急性冠状动脉综合征患者Th17细胞检测及意义   总被引:3,自引:0,他引:3  
目的:探讨急性冠状动脉综合征患者外周血辅助性T细胞(T help,Th)17的水平及意义.方法:将85例患者分为4组:急性心肌梗死(AMI)组26例,不稳定型绞痛(UA)组17例,稳定型心绞痛(SA)组22例和胸痛综合征(CPS)组20例.采用流式细胞分析法,检测各组患者外周血Th17细胞占CD4 T细胞比例.应用酶联免疫吸附方法(ELISA)检测患者外周血血浆Th17相关细胞因子白细胞介素(IL)-17和IL-23的表达.结果:AMI组和UA组患者外周血Th17/CD4 T细胞比例分别为(2.3±1.3)%、(2.1±1.1)%,明显高于SA组(0.5±0.3)%和CPS组的(0.3±0.2)%(P<0.01).AMI组和UA组患者IL-17和IL-23水平[IL-17:AMI组(72.3±50.2)ng/L、UA组(64.6±38.7)ng/L;IL-23:AMI组(615±254)ng/L,UA组(571±252)ng/L]亦明显高于SA组[IL-17;(24.2±15.3)ng/L,IL-23:(136±67)ng/L]和CPS组[IL-17:(20.8±13.3)ng/L,IL-23:(128±70)ng/L],P<0.05;而SA组和CPS组间差异无统计学意义(P>0.05).患者血浆IL-17水平与血浆IL-23水平呈正相关(r=0.631,P<0.01).结论:急性冠状动脉综合征患者外周血Th17细胞比例增加,IL-17、IL-23水平升高,Th17细胞可能参与动脉粥样斑块不稳定和急性冠状动脉综合征的发病.  相似文献

目的 探讨核因子-κB(NF-κB)蛋白表达水平的变化与急性冠状动脉综合征(ACS)患者冠状动脉病变特征的关系.方法 初步诊断为冠心病并行选择性冠状动脉造影(CAG)患者共81例.根据病情将患者分为ACS组50例,包括急性心肌梗死21例,不稳定型心绞痛29例;稳定型心绞痛(SAP)组31例;选择同期CAG正常者14例作为对照组.采用免疫印迹法检测外周血单个核细胞(PBMC)NF-κB蛋白表达水平的变化,并结合Gensini冠状动脉病变积分标准比较NF-κB蛋白表达水平与冠状动脉管腔病变程度的关系.结果 ACS组NF-κB蛋白表达水平较SAP组和对照组明显增高,分别为0.85±0.18、0.75±0.21和0.71±0.23(F=3.72,P<0.05);NF-κB蛋白表达水平与Gensini冠状动脉病变积分无相关性(r=0.07,P>O.05).结论 ACS患者存在NF-κB的激活,且NF-κB蛋白表达水平与冠状动脉病变狭窄程度无相关性.  相似文献

目的观察急性冠脉综合征(acute coronary syndrome,ACS)患者外周血单核细胞五聚蛋白-3(pentraxin-3,PTX-3)的浓度变化,探讨其对ACS早期诊断的意义。方法将90例ACS患者分为不稳定型心绞痛(unstable angina group,UAP)组47例,急性心肌梗死(acute myocardial infarction,AMI)组43例;另外,选同期相匹配的稳定型心绞痛(stable angina group,SAP)组42例和健康体检者33名作为对照组。采用酶联免疫吸附法(ELISA)测定各组外周血单核细胞PTX-3浓度。结果 UAP组、AMI组外周血单核细胞PTX-3浓度均高于SAP组和健康对照组,差异有统计学意义(P<0.05);且AMI组明显高于UAP组,差异有统计学意义[(8.32±2.79)μg/L vs.(5.37±1.83)μg/L,P<0.01];SAP组与健康对照组比较,差异无统计学意义[(4.28±1.70)μg/L vs.(3.69±3.02)μg/L,P>0.05]。结论 ACS患者外周血单核细胞中可检测PTX-3的存在,且外周血单核细胞PTX-3浓度可以作为识别和判断ACS发作的临床指标。  相似文献

