We examined the effect of dietary conjugated linoleic acid (CLA) on the growth of injected hepatoma dRLh-84 in Donryu rats. After experimental diets containing 0% or 2% CLA were given to male Donryu rats for 3 wk, dRLh-84 cells were injected into the left lobe of the hepatic capsule, and the experimental diet was continued. The cells formed a solid tumor > or = 1 wk after the injection, and thereafter the tumor grew with feeding duration. In a morphological study, this tumor appeared to be a low-differentiated hepatoma, and there was no remarkable difference in the morphology of the tumor between 0% and 2% CLA groups. Tumor weight was significantly higher in the 2% CLA group than in the 0% CLA group throughout the feeding period after the injection. Serum glutamic-oxaloacetic transaminase and glutamic-pyruvic transaminase activities were significantly higher in 2% CLA-injected rats than in 0% CLA-injected rats at 3 wk after the injection. CLA upregulated acyl-CoA oxidase activity, especially 1 wk after the injection. However, dietary CLA did not activate carnitine palmitoyl transferase II, which is a rate-limiting enzyme in the mitochondrial beta-oxidation pathway. Natural killer cell activity in the spleen tended to be higher in injected rats, but a significant effect of dietary CLA was not recognized. Serum interferon-gamma and tumor necrosis factor-alpha levels were higher in injected than in sham rats. Moreover, these levels were higher in 2% CLA groups than in the respective 0% CLA groups.  相似文献

The effects of a combination of dietary conjugated linoleic acid (CLA) supplemented with sesamin on hepatic ketogenesis and triacylglycerol secretion were compared using the livers of rats fed diets containing 1% CLA or linoleic acid (LA) in combination with 0.2% sesamin for 14 d, respectively. The feeding of CLA, as compared to LA, caused a significant reduction in the weight of perirenal adipose tissue but not that of epididymal adipose tissue, and affected neither growth parameters nor hepatic lipid concentration. Hepatic production of ketone bodies was consistently higher in rats fed CLA than in those fed LA, while triacylglycerol secretion was reversed. No significant difference was noted in the hepatic secretion of cholesterol among the groups. Although there was no effect of the dietary combination of CLA with sesamin on adipose tissue weight, hepatic lipid parameters and ketone body production were observed: i.e., triacylglycerol secretion tended to be reduced. These results suggest that the dietary combination of CLA with sesamin may be an effective approach for lowering serum triacylglycerol levels. The decreased hepatic secretion of triacylglycerol is, in part, due to enhanced fatty acid oxidation in the liver.  相似文献

ObjectiveOur aim was to investigate the effects of dietary conjugated linoleic acid (CLA) at high-fat (HF) levels on parameters related to triacylglycerol (TG) regulation and some potential impacts on liver damage.MethodsGrowing mice were fed a control diet (7% corn oil), an HF diet containing 20% corn oil, or an HF diet containing 3% CLA (HF + CLA) for 30 d. Tissue and organ weights, plasma and tissue TG levels, and parameters related to their regulation were evaluated. Liver oxidative status was also assessed.ResultsDietary CLA showed detrimental and beneficial effects. CLA added to the HF diet caused hepatomegaly (+32%) and exacerbated the hepatic TG accumulation (+168%) observed with the HF diet without inducing liver damage; however, it significantly reduced plasma TG concentrations (?37%) and normalized muscular TG content. An increase in glutathione was associated with total normalization of liver lipid peroxidation. In addition, HF + CLA caused dystrophy of epididymal fat pads, even when the HF diet had increased the adipose tissue mass (30%). The biochemical mechanisms involved in the regulation of lipid levels were related to reduced (?20%) hepatic very low-density lipoprotein-TG secretion and decreased muscle (?35%) and adipose (?49%) tissue contributions to the removal of plasma TG by lipoprotein lipase enzymes.ConclusionExamination of CLA at HF levels showed hepatomegaly and exacerbation of lipid accretion as a negative impact; however, some positive aspects such as hypotriglyceridemia and protection against oxidative stress were also induced. Even the fat reduction is nutritionally important for weight control; the biochemical mechanisms whereby CLA mediates the potential effects could produce undesirable metabolic alterations.  相似文献

