Ammonia is a natural variable in ponds and lakes. Although an important source of nitrogen for microalgae, at high concentrations ammonia can affect the density and diversity of cladocerans. Using the cladocerans Ceriodaphnia dubia, and Moina macrocopa, the effect of nominal concentrations of ammonium chloride under acute and chronic exposures at different levels of algal food was tested. Regardless of food level, C. dubia was more sensitive than M. macrocopa to ammonia. In the absence of food, the median lethal concentration of ammonia (LC(50) 24 h) for C. dubia was (112 mg L(-1)) less than half that of M. macrocopa (232 mg L(-1)). When algal food (0.5 x 10(6) and 1.5 x 10(6) cells ml(-1) of Chlorella) was used, the LC(50) values were much higher for both cladoceran species. Based on the population growth studies, it was found that in controls of M. macrocopa an increase in the availability of Chlorella from 0.5 to 1.5 x 10(6) cells mL(-1) led to an increase in the maximum density from 4.7 +/- 0.2 to 16.4 +/- 1.2 ind.mL(-1), while in C. dubia the peak population density decreased from 7.9 +/- 0.6 to 5.0 +/- l.0 ind.mL(-1). An increase in ammonia concentration (10 to 40 mg L(-1) for C. dubia and 20 to 120 mg L(-1) for M. macrocopa) resulted in a corresponding decrease in peak population densities of the tested cladocerans. The rate of population increase (r) values for M. macrocopa in the controls ranged from 0.21 +/- 0.001 and 0.25 +/- 0.02 at 0.5 and 1.5 x 10(6) cells mL(-1) of Chlorella, respectively. The corresponding values of C. dubia in controls were 0.21 +/- 0.004 and 0.18 +/- 0.01. At 0.5 x 10(6) cells mL(-1) of algal food, the r values became negative under 40 and 120 mg L(-1) of ammonia for C. dubia and M. macrocopa, respectively. We studied the effect of fructose on hepatic conversion of amino-N to urea-N as quantified by the Capacity of Urea-N Synthesis (CUNS) determined in rats during alanine loading. There were 2 control groups, one without and one with infusion of somatostatin, in order to control the effects of insulin and glucagon. Somatostatin reduced CUNS from 8.5 +/- 0.5 mumol/(min x 100 g BW) to 6.3 +/- 0.3 mumol/(min x 100 g BW) (mean +/- SEM) (p < 0.01) and reduced glucagon concentrations by 75% (p<0.05). Insulin and glucose concentrations did not change. Fructose, at blood concentrations of about 1 mmol/l further reduced CUNS to 3.6 +/- 0.3 mumol/(min x 100 g BW) (p < 0.01). Insulin increased slightly (p < 0.05), but neither glucose nor glucagon changed. At increasing fructose concentrations up to 2 mmol/l there was no further effect on CUNS. Fructose in concentrations as used for parenteral nutrition and independent of glucoregulatory hormones, decreased hepatic amino acid catabolism.  相似文献

