比较了本院基因医学研究所和北京医科大学提供的两株7TD1细胞。选择北京医科大学提供的细胞株进行克隆,筛选出对IL-6增殖反应性和依赖性较强的3株细胞(7TD1MC1、7TD1MC4、7TD1MC5)。其中7TD1MC5细胞的反应曲线呈典型的“倒S”形,对IL-6的反应性和依赖性很好。MTT比色法显示了与T3H-TdR掺入法一致的结果,采用了7TD1MC5细胞及MTT比色法测定IL-6生物活性,可减  相似文献

作者报道了国内重组人白介素-6(rhIL-6)对6.5Gy照射Cs7BL/6J小鼠后10天造血功能(主要是造血干细胞、粒系和红系祖细胞)恢复的影响.结果表明;rhIL-6对照后早期造血的影响与白细胞介素-6(IL-6)剂量有关,且作用是双向的.当IL-6剂量在200~1000μg·kg-1·d-1时,脾重和内源性肿结节(CFU-S)数随IL-6剂量增大而逐渐增加,与对照组相比,CFU-S数的增加统计学意义更明显.与此同时骨髓有核细胞数和CFU-GM产率以及CFU-GM/股骨也明显增加,但1000μg·kg-1·d-1组的效果反而下降,因此,rhIL-6促进照后早期造血恢复的最佳剂量可能在500μg·kg-1·d-1左右.当IL-6剂量为10μg·kg-1·d-1时,对照射小鼠造血恢复起抑制作用。这些发现对rhIL-6的应用是很有实际意义的.  相似文献

目的探讨头低位(-6°)卧床对人淋巴细胞增殖能力和某些细胞因子产生的影响。方法6名健康男性青年头低位(-6°)卧床6d,卧床前1d、3d、6d各采血一次,测定免疫指标。结果卧床3d,α干扰素(interferonα,IFNα)产生明显降低(P<0.05),NK细胞(naturalkilercel,NK)杀伤活性明显增强(P<0.05),γ干扰素(interferonγ,IFNγ)的产生和白细胞介素2受体(interleukin2receptor,IL2R,又称CD25)表达功能均呈降低趋势。卧床6d,IFNα产生和NK杀伤活性均恢复至卧床前的水平,IFNγ产生和CD25则明显降低(P<0.05)。随着卧床时间的延长,T淋巴细胞增殖能力和IL2的产生则逐渐降低,而IL6的产生则逐渐增加。结论头低位(-6°)卧床对细胞免疫功能有一定的影响。  相似文献

报道了应用白介素6(IL-6)依赖细胞株7TD1,以及MTT比色法测定IL-6生物活性的简便方法。本法测定结果和 ̄3H-TdR掺入法一致;检测灵敏度为1pg或1U/ml以下;可用于正常人或病人血清中IL-6水平的测定。采用本法检测了我国正常成人55例,血清IL-6活性为6.75±2.98U/ml;而11例肺结核排菌者和12例肺癌患者的血清IL-6活性分别为14.29±8.46和17.14±9.12U/ml,与正常人有显著差别(P<0.01和P<0.001),为进一步临床应用奠定了基础。  相似文献

目的探讨流式细胞仪在造血干细胞基因转移效率测定中的应用价值。方法以干细胞因子(SCF)、白细胞介素(IL)3、IL6单独或联合作用(SCF/IL3、SCF/IL6、IL3/IL6、SCF/IL3/IL6)后的Balb/c小鼠造血干细胞作为靶细胞,应用含有绿色荧光蛋白(GFP)基因的逆转录病毒载体GCGPXSN介导的基因转移技术实施基因转移,采用FACS型流式细胞仪测定基因转移效率。结果病毒上清转染的阳性对照组基因转移率为0.06%,以不同细胞因子作用后基因转移效率提高至007%~0.20%,细胞因子作用前后比较,除SCF/IL6组外,差异均有显著或非常显著意义(P<005或P<001)。其中以SCF/IL3组与SCF/IL3/IL6组的提高最为明显,均达020%,与其他各细胞因子组比较,差异也有非常显著意义(P<001)。结论应用流式细胞仪可测定出SCF、IL3、IL6单独或联合作用对小鼠造血干细胞基因转移效率的不同影响,具有快速、方便、准确的优点。  相似文献

