目的对血清样品miRNA表达水平变化的实时定量PCR检测进行观察,同组织样品的miRNA的变化趋势进行比较,以评价血清miRNA在原发性肝癌早期诊断中的价值。方法通过荧光定量PCR检测miRNA-125a、-200a、-199a等3个有代表性的miRNA指标在正常和癌变样品中的变化,验证组织及血清两类样品中miRNA的变化趋势,分别取癌旁组织和癌组织以及正常人与癌症患者血清为材料。结果肝癌患者血清是正常对照血清中miRNA的含量的10%~15%,肝癌组织中的miRNA含量是癌旁组织的10%~20%,而且血清miRNA的变化趋势与组织miRNA较一致。结论血清miRNA可以反映原发性肝癌的发生,说明血清miRNA非常有可能成为一种新的生物标记物用于原发性肝癌的早期诊断。 相似文献

肝癌是世界范围内常见的恶性肿瘤。 miRNA是一类非编码调控RNA,通过转录后来调控靶基因表达,参与细胞发育、增殖、分化、凋亡等一系列重要生物学进程。近期的研究发现,miRNA具有癌基因和抑癌基因的作用,在肿瘤的发生和发展中起着重要的作用。已发现若干miRNA直接参与肝细胞癌的发生和发展, miRNA表达谱与肝细胞癌的诊断、分期、进展和预后等相关。本文就miRNA及其靶基因在肝癌发生发展中发挥的重要作用作一综述。 相似文献

利用放射免疫分析(RIA)方法,对72例健康人,48例原发性肝癌病人(HCC)及40例肝硬化病人血清,分别检测了甲胎蛋白分泌型免疫球蛋白A,β2微球蛋白,酸性铁蛋白,铜蓝蛋白和免疫球蛋白E等六种肿瘤标志物,结果表明,原发性肝癌病人,血清各种瘤标志物含量均显著增高(与对照组比较呈显著性差别,P〈0.01),部分项目与肝硬化一比较显著性差异,而它们单项目的阳性率分别为72.9%,75%,77.1%,8 相似文献

目的 通过检测血清肿瘤标志物PIVKA-Ⅱ、AFP和GGT的水平,探讨这3个检测项目在原发性肝癌中的诊断价值,进而为原发性肝癌的早期诊断提供有力的依据.方法 研究对象为已证实患原发性肝癌(PHC)的住院患者,对照为健康体检人员与患肝良性疾病患者.分别测定血清PIVKA-Ⅱ、AFP和GGT水平,绘制受试者工作特征曲线(receiver operating characteristic curve,ROC),明确最佳临界值(cut-off value),评价3项指标对原发性肝癌的诊断价值.结果 血清PIVKA-Ⅱ、AFP和GGT水平在原发性肝癌与肝胆良性疾病和健康体检者比较时,3项指标的差异均具有统计学意义(P<0.01).3项指标用于诊断原发性肝癌绘制的ROC曲线,AUCPIVKA-Ⅱ=0.882,AUCAFP=0.833,AUCGGT=0.703,可见,在诊断PHC方面,单项指标中PIVKA-Ⅱ具备最高的诊断价值,其次为AFP,诊断价值最低的为GGT.分析PIVKA-Ⅱ、AFP与GGT血清水平对PHC的诊断效率,灵敏度PIVKA-Ⅱ最高为73.0%(AFP:55.7%,GGT:47.8%),特异性AFP最高为97.9%(PIVKA-Ⅱ:96.5%,GGT:73.4%),阳性预测值AFP最高为95.5%(PIVKA-Ⅱ:94.4%,GGT:59.1%),阴性预测值PIVKA-Ⅱ最高为81.7%(AFP:73.3%,GGT:63.6%),准确性PIVKA-Ⅱ最高为86.0%(AFP:79.1%,GGT:62.0%).当3项指标联合检测时,特异性和阳性预测值略有下降,而灵敏度上升到80.9%(单项检测最高的PIVKA-Ⅱ为73.0%),阴性预测值上升到86.0%(单项检测最高的PIVKA-Ⅱ为81.7%),准确性上升到88.4%(单项检测最高的PIVKA-Ⅱ为86.0%).结论 单项指标中PIVKA-Ⅱ有较好的灵敏度和准确性,AFP具有较好的特异性,而AFP和GGT水平对于PIVKA-Ⅱ阴性的原发性肝癌的诊断具有较好的补充作用.因此,3项指标联合测定,能够使PHC的诊断准确性与灵敏度提升. 相似文献

目的:通过研究SMARCB1在肝细胞癌(hepatocellular carcinoma,HCC)组织的表达,阐明其对HCC的早期诊断及预后的作用。方法:在癌症基因组图集(The Cancer Genome Atlas,TCGA)数据库中筛选出SMARCB1基因,运用免疫组织化学(immunohistochemistry,IHC)技术和TCGA分析SMARCB1在HCC组织和正常组织的表达情况,阐述其在HCC发生、发展进程中的作用。结果:IHC结果证实,与正常肝组织相比,HCC中SMARCB1的蛋白表达量显著上升(P0.01)。IHC的结果显示SMARCB1的蛋白表达量与原发肿瘤分期呈正相关(P0.05),即SMARCB1表达量越高,原发肿瘤分期越趋向晚期。TCGA的结果显示SMARCB1的高表达是HCC的独立预后因素(P0.05)。结论:SMARCB1可能起着促癌基因的作用,临床上根据其在组织中的表达差异,可鉴别早期的HCC及良性组织,并可能有效地进行预后判断。 相似文献