The extent of mercury contamination in Surinamese food fishes due to small-scale gold mining was investigated by determination of the total mercury concentration in 318 freshwater fishes, 109 estuarine fishes, and 110 fishes from the Atlantic Ocean. High background levels were found in the piranha Serrasalmus rhombeus (0.35 microg Hg x g(-1) muscle tissue, wet mass basis) and the peacock cichlid Cichla ocellaris (0.39 microg x g(-1)) from the Central Suriname Nature Reserve. Average mercury levels in freshwater fishes were higher in piscivorous species than in nonpiscivorous species, both in potentially contaminated water bodies (0.71 and 0.19 microg x g(-1), respectively) and in the control site (0.25 and 0.04 microg x g(-1), respectively). Mercury concentrations in piscivorous freshwater fishes were significantly higher in rivers potentially affected by gold mining than in the control site. In 57% of 269 piscivorous freshwater fishes from potentially contaminated sites, mercury levels exceeded the maximum permissible concentration of 0.5 microg Hg x g(-1). The highest mercury concentrations (3.13 and 4.26 microg x g(-1)) were found in two piranhas S. rhombeus from the hydroelectric reservoir Lake Brokopondo. The high mercury levels in fishes from Lake Brokopondo were to some extent related to gold mining because fishes collected at eastern sites (i.e., close to the gold fields) showed significantly higher mercury concentrations than fishes from western localities. In the estuaries, mercury levels in ariid catfish (0.22 microg x g(-1)) and croakers (0.04-0.33 microg x g(-1)) were distinctly lower than those in piscivorous fishes from contaminated freshwater sites. In the isolated Bigi Pan Lagoon, the piscivores snook Centropomus undecimalis (0.04 microg x g(-1)) and tarpon Megalops atlanticus (0.03 microg x g(-1)) showed low mercury levels. Mercury levels were significantly higher in marine fishes than in estuarine fishes, even with the Bigi Pan fishes excluded. High mercury concentrations were found in the shark Mustelus higmani (0.71 microg x g(-1)), the crevalle jack Caranx hippos (1.17 microg x g(-1)), and the barracuda Sphyraena guachancho (0.39 microg x g(-1)), but also in nonpiscivorous species such as Calamus bajonado (0.54 microg x g(-1)), Haemulon plumieri (0.47 microg x g(-1)), and Isopisthus parvipinnis (0.48 microg x g(-1)). Mercury levels were positively correlated with the length of the fish in populations of the freshwater piscivores S. rhombeus, Hoplias malabaricus, and Plagioscion squamosissimus, in estuarine species (Arius couma, Cynoscion virescens, and Macrodon ancylodon), and in S. guachancho from the Atlantic Ocean.  相似文献

Certain algae contain the (n-3) fatty acid DHA, yet the relation between algal oil supplementation and cardiovascular disease risk factors has not been systematically examined. Our objective was to examine the relation between algal oil supplementation and cardiovascular disease risk factors. We conducted a systematic review of randomized controlled trials published between 1996 and 2011 examining the relation between algal oil supplementation and cardiovascular disease risk factors and performed a meta-analysis of the association between algal oil DHA supplementation and changes in the concentrations of TG, LDL-cholesterol (LDL-C), and HDL-cholesterol (HDL-C). We identified 11 randomized controlled trials with 485 healthy participants that evaluated the relation between algal oil DHA supplementation and TG, LDL-C, and HDL-C. The median dose of algal DHA was 1.68 g/d. The pooled estimate for the change in TG concentration was -0.20 mmol/L (95% CI: -0.27 to -0.14), 0.23 mmol/L (95% CI: 0.16-0.30) for LDL-C, and 0.07 mmol/L (95% CI: 0.05-0.10) for HDL-C. DHA supplementation from algal oil, a marine source of (n-3) fatty acids not extracted from fish, may reduce serum TG and increase HDL-C and LDL-C in persons without coronary heart disease.  相似文献

We initially investigated effects and uptake of cadmium and copper on Pseudokirchneriella subcapitata (primary producer; formerly known as Selenastrum capricornutum). Our major efforts focused on the effects of metal-laden (i.e., Cd, Cu) P. subcapitata when used to provide dietary metal exposure to Ceriodaphnia dubia (primary consumer). Major test endpoints included feeding rate, survival, and reproduction. Metal body burden in C. dubia increased dose dependently at dietary exposure concentrations of 0.6 microg/g algal dry weight or less for cadmium and 74.7 microg/g algal dry weight or less for copper. However, it decreased sharply when dietary concentrations exceeded 0.6 microg/g algal dry weight for cadmium and 74.7 microg/g algal dry weight copper. Both dietary cadmium and copper affected all the major test endpoints. Whereas reproduction was observed to be the most sensitive endpoint, survival was observed to be the least sensitive endpoint. These results demonstrated the potential importance of dietary exposure for consideration in metal regulations or risk assessment.  相似文献

