Mode of action and aquatic exposure thresholds of no concern   总被引:1,自引:0,他引:1  
Threshold concepts of toxicological concern are based on the possibility of establishing an exposure threshold value for chemicals below which no significant risk is to be expected. The objective of the present study is to address environmental thresholds of no toxicological concern for freshwater systems (ETNCaq) for organic chemicals. We analyzed environmental toxicological databases (acute and chronic endpoints) and substance hazard assessments. Lowest numbers and 95th-percentile values were derived using data stratification based on mode of action (MOA; 1 = inert chemicals; 2 = less inert chemicals; 3 = reactive chemicals; 4 = specifically acting chemicals). The ETNCaq values were derived by multiplying the lowest 95th percentile values with appropriate application factors; ETNCaq,MOA1-3 is approximately 0.1 microg/L. A preliminary analysis with complete MOA stratification of the databases shows that in the case of MOA1 or MOA2, the ETNCaq value could be even higher than 0.1 microg/L. A significantly lower ETNCaq,MOA4 value was observed based on the long-term toxicity information in the European Centre for the Ecotoxicology and Toxicology of Chemicals database. Application of the ETNCaq value in a tiered risk-assessment scheme may help chemical producers to set data-generation priorities and to refine or reduce animal use. It also may help to inform downstream users concerning the relative risk associated with their specific uses and be of value in putting environmental monitoring data into a risk-assessment perspective.  相似文献

Although a standard genome‐wide significance level has been accepted for the testing of association between common genetic variants and disease, the era of whole‐genome sequencing (WGS) requires a new threshold. The allele frequency spectrum of sequence‐identified variants is very different from common variants, and the identified rare genetic variation is usually jointly analyzed in a series of genomic windows or regions. In nearby or overlapping windows, these test statistics will be correlated, and the degree of correlation is likely to depend on the choice of window size, overlap, and the test statistic. Furthermore, multiple analyses may be performed using different windows or test statistics. Here we propose an empirical approach for estimating genome‐wide significance thresholds for data arising from WGS studies, and we demonstrate that the empirical threshold can be efficiently estimated by extrapolating from calculations performed on a small genomic region. Because analysis of WGS may need to be repeated with different choices of test statistics or windows, this prediction approach makes it computationally feasible to estimate genome‐wide significance thresholds for different analysis choices. Based on UK10K whole‐genome sequence data, we derive genome‐wide significance thresholds ranging between 2.5 × 10?8 and 8 × 10?8 for our analytic choices in window‐based testing, and thresholds of 0.6 × 10?8–1.5 × 10?8 for a combined analytic strategy of testing common variants using single‐SNP tests together with rare variants analyzed with our sliding‐window test strategy.  相似文献

Distinguishing true from false positive findings is a major challenge in human genetic epidemiology. Several strategies have been devised to facilitate this, including the positive predictive value (PPV) and a set of epidemiological criteria, known as the “Venice” criteria. The PPV measures the probability of a true association, given a statistically significant finding, while the Venice criteria grade the credibility based on the amount of evidence, consistency of replication and protection from bias. A vast majority of journals use significance thresholds to identify the true positive findings. We studied the effect of p value thresholds on the PPV and used the PPV and Venice criteria to define usable thresholds of statistical significance. Theoretical and empirical analyses of data published on AlzGene show that at a nominal p value threshold of 0.05 most “positive” findings will turn out to be false if the prior probability of association is below 0.10 even if the statistical power of the study is higher than 0.80. However, in underpowered studies (0.25) with a low prior probability of 1 × 10?3, a p value of 1 × 10?5 yields a high PPV (>96 %). Here we have shown that the p value threshold of 1 × 10?5 gives a very strong evidence of association in almost all studies. However, in the case of a very high prior probability of association (0.50) a p value threshold of 0.05 may be sufficient, while for studies with very low prior probability of association (1 × 10?4; genome-wide association studies for instance) 1 × 10?7 may serve as a useful threshold to declare significance.  相似文献