目的观察外源性重组IL-1受体拮抗剂(IL-1ra)对实验性脑创伤的治疗作用,以及对脑组织中NTFα、IL-6两种炎性细胞因子水平的影响。方法采用液压型脑创伤装置造成大鼠右顶部脑挫伤。治疗组动物分别于脑创伤后15分钟,2,4,6,8,24,48小时左侧侧脑室内注射10μg的IL-1ra,总剂量为每只70μg。动物于创伤后3天处死,脑组织连续切片染色,计算出创伤体积,通过比较动物创伤灶体积大小观察IL-1ra的疗效。结果IL-1ra治疗组创伤体积为(21.6±5.9)mm3,对照组为(29.4±5.7)mm3,IL-1ra治疗组脑创伤灶体积明显小于对照组(P<0.05)。另外,创伤后8小时观察,IL-1ra治疗组创伤侧皮层IL-6的水平为(243.1±37.8)U/mg,对照组为(298.8±49.2)U/mg,IL-1ra治疗组动物脑组织内IL-6的水平低于对照组(P<0.05)。结论脑室内注射外源性IL-1ra可有效减轻大鼠脑创伤的损伤程度和范围,IL-1ra除能拮抗IL-1的直接作用外,可能还通过影响其他炎性因子(如IL-6)的生成间接抑制炎性反应,从而对缓解脑创伤后继发炎性损害有十分重要的作用  相似文献

本应用碱性磷酸酶抗碱性磷酸酶(APAAP)法检测了9例多发性骨髓瘤(MM)患和11例健康人外周血IL-4产生细胞。结果表明:MM患外周血IL-4产生细胞(7.6%±2.1%)显少于正常人(15.36%±4.1%),(P<0.001),并与患外周血免疫球蛋白水平的减少呈显相关。提示IL-4产生细胞减少与MM患免疫球蛋白的分泌抑制有关。  相似文献

The Knee injury and Osteoarthritis Outcome Score (KOOS) is a self-administered instrument measuring outcome after knee injury at impairment, disability, and handicap level in five subscales. Reliability, validity, and responsiveness of a Swedish version was assessed in 142 patients who underwent arthroscopy because of injury to the menisci, anterior cruciate ligament, or cartilage of the knee. The clinimetric properties were found to be good and comparable to the American version of the KOOS. Comparison to the Short Form-36 and the Lysholm knee scoring scale revealed expected correlations and construct validity. Item by item, symptoms and functional limitations were compared between diagnostic groups. High responsiveness was found three months after arthroscopic partial meniscectomy for all subscales but Activities of Daily Living.  相似文献

Acute limping may be the result of multiple pathologies in children. The differential diagnosis varies based on the age of the child. Irrespective of age, the initial imaging work-up includes AP and frog leg radiographs of the pelvis and ultrasound; MRI may sometimes be helpful. In children less than 3 years, infections and trauma are most frequent. MRI is the imaging modality of choice when osteomyelitis is clinically suspected. Between the ages of 3 and 10 years, transient synovitis of the hip and Legg-Calvé-Perthes disease are main considerations but infection, inflammation and focal bony lesions are also considered. Objective To investigate endovascular treatment of traumatic direct carotid-cavernous fistulas (CCF) and their complications such as pseudoaneurysms. Methods: Over a five-year period, 22 patients with traumatic direct CCFs were treated endovascularly in our institution. Thirteen patients were treated once with the result of CCF occluded, 8 twice and 1 three times. Treatment modalities included balloon occlusion of the CCF, sacrifice of the ipsilateral internal carotid artery with detachable balloon, coll embolization of the cavernous sinus and secondary pseudoaneurysms, and covered-stem management of the pseudoaneurysms. Results All the direct CCFs were successfully managed endovascularly. Four patients developed a pseudoaneurysm after the occlusion of the CCF with an incidence of pseudoaneurysm formation of 18.2% (4/22). A total number of 8 patients experienced permanent occlusion of the ICA with a rate of ICA occlusion reaching 36.4% (8/22). Followed up through telephone consultation from 6 months to 5 years, all did well with no recurrence of CCF symptoms and signs. Conclusion Traumatic direct CCFs can be successfully managed with endovascular means. The pseudoaneurysms secondary to the occlusion of the CCFs can be occluded with stent-assisted coiling and implantation of covered stents.  相似文献