Saturated water solutions of calcium chloride, calcium bromide (densities 1.30 kg x dm(-3) and 1.61 kg x dm(-3), respectively) and their 1:1 mixture have been commonly used as oil industry "high-density brines." In our experiment they were added to tap water in amounts appropriate to achieve concentrations of 0.025, 0.05, 0.075, and 0.1 mol x dm(-3) to study their cytotoxic effect on the root tip cells of shallot ( Allium cepa L. var. ascalonicum). All tested solutions in concentrations of 0.075 and 0.1 mol x dm(-3) caused significant inhibition of shallot root growth. CaBr (2) showed this effect in concentration 0.05 mol x dm(-3). The investigated solutions in all concentrations applied decreased mitotic activity in root tip cells. The most of mitotic abnormalities were the consequence of spindle failure and chromosome stickiness. Furthermore, the cell microtubules were investigated by indirect immunofluorescence to confirm that most abnormalities observed were the consequence of spindle failure. The present study, as well as previously done Lemna tests and Chlorella tests showed that investigated samples have certain effects on plants, so constant control of their presence in the environment is needed.  相似文献

We examined the effect of daily consumption of dietary diacylglycerol (DG) oil on serum lipid concentrations in patients with diabetes whose serum triacylglycerol (TG) levels were persistently increased despite continuous nutritional counseling at the outpatient clinic. Patients (n = 16) were divided into DG and control groups (n = 8 each). DG was incorporated (target dose 10 g/d) by substituting DG oil (80 g DG/100 g oil) for the ordinary TG cooking oil used at home for 12 wk. The control group continued consuming ordinary TG cooking oil. Dietary records indicated that there were no differences between groups in total energy intake or percentage of energy from fat. In the DG group, TG intake decreased from 26.8 +/- 9.3 to 15.7 +/- 8.9 g/d, whereas DG intake increased from 0.3 +/- 0.1 to 10.6 +/- 3.9 g/d. No differences between groups were observed in body weight, total fat intake or total oil consumption throughout the study period. In the DG group, serum TG levels decreased 39.4% from 2.51 +/- 0.75 mmol/L to 1.52 +/- 0.28 mmol/L. Serum glycohemoglobin A(1c) (HbA(1c)) concentration also decreased 9.7%. In contrast, there were no changes in these variables in the control group. Serum total and HDL cholesterol were not affected in either group. These results indicate that DG oil may be useful as an adjunct to the standard diet therapy of fat restriction in the management of diabetics with hypertriglyceridemia.  相似文献

Using a random crossover design, we examined the effects of glyburide for 4 wk on glucose, insulin, lipid, and lipoprotein metabolism in 10 men with non-insulin-dependent diabetes (NIDDM) receiving dietary fish-oil concentrates containing omega 3 (n-3) fatty acids (8 g/d). Compared with glyburide alone, fasting plasma glucose concentrations increased with fish oil. Although glyburide with fish oil decreased fasting glucose concentrations, they did not return to baseline. Basal insulin concentrations were unaltered by fish oil without or with glyburide; however, postprandial insulin concentrations were decreased by fish oil. Although total cholesterol and triglyceride concentrations were unchanged, very-low-density-lipoprotein cholesterol concentrations decreased and low-density-lipoprotein cholesterol rose and apolipoprotein B concentrations trended higher. Thus, glyburide only partially rectified the impaired fuel homeostasis associated with fish-oil supplements in patients with NIDDM. Therefore, we do not recommend intake of fish oil concentrates containing n-3 fatty acids in patients with NIDDM.  相似文献

OBJECTIVES: To determine the sensitivity of air displacement plethysmography (APD) for evaluation of changes in body composition in normal subjects. DESIGN: Comparison of measurements with and without oil or water loads. SUBJECTS AND METHODS: Ten healthy volunteers were analyzed, without and with 1 l and 2 l of oil or water. The measured and true changes in fat mass and fat-free mass were compared by paired t-tests. A correlation study and a Bland & Altman procedure was performed on the 60 measurements of adiposity changes in 30 subjects carrying 0.5 l (n=8 x 2), 1 l (n=10 x 2) and 2 l (n=12 x 2) oil and water loads. RESULTS: Fat-free mass increased when the 10 subjects were carrying water. When they carried oil, fat mass increased, however, a approximately 0.5 kg increase of fat-free mass was also detected. Two liters loads led to distinct changes: +1.49+/-0.59 kg fat and +0.50+/-0.60 kg fat-free with oil and +0.37+/-0.57 kg fat and +1.70+/-0.56 kg fat-free with water (both P<0.001). Mixed loads (+1 l oil and 1 l water) led to detect +0.85+/-0.48 kg fat and +1.09+/-0.45 kg fat-free (both P<0.005 vs without load). For the 30 subjects analyzed thrice, measured changes in fat and fat-free mass were slightly underestimated (-15%, NS) but correlated with the true changes. Measured changes in adiposity were correlated with the true changes, with no bias as indicated by the Bland & Altman procedure. CONCLUSION: APD detects approximately 2 kg changes in fat or fat-free mass in small populations.  相似文献

