A cytogenetic analysis was performed in 28 mercury-exposed subjects as well as eight control subjects from the same chloralkali plant and in 12 healthy controls from the “general population.” No chromosomal effects could be demonstrated in the mercury-exposed population in comparison with both control groups. Also no effect of smoking was observed. From the study it seems obvious that mercury, although it is capable of producing chromosomal damage even at low exposure levels, will not be harmful in this respect when adequate protection measures are taken.  相似文献

Cytogenetic analyses of mice exposed to dichloromethane   总被引:2,自引:0,他引:2  
Chromosome damage was studied in female B6C3F1 mice exposed to dichloromethane (DCM) by subcutaneous or inhalation treatments. No increase in the frequency of either sister chromatid exchanges (SCEs) or chromosome aberrations (CAs) in bone marrow cells was observed after a single subcutaneous injection of 2,500 or 5,000 mg/kg DCM. Inhalation exposure to DCM for 10 days at concentrations of 4,000 or 8,000 ppm resulted in significant increases in frequencies of SCEs in lung cells and peripheral blood lymphocytes, CAs in lung and bone marrow cells, and micronuclei (MN) in peripheral blood erythrocytes. Lung cell CAs and blood erythrocyte MN reached frequencies of approximately two times control levels. Following a 3-month inhalation exposure to 2,000 ppm DCM, mice showed small but significant increases in lung cell SCEs and peripheral blood erythrocyte MN. These findings suggest that genotoxicity may play a role in the carcinogenicity of DCM in the lungs of B6C3F1 female mice.  相似文献

A cytogenetic study was performed in lymphocytes of hospital workers occupationally exposed to X- and gamma-rays using the micronucleus centromere assay. A comparison of the data for the exposed group and an age-matched group of non-exposed hospital workers showed a significant (P < 0.05) increase in centromere-positive micronuclei for the radiation workers, while no effect on centromere-negative micronuclei was present. The observed systematic increase in micronucleus frequency with age was mainly due to increased chromosome loss, reflected in the centromere-positivity of the micronuclei. The micronucleus frequencies were 40% higher in females than in males, which can again be attributed to higher chromosome loss. Two exposed individuals showed exceptionally high micronucleus yields, 90% of which were centromere-positive. In situ hybridization with a centromeric probe for chromosome X shows that X chromosome loss is responsible for these high micronucleus yields. In the studied population, smoking had no significant effect on the micronucleus yields. The results obtained indicate that in contrast to the predominantly clastogenic action of acute exposure to ionizing radiation, the aneugenic properties of radiation may be important after long-term chronic low dose exposure.  相似文献

The genotoxic effects of occupational exposure to ionizing and non-ionizing radiation were investigated in 25 physicians and nurses working in hospitals and in 20 individuals working at radio-relay stations. Examination was conducted by chromosome aberration analysis of peripheral blood lymphocytes. The data showed that total number of chromosome aberrations in people exposed to ionizing and radio-frequency radiation (4.08 +/- 0.37 and 4.35 +/- 0.5 on 200 scored metaphases, respectively) were almost equally higher than those of non-irradiated subjects. The increase was in proportion to the number of individuals having more that 5-aberration/200 metaphases. Acentric fragments comprised the most frequently seen type of aberration. The average numbers in examined groups (11.8 x 10(-3) and 14.8 x 10(-3) per cell, respectively), were significantly higher than 4.2 x 10(-3), which was observed in controls, unexposed individuals. Dicentric fragments were also frequent (4.8 x 10(-3) and 6.25 x 10(-3), respectively, vs. 0.52 x 10(-3) in control). In contrast, the frequency of chromatid breaks increased only after ionizing radiation (3.8 x 10(-3) vs. 0.26 x 10(-3) in control). A positive correlation between the total number of chromosome aberrations and cumulative 6-years dosage was also found. The data emphasized the dangerous effects of prolonged exposure to both types of radiation and indicated that chromosomal aberration analysis should be obligatory for individuals working at radio-relay stations.  相似文献

