The lipid lowering activity of fenugreek seeds ( Trigonella foenum graecum L.) was studied in 60 non-insulin dependent diabetic subjects. Isocaloric diets without and with fenugreek were given for 7 days and 24 weeks respectively. Ingestion of an experimental diet containing 25 g fenugreek seed powder resulted in a significant reduction of total cholesterol, LDL and VLDL cholesterol and triglyceride levels. These results indicate the beneficial effect of fenugreek seeds in diabetic subjects.  相似文献

The hypocholesterolaemic and hypotriglyceridaemic properties of fenugreek seed subfractions were studied in alloxan diabetic dogs. These subfractions were added to the diet of animals for 21 days. Subfraction a containing testa + endosperm (fibres 79.4%) induced a clear reduction in hyperglycaemia and in elevated plasma cholesterol levels. Subfraction b containing cotyledons + axis (proteins 52.8%, saponins 7.2%) caused a diminution in elevated plasma cholesterol and triglyceride levels. No beneficial effect was observed with subfraction P (proteins 70.50%). Subfraction S (saponins 22.2%) induced, like subfraction b, a reduction in hypercholesterolaemia and hypertriglyceridaemia of the diabetic dogs without inducing significant changes in HDL cholesterol concentrations. Our results show that the hypolipidaemic effects of fenugreek seeds are due to the fraction rich in fibres (hypocholesterolaemic effect) and to the fraction rich in saponins (hypocholesterolaemic and hypotriglyceridaemic effects).  相似文献

The effect of fenugreek seeds on the activities of beta-glucuronidase and mucinase during 1,2-dimethylhydrazine (DMH)-induced colon carcinogenesis in rats was studied. Rats were given a weekly subcutaneous injection of DMH at a dose of 20 mg/kg body weight, for 15 weeks. Fenugreek seed powder was weighed depending upon the weight of individual rats and incorporated in the powdered pellet diet at a dose of 2 g/kg body weight. After an experimental period of 30 weeks the activity of beta-glucuronidase significantly increased in the colon, intestine, liver and colon contents in DMH administered rats when compared to an untreated control group. Increase in beta-glucuronidase may increase the hydrolysis of carcinogen-glucuronide conjugate, liberating carcinogen and/or co-carcinogen within the colonic lumen. Inclusion of fenugreek seed powder in the diet significantly decreased the activity of beta-glucuronidase in all the tissues studied. This may prevent the free carcinogens from acting on colonocytes. Mucinase helps in hydrolysing the protective mucin. Mucinase activity was increased in the colon content and fecal content of animals given DMH when compared to control, while the activity was significantly reduced in animals given DMH + fenugreek when compared to animals given DMH only. Our study shows that supplementation of fenugreek seeds in the diet inhibits colon carcinogenesis, by modulating the activities of beta-glucuronidase and mucinase. The beneficial effect may be attributed to the presence of fibre, flavonoids and/or saponins.  相似文献

The hypoglycaemic activity of fenugreek seeds (Trigonella foenum graecum) in experimental animals and humans has been well documented. Fenugreek has been shown to reduce fasting and postprandial blood glucose levels in diabetic patients. However, it is not clear whether the improvement in glucose tolerance is due to the effect of fenugreek on the absorption or metabolism of glucose. A metabolic study was carried out, and diets with or without 25 g fenugreek were given randomly to 10 non-insulin dependent diabetics, each for 15 days, in a crossover design. An intravenous glucose tolerance test (GTT) at the end of each study period indicated that fenugreek in the diet significantly reduced the area under the plasma glucose curve (AUC), half-life, and increased the metabolic clearance rate. In addition, it increased erythrocyte insulin receptors. These results suggest that fenugreek can improve peripheral glucose utilization which contributes to an improvement in glucose tolerance. Thus, fenugreek may exert its hypoglycaemic effect by acting at the insulin receptor as well as at the gastrointestinal level.  相似文献