OBJECTIVE: Spot urine samples were investigated to determine correlations between urinary creatinine and specific gravity, and intra- and inter-day variations other than gender- and age-dependence of urinary concentrations. METHODS: Urinary creatinine concentrations and specific gravity were determined in 534 spot samples (385 from men and 149 from women). Subjects' ages ranged between 18 and 68 years. Spot urine samples were also collected from 14 male subjects before and after a 1-week work-shift for the evaluation of intra- and inter-day variations of creatinine and specific gravity. RESULTS: In spot samples, creatinine concentrations ranged between 0.16 and 4.36 g/l and specific gravity between 1.002 and 1.037. A high correlation (r = 0.82, P < 0.001) was observed between creatinine and specific gravity; male subjects showed significantly higher values of creatinine (P < 0.001) than did female subjects (1.90 +/- 0.74 and 1.41 +/- 0.72 g/l, respectively) and specific gravity (1.023 +/- 0.006 and 1.020 +/- 0.007, respectively). In addition, creatinine but not specific gravity significantly decreased (P < 0.02) in subjects older than 50 years, compared with those under 40. CONCLUSIONS: Results confirm the gender-dependence of creatinine concentrations in spot specimens and also show age-dependence, indicating the need for these aspects to be considered when the range of acceptable samples is to be set. No significant intra- or inter-day variations were observed for the two parameters. Lastly, the possibility of a comparison of differently adjusted values was indicated by a conversion formula derived from adjustments to creatinine and the corresponding specific gravity of a hypothetical urinary value, as follows: specific gravity adjusted values = 1.48 x creatinine adjusted values.  相似文献

The aim of this study was to determine the metabolism and the tolerance of a new amino acid (AA) solution administered under conditions mimicking cyclical parenteral nutrition (PN) in humans. Eight healthy volunteers received peripheral PN for 10 h providing 10.5 mg N x kg(-1) x h(-1) and 2.0 kcal x kg(-1) x h(-1) (glucose-to-lipids ratio: 70/30%). For adaptation, a non-protein energy intake was increased progressively for 90 min; thereafter, AA infusion was started and maintained at a constant rate for 10 h. Plasma and urine concentrations of all the AAs were measured before, during and after the PN. For each given AA, the relation between plasma variations at the steady-state and infusion rate, plasma clearance (Cl), renal clearance (Clr), re-absorption rate (Reab) and, retention rate (Reten) were determined. The nitrogen balance (DeltaN) was calculated during the PN period. The results are presented as means+/-sem. All plasma AA concentrations decreased during the starting period of non-protein energy intake. The plasma AA concentrations reached a steady-state within 3 h upon AA infusion, except for glycine and lysine (6 h). At the steady state, the plasma concentrations of the infused AAs were closely correlated to their infusion rate (y= -18.3+1.5x, r(2)=0.92). The plasma glutamine concentration was maintained during the PN, which indicates that the solution might stimulate the de novo synthesis of this AA. When the PN was stopped, plasma levels of the AAs decreased, most of them returning to their basal levels, or significantly below for lysine (P<0.05), alanine (P<0.05), proline (P<0.01) and glutamine (P<0.05). No volunteer showed any adverse effect during the infusion period. DeltaN was: 0.8+/-0.5 gN/10 h. Metabolic characteristics for essential AAs were: Cl<0.5 l min(-1), Clr <1.5 ml x min(-1) Reab >or= 99%, Reten >or=99% and for non-essential AAs: Cl <0.6 l x min(-1) except aspartate (2.8+/-0.3 l x min(-1)), Clr < 3 ml x min(-1) except glycine (6.8+/-0.7), aspartate (8.2+/-3.5) and histidine (8.8+/-1.3); Reab >or= 98% except glycine (95+/-1), aspartate (94+/-2) and histidine (94+/-1), Reten >or=97% except histidine (94+/-1), glycine (95+/-3). These results indicate that in healthy subjects, the amounts of AAs provided by the new solution were well balanced for an intravenous administration, and so were well utilized without excessive urinary excretion. The present study provides useful metabolic parameters for a further evaluation in disease.  相似文献