This study was conducted to evaluate the cytogenetic effects in male workers exposed to mercury fulminate. A total of 29 male workers and 29 age- and sex-matched controls were examined. The mean mercury level in urine from the exposed workers was 123.2 +/- 54.1 micrograms/l compared with 39.2 +/- 11.1 micrograms/l in the control group. The difference was statistically significant (P less than 0.001). Metaphase chromosomes were studied. Micronucleated peripheral blood lymphocytes were also analyzed in cytochalasin B blocked binucleated lymphocytes. The percentage of metaphases with chromosomal aberrations was significantly higher (P less than 0.001) in the exposed group (6.1 +/- 2.3) compared to the control group (2.8 +/- 0.7). The chromosomal aberrations detected were in the form of gaps, breaks and fragments. A significant increase in the incidence of micronucleated lymphocytes was found among the exposed group (7.1 +/- 4.2) compared to the control group (5.4 +/- 2.2) (P less than 0.01). The detected chromosomal damage correlated neither with the duration of exposure nor with the urinary mercury level.  相似文献

Bhawalpur is a major cotton-growing area in Pakistan. Cotton picking in Pakistan is carried out by females and as a result of the intensive use of pesticides during the growing season these females are exposed to pesticide residues in the picking season. In the present study, peripheral blood was obtained from 69 cotton pickers and 69 unexposed females and used to assess the effect of pesticide exposure on genetic damage as well as on hepatic enzymes and serum cholinesterase. The subjects were of similar average age in workers and control groups (37.55 +/- 12.75 vs. 37.52 +/- 13.47, P > 0.05). Average exposure time of the picker females was 10.26 +/- 6.14 years. Subjects from the exposed group did not use any protective measures during their work activities. Levels of serum cholinesterase were lower, and levels of alkaline phosphatase, alanine aminotransferase, and aspartate aminotransferase were higher in the exposed workers as compared with the control group (P < 0.001). The exposed group exhibited significantly increased frequencies of binucleated cells with micronuclei (12.72 +/- 3.48 vs. 4.35 +/- 2.44, P < 0.001) and total number of micronuclei in binucleated lymphocytes (16.51 +/- 4.27 vs. 5.86 +/- 3.09, P < 0.001) in comparison with subjects of the control group. The binucleated cells with micronuclei frequency also seemed to increase with age in both the groups, however, the magnitude of increase was greater in exposed group than the control. Results from the present study indicate that occupational exposure to pesticide mixtures results in cytogenetic damage in exposed females.  相似文献

Among various cytogenetic changes stable chromosome aberrations (SCHA) seem to be the most significant for ageing and carcinogenesis. Being nonlethal they can persist through cell divisions and accumulate in time. We studied the age response of SCHA (translocations and insertions) in normal and radiation exposed human populations. Two cohorts of people at the age range of 3--72 years were studied: control (43 persons) and exposed to low doses of accidental irradiation due to Chernobyl accident and atomic bomb testing in Semipalatinsk (67 persons). FISH method was used for visualisation of chromosome aberrations. Metaphases from cultured lymphocytes were hybridised with biotinilated whole chromosome specific DNA probes for 1, 4 and 12 chromosomes, and with pancentromeric probe labelled with digoxigenin. The frequency of SCHA in lymphocytes increased as a quadratic function of donor age in both populations studied, being higher in exposed cohort as compared with control one. No age dependence for dicentrics was observed. The frequency of SCHA is a reliable biomarker of ageing in humans. Quadratic model of their age-response gives reasons to suggest that their increase is due to lower level of DNA repair or/and the genomic instability in older people. The exposure of people to low doses of ionising radiation accelerates the age-related increase of SCHA frequency.  相似文献

Widespread use of pesticides in agriculture represents a threat not only to the environment but also to human populations exposed to them. Many of these compounds are capable of inducing mutations in DNA and lead to several diseases including cancer. In the present study, cytogenetic damage in peripheral lymphocytes from 33 farmers of Oporto district (Portugal) exposed to pesticides was evaluated by means of micronuclei (MN), sister chromatid exchange (SCE) and chromosomal aberrations (CA). In addition, effect of polymorphic genes of xenobiotic metabolizing enzymes (GSTM1, GSTT1, GSTP1, CYP2E1 and EPHX1) was also evaluated. A non-exposed group from the same area and with same demographic characteristics without exposure to genotoxic compounds was studied and data obtained from both groups was compared. MN and SCE frequencies were significantly higher in the exposed group (P < 0.005). In what concerns CA results, no significant differences were observed. It was possible to relate a specific working environment (greenhouses) with higher levels of genetic damage. Use of personal protective equipment revealed to be important to prevent exposure and diminish genetic damage inflicted by pesticides. Allele frequencies of studied polymorphic genes obtained in this study are similar to the ones described by other authors for Caucasian populations. Despite the low number of subjects, results suggest that low mEH (microsomal epoxide hydrolase) activity as well as GSTT1 positive genotype are associated with increased cytogenetic damage.  相似文献