[目的 ] 研究共轭亚油酸对人体的减肥作用。  [方法 ]  3 0名单纯性肥胖的志愿者连续食用共轭亚油酸胶囊 3 5d ,于试验开始及结束时均测试各项安全性指标及功效性指标。  [结果 ] 受试者连续食用共轭亚油酸胶囊 3 5d后 ,体重平均下降 1.9kg ,体脂总量平均下降 1.2kg ,体脂百分率平均下降 0 .7% ,三角肌处脂肪厚度减少 2 .1mm ,肩胛下脂肪厚度减少 4.1mm ,脐旁脂肪厚度减少 3 .5mm ,髂棘处脂肪厚度减少 3 .6mm ,腰围减少 4.3cm ,臀围减少 1.5cm。试验前、后受试者各项临床指标均未见异常 ,并且试验后运动耐力未下降。  [结论 ] 共轭亚油酸对人体具有明显的减肥作用 ,且对机体健康无明显损害。  相似文献

目的研究不同共轭亚油酸对人类胃癌细胞AGS、肝癌细胞Bel7402和肠癌细胞Lovo细胞增殖的影响。方法采用体外培养的AGS、Bel7402和Lovo细胞为模型,测定不同处理后活细胞数量。结果研究采用的5种共轭亚油酸异构体都对AGS、Bel7402和Lovo细胞增殖表现出了抑制作用,对AGS的抑制作用要普遍高于对Bel7402和Lovo的抑制作用,并且浓度不同,抑制作用也具有差异。结论五种共轭亚油酸异构体都具有抑制癌细胞增殖的作用,并且这种作用受到共轭亚油酸浓度的影响。  相似文献

Conjugated linoleic acid (CLA) refers to a group of positional and geometric isomers of linoleic acid. Studies using animal models have shown that CLA reduces adiposity, improves plasma lipoprotein metabolism and insulin sensitivity and reduces arteriosclerosis. Whilst CLA may have therapeutic potential with regard to coronary artery disease risk factors in human subjects, there has been little investigation into its effects in human subjects. This current study investigated the effects of dietary supplementation using two isomeric blends of CLA on triacylglycerol (TAG)-rich lipoprotein metabolism and reverse cholesterol transport in human subjects and evaluates whether CLA modulated cardiovascular disease risk factors. Fifty-one normolipidaemic subjects participated in this randomised double-blind placebo-controlled intervention trial. Subjects were randomly assigned to receive 3 g cis-9,trans-11-trans-10,cis-12 isomeric blend (50 : 50) or a cis-9,trans-11-trans-10,cis-12 isomeric blend (80 : 20) CLA or linoleic acid (control)/d for 8 weeks. Metabolic effects of dietary conjugated linoleic acid (CLA) isomers in rats   总被引:3,自引:0,他引:3  
To know the effect of individual conjugated linoleic acid (CLA) isomers on lipid metabolism, male rats were fed diets containing either linoleic acid (LA, a control fatty acid), CLA containing mainly 9c,11t-isomer, 10t,12c-isomer (at the 0.8% level) or an equivalent mixture of these isomers (at the 0.4% level) for 26 days. Although there were no differences in food intake and body weight gain among the groups, the food efficiency was low in rats fed the CLA containing 10t,12c-isomer in comparison with those fed 9c,11t-CLA. The weight of white (epididymal and perirenal) adipose tissues decreased in all CLA groups, while that of brown adipose tissue increased in rats fed 10t,12c-CLA, the difference between 9c,11t- and 10t,12c-isomers being significant. The concentration of serum total cholesterol, triacylglycerol and phospholipid tended to be lower in rats fed CLA preparations containing 10t,12c-CLA, while there was a trend toward an increasing concentration of serum HDL-cholesterol in all CLA groups. The rate of fatty acid β-oxidation in the liver peroxisomes was slightly higher in rats fed CLA than in those fed LA, and the activity of mitchondrial carnitine parmitoyltranceferase (CPT) in the CLA-mix group significantly increased compared to the LA group. The effect of CLA on the concentration of spleen and serum IgE, IgG and IgA level was variable. No CLA effect was observed on the concentration of serum leptin and TNF-. Thus, individual isomers of CLA may have different effects on some metabolic parameters in rats. The results also suggested a probable interaction of different isomers on lipid metabolism.  相似文献

