, mechatronics and artificial-intelligence factors, including machine learning related to movement phenotypes and applications.  相似文献

Anew research study evaluated the mechanisms of prostate cancer (PCa)resistance to androgen deprivation therapy (ADT) and progression to castrate-resistant disease. Using a codinical approach, the investigators were able to identify key gen-etic determinants of ADT resistance, gain insight into the molecular pathways that play a key role in this transition and pro-pose a potential management strategy to overcome ADT resistance in select cancers. This represents a step forward towards per-sonalized medicine guided by specific molecular markers of prostate cancer enab-ling more effective therapies targeted towards altered metabolic pathways.  相似文献

Nutritional insufficiency during pregnancy has been shown to alter the metabolism of the offspring and can increase the risk of type 2 diabetes. The phenotype in the offspring involves changes to the morphology and functional capacity of the endocrine pancreas, and in the supporting islet microvasculature. Pancreatic β-cells possess a plastic potential and can partially recover from catastrophic loss. This is partly due to the existence of progenitors within the islets and the ability to generate new islets by neogenesis from the pancreatic ducts. This regenerative capacity is induced by bone marrow-derived stem cells, including endothelial cell progenitors and is associated with increased angiogenesis within the islets. Nutritional insults in early life, such as feeding a low protein diet to the mother, impair the regenerative capacity of the β-cells. The mechanisms underlying this include a reduced ability of β-cells to differentiate from the progenitor population, changes in the inductive signals from the microvasculature and an altered presence of endothelial progenitors. Statin treatment within animal models was associated with angiogenesis in the islet microvasculature, improved vascular function and an increase in β-cell mass. This demonstrates that reversal of the impaired β-cell phenotype observed following nutritional insult in early life is potentially possible.  相似文献