This study examined the effect of a Du-zhong (Eucommia ulmoides Oliver) leaf extract (0.175 g/100 g diet) that was supplemented with a high-fat diet (10% coconut oil, 0.2% cholesterol, wt/wt) on hyperlipidemic hamsters. Hamsters fed with Du-zhong leaf extract for 10 weeks showed a smaller size of epididymal adipocytes compared to the control group. The supplementation of the Du-zhong leaf extract significantly lowered the plasma levels of triglyceride, total cholesterol, LDL-cholesterol, non HDL-cholesterol, and free fatty acid, whereas it elevated the HDL-cholesterol/total cholesterol ratio and apolipoprotein A-I levels. The hepatic cholesterol concentration was lower in the Du-zhong group than in the control group. The plasma total cholesterol concentration was positively correlated with hepatic HMG-CoA reductase activity (r = 0.547, p < 0.05) and hepatic cholesterol concentration (r = 0.769, p < 0.001). The hepatic fatty acid synthase and HMG-CoA reductase activities were significantly lowered by a Du-zhong leaf extract supplement in high fat-fed hamsters. Hepatic fatty acid synthase activity was positively correlated with plasma fatty acid concentration (r = 0.513, p < 0.05) that was lower in the Du-zhong group. These results demonstrate that the Du-zhong leaf extract exhibits antihyperlipidemic properties by suppressing hepatic fatty acid and cholesterol biosynthesis with the simultaneous reduction of plasma and hepatic lipids in high fat-fed hamsters.  相似文献

Effects of Panax ginseng on plasma and hepatic lipids were investigated in the high cholesterol diet-fed rats and in patients with hyperlipidemia. Oral administration of red ginseng powder reduced plasma total cholesterol, triglyceride and NEFA, while plasma HDL-cholesterol was elevated. Platelet adhesiveness was also reduced by ginseng administration. The plasma lipid-improving actions were also observed in patients with hyperlipidemia. Hepatic cholesterol and triglyceride contents were decreased and phospholipid increased by ginseng administration in the high cholesterol diet-fed rats, corresponding to improvement of the fatty liver.  相似文献

The seeds of Trigonella foenum graecum (fenugreek) have been reported to have antidiabetic and hypocholesterolaemic properties in both animal models and humans. Activity has been attributed largely to fenugreek's saponin and high fibre content, and is probably not related to its major alkaloid trigonelline. Antihyperglycaemic effects have been linked to delayed gastric emptying caused by the fibre content, and to (unidentified) components that inhibit carbohydrate digestive enzymes. Fenugreek administration may increase plasma insulin levels in vivo. Its major free amino acid, 4-hydroxyisoleucine, stimulates insulin secretion from perfused pancreas in vitro. The hypocholesterolaemic effect has been attributed to increased conversion of hepatic cholesterol to bile salts due to loss, in the faeces, of complexes of these substances with fenugreek fibre and saponins. Fenugreek treatment selectively reduces the LDL and VLDL fractions of total cholesterol, and HDL-cholesterol has also been reported to increase in alloxan-induced diabetic rats and type II diabetic individuals following treatment with fenugreek. Fenugreek administration has not been reported to cause any toxicological effects. Its regular consumption may therefore be beneficial in the management of diabetes and the prevention of atherosclerosis and coronary heart disease. © 1998 John Wiley & Sons, Ltd.  相似文献

The hypolipidaemic effect of fenugreek seeds (Trigonella foenum graecum L.) was evaluated in ten hyperlipidaemic subjects. Isocaloric diets with and without fenugreek were each given for 20 days. Ingestion of experimental diets, containing 100 g of debitterized fenugreek powder, resulted in a significant reduction in the serum total cholesterol, LDL and VLDL cholesterol and triglyceride levels. HDL cholesterol levels were not altered but the ratio with total cholesterol and LDL and VLDL cholesterol were significantly increased. These results indicate the beneficial effect of fenugreek seeds in hyperlipidaemic patients.  相似文献