Evidence from animal studies suggests that conjugated linoleic acid (CLA) modulates plasma and tissue appearance of newly synthesized PUFA. The effects of a 1.2g (0.5 % energy) daily intake of the cis-9,trans-11 (c9,t11) isomer of CLA, trans-10,cis-12 (t10,c12) isomer of CLA or olive oil (placebo) on linoleic acid (LA) and linolenic acid (LNA) metabolism in healthy human volunteers was investigated. Fifteen subjects were fed an experimental diet and supplemented with c9,t11-CLA, t10,c12-CLA or placebo for 7 d before consuming a tracer dose of U-[(13)C]LA (50 mg) and U-[(13)C]LNA (50 mg). Blood samples were taken at 0, 2, 4, 6, 8, 24, 48, 72 and 168 h and analysed using high-precision MS. No differences between the groups in peak plasma [(13)C]LA (10.3-11.6 % of dose), [(13)C]LNA (2.5-2.9 % of dose), [(13)C]arachidonic acid (0.09-0.12 % of dose), [(13)C]EPA (0.04-0.06 % of dose) or [(13)C]DHA (0.06-0.10 % of dose) were detected. Concentration v. time curves (area under the curve) also showed no significant differences between groups. This suggests that, in healthy human subjects consuming a diet with adequate intake of essential fatty acids, CLA does not affect metabolism of LA or LNA.  相似文献

We examined the effect of dietary conjugated linoleic acid (CLA) on liver regeneration after a partial hepatectomy (PH) in Sprague-Dawley rats. PH was performed on rats fed a 0 or 1 wt.% CLA diet for 3 wk. Average liver weight in the CLA fed rat population was heavier than the control rat population at the time of PH and 1-d after PH. Conversely. CLA fed rats' liver weight was significantly lower than control rats at 7-d after PH. This suggests that dietary CLA reduced liver weight gain after PH. Dietary CLA did not affect serum aspartate aminotransferase (AST) or alanine aminotransferase (ALT) activities. However. CLA significantly reduced serum albumin levels at 1-d but not at 7-d after PH. 5-Bromo- and 5-iododeoxyuridine incorporation into hepatocytes 1-d post PH was lower in the CLA group. In conclusion, the data suggests that dietary CLA inhibits DNA synthesis after PH, which results in hepatocyte proliferation inhibition.  相似文献

The effect of dietary conjugated linoleic acid (CLA) on body composition and energy metabolism was investigated in broiler chickens. Male broiler chicks were assigned to receive either a control diet (1 % sunflower oil) or a diet containing CLA (1 % of a 1:1 mixture of trans-10, cis-12 and cis-9, trans-11 isomers of octadecadienoic acid). The diets were fed ad libitum for 3 weeks and there were eight replicates per diet, each replicate including four chickens so that each treatment had thirty-two animals. The proportion of body fat was lower in the control group than in the CLA group. No significant differences as to the proportions of body water, ash and protein were observed. Feed and energy intake were significantly lower in CLA-fed birds. The percentage of ingested energy lost in excreta was higher after CLA feeding and heat expenditure as a percentage of ingested energy was lower in the CLA-fed group. The CLA-fed group showed a higher percentage of SFA and lower percentages of MUFA and PUFA in carcass fat. It is concluded that CLA stimulated de novo fatty acid synthesis and lowered desaturase activity.  相似文献

