自噬现象是一个广泛存在于真核细胞中的生命过程,最早于1962年由Ashford和Porter发现的.近年来,随着对自噬作用研究的深入,发现自噬在多种疾病中都扮演着十分重要的角色.慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)逐渐成为一个不容忽视的公共健康问题,据世界卫生组织/世界银行研究推测,截至2020年该病将成为全球经济负担的第五大疾病.但是,COPD的发病机制尚不明确,随着自噬现象及其与COPD的关系研究进一步深入,为COPD的防治寻找新的治疗策略.因此,该文着重讨论自噬在COPD发病机制中的作用及其在COPD中的临床运用价值. 相似文献

自噬(autophagy)又称细胞的自体溶解,是指隔离膜包裹胞浆蛋白和细胞器形成自噬体(autophagosome),然后与溶酶体融合成自噬溶酶体(autolysosome)并在其中降解的过程。自噬现象普遍存在于大部分真核细胞中,通过对细胞内变性蛋白或受损细胞器进行降解,以维持细胞内物质循环及代谢调节的稳定,在细胞生长、分化、功能执行、自我更新、衰老及死亡中发挥着重要调控功能。近期有大量研究表明,细胞自噬与肝炎病毒感染的发生、发展及预后有相关性。本文就自噬发生、演变过程中的分子机制及其与肝炎病毒感染的关系作一概述。 相似文献

自噬是一个高度保守的蛋白质和细胞器分解过程,这个过程可使蛋白质再循环及清除衰老的细胞器,自噬与细胞内环境稳定及组织器官发育等多种细胞活动有关。近年来有研究表明自噬与多种疾病有关,如神经肌肉变性疾病、癌症及心脏疾病。在正常心脏中,存在着低水平的自噬活动,若自噬缺陷可引起心功能不全甚至心力衰竭,在心脏后负荷过重、缺血及缺血再灌注损伤中也可观察到自噬活动的增加,自噬的缺陷与增多都与疾病的进展有关,该文就自噬在心脏疾病中的相关研究作一综述。 相似文献

Ethanol metabolism in hepatocytes causes the generation of reactive oxygen species, endoplasmic reticulum stress and alterations in mitochondrial energy and REDOX metabolism. In ethanol-exposed liver disease, autophagy not only acts as a cleanser to remove damaged organelles and cytosolic components, but also selectively clears specific targets such as lipid droplets and damaged mitochondria. Moreover, ethanol appears to play a role in protecting hepatocytes from apoptosis at certain concentrations. This article describes the evidence, function and potential mechanism of autophagy in ethanol-exposed liver disease and the controversy surrounding the effects of ethanol on autophagy. 相似文献

The epidemic of heart failure continues apace, and development of novel therapies with clinical efficacy has lagged. Now, important insights into the molecular circuitry of cardiovascular autophagy have raised the prospect that this cellular pathway of protein quality control may be a target of clinical relevance. Whereas basal levels of autophagy are required for cell survival, excessive levels - or perhaps distinct forms of autophagic flux - contribute to disease pathogenesis. Our challenge will be to distinguish mechanisms that drive adaptive versus maladaptive autophagy and to manipulate those pathways for therapeutic gain. Recent evidence suggests this may be possible. Here, we review the fundamental biology of autophagy and its role in a variety of forms of cardiovascular disease. We discuss ways in which this evolutionarily conserved catabolic mechanism can be manipulated, discuss studies presently underway in heart disease, and provide our perspective on where this exciting field may lead in the future. There are little data concerning the prevalence of pain and its intensity in the population suffering from kidney diseases. However, according to number of pathologies with a kidney impact, the sequellae of the kidney disease itself and the technology in this area, there is a risk for a high prevalence rate of either acute and/or chronic pain. This can be responsible for some degree of psychologic disturbances as a major impact on the quality of life.