The effect of fenugreek seeds (Trigonella foenum graecum) on blood lipid peroxidation and antioxidant status in alloxan diabetic rats was studied. Increased lipid peroxidation and alterations in circulating antioxidants were observed in the diabetic state. The levels of glutathione, ascorbic acid and β‐carotene in blood were significantly lowered and α‐tocopherol content was increased. Supplementation of fenugreek seeds in the diet lowered lipid peroxidation. The contents of glutathione and β‐carotene were increased and the α‐tocopherol content was lowered. The level of ascorbic acid was unaltered. The level of antioxidants were higher in normal rats which were fed with the fenugreek supplemented diet compared with control animals which were fed commercial rat chow. The study shows that disrupted free radical metabolism in diabetic animals may be normalized by fenugreek seed supplementation in the diet.­Copyright © 1999 John Wiley & Sons, Ltd.  相似文献

In the present study, an aqueous extract from Erica multiflora L. (Ericaceae) flowers was evaluated for its hypocholesterolaemic and hypotriglyceridaemic activities using Triton WR-1339 induced hyperlipemic rats as experimental model. Hyperlipideamia was developed by intraperitonial injection of Triton (200mg/kg body weight). The animals were divided into control (CG), hyperlipidaemic (HG), hyperlipidaemic plus herb extract (HG+EmE) and hyperlipidaemic plus fenofibrate (HG+FF) treated groups. Intragastric administration of Erica multiflora extract (0.25 g/100g body weight) to the rats caused a significant decrease on their plasma lipid levels (quantified using enzymatic kits). At 7h after treatment, plasma total cholesterol, triglycerides and LDL-cholesterol were decreased by 47%, 95% and 67%, respectively, but the change of HDL-cholesterol level was not significant. However, the hypolipidaemic effect of fenofibrate was limited to triglycerides and LDL-cholesterol, which were lowered by about 92% and 41%, respectively. At 24h after treatment, Erica multiflora extract reduced plasma total cholesterol by 68.5% and triglycerides by 91%. HDL-cholesterol was not significantly increased and LDL-cholesterol was lowered by 80.5%. In fenofibrate treated rats, only plasma triglyceride concentrations were lowered by 82%, while the other lipid parameters were not significantly changed indicating that this aqueous herb extract may contain products that lower plasma lipid concentrations and might be beneficial in treatment of hyperlipideamia.  相似文献

Fenugreek (Trigonella foenum-graecum) is used as a spice, vegetable and a medicinal plant. Since antioxidant properties have been linked to health benefits of natural products, such properties were studied in germinated fenugreek seeds which are considered to be more beneficial than dried seeds. Different fractions of the germinated seeds were used to determine their antioxidant potential at different levels. The assays employed were ferric reducing antioxidant power, radical scavenging by 1,1-diphenyl-2-picrylhydrazyl, ferrylmyoglobin/2,2'-azobis-3-ethylbenzthiazoline-6-sulfonic acid, pulse radiolysis, oxygen radical absorbance capacity and inhibition of lipid peroxidation in mitochondrial preparations from rat liver. An aqueous fraction of fenugreek exhibited the highest antioxidant activity compared with other fractions. As the quantity of phenolic and flavonoid compounds can be related to antioxidant activity, the contents from these extracts were measured. HPLC analysis was carried out to detect polyphenols, flavonoids and other components. This study reveals significant antioxidant activity in germinated fenugreek seeds which may be due partly to the presence of flavonoids and polyphenols.  相似文献

The present study was designed to evaluate the effect of supplementation of fenugreek leaves, an indigenous plant widely used in Indian Ayurvedic medicine for the treatment of diabetes mellitus, in streptozotocin induced diabetic rats. Supplementation of the diet with fenugreek leaves showed a significant effect on hyperglycaemia, hypoinsulinaemia and glycosylated haemoglobin in streptozotocin diabetic rats. Fenugreek leaves improved the body weight and liver glycogen. Fenugreek leaves also showed a significant effect on key carbohydrate metabolic enzymes in diabetic rats. The effect of fenugreek leaves was found to be similar to that of glibenclamide. Thus, fenugreek leaves exhibited antidiabetic action in streptozotocin-induced diabetic rats. Insulin restored all the parameters to near normal levels in diabetic rats.  相似文献