OBJECTIVE: To determine if dietary conjugated linoleic acid (CLA) is associated with bone mineral density (BMD) of different skeletal sites in postmenopausal women. METHODS: A cross-sectional analysis in 136 Caucasian, healthy, postmenopausal women, mean age 68.6 years. BMD and soft tissue were assessed by dual energy x-ray absorptiometry (DXA). Energy, calcium, protein, fat, CLA and other relevant nutrients were estimated using 3 day dietary records. Supplement use was recorded as well. Current and past physical activity were determined using the Allied Dunbar National Fitness Survey for older adults. RESULTS: CLA (63.1 +/- 46.8 mg, mean +/- SD) was a significant predictor of Ward's triangle BMD (p = 0.040) in a multiple regression model containing years since menopause (18.5 +/- 8.4 y), lean tissue, energy intake (1691 +/- 382 kcal/day) dietary calcium (873 +/- 365 mg), protein (70.6 +/- 18.6 g), fat (57.9 +/- 23.9 g), zinc (19.2 +/- 13.6 mg), and current and past physical activity, with R(2)(adj) = 0.286. Subjects were also divided into groups below (Group 1) and above (Group 2) the median intake for CLA. Group 2 had higher BMD in the forearm, p = 0.042, and higher BMD in the hip, lumbar spine and whole body, however statistical significance was not reached. CONCLUSION: These findings indicate dietary CLA may positively benefit BMD in postmenopausal women. More studies are warranted examining the relationship between dietary CLA and BMD.  相似文献

Conjugated linoleic acid (CLA) is a collective term used for fatty acids with a conjugated double bond that are geometrical and positional isomers of linoleic acid. Anti-obesity and anti-cancer properties, an immunopotentiation effect, and promotion of bone formation by CLA have been shown in cell culture and animal studies. A mixture of 9c11t- and 10t12c-CLA is now used as a health food supplement after testing in clinical trials. These trials focused on improvement of lipid metabolism by CLA, whereas few studies have examined absorption and metabolism of CLA in humans. In addition, there is no report concerning absorption and metabolism of CLA in Japanese. This study was designed to examine CLA concentration in blood, the elimination rate of CLA, and metabolic differences between 9c11t-CLA and 10t12c-CLA in blood in Japanese who ingested CLA (about 2 g/d, equal weights of 9c11t-CLA and 10t12c-CLA) for 3 wk. Blood samples were collected 1 wk before the 3-wk period, on the first and last days of the period, and 1 wk after the end of the period, and the CLA concentration and distribution in blood were investigated. The CLA concentration in blood was significantly increased by CLA ingestion and reached 36 μmol/L. The CLA concentration in blood one week after the intake period was significantly lower than that at the end of CLA intake. The 10t12c-CLA level in plasma decreased faster than that of 9c11t-CLA. This suggests faster metabolism (fatty acid β oxidation) of 10t12c-CLA compared with 9c11t-CLA.  相似文献

The main objectives of the present study were to determine the effect of dietary conjugated linoleic acid (CLA) isomers on the fatty acid composition and cholesterol content of egg-yolk lipids. Forty-five 25-week-old laying hens were randomly distributed into five groups of nine hens each and maintained in individual laying cages, throughout 12 weeks of the experiment. They were assigned to the five treatments that consisted of commercial layer diets containing 0, 5, 10, 15 or 20 g pure CLA/kg. Feed intake of hens varied little and insignificantly. Egg mass was uniformly lower (P<0.05) in the hens fed the CLA-enriched diets. Feed conversion efficiency, when expressed per kg eggs, was impaired (P<0.05), although without obvious relation to the dietary CLA concentration. Feeding the CLA-enriched diets resulted in gradually increasing deposition of CLA isomers (P<0.01) in egg-yolk lipids. Saturated fatty acids were increased (P<0.01) and monounsaturated fatty acids decreased (P<0.01). Polyunsaturated fatty acids (PUFA), when expressed as non-CLA PUFA, were also significantly decreased (P<0.01). The most striking effects (P<0.01) were observed for palmitic (16 : 0) and stearic (18 : 0) acids, which increased from 23.6 to 34 % and from 7.8 to 18 %, respectively. On the other hand, oleic acid (18 : 1n-9) decreased from 45.8 to 24.3 %. Among non-CLA PUFA, linoleic (18 : 2n-6) and alpha-linolenic (18 : 3n-3) acids were strongly (P<0.01) decreased, from 14.2 to 7.7 % and from 1.3 to 0.3 %, respectively. The same was true for arachidonic (20:4n-6) and docosahexaenoic (22 : 6n-3) acids. The cholesterol content of egg yolks, when expressed in mg/g yolk, was not affected by the dietary CLA concentrations. In conclusion, unless the adverse effects of CLA feeding to laying hens on the fatty acid profile of egg yolks are eliminated, the CLA-enriched eggs cannot be considered functional food products.  相似文献