It has long been known that relatively high-dose ionising radiation exposure (>?1 Gy) can induce cataract, but there has been no evidence that this occurs at low doses (<?100 mGy). To assess low-dose risk, participants from the US Radiologic Technologists Study, a large, prospective cohort, were followed from date of mailed questionnaire survey completed during 1994–1998 to the earliest of self-reported diagnosis of cataract/cataract surgery, cancer other than non-melanoma skin, or date of last survey (up to end 2014). Cox proportional hazards models with age as timescale were used, adjusted for a priori selected cataract risk factors (diabetes, body mass index, smoking history, race, sex, birth year, cumulative UVB radiant exposure). 12,336 out of 67,246 eligible technologists reported a history of diagnosis of cataract during 832,479 person years of follow-up, and 5509 from 67,709 eligible technologists reported undergoing cataract surgery with 888,420 person years of follow-up. The mean cumulative estimated 5-year lagged eye-lens absorbed dose from occupational radiation exposures was 55.7 mGy (interquartile range 23.6–69.0 mGy). Five-year lagged occupational radiation exposure was strongly associated with self-reported cataract, with an excess hazard ratio/mGy of 0.69?×?10?3 (95% CI 0.27 ×?10?3 to 1.16 ×?10?3, p?<?0.001). Cataract risk remained statistically significant (p?=?0.030) when analysis was restricted to?<?100 mGy cumulative occupational radiation exposure to the eye lens. A non-significantly increased excess hazard ratio/mGy of 0.34?×?10?3 (95% CI ??0.19?×?10?3 to 0.97?×?10?3, p?=?0.221) was observed for cataract surgery. Our results suggest that there is excess risk for cataract associated with radiation exposure from low-dose and low dose-rate occupational exposures.  相似文献

Estimated 8-hr LC50 values of zinc and cadmium were less than 1 ppm in the absence of soluble calcium and magnesium, but were raised, for example, to 24 ppm of zinc and 19 ppm of cadmium by the addition of calcium plus magnesium at a concentration (500 ppm) equivalent to that of very hard water. Experiments with 65Zn showed that uptake of this element from sublethal concentrations over 8 hr was reduced from approximately 50 × 10?2 μg liter?1 to 10 × 10?2 μg liter?1 per 106 cells by the addition of 500 ppm of calcium and magnesium. Diminished uptake may partly explain the antagonism observed in the toxicity tests.  相似文献

To date, pesticides, especially organophosphate pesticide such as chlorpyrifos, have been frequently applied to paddy fields over time to maintain product quality, protect agricultural crops from various pests, and increase yield. This study evaluates dermal exposure to chlorpyrifos in rice farmers along with providing a health risk assessment. Thirty-five rice farmers participated and completed an in-person interview, and patch technique was used to evaluate dermal exposure to chlorpyrifos. The chlorpyrifos residue was extracted from the gauze patches and quantified by gas chromatography equipped with flame photometric detector (GC-FPD). The results showed that chlorpyrifos concentrations were greater in males (526.34 ± 478.84 mg/kg) than females (500.75 ± 595.15 mg/kg). Average daily dose sampled from seven points on male and female farmers were 31.72 × 10?4, 193.32 × 10?4, 5.38 × 10?4, 190.48 × 10?4, 170.47 × 10?4, 465.91 × 10?4, and 43.04 × 10?4 mg/kg-day. The hazard quotient (HQ) at the mean and 95th percentile level was found to be greater than acceptable (HQ > 1). Rice-growing farmers in this area may be at risk for adverse health effects due to continuous dermal exposure to chlorpyrifos from their improper use of personal protective equipment (PPE).  相似文献