The inhibitory effect of the MeOH extract of Pleurospermum kamtschaticum (Umbelliferase) and its fractions were tested in hyperlipidemic and hypercholesterolemic rats using four animal models induced using poloxamer-407 or using Triton WR-1339 as intrinsic inducers and by 30% corn oil or high cholesterol diet as extrinsic inducers. We measured serum triglyceride, total cholesterol, HDL-cholesterol and LDL-cholesterol levels as measures of its hypocholesterolemic or hypolipidemic effects. Since the MeOH extract and the BuOH fraction of Pleurospermum kamtschaticum were found to be active using these four hypolipidemic assays, its major saponin buddlejasaponin IV {BS(IV)} isolated from the BuOH fraction were also tested to demonstrate the active components. BS(IV) was found to significantly inhibit hypercholesterolemia and hyperlipidemia by extrinsic and intrinsic inducers. In particular, BS(IV) reduced the blood thiobarbituric acid reactive substance (TBARS) and hydroxy radical levels, and increased superoxide dismutase activity in high cholesterol diet-induced rats, thus suggesting that BS(IV) reduces oxidative stress caused by a high cholesterol diet. Moreover, these effects of BS(IV) were comparable to probucol, which was used as a positive control. These results suggested that Pleurospermum kamtschaticum which is traditionally used to treat atherosclerosis and its active major saponin BS(IV) could be used to treat hypercholesterolemia or hyperlipidemia.  相似文献

目的:研究草苁蓉提取物(BRE)对实验性高脂血症兔血浆脂蛋白抗脂质过氧化作用的影响。方法:高脂饲料喂养法建立兔高脂血症模型,连续给药8周后分离血浆脂蛋白,观察各血浆脂蛋白过氧化脂质(LPO)含量及低密度脂蛋白(LDL)氧化易感性的变化。结果:饲喂BRE后高脂血症兔血浆总胆固醇(TC)、极低密度脂蛋白胆固醇(VLDL-CH)和低密度脂蛋白胆固醇(LDL-CH)水平明显降低;血浆总LPO和脂蛋白LPO含量明显减少;在体外进行氧化修饰时,LDL氧化延滞时间明显延长。结论:BRE可降低高脂血症兔血脂水平和血浆脂蛋白脂质过氧化程度,增强LDL抗氧化修饰能力,因此能够起到防治动脉粥样硬化症的积极作用。  相似文献

Ethanolic extract of seeds of Cassia tora L. and its fractions were investigated for hypolipidemic activity on triton induced hyperlipidemic profile. Ethanolic extract and its ether soluble and water soluble fraction decreased serum level of total cholesterol by 42.07, 40.77 and 71.25%, respectively. On the other hand ethanolic extract, ether soluble fraction and water soluble fraction increased the serum HDL-cholesterol level by 6.72, 17.20 and 19.18%, respectively. Ethanolic extract, ether fraction and water fraction decreased triglyceride level by 26.84, 35.74 and 38.46%, respectively. The reduction in LDL-cholesterol level by ethanolic extract, ether soluble fraction and water soluble fraction were 69.25, 72.06 and 76.12%, respectively.  相似文献

Phytoestrogens with a biological activity like estradiol are naturally found in many plants. This study was designed to investigate the effect of red clover, a phytoestrogen-rich member of the legume family (Trifolium pratense) on the development of atherosclerosis in male hyperlipidemic rabbits. Twenty rabbits were semi-randomly distributed into four groups of five each. Two groups received either normal diet or normal diet supplemented with red clover. Two other groups received similar diets to both of which 1% cholesterol was added. Dietary use of red clover (RC) in hyperlipidemic rabbits significantly decreased C-reactive protein (CRP), triglyceride (TG), total cholesterol and LDL-cholesterol (LDL-C) whereas, HDL-cholesterol (HDL-C) was significantly increased in those animals (p < 0.05). Fatty streak formation was also significantly lower in aorta and left and right coronary arteries in the same animals due to use of dietary RC supplementation. These findings suggest that dietary RC may reduce cardiovascular risk factors.  相似文献