目的通过研究共轭亚油酸(CLA)对饮食诱导胰岛素抵抗大鼠脂肪酸连接蛋白(ap2)基因表达的影响,探讨CLA抗糖尿病作用的机制。方法选用雄性Wistar大鼠,随机分为对照组、高脂组、高脂+CLA组(每100g饲料含CLA分别为0·75g、1·50g、3·00g),每组动物10只,观察CLA对胰岛素抵抗大鼠胰岛素、血糖水平的影响,并应用RT-PCR的方法检测ap2、过氧化物酶体增殖物活性受体γ(PPARγ)的表达水平。结果高脂组大鼠血清游离脂肪酸(FFA)、胰岛素和糖血水平显著高于基础组,CLA可降低胰岛素抵抗大鼠血清FFA、血糖、胰岛素水平,并可增加其脂肪组织ap2、PPARγmRNA的表达水平。结论CLA可通过激活PPARγ上调脂肪酸连接蛋白基因的表达,改善肥胖大鼠的胰岛素抵抗。  相似文献

Past research showed that mammary gland morphogenesis in the pubescent rat was retarded by the feeding of conjugated linoleic acid (CLA). A major objective of the present study was to examine the proliferative activity and the expression of cell cycle regulatory proteins in the developing mammary epithelium of rats fed a mixture of CLA isomers (primarily as free fatty acid c9, t11-CLA and t10,c12-CLA) or a highly enriched natural source of c9,t11-CLA (as triacylglycerol in butterfat). In both experiments, the diets, with or without CLA, were started at weaning and continued for four weeks. The two CLA preparations were equally effective in suppressing bromodeoxyuridine labeling and the expression of cyclin D1 and cyclin A (determined by immunohistochemistry) in the terminal end buds and alveolar clusters of the mammary epithelium while it undergoes extensive ductal branching during pubescence. There was a trend of an increase, although not statistically significant, in the proportion of cells expressing the p16 and p27 cdk inhibitors. A separate experiment was designed to evaluate the effect of c9,t11-CLA (as a free fatty acid of > 90% purity) treatment on the rate of proliferation of the mammary epithelium as the animal matured from weanling to adult. The bromodeoxyuridine labeling data indicated that the mammary epithelium appeared to lose its sensitivity to CLA control of proliferation as it completely filled the fat pad and became quiescent. These observations suggest that the responsiveness of mammary epithelial cells to CLA intervention may be dependent on their proliferative status.  相似文献