由SARS-CoV-2感染引起的COVID-19为全球流行病,严重威胁全球公共卫生,感染无特异性免疫力人群影响人们改变社交行为和习惯,产生全球经济负担,目前尚无有效治疗药物。因此,自然感染诱导的适应性免疫应答备受关注。自然感染SARS-CoV-2可诱导绝大多数成人产生体液免疫应答和细胞免疫应答,形成高滴度S蛋白特异性IgM、IgG和IgA类抗体、高水平中和抗体、强CD4+T细胞应答、特异性记忆B细胞和记忆T细胞,可获得抗SARS-CoV-2感染的免疫保护,但免疫保护持续时间仍不确定。对SARS-CoV-2感染诊断、血清学监测、免疫策略、感染防控及疫苗开发等有重要意义。  相似文献

A 57-year-old female developed myelodysplastic syndrome (MDS) that terminated as a biphenotypic leukemia after exposure to chemoradiotherapy. Double staining of blast cells, using monoclonal antibodies specific for myeloid and lymphoid lineage, demonstrated that one-third of the leukemic cells simultaneously expressed the E rosette-associated antigen (OKT11) and myeloid-associated antigen (MY7). This finding suggests the possibility that some cases of MDS are clonal disorders that arise in a pluripotent stem cell that can also differentiate to T cell lineage.  相似文献

目的探讨洛伐他汀对急性冠脉综合征(ACS)患者外周血单核细胞ABCA1蛋白表达的影响。方法选取急性冠脉综合征患者21例,密度梯度法分离外周血单核细胞。洛伐他汀分别在同一浓度(10p,mol/L)、不同时间(0、6、12、24h),同一时间(24h)、不同浓度(0、1、10、100p,mol/L)条件下与单核细胞在细胞培养箱中孵育,孵育结束后用流式细胞技术检测外周血单核细胞ABCA1蛋白。结果与对照组比,洛伐他汀作用单核细胞后,ABCA1蛋白表达无明显变化(P〉0.05)。结论洛伐他汀不能诱导ABCA1蛋白表达增加,其升高高密度脂蛋白(HDL)的机制可能与ABCA1蛋白无关。  相似文献

目的:探讨在急性冠脉综合征(acute coronary syndrome,ACS)患者外周血单核-巨噬细胞中Notch1分子及炎性细胞因子的表达。方法: 以密度梯度离心法分离36例ACS患者、14例稳定型心绞痛患者及30例健康对照者外周血单核细胞,并以氟波酯(PMA)使之转化为巨噬细胞。分别采用实时(RT)-PCR和Western blot测定巨噬细胞中Notch1 mRNA和其蛋白的表达;采用RT-PCR和ELISA测定炎性细胞因子血管细胞黏附分子-1(VCAM-1) mRNA和其蛋白及单核趋化蛋白-1(MCP-1) mRNA和其蛋白的表达。结果: 与对照组比较,ACS患者巨噬细胞中Notchl mRNA和其蛋白的表达明显升高(P<0.05),ACS诱导的VCAM-1和MCP-1 mRNA和二者的蛋白浓度明显增高(P<0.05)。结论: Notch1信号通路及其介导的巨噬细胞天然免疫应答在动脉粥样硬化易损斑块的发生发展中发挥着重要的调节作用。  相似文献

目的观察类风湿关节炎(RA)患者外周血CD4 T细胞TIM-3mRNA的表达,探讨TIM-3在RA发病中的作用。方法应用实时定量反转录-聚合酶链反应(RT-PCR)技术检测25例RA患者和18名健康对照组外周血CD4 T细胞TIM-3mRNA的表达,并分析TIM-3mRNA的表达与类风湿因子(RF)、C反应蛋白(CRP)的相关性。结果RA患者外周血CD4 T细胞TIM-3mRNA的相对表达较健康对照组明显增多(0.84±0.09vs0.55±0.07,t=11.73,P<0.01),且与RF水平呈正相关(r=0.84,P<0.01);高度活动组RA患者TIM-3mRNA的表达高于低中度活动组(0.89±0.06vs0.81±0.09,t=2.49,P<0.05)。结论RA患者CD4 T细胞TIM-3mRNA的表达异常增多并与RF水平呈正相关,高度活动组TIM-3mRNA的表达高于轻中度活动组,提示TIM-3mRNA的表达与RA的病情活动有关,TIM-3可能在RA的发病中起重要作用。  相似文献