The rationale for adjusting occupational exposure limits for unusual work schedules is to assure, as much as possible, that persons on these schedules are placed at no greater risk of injury or discomfort than persons who work a standard 8 hr/day, 40 hr/week. For most systemic toxicants, the risk index upon which the adjustments are made will be either peak blood concentration or integrated tissue dose, depending on what chemical's presumed mechanism of toxicity. Over the past ten years, at least four different models have been proposed for adjusting exposure limits for unusually short and long work schedules. This paper advocates use of a physiologically-based pharmacokinetic (PB-PK) model for determining adjustment factors for unusual exposure schedules, an approach that should be more accurate than those proposed previously. The PB-PK model requires data on the blood:air and tissue:blood partition coefficients, the rate of metabolism of the chemical, organ volumes, organ blood flows and ventilation rates in humans. Laboratory data on two industrially important chemicals--styrene and methylene chloride--were used to illustrate the PB-PK approach. At inhaled concentrations near their respective 8-hr Threshold Limit Value-Time-weighted averages (TLV-TWAs), both of these chemicals are primarily eliminated from the body by metabolism. For these two chemicals, the appropriate risk indexing parameters are integrated tissue dose or total amount of parent chemical metabolized. Since methylene chloride is metabolized to carbon monoxide, the maximum blood carboxyhemoglobin concentrations also might be useful as an index of risk for this chemical.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

There is accumulating epidemiological evidence that exposure to some solvents, metals, asphyxiants and other substances in humans is associated with an increased risk of acquiring hearing loss. Furthermore, simultaneous and successive exposure to certain chemicals along with noise can increase the susceptibility to noise-induced hearing loss. There are no regulations that require hearing monitoring of workers who are employed at locations in which occupational exposure to potentially ototoxic chemicals occurs in the absence of noise exposure. This project was undertaken to develop a toxicological database allowing the identification of possible ototoxic substances present in the work environment alone or in combination with noise exposure. Critical toxicological data were compiled for chemical substances included in the Quebec occupational health regulation. The data were evaluated only for noise exposure levels that can be encountered in the workplace and for realistic exposure concentrations up to the short-term exposure limit or ceiling value (CV) or 5 times the 8-h time-weighted average occupational exposure limit (TWA OEL) for human data and up to 100 times the 8-h TWA OEL or CV for animal studies. In total, 224 studies (in 150 articles of which 44 evaluated the combined exposure to noise and a chemical) covering 29 substances were evaluated using a weight of evidence approach. For the majority of cases where potential ototoxicity was previously proposed, there is a paucity of toxicological data in the primary literature. Human and animal studies indicate that lead, styrene, toluene and trichloroethylene are ototoxic and ethyl benzene, n-hexane and p-xylene are possibly ototoxic at concentrations that are relevant to the occupational setting. Carbon monoxide appears to exacerbate noise-induced hearing dysfunction. Toluene interacts with noise to induce more severe hearing losses than the noise alone.  相似文献

Background

It can be difficult for occupational health professionals to assess which toxicological databases available on the Internet are the most useful for answering their questions. Therefore we evaluated toxicological databases for their ability to answer practical questions about exposure and prevention. We also propose recommended practices for searching for toxicological properties of chemicals.

Methods

We used a systematic search to find databases available on the Internet. Our criteria for the databases were the following: has a search engine, includes factual information on toxic and hazardous chemicals harmful for human health, and is free of charge. We developed both a qualitative and a quantitative rating method, which was used by four independent assessors to determine appropriateness, the quality of content, and ease of use of the database. Final ratings were based on a consensus of at least two evaluators.

Results

Out of 822 results we found 21 databases that met our inclusion criteria. Out of these 21 databases 14 are administered in the US, five in Europe, one in Australia, and one in Canada. Nine are administered by a governmental organization. No database achieved the maximum score of 27. The databases GESTIS, ESIS, Hazardous Substances Data Bank, TOXNET and NIOSH Pocket Guide to Chemical Hazards all scored more than 20 points. The following approach was developed for occupational health professionals searching for the toxicological properties of chemicals: start with the identity of the chemical; then search for health hazards, exposure route and measurement; next the limit values; and finally look for the preventive measures.