Summary. Background: Conjugated linoleic acid (CLA) has been reported to decrease fat deposition, and increase lean body mass. This has been broadly inferred to mean that CLA alters protein turnover. However, data to test the effects of CLA on protein turnover are lacking. An enhancement in immune responses by CLA has also been demonstrated. Aim of the study: The objective of this study was to determine the potential for dietary CLA and protein intervention to improve nutritional and functional recovery in an animal model of catabolic stress and immunodepletion. Methods: Diets varying in their protein levels in the presence or absence of CLA were tested for their effects on the recovery of glucocorticoid (intraperitoneal injection of dexamethasone, 120 mg/kg) treated rats. Following steroid injection, rats were fed 4 dietary treatments for 4 d. The diets contained 10 or 20 g/100 g protein with or without 0.5 g/100 g CLA. Results: Dexamethasone treatment resulted in a decreased food intake and loss of weight, independent of dietary treatment. A higher number of blood monocytes occurred in rats fed the high CLA diets. The protein fractional synthesis rate in spleens of rats fed the diets containing either high proteins or CLA were higher compared to those fed diets with low protein content or without CLA, respectively. CLA, consumed post-dexamethasone treatment, did not improve protein turnover in the other tissues studied, including gut mucosa, liver, muscle and thymus. Conclusions: The present study was performed to determine the effect of CLA in acute conditions, as opposed to a preventive approach, on the recovery from a catabolic stress with immunodepletion. Dietary conjugated linoleic acid (CLA) decreases yolk 18:1(n-9), induces chick embryonic mortality and alters egg quality. A study was conducted to determine whether olive oil would prevent these adverse effects of CLA. Hens (15 per treatment) were fed diets containing 0.5 g corn oil/100 g (CO), 0.5 g CLA/100 g (CLA), 0.5 g corn oil plus 10 g olive oil/100 g (CO + OO) or 0.5 g CLA plus 10 g olive oil/100 g (CLA + OO). After 74 d of feeding, hens were placed on CO for 10 d. Hens were artificially inseminated weekly. For hatchability studies, fertile eggs were collected daily, stored at 15 degrees C for 24 h and then incubated. After 6 d of feeding, embryonic mortality rates were 15, 100, 8 and 16% in the CO, CLA, CO + OO and CLA + OO groups, respectively. When CLA-fed hens were fed the CO diet, hatchability improved to that of the CO group within 7 d. For fatty acid analysis, three eggs were obtained at the 7 d of feeding. Relative CLA levels of yolk from CO-, CLA-, CO + OO- and CLA + OO-fed hens were 0.11 +/- 0.01, 1.91 +/- 0.16, 0.08 +/- 0.04 and 0.69 +/- 0.07 g/100 g fatty acids, respectively. The ratios of 16:0/16:1(n-7) and 18:0/18:1(n-9) of yolk from CLA-fed hens were approximately 1- and approximately 1.5-fold greater, respectively, compared with those fed CO. OO prevented CLA-induced increases in 16:0 and 18:0 and the decrease in 18:1(n-9) in yolk. Fertile eggs were stored at 4 degrees C for 2 or 10 wk and analyzed for pH or mineral levels. Dietary CLA caused abnormal pH changes of albumen and yolk when eggs were stored at 4 degrees C. The pH of yolk and albumen from CO-fed hens after 10 wk of storage was 6.12 +/- 0.12 and 9.06 +/- 0.03, respectively, versus 7.89 +/- 0.25 and 8.32 +/- 0.16, respectively, in eggs from CLA-fed hens. OO prevented CLA-induced abnormal changes in the pH of albumen and yolks. Eggs from CLA-fed hens had greater iron, calcium and zinc concentrations and lower magnesium, sodium and chloride concentrations in albumen relative to those from hens fed CO. OO prevented CLA-induced mineral exchange between yolk and albumen, presumably by reducing the yolk saturated fatty acids, which are believed to disrupt the vitelline membrane during cold storage. This study suggests that the adverse effects of CLA may be due to the increased level of saturated fatty acids. However, because the addition of olive oil also lowered egg CLA content, the direct role of egg CLA on egg hatchability and quality cannot be ruled out.  相似文献

New results on the physiological properties of conjugated linoleic acid have been published by several working groups, especially showing the effects of single conjugated linoleic acid isomers on carcinogenesis and body composition. Recently, other studies have shown that conjugated linoleic acid has an influence on diabetes mellitus, platelet aggregation and the immune system. Conjugated linoleic acid was found to modify prostaglandin metabolism and delta9-desaturase activity and influence apoptosis. Furthermore, improved analytical methods using 13C nuclear magnetic resonance and silver ion high performance liquid chromatography are available to investigate the composition of conjugated linoleic acid mixtures and the exact structure of separated isomers. Also, the synthesis of isolated isomers is described, as published by different authors, in order to determine further the effects of each single conjugated linoleic acid isomer. In addition, new data on the contents of conjugated linoleic acid in foods, human adipose tissue and fluids are given in this review.
