目的 探讨超声造影剂介导下重组腺相关病毒载体(rAAV2-GFP)心肌靶向转染前后大鼠左室心肌功能的变化.方法 健康成年SD大鼠20只,随机分为对照组与实验组:对照组尾静脉注射超声造影剂+超声照射;实验组注射携带rAAV2-GFP造影剂+超声照射.分别于实验前和实验后14 d利用斑点追踪技术测量大鼠周向应变、收缩期应变率,舒张期应变率、扭转角度,并同时测量大鼠射血分数、短轴缩短率、舒张末期容积及收缩末期容积等左心功能指标.行心肌组织冰冻切片,荧光显微镜下观察GFP的表达情况.结果 超声介导重组腺相关病毒心肌靶向转染前后大鼠左室心肌功能指标及相对于心底水平的扭转角度及达峰时间的差异均无统计学意义(P>0.05).结论 超声及超声造影剂介导重组腺相关病毒载体靶向转染心肌组织对大鼠左室心肌功能无影响.  相似文献

目的:应用超声斑点追踪显像(STE)评价腹主动脉缩窄性高血压左室肥厚兔和舒张性心力衰竭兔模型的左室纵向、径向、圆周应变及扭转运动。方法:34只雄性新西兰兔随机分为3组:假手术组(对照组,n=10),左室肥厚组(LVH组,n=12),舒张性心力衰竭组(DHF组,n=12)。LVH、DHF兔以肾上腹主动脉缩窄的方法建立,模型建立后测量左室应变及扭转,并行左室导管检查。结果:LVH组纵向应变(LS)减低,DHF组的LS和径向应变(RS)均减低,LVH组的左室峰值扭转角度(Ptw)增加,差异有显著统计学意义,DHF组的Ptw仅有增加趋势。结论:STE可准确评价慢性压力超负荷对兔左室功能的影响。  相似文献

目的探讨心肌细胞移植对Wistar大鼠心肌梗死后左室重构及心功能的影响。方法50只雄性Wistar大鼠结扎冠状动脉左前降支成功建立慢性心肌梗死模型,随机分为试验组,即新生鼠心肌细胞移植组25只;对照组,即细胞培养液基质移植组25只。另设假手术组15只为正常对照。心肌梗死后4周和细胞移植后4周,采用高频超声心动图评价大鼠左室形态和功能。用免疫组化技术检测试验组移植细胞的存在。结果细胞移植后4周,与对照组相比,试验组左室收缩末期内径、舒张末期内径、收缩末期容积和舒张末期容积均显著缩小(P〈0.01),左室前壁舒张末期厚度明显增厚(P〈0.01),左室射血分数和缩短率显著升高(P〈0.01)。免疫组化显示试验组心肌瘢痕区边缘有BrdU阳性细胞存在。结论心肌细胞移植可逆转心肌梗死大鼠左室重构,改善左室功能。高频超声心动图为心肌细胞移植效果评价提供了有效实用的方法。  相似文献

目的 应用超声斑点追踪成像(STI)评价正常人左室心肌周向收缩功能.方法 35例正常人取二尖瓣、乳头肌及心尖水平短轴连续3个心动周期的二维灰阶图像,使用Qlab 6.0工作站进行脱机分析,系统自动将左室短轴分为前间隔、前壁、侧壁、后壁,下壁、室间隔6段,共计18个节段,将左室壁等分为心内膜下感兴趣区及心外膜下感兴趣区,应用STI技术分析感兴趣区的周向收缩期峰值应变.结果 心内膜层心肌各室壁节段周向收缩期峰值应变均高于心外膜层心肌(P<0.05),尤以前间隔和后壁节段差异更为显著(P<0.01).结论 超声斑点追踪成像技术不受声束角度的限制,可用于评价左室局部心肌的周向收缩功能.  相似文献

目的应用实时三维超声心动图(RT3DE)绘制正常人左室节段容量-时间曲线(VTC),探讨这一新技术评价左室节段舒张功能的可行性.方法应用RT3DE检查35例健康志愿者,获得节段与整体VTC曲线,比较各部分舒张功能间差异性和相关性.结果所有研究对象均获得满意RT3DE和2DE图像;可直接观察左室节段及整体运动;旋转切割全容积图像可立体显示心脏腔室的形态、结构;分析三维数据绘制动态rVTC.结论 RT3DE技术绘制节段VTC方法评价左室局部舒张功能是可行的,能够为临床工作提供详细的定量资料,具有广阔的应用前景.  相似文献