Conclusion

A rating system of toxicological databases to assess their value for occupational health professionals discriminated well between databases in terms of their appropriateness, quality of information, and ease of use. Several American and European databases yielded high scores and provide a valuable source for occupational health professionals.  相似文献

Abstract

To investigate the contamination levels of respirable dust released in the work environment and the induced workers’ health risk at a coal-fired power plant, we collected 405 dust samples from different dusty workstations by personal sampling during the coal-fired power generation process. Then, an inhalation risk assessment model from the USEPA was combined with the Monte Carlo simulation method to quantitatively evaluate the health risk caused by dust inhalation. Of 10 workstations researched, the dust concentration in the most workstations exceeded the prescribed occupational exposure limit. Workers engaged in ash removal suffered the highest health risk at 4.08?×?10?6 ± 2.85?×?10?6 (95% CI), closely followed by those involved in other job categories. The results can contribute to the formulation of targeted dust prevention measures and implementation of risk management for the coal-fired power sector.  相似文献

One contaminant produced unintentionally during the manufacture of chlorophenols and phenoxy herbicides is 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD). The resulting TCDD-containing wastes have been detected at many hazardous waste sites which in recent years have been in the process of remediation. Concerns about worker exposure to TCDD-contaminated soil (dust) during remediation of hazardous waste sites have produced a need for an occupational exposure limit (OEL) for TCDD. The animal toxicology data and human experience with TCDD are reviewed, and an occupational exposure limit for TCDD is proposed. The animal data support risk estimations which are based on TCDD as a nongenotoxic carcinogen. Studies on human populations have failed to demonstrate clearly any significant long-term health effects at levels to which humans have been exposed. The data indicate that an 8-hr time-weighted average limit of 2 ng/m3 is appropriate, and the associated risk would be consistent with other carcinogens at their corresponding OELs. A preliminary OEL of 0.2 ng/m3 (200 pg/m3) is recommended, however, in light of other sources of exposure because of TCDD's ubiquitousness in the environment, its unclear mechanism of action, and its rather long biological half-life in humans. This limit provides an ample margin of safety to prevent chloracne following repeated, acute exposure, and it addresses those chronic effects of TCDD observed in animal studies as well as those observed after accidental human exposure. The resulting body burden caused by chronic exposure to TCDD at the proposed OEL is examined. Information concerning the fundamental mechanisms of action of both natural and environmental hormones, combined with information concerning endogenous hormone concentrations, reveals how endocrine-disrupting chemicals with estrogenic activity (EEDCs) can be active at concentrations far below those currently being tested in toxicological studies. Using only very high doses in toxicological studies of EEDCs thus can dramatically underestimate bioactivity. Specifically: a) The hormonal action mechanisms and the physiology of delivery of EEDCs predict with accuracy the low-dose ranges of biological activity, which have been missed by traditional toxicological testing. b) Toxicology assumes that it is valid to extrapolate linearly from high doses over a very wide dose range to predict responses at doses within the physiological range of receptor occupancy for an EEDC; however, because receptor-mediated responses saturate, this assumption is invalid. c) Furthermore, receptor-mediated responses can first increase and then decrease as dose increases, contradicting the assumption that dose-response relationships are monotonic. d) Exogenous estrogens modulate a system that is physiologically active and thus is already above threshold, contradicting the traditional toxicological assumption of thresholds for endocrine responses to EEDCs. These four fundamental issues are problematic for risk assessment methods used by regulatory agencies, because they challenge the traditional use of extrapolation from high-dose testing to predict responses at the much lower environmentally relevant doses. These doses are within the range of current exposures to numerous chemicals in wildlife and humans. These problems are exacerbated by the fact that the type of positive and negative controls appropriate to the study of endocrine responses are not part of traditional toxicological testing and are frequently omitted, or when present, have been misinterpreted.  相似文献