Toxicological evaluation of fenugreek seeds was made in 60 NIDDM subjects in a 24 week study in which changes in body weight, clinical signs and symptoms and serum parameters such as SGOT, SGPT, alkaline phosphatase, bilirubin, creatinine and blood urea, together with haematological parameters, was studied. Ingestion of an experimental diet containing 25 g fenugreek seed powder resulted in no renal or hepatic toxicity, but interestingly blood urea levels were seen to decrease after week 12.  相似文献

Shengmai San (SMS), which is comprised of the medicinal herbs of Panax ginseng C.A. Meyer, Schisandra chinensis Baill., and Ophiopogon japonicus Ker-Gawl (2:1:2)., is a traditional Chinese medicine being used for treating coronary heart disease. The aim of this study was to investigate the effects of SMS on the plasma and liver lipids, lipid peroxidation and antioxidant systems in liver and heart of cholesterol-fed rats. Rats were fed on a high-cholesterol (0.5%) diet (control group), high-cholesterol diet containing 2% SMS (2% SMS group) and 4% SMS (4% SMS group) for four weeks. The oxidative stress marker (thiobarbituric acid reactive substances, TBARS) and antioxidant defense systems including glutathione (GSH), glutathione peroxidase (GSH-Px), glutathione-S-transferase (GST) and superoxide dismutase (SOD) activities in rat liver and heart were evaluated. Results showed that rats fed with SMS-containing diet had reduced the H(2)O(2)-induced erythrocytes susceptibility to hemolysis, and 4% SMS feeding rats had higher plasma GSH concentration compared to the animals fed with the control diet. However, SMS had no effect on plasma lipids (total cholesterol, triglyceride and high-density lipoprotein cholesterol) and TBARS concentration. On the other hand, rats fed with the 4% SMS diet reduced the hepatic cholesterol and triglyceride contents. Fecal bile acid excretion was significantly increased in rats fed with the SMS-containing diet. Higher hepatic GSH and lower TBARS concentrations were observed in rats fed with the 4% SMS diet compared with the rats fed with the control diet. No significant difference in activities of GSH-Px, GST and SOD was found in liver and heart after the SMS treatment. Results from this study indicate that the SMS may reduce hepatic lipids and lipid peroxidation in rats.  相似文献

In this study the effect of Padma 28 on the level of ascorbate free radicals and lipids in animals receiving a high-fat diet (HFD) was measured. Male Wistar rats were divided into four groups: (I) controls, (II) animals given HFD containing cholesterol and coconut oil, (III) rats treated with Padma 28, (IV) HFD + Padma 28, over a period of 14 days. Electron spin resonance (ESR) spectra, corresponding to ascorbate free radicals, were measured from samples of the whole blood. The number of these radicals was significantly increased in animals of group IV (HFD + Padma 28), whilst the elevation of serum cholesterol, triglyceride and β-lipoproteins was in this group significantly suppressed; HDL cholesterol content was normalized.  相似文献

Aim

The present study is designed to investigate the effect of ShanZha (Crataegus pinnatifida) on obesity or dyslipidemia induced by high-fat diet in hamsters and characterize the role of PPARα in this action of ShanZha.

Materials and methods

We induced dyslipidemia and obesity in hamsters using high-fat diet and treated hamsters with ShanZha or vehicle for 7 days. We measured the body weight, adipose tissue weights, and food intake of hamsters. Plasma total cholesterol (TC), triglyceride (TG), LDL-cholesterol (LDL-C) and HDL-cholesterol (HDL-C) were determined at the beginning and end of this treatment. Effect of ShanZha on adipogenesis was examined in vitro and change of PPARα was analyzed using Western blot.

Results

The food intake, body weights, and weights of both brown and white adipose tissues were markedly reduced in hamsters receiving ShanZha as compared with the vehicle-treated control. Plasma levels of TC, TG and LDL-C were decreased by this ShanZha treatment while HDL-C was elevated. The effects of ShanZha were reversed by the combined treatment with PPARα antagonist, MK886. ShanZha inhibited the fat droplet accumulation in 3T3-L1 adipocytes in a dose-dependent manner and this effect was abolished by MK886. Western blot results showed activation of PPARα by ShanZha in hamster adipose tissue.

Conclusion

We suggest that ShanZha could activate PPARα to improve dyslipidemia or obesity.  相似文献