目的 观察醋酸铅染毒对大鼠海马和小脑c fos基因表达及学习记忆功能的影响 ,同时观察阿胶、天麻的拮抗作用。方法 大鼠经亚慢性醋酸铅染毒 (高剂量 0 .2g·kg- 1 ·d- 1 ,低剂量 0 .1g·kg- 1 ·d- 1 ) ,同时设立天麻 (4g·kg- 1 ·d- 1 )、阿胶 (1g·kg- 1 ·d- 1 )单独和联合作用组 ,监测各组血铅浓度 ,观察各组大鼠学习记忆功能的变化 ,最后利用原位杂交技术观察学习记忆过程中各组海马CA3区及小脑c fosmRNA表达状况的变化。结果  (1 )高、低剂量的铅染毒均使大鼠血铅浓度增加 ,与对照组比较 ,差异均有显著性 (P <0 .0 1 )。 (2 )Y型迷宫试验中 ,染铅使大鼠学习达标前所受电击次数增加 ,与对照组比较 ,差异有显著性 (P <0 .0 1 ) ;天麻和阿胶单用可减少染铅大鼠学习达标前所受电击次数 ,与染铅组比较 ,差异有显著性 (P <0 .0 5) ,二者联合应用较之单独应用使电击次数减少更明显 ,与染铅组比较 ,差异有显著性 (P <0 .0 1 )。 (3)低铅 +天麻 +阿胶组标本切片上海马CA3区着色较深的c fos阳性细胞最为密集 ,与对照组接近 ,高铅 +天麻 +阿胶组也有较多染色较深的c fos阳性细胞 ,但分布不及低铅 +天麻 +阿胶组密集 ,高铅组、低铅组则罕见c fos阳性细胞 ,各天麻、阿胶单独拮抗组着色及密度分别介于各单纯  相似文献

目的应用上海水源水淀山湖藻类样品,优化微囊藻毒素(microcystin-RR,-YR,-LR)提取、纯化和HPLC检测方法。方法分别以5%乙酸和75%甲醇为提取溶剂,80%甲醇(加三氟乙酸)为洗脱溶剂,采用不同温度(室温和56℃)和离心条件(10000×g,30min和3500r/min,10min),提取和纯化样品中的微囊藻毒素。应用HPLC,以乙腈、水和TFA为流动相,梯度洗脱检测微囊藻毒素。结果5%乙酸提取冻干藻细胞时,56℃水浴加低速离心是一个简单有效提取MC-RR的方法;80%甲醇洗脱溶剂中加入三氟乙酸能明显增加微囊藻毒素的洗脱;淀山湖藻类样品中微囊藻毒素含量由高到低依次为MC-RR,MC-LR和MC-YR。结论提取冻干藻细胞中的微囊藻毒素,以75%甲醇为提取溶剂,80%甲醇+0·05%TFA为洗脱溶剂MC-YR和MC-LR提取效率高;5%乙酸提取MC-RR效率高。  相似文献