In response to the intention of the Workers' Compensation Board of British Columbia (WCB of BC) to eliminate made-in-BC occupational exposure limits (OELs) and adopt threshold limit values (TLVs), this study assessed the potential health impacts on healthcare workers (HCWs) of the proposed change, by (1) reviewing the processes used to establish the OELs and TLVs, (2) selecting of substances of health concern for HCWs, (3) identifying chemicals with discordances between existing OELs and the 2002 TLVs, and 4) reviewing the discordances and assessing the potential health implications. Differences in philosophies, policies and processes that influenced the setting of OELs and TLVs were substantial. The TLV process involves U.S. and international priorities; in BC, a tripartite committee determined OELs taking into consideration how OELs should be interpreted in the local context. 47 chemicals of concern to BC HCWs were discordant, with significant discordances totalling 57; 15 compounds had BC 8-hour OELs lower than their respective TLVs and three TLVs were lower than the 8-hour BC OELs. Review of six chemicals with discordances suggested a potential for increased risks of adverse health effects. Eliminating the local capacity and authority to set OELs is unlikely to cause major health problems in the short run, but as chemicals in use locally may not have up-to-date TLVs, eliminating the capacity for local considerations should be undertaken with great caution. While the WCB of BC did implement the change, the present report resulted in procedural changes that will provide better protection for the workforce.  相似文献

Background: Large numbers of chemicals require evaluation to determine if their production and use pose potential risks to ecological and human health. For most chemicals, the inadequacy and uncertainty of chemical-specific data severely limit the application of exposure- and risk-based methods for screening-level assessments, priority setting, and effective management.Objective: We developed and evaluated a holistic, mechanistic modeling framework for ecological and human health assessments to support the safe and sustainable production, use, and disposal of organic chemicals.Methods: We consolidated various models for simulating the PROduction-To-EXposure (PROTEX) continuum with empirical data sets and models for predicting chemical property and use function information to enable high-throughput (HT) exposure and risk estimation. The new PROTEX-HT framework calculates exposure and risk by integrating mechanistic computational modules describing chemical behavior and fate in the socioeconomic system (i.e., life cycle emissions), natural and indoor environments, various ecological receptors, and humans. PROTEX-HT requires only molecular structure and chemical tonnage (i.e., annual production or consumption volume) as input information. We evaluated the PROTEX-HT framework using 95 organic chemicals commercialized in the United States and demonstrated its application in various exposure and risk assessment contexts.Results: Seventy-nine percent and 97% of the PROTEX-HT human exposure predictions were within one and two orders of magnitude, respectively, of independent human exposure estimates inferred from biomonitoring data. PROTEX-HT supported screening and ranking chemicals based on various exposure and risk metrics, setting chemical-specific maximum allowable tonnage based on user-defined toxicological thresholds, and identifying the most relevant emission sources, environmental media, and exposure routes of concern in the PROTEX continuum. The case study shows that high chemical tonnage did not necessarily result in high exposure or health risks.Conclusion: Requiring only two chemical-specific pieces of information, PROTEX-HT enables efficient screening-level evaluations of existing and premanufacture chemicals in various exposure- and risk-based contexts. https://doi.org/10.1289/EHP9372  相似文献

This study aimed to assess workers’ exposure to benzene, toluene, ethylbenzene, and xylene (BTEX) compounds in refueling stations of Ardabil city (Iran). Twenty-four refueling stations including 15 petrol and 9 compressed natural gas (CNG) stations from different regions were selected and monitored for ambient BTEX concentrations. Air samples were taken based on NIOSH Manual of Analytical Method no 1501. Target compounds were extracted using CS2 and analyzed by GC equipped with FID. Average concentrations of benzene, toluene, ethylbenzene, and xylene were obtained 2.01, 1.80, 2.72, and 1.65 mg/m3, respectively. Benzene concentrations exceeded the occupational exposure limit set by the Iran Ministry of Health and Medical Education. Its concentrations were significantly higher in commercial areas (2.72 mg/m3) compared to suburban areas (1.89 mg/m3). BTEX concentrations in gasoline stations were slightly, but not significantly, higher than those in CNG stations. Long-term exposure cancer risk of 1884?×?10?6?±?390?×?10?6 and hazard index of 22.83?±?3.66 were estimated for benzene and BTEX compounds, respectively. The results declare the necessity for controlling BTEX emission (mainly benzene) and monitoring employee’s exposure in refueling stations.  相似文献