目的:观察儿童左心室正常发育的超声心动图测值。判定其对心功能的评估价值。方法:2002—03/2003—11上海市杨浦区黑山路幼稚园及同期解放军第二军医大学长海医院儿科住院儿童共1005例。纳入标准:无心脏疾患,体格发育匀称的儿童。排除标准:儿童监护人不同意参加研究者。符合上述标准儿童134例,男62例,女72例。随机将其分为4组:1-3岁组30例,4—6岁组35例,7—9岁组35例,10—12岁组34例。利用Simpon法和M型测量各组左心室舒张末容积(end diastolic volume。EDV)及舒张末容积指数(end diastolic volume index。EDVI=EDV/体表面积);测量左心室室间隔、后壁的厚度;计算左心室心肌质量及容积与心肌质量的比值。结果:Simpson法的测值较M型小。两种检测方法的测量结果间有差异有显著性意义(P&;lt;0.05)。左室EDV和心肌质量随年龄增长有明显增大,而各组EDVI两两比较,差异无显著性意义(P&;gt;0.05)。各年龄组间利用Simpon法和M型测量获得总体EDVI平均值[(41,4&;#177;7.2)。(54.4&;#177;8.7)mL/m^2]差异无显著性意义(P&;gt;0.05)绪论:超声心动图观察儿童左心室发育指标以容积指数较为简便和合理。  相似文献

目的 探讨超声促进携带增强型绿色荧光蛋白报告基因(EGFP)的重组腺相关病毒载体2(rAAV2)心肌转染过程中rAAV2的最佳滴度.方法 健康成年SD大鼠21只随机分为实验组与对照组.实验组给予连接于SonoVue微泡表面的rAAV2-EGFP尾静脉注入,滴度为1.5×109~3.0×101111 vg/ml(virus genome/ml),超声爆破造影剂,促进rAAV2-EGFP心肌内转染.对照组只给予尾静脉注射造影剂并超声爆破(无rAAV2-EGFP).2周后处死大鼠并制作心肌组织冰冻切片.荧光显微镜下检测荧光蛋白的表达量,代表rAAV2转染率.结果 随着注入的rAAV2滴度的增加,心肌内荧光表达量亦增加,至滴度达到1.5×1011vg/ml时荧光的表达量显著增加(P<0.01).结论 促进心肌组织转染的rAAV2的最佳滴度为1.5×1011vg/ml.  相似文献

目的 探讨无创性估测左室心肌松弛时间常数的新方法。方法 在30 例二尖瓣返流患者中,应用返流压差法测量左室松弛时间常数(T) ,应用脉冲波多普勒测量二尖瓣瓣尖及瓣下1 cm 、2 cm 及3cm 取样点的E 波流速、瓣下各点与瓣尖E 波流速比值,各点心电图R 波顶峰至E 波顶峰的时间( RE) 、瓣下各点与瓣尖RE0 时间比值。结果 在各项指标中,RE2/RE0 和RE3/ RE0 两项指标与T 值相关最佳(r = 0 .87 和r = 0 .89 , P < 0 .001) 。结论 左室松弛减退可导致E 波传播时间的延长,而左室中部与二尖瓣瓣尖RE 比值为无创性估测T 值提供了一个新的方法。  相似文献

目的:采用压力-应变环(PSL)无创技术参数定量评估晚孕期孕妇不同糖代谢异常环境下左室心肌做功变化特点。方法:收集2020年10月—2021年6月77例糖耐量异常孕妇和46例健康孕妇,并根据2020年美国糖尿病学会(ADA)诊疗指南分为妊娠期糖尿病(GDM)组和糖耐量受损(IGT)组,分别为45例和32例。在孕36周以后分娩前进行母体超声心动图检查,分别留存至少3个心动周期的心尖四腔、三腔、两腔标准切面动态图像,导入工作站并分析获取左心室压力-应变曲线及其具体参数,如整体做功指数(GWI)和效率(GWE)、整体有用功(GCW)和无用功(GWW)。并通过三维全容积成像技术获取左室的质量(LVM)、质量指数(LVMI)及左室射血分数(LVEF),并将以上参数进行比较,以评估三组参数间的差异。结果:三组间对照组GLS绝对值、GWI、GCW高于IGT组,IGT组又高于GDM组;GDM组LVM、LVMI较IGT组和对照组均增高(均P<0.05)。结论:GDM组及IGT组孕妇晚孕期左室GLS、GWI、GCW均较正常孕妇下降,且在糖尿病前期较LVM、LVMI更敏感。  相似文献

Methods Twenty one adult SD rats were divided into seven groups. SonoVue attached with different titer [1.5 × 109~ 3.0 × 1011 vg/ml (virus genome/ml)] of rAAV2-EGFP was infused into the tail vein of rats, following ultrasound mediated microbubbles destruction,as experiment groups. Normal saline was infused into the tail vein of rats as the control group (without rAAV2). Rats were killed after 14 days and hearts were harvested. GFP protein expression which showed rAAV2 transfer was observed under fluorescence microscope in frozen section and integrate optical density(IOD) was measured by Image Pro Plus software. Results When the titer of rAAV2 was 1.5 × 1011 vg/ml infused into the tail vein of rats there was much more GFP expression in myocardium than lower titers (P <0.01).