Newly hatched mummichog (Fundulus heteroclitus) were exposed in a 96-h static renewal assay to water-accommodated fractions of dispersed crude oil (DWAF) or crude oil (WAF) to evaluate if the dispersant-induced changes in aqueous concentrations of polycyclic aromatic hydrocarbons (PAH) affected larval survival, body length, or ethoxyresorufin-O-deethylase (EROD) activity. Weathered Mesa light crude oil (0.05-1 g/L) and filtered seawater with or without the addition of Corexit 9500 were used to prepare DWAF and WAE At 0.2 g/L, the addition of dispersant caused a two- and fivefold increase in the concentrations of total PAH (sigmaPAH) and high-molecular-weight PAH (HMWPAH) with three or more benzene rings. Highest mortality rates (89%) were observed in larvae exposed to DWAF (0.5 g/L; sigmaPAH, 479 ng/ml). A reduction in body length was correlated with increased levels of sigmaPAH (r2 = 0.65, p = 0.02) and not with HMWPAH. The EROD activity increased linearly with HMWPAH (r2 = 0.99, p = 0.001) and not with sigmaPAH. Thus, chemical dispersion increased both the sigmaPAH concentrations and the proportion of HMWPAH in WAF. Dispersed HMWPAH were bioavailable, as indicated by a significantly increased EROD activity in exposed mummichog larvae, and this may represent a significant hazard for larval fish.  相似文献

A total of 29 tank cleaners and 31 referent controls participated in the study. In most cases, the tank cleaners were employed in small companies, usually specialized subcontractors such as firms only working in refineries cleaning oil tanks and handling oil spills. The air concentrations of hydrocarbons (HCs) in tanks containing residuals from heavy fuel oil were generally low, unless the oil was still warm. Addition of light fuel oil to facilitate the cleaning of tanks containing viscous, heavy fuel oils resulted in total airborne HC levels of 1000-1500 mg/m3. High levels of HC were measured in tanks with low-boiling petroleum fractions (naphtha and light fuel oils) of 1000-2600 mg/m3 (maximum). Today, most cleaners use air-supplied respirators or air-purifying respirator cartridges inside tanks with petroleum products or other chemicals. The exception is small firms handling fuel oils for heating purposes where only 50% of the workers use protective equipment regularly; the other workers only occasionally use protective equipment even if the air concentrations of HC are high. Protective equipment is rarely used in small, domestic tanks. Measurements of heart rate showed that tank cleaning is, at times, a highly strenuous job. No differences between tank cleaners and controls were found with respect to spirometry, liver enzymes, or frequency of micronuclei. Acute intoxications were not frequently reported in this group. However, this investigation may underestimate the true risk, as it is a cross-sectional study that found that exposures were highly variable, both quantitatively and qualitatively. We evaluated the susceptibility of Galerucella calmariensis, a species used to control purple loosestrife (Lythrum salicaria), to three mosquito control larvicides. Larvae and adults were fed loosestrife cuttings dipped in Abate (< or = 375 g x L(-1)), Altosid (< or = 250 g x L(-1)), and Bacillus thuringiensis var israeliensis (Bti) (< or = 110 g x L(-1)). Eggs on cuttings were dipped in the same concentrations. Pupae were immersed in Abate and Altosid solutions (< or = 474.4 microg x L(-1) and < or = 1,169.2 microg x L(-1), respectively). Hatching success of eggs dipped in Abate (> or = 3.75 g x L(-1)) was reduced significantly and survival was significantly lower among larvae and adults eating cuttings dipped in Abate (> or = 0.17 g x L(-1) and > or = 2.27 g x L(-1), respectively). Hatching success of eggs dipped in Altosid (> or = 2.52 g x L(-1)) was reduced significantly. With exposure to Altosid, larval survival to pupation and adult emergence was reduced significantly at concentrations of > or = 2.92 g x L(-1) and > or = 0.63 g x L(-1), respectively. Altosid (> or = 0.23 g x L(-1)) also delayed the onset of pupation and adult emergence among larvae that survived to pupate. Larvae that survived with exposure to Altosid (> or = 1.72 g x L(-1)) grew to 70% larger than those exposed to lower concentrations. Pupal survival was unaffected with exposure to Abate and Altosid and adult survival was unaffected with exposure to Altosid. Bacillus thuringiensis var israeliensis did not adversely affect any life stage of G. calmariensis. The mean Abate concentration on cuttings exposed to operational spraying was in the range that reduced egg hatchability and adult survival but was higher than concentrations that caused complete mortality of larvae. The mean Altosid concentration on cuttings exposed to operational spraying was in the range that reduced hatching success in eggs and delayed pupation and adult emergence of larvae.  相似文献