In response to the ENFSI and EDNAP groups’ call for new STR multiplexes for Europe, Promega® developed a suite of four new DNA profiling kits. This paper describes the developmental validation study performed on the PowerPlex® ESI 16 (European Standard Investigator 16) and the PowerPlex® ESI 17 Systems. The PowerPlex® ESI 16 System combines the 11 loci compatible with the UK National DNA Database®, contained within the AmpFlSTR® SGM Plus® PCR Amplification Kit, with five additional loci: D2S441, D10S1248, D22S1045, D1S1656 and D12S391. The multiplex was designed to reduce the amplicon size of the loci found in the AmpFlSTR® SGM Plus® kit. This design facilitates increased robustness and amplification success for the loci used in the national DNA databases created in many countries, when analyzing degraded DNA samples. The PowerPlex® ESI 17 System amplifies the same loci as the PowerPlex® ESI 16 System, but with the addition of a primer pair for the SE33 locus. Tests were designed to address the developmental validation guidelines issued by the Scientific Working Group on DNA Analysis Methods (SWGDAM), and those of the DNA Advisory Board (DAB). Samples processed include DNA mixtures, PCR reactions spiked with inhibitors, a sensitivity series, and 306 United Kingdom donor samples to determine concordance with data generated with the AmpFlSTR® SGM Plus® kit. Allele frequencies from 242 white Caucasian samples collected in the United Kingdom are also presented. The PowerPlex® ESI 16 and ESI 17 Systems are robust and sensitive tools, suitable for the analysis of forensic DNA samples. Full profiles were routinely observed with 62.5 pg of a fully heterozygous single source DNA template. This high level of sensitivity was found to impact on mixture analyses, where 54–86% of unique minor contributor alleles were routinely observed in a 1:19 mixture ratio. Improved sensitivity combined with the robustness afforded by smaller amplicons has substantially improved the quantity of data obtained from degraded samples, and the improved chemistry confers exceptional tolerance to high levels of laboratory prepared inhibitors.  相似文献

The purpose of this study was twofold: (a) to investigate the prevalence of hip and groin pain in sub‐elite male adult football in Denmark and (b) to explore the association between prevalence and duration of hip and groin pain in the previous season with the Copenhagen Hip and Groin Outcome Score (HAGOS) in the beginning of the new season. In total 695 respondents from 40 teams (Division 1–4) were included. Players completed in the beginning of the new season (July–Sept 2011) a self‐reported paper questionnaire on hip and/or groin pain during the previous season and HAGOS. In total 49% (95% CI: 45–52%) reported hip and/or groin pain during the previous season. Of these, 31% (95% CI: 26–36%) reported pain for >6 weeks. Players with the longest duration of pain during the previous season had the lowest HAGOS scores, when assessed at the beginning of the new season, P < 0.001. This study documents that half of sub‐elite male adult football players report pain in the hip and/or groin during a football season. The football players with the longest duration of pain in previous season displayed the lowest HAGOS scores in the beginning of the new season.  相似文献

Objective To evaluate the preliminaily clinical efficacy and retrievability of a retrievable hinged covered metallic stent in the treatment of the bronchial stump fistula (BSF). Methods Between April 2003 and March 2005, 8 patients with bronchial stump fistula after pneumonectomy or lobectomy were treated with two types (A and B) of retrievable hinged covered metallic stents. Type A stent was placed in 6 patients and type B in 2 under fluoroscopic guidance. The stent was removed with a retrieval set when BSF was healed or complications occurred. Results Stent placement in the bronchial tree was technically successful in all patients, without procedure-related complications. Immediate closure of the BSF was achieved in all patients after the procedure. Stents were removed from all patients but one. Removal of the stents was difficult in two patients due to tissue hyperplasia. Patients were followed up for 6 - 21 months. Placement of the stents remained stable in all patients except one due to severe cough. Permanent closure of BSF was achieved in 7 (87.5%) of 8 patients. Conclusion Use of a retrievable hinged covered expandable metallic stent is a simple, safe, and effective procedure for closure of the BSF. Retrieval of the stent seems to be feasible.