Evaluation of PRL-3 expression, and its correlation with angiogenesis and invasion in hepatocellular carcinoma 总被引:2,自引:0,他引:2
Protein phosphatase of regenerating liver 3 (PRL-3) is a metastasis-associated phosphatase. Studies have shown that its overexpression increases cell motility and invasiveness. In this study, we aimed to investigate the expression of PRL-3 in hepatocellular carcinoma (HCC) tumor tissues and determine its correlations with matrix metalloproteinases (MMP-2, MMP-9) and E-cadherin in HCC. Paired cancerous and non-cancerous tissues were freshly collected from 42 primary HCC patients. PRL-3 expression at both mRNA and protein level was evaluated by real-time PCR, Western blot analysis and immunohistochemistry. The microvessel density (MVD) in HCC was detected with immunohistochemistry. The mRNA expression of MMP-2, MMP-9 and E-cadherin was analyzed by real-time PCR in search of correlations with PRL-3. We found that PRL-3 was significantly up-regulated in the HCC tumor tissues compared with corresponding noncancerous liver tissues (0.664+/-0.053 vs. 0.024+/-0.003, P<0.001). The mRNA level of PRL-3 in tissues was correlated with serum alpha-fetoprotein level, vascular invasion and metastasis (P<0.001). PRL-3 expression was closely related to MVD. Furthermore, we found a significant correlation between PRL-3 mRNA expression and MMP-2, MMP-9 and E-cadherin. Our results demonstrated that PRL-3 is up-regulated in HCC. It is strongly suggested that PRL-3 plays a key role in the angiogenesis and invasion of HCC. MMP-2, MMP-9 and E-cadherin might be involved in PRL-3 functions in HCC. 相似文献

The aim of this study was to investigate the relationship among MTA2, Ki-67 and HCC patient prognosis. Tissue microarray and immunohistochemistry were used to detect the expression of MTA2 and Ki-67 in HCC samples and corresponding adjacent samples. We found MTA2 and Ki-67 were both increased in HCC tissues than those in adjacent tissues and nuclear MTA2 was associated with Ki-67 (P = 0.019). Moreover, nuclear MTA2 was a risk factor of distant metastasis in patients with HCC andKi-67 showed a negative correlation with histological grade (P < 0.05, P < 0.01, respectively). Multivariate Cox model analysis revealed that Ki-67 expression was an independent prognosis factor in HCC patients (P = 0.020). These results indicated there might be a tight correlation among MTA2, Ki-67 and HCC prognosis. MTA2 combined with Ki-67 might be used to predict HCC patient prognosis. 相似文献

The aim of this study was to identify a microRNA (miRNA) expression signature for predicting HCC (hepatocellular carcinoma) survival. A total of 322 HCC patients in The Cancer Genome Atlas (TCGA) database were randomly divided into training and testing set. miRNAs, associated with survival time in the training set, were identified by using univariate Cox regression analysis. The risk score was formulated based on the expression levels of these miRNAs. Then the miRNA signature was validated in testing set through Kaplan–Meier analysis and log‐rank test. hsa‐miR‐301a, hsa‐miR‐132, hsa‐miR‐212, hsa‐miR‐489, and hsa‐miR‐1468 were identified to formulate risk score in training set and used to calculate the risk score of each patients in testing set. About 161 patients in testing set were segregated into high‐ and low‐risk group according to the median risk score. The survival time of high‐risk group was significantly shorter (p = 0.0248) than low‐risk group in testing test. The target genes of five miRNAs were significantly enriched in valine, leucine, and isoleucine degradation pathway and PPAR signaling pathway. hsa‐miR‐1468 had an up‐regulated tendency in HCC tissues compared to adjacent tumor tissues. The expression of hsa‐miR‐301a, hsa‐miR‐132, hsa‐miR‐212, hsa‐miR‐489, and hsa‐miR‐1468, which might be potential biomarkers to evaluate HCC patients' prognosis. 相似文献

背景:星形胶质细胞上调基因1是肿瘤发生转移早期的重要标记分子之一,很有可能成为肝癌临床治疗的分子靶标。
目的:探索原发性肝细胞癌患者星形胶质细胞上调基因1在其癌组织中的表达水平,并推测其在原发性肝细胞癌患者微创疗效中的地位。
方法:采用组织芯片方法和免疫组织化学染色方法检测120例原发性肝细胞癌患者肿瘤组织中星形胶质细胞上调基因1蛋白表达水平,并与其临床病理资料进行相关性分析。
结果与结论:原发性肝细胞癌患者星形胶质细胞上调基因1蛋白表达水平与其临床分期和组织分化程度密切相关,且其星形胶质细胞上调基因1蛋白高表达水平与Ki-67阳性率和肿瘤微血管密度有关,但该蛋白高表达与患者的性别及年龄无相关性。说明星形胶质细胞上调基因1蛋白表达水平与常规恶性检测指标相关,检测该蛋白表达水平对于判断原发性肝细胞癌具有重要意义。 相似文献