The object of the present study was to evaluate the physiologic mechanisms regulating erythropoiesis by monitoring the changes in reticulocyte count in the circulation and the formation of erythroblastic islands in the bone marrow following hypertransfusion and dehydration. Forty-eight male Wistar rats (110-130 grams) were transfused with 70% isogenic suspension of washed and packed red cells at a dose of 2.5 ml/100 g body weight intravenously on two consecutive days (absolute erythrocytosis) or deprived of water for three days to produce relative erythrocytosis and then injected i.v. with saline at a dose of 2.5 ml/100 g body weight with water provided at libitum. Hematocrit was measured using the microhematocrit method. Reticulocytes were enumerated using the new methylene blue stain. Plasma EPO levels were measured radioimmunologically and a bone marrow suspension was prepared for morphological examination and absolute erythroblastic islands determination. We observed a marked suppression of erythropoiesis following the transfusion as evidenced by the elevation in hematocrit values up to 0.64 +/- 0.04, a decrease in reticulocyte count and plasma EPO reduction--13.5 +/- 3.4 mU/ml (P < 0.05) and a suppression of EO formation in the bone marrow (P < 0.01). The hematocrit of the dehydrated rats was elevated during the first 24 hours (P < 0.001). Reticulocytes, as well as EO, decreased on day 3 down do 53.6 x 10(9)/l +/- 9.8 x 10(9)/l (P < 0.001) and 153.6 x 10(3) +/- 21.5 x 10(3)/femur (P < 0.001), respectively. The EPO level was measured simultaneously--16.3 +/- 4.2 mU/ml versus 24.6 +/- 5.3 mU/ml at day 1 (P < 0.001). Following rehydration, a fast increase in the hematocrit value with a concomitant increase in EO and the reticulocyte counts was observed. Our results show that the absolute and relative changes in hematocrit values are paramount in the feedback mechanism for erythroid homeostasis.  相似文献

Cadmium is a toxic element ubiquitous in the environment, which damages biological systems in various ways. The major source of cadmium exposure is food. High cadmium content in the soil leads to high cadmium concentrations in certain plants such as grains (above all surface layers and germs), oil or non-oil seeds, fruit and vegetables. These food commodities are the crucial components of a vegetarian nutrition. Blood cadmium concentrations were measured in two non-smoking population groups: the vegetarian group (n = 80) and the non-vegetarian (control) group of general population on traditional mixed diet (n = 84). The significantly higher blood cadmium content (1.78 +/- 0.22 vs. 0.45 +/- 0.04 microg/l) was measured in vegetarian group. Healthy risk values > 5 microg/l were found in 6 vegetarians vs. no non-vegetarian. The highest cadmium concentration (3.15 +/- 0.77 microg/l) was measured in vegan subgroup (plant food only, n = 10) and that value decreased with increasing animal food consumption (1.75 +/- 0.36 microg/l, lactovegetarian and lactoovovegetarian subgroup/added dairy products and eggs, n = 41/, 1.34 +/- 0.21 microg/I, semivegetarian subgroup /as a previous subgroup and added white meat, n = 291). Risk vegetarians vs. non-risk vegetarians consume significantly higher amounts of whole grain products, grain sprouts and oil seeds. Blood cadmium content is directly influenced by age (r = 0.32, p < 0.001), by whole grain product intake (r = 0.66, p < 0.001) and by duration of vegetarianism (r = 0.5, p < 0.001). Oxidative stress plays a major role in chronic cadmium induced hepatic and renal toxicity as well as in other consequences of cadmium injuries. Vegetarians have significantly higher plasma concentrations of natural antioxidants. The sufficient antioxidative protection against cadmium induced free radical formation in vegetarians may inhibit the harmful effects of greater cadmium intake from plant food.  相似文献