Stroke is a rare but severe event after acute coronary syndrome. Relations between both arterial territories are still discussed but prevalence of asymptomatic carotid stenosis potentially implicated in the mechanism of stroke is under-investigated. This study aimed to determine the prevalence of asymptomatic carotid stenosis in that population and review the value of screening in a view to potential surgical correction or other preventive therapies. Systematic carotid Doppler ultrasound screening was implemented on 152 consecutive patients admitted to the intensive care unit due to acute coronary syndrome. Fifty-two percent of patients had at least one carotid artery stenosis determined using the NASCET method. Forty-three percent had at least one carotid stenosis <30%, 6% had mild (30–60%) stenosis, and 2.6% had high-grade (>60%) stenosis. Existence of carotid stenosis was associated with age, diabetes mellitus, hypertension, and with the extent of the coronary atherosclerosis (left main artery stenosis). In multivariate analysis, age and diabetes were independently associated with existence of a carotid stenosis. When ultrasound screening was restricted to patients identified by multivariate analysis, the prevalence of carotid stenosis potentially requiring surgical treatment rose to 4.6%. The frequency of asymptomatic carotid atherosclerosis was high in this population but lesions were mild. While the small number of high-grade lesions indicates that systematic screening is not to be recommended, screening of patients aged >65 years or with diabetes, or both, with a view to surgery may be envisaged, since the role of mild stenosis in the occurrence of stroke remains debatable.  相似文献

目的 观察不同类型冠心病患者外周血单核细胞核因子 κB (NF κB)活性水平变化及其与血浆可溶性细胞间黏附分子 1(sICAM 1)的关系 ,以探讨识别与预测急性冠状动脉综合征 (ACS)的炎性指标。方法 采用夹心酶联免疫吸附法和免疫组织化学染色测定。 6 0例冠心病患者 [30例ACS ,30例稳定性冠心病 (SCHD) ]及 30例对照组的血浆sICAM 1浓度和外周血单核细胞NF κB活性。同时采用直线相关分析法分析NF κB活性与sICAM 1浓度相关性。结果 ACS组NF κB活性和sICAM 1浓度显著高于SCHD组和对照组 (P <0 .0 1) ,而且NF κB活性与sICAM 1浓度呈显著正相关。但SCHD组NF κB活性和sICAM 1浓度与对照组比较 ,差异无显著性意义。结论 ACS组NF κB活性和sICAM 1浓度升高 ,提示其可能与ACS的发生有关 ,血浆sICAM 1浓度水平升高可能是通过NF κB调节途径而实现。  相似文献

Our and others' previous studies have shown that Schistosoma japonicum (SJ) infection can inhibit allergic reactions. Moreover, we found that adoptive transfer of dendritic cells (DCs) from inhibited mice showed a similar inhibitory effect on allergy, suggesting a critical role of DCs in SJ-infected mediated inhibition of allergy. In this study, we further examined the mechanism by which DCs contribute to inhibition of allergy. Our results showed that DCs from SJ-infected mice (SJDCs) produced significantly higher levels of IL-10 compared to those from naive control mice (NDCs). Adoptive transfer of SJDCs, unlike NDCs, significantly increased CD4+CD25+Foxp3+ T cells and CD4+CD25+IL-10+ T cells regulatory T-cell responses in vivo. This was correlated with significantly reduced production of IL-4 and IL-5 by CD4+ T cells, eotaxin in lung tissues and reduced airway allergic inflammation in the SJDC recipients following allergen sensitization and challenge. These data suggest that helminth infection may induce tolerogenic DCs that can inhibit the development of airway allergic inflammation through enhancing T regulatory cell responses.  相似文献