Using the transmission electron microscope, asbestos fibers have been assessed in lymph fluid collected from the thoracic lymph duct in five groups of rats previously exposed to asbestos fibers (by ingestion). Ten rats were gavaged a single dose weighing approximately 20 mg. Five were given pure UICC chrysotile A while another group of five had pure UICC crocidolite. All the rats of the chrysotile group were positive animals with recovery rate values ranging from 6.9 × 10?7 to 3 × 10?5 (90% of the fibers being recovered during the first 16 hr following the gavage). The crocidolite group had only three positive animals and lower recovery rate values of 5.7 × 10?8 to 5.6 × 10?7. A third group was fed a synthetic diet containing 1%, by weight, chrysotile with a majority of short fibers (90% below 4 μm). Of the 15 rats comprising this group, 13 were positive with maximum daily recovery rates ranging from 2.1 × 10?7 to 2.1 × 10?6. A group of eight rats fed the same kind of diet but containing a higher proportion of long fibers, showed only four positive animals, however, they had higher daily recovery rates ranging from 1.9 × 10?5 to 2.1 × 10?4. No fibers were encountered in the samples of the two control rats. This study demonstrates the passage of chrysotile and crocidolite fibers across the gastrointestinal wall, with the passage rate being higher for long fibers than short ones.  相似文献

目的研究21名打磨工9个月内纯音听阈测试结果的变化,为指导企业做好噪声防护提供依据。方法根据2年的现场检测和职业性健康体检的资料,对噪声检测值和纯音听阈测试结果进行统计分析。结果9个打磨岗位的噪声值有8个超过职业接触限值,21名打磨工听阈平均修正值曲线符合职业性噪声聋的特点。双耳语频段的听阈平均修正值均小于高频段,2008年差别均有显著性(P〈0.05),2009年两者之间的差别均无显著性(P〉0.50),9个月以后双耳12个频率的听阈平均修正值中有8个的差别有显著性(P〈0.05)或高度显著性(P≤0.001),其中有5个是语频段的,说明噪声对语频段的影响已超过高频段。结论噪声对21名打磨工语频段的影响已超过了高频段,企业应在做好打磨工的个人防护的同时,减少他们接触噪声的时间,否则将会出现职业性噪声聋。  相似文献

In this work, statistics and geospatial analysis are used to investigate the hypothesis that either bauxitic soils or underlying geology increases the carcinogenic health risks to the population due to exposure from naturally occurring radioactive materials (NORM) in the soil environment over which they reside. The rationale for this study is the large distribution of bauxitic soils in Jamaica and a review of the last published 5-year cancer survey study of Jamaica. A negative health environment was defined as one where the excess lifetime cancer risk (ELCR) value exceeded the world mean value of 2.9?×?10?4 and the annual gonadal dose equivalent (AGDE) was a multiple of the acceptable level of 300 μSv/year. The excess lifetime cancer risk index in Jamaica ranges from 0.16?×?10?5 to 7.92?×?10?4 with a mean value of 1.63?×?10 ?4. The mean value of the ELCR index in Jamaica was statistically significantly lower than the world mean value. The main areas of concern were the sections of the parishes of Manchester and St. Elizabeth which exhibited value of up to five (5) times the acceptable level of the ELCR index. The AGDE index showed a moderate correlation of 0.511 with a reclassified geology group (limestone and non-limestone) introduced in this study. The soils overlaying limestone formations in Jamaica were characterized by a statistically significantly higher dose rate exposure to the gonads of the population. Geological explanations for the levels of the primordial radionuclides are also discussed in this paper. The results suggested that limestone geology in Jamaica had a significantly higher mean value for both indices (ELCR and AGDE) when compared to other geologies. Since bauxitic soils predominantly overlay these rocks in Jamaica, the research hypothesis regarding the impact bauxitic soils is supported by the results of this study. The impact of geology was not significant, and probable causes are discussed.  相似文献