The intake of (n-3) long-chain PUFA is associated with a decreased risk of fatal myocardial infarction. Whether this effect is attributable to the effects of docosahexaenoic acid [22:6(n-3) (DHA)] on vascular function, particularly at intakes <1 g/d, is unknown. We report a randomized, double-blind, crossover, placebo controlled trial of 0.7 g DHA/d as a purified algal derived triacylglycerol (1.5 g/d) vs. placebo (1.5 g olive oil/d) on vascular function and biochemical indices of endothelial dysfunction in 38 healthy men and women, aged 40-65 y. Each treatment phase lasted 3 mo, separated by a 4 mo washout period. Supplementation increased the proportion of DHA in erythrocytes lipids by 58%, compared with placebo. Arterial compliance and endothelium independent and dependent responses, plasma concentrations of C-reactive protein, soluble thrombomodulin, E-selectin, von Willebrand factor antigen, and urinary microalbumin and isoprostane excretion were unaffected by treatment. Diastolic blood pressure decreased by 3.3 mm Hg (95% CI -6.1 to -0.6; P = 0.01). Heart rate tended to be 2.1 beats/min lower after DHA treatment than after the placebo period (P = 0.15). The results indicate that a moderate increase in the daily intake of DHA to approximately 0.7 g DHA lowers diastolic BP but does not influence indices of endothelial function or arterial stiffness in the short term.  相似文献

BACKGROUND: The intake of 10 g/day of short-chain-fructo-oligosaccharides (sc-FOS) has been shown to increase significantly bifidus counts and to produce high amounts of short-chain fatty acids (SCFA), presumed to influence glucose and lipid metabolism. AIM: To evaluate the effects of moderate intake of sc-FOS on glucose and lipid metabolism in individuals with mild hypercholesterolaemia. Design: A randomized double-blind sequential cross-over study. SUBJECTS AND METHODS: Thirty subjects of both genders (20 M/10 F), mean age 45.5+/-9.9 years (M+/-SD), BMI 26.6+/-2.2 kg/m(2), with plasma cholesterol >5.17 and <7.76 mmol/l and plasma triglycerides <3.45 mmol/l, participated in the study. The study was performed after a wash-out period of 1 month and a run-in period of 1 month to stabilize patients on a standard diet (CHO 50%, fat 30%, protein 20%, fibre 20 g/day) plus placebo (maltodextrine plus aspartame 15 g/day). At the end of run-in, subjects were randomly assigned to receive sc-FOS (Actilight) (10.6g/day) or placebo (maltodextrine plus aspartame 15 g/day) with tea and/or coffee for a duration of 2 months and thereafter switched to the other treatment for additional 2 months. Plasma glucose, total and lipoprotein (VLDL, LDL, HDL) cholesterol and triglyceride concentrations were measured in the fasting state at the end of run-in and of each treatment period. At the end of the two treatment periods, patients consumed a standard test meal (protein 15%, carbohydrate 34%, fat 51%, kJ 3988) 1h after the administration of 5.3g of sc-FOS or placebo; plasma glucose, insulin, free fatty acid (FFA) and triglyceride responses to the test meal were evaluated. RESULTS: No significant difference in fasting parameters was detected between the two treatments. After sc-FOS and placebo plasma cholesterol levels were, respectively, 6.47+/-0.70 and 6.44+/-0.78 mmol/l (n.s.) and plasma triglycerides were 1.53+/-0.71 and 1.56+/-0.53 mmol/l (n.s.). No significant differences were observed in cholesterol and triglyceride content of VLDL, LDL and HDL and in plasma Apo A1 levels; conversely, fasting plasma Lp(a) concentrations were significantly increased after sc-FOS (37+/-38 vs. 33+/-35 mg/dl; P<0.005). Postprandial responses of glucose, FFA and triglycerides were not significantly different between sc-FOS and placebo, while postprandial insulin response (incremental area) was significantly reduced after sc-FOS compared to placebo (14,490+/-7416 vs. 17,760+/-7710 pmol/l x 300 min; P<0.02). CONCLUSIONS: A moderate intake of sc-FOS has no major effects on lipid metabolism, both in the fasting and in the postprandial period, in individuals with mild hypercholesterolaemia. A small but significant increase of Lp(a) concentrations was observed with sc-FOS consumption together with a reduction of the postprandial insulin response; however, the clinical relevance of these small effects is unclear.  相似文献