The use of agricultural chemicals without correct protection may lead to alterations in the genetic material of cells and the possible development of several types of tumors. The individual genetic variability in the enzymes which metabolize agricultural chemicals is also involved in this process, such as when the enzymes are not efficient in the detoxifying process of the organism, the metabolic subproducts accumulate, contributing to the tumorigenic process. Cytogenetic monitoring was carried out on a group of 20 male workers occupationally exposed to a mixture of pesticides in the town of S?o Jer?nimo da Serra, PR (Brazil). Student's t = test and Wilcoxon's test showed, respectively, that there was no significant difference between the chromosome aberration frequencies between the exposed and control groups and between the paired individuals. However, there was a significant difference in the two analyses regarding the mitotic index of the sampled individuals. Smoking and time of exposure to agricultural chemicals did not influence the cytogenetic responses obtained, but the mitotic index of the control individuals was higher than that of the exposed individuals from the different age groups. The GSTM1 gene polymorphism was 33% null. When statistical tests were carried out to assess the relationship of the GSTM1 genotypes with the chromosome aberrations and mitotic indexes, there was no significant difference. The VIII Region of Bio-Bio is a major fruit-growing area of Chile that makes intensive use of agricultural pesticides. The cytogenetic damage associated with exposure to mixtures of pesticides was evaluated by comparing peripheral blood lymphocyte micronucleus (MN) frequencies in a group of 64 female agricultural workers and 30 female controls. The exposed subjects worked during the spring and summer in thinning and pruning fruit trees and in harvesting and packing different fruits, such as raspberries, grapes, apples, and kiwis. They did not use any protective measures during their work activities. A significant increase in the frequency of binucleated cells with micronuclei (BNMN) was found in the exposed women as compared with the controls (36.94 +/- 14.47 vs. 9.93 +/- 6.17 BNMN/1000 BN cells; P < 0.001). The frequency of BNMN varied as a function of age in both the exposed and control groups, but no correlation was found between BNMN frequency and the duration of exposure. Also, smoking and other habits had no effect on MN frequency. Our study confirms that occupational exposure to pesticide mixtures results in cytogenetic damage.  相似文献

Sister chromatid exchanges (SCE) and chromosome aberrations(CA) were studied in the lymphocytes of 70 male agriculturalworkers occupationally exposed to several pesticides and 69matched controls, without indication of exposure to pesticides,from ‘El Maresme’ (Barcelona, Spain). Cytogenetic study of 30 bladder carcinomas confirmed the heterogeneity and the complexity of the karyotypic picture in this type of tumor. Presence of numerical and/or structural chromosome aberrations was observed in all tumors. Clonal abnormalities were found in 19 cases. Chromosomes most frequently involved in changes were chromosome #1, #3 and #11(36.6%, 26.6%, and 20% of the cases respectively). Trisomy 7 and monosomy 9 were the sole abnormalities in one case each.  相似文献

The effect of carbon dioxide (CO2) and Nd-YAG high-energy lasers on the formation of peritoneal adhesions was studied using light, scanning, and transmission electron microscopy in albino rats. Both types of lasers initiate the development of peritoneal adhesions similar in structure. The intensity of adhesion formation is markedly lower in the case of Nd-YAG laser as compared to the action of CO2 laser of the same power. It is well known that Fanconi anemia (FA) patients show a hypersensitivity to the effect of cross-linking agents such as mitomycin C and diepoxybutane, while the sensitivity of these patients to ionizing radiation is controversial. Fanconi anemia heterozygotes do not show a hypersensitivity to the above-mentioned agents. However, bleomycin it is used to identify mutagen sensitive individuals, especially among head and neck cancer patients. We present here a preliminary study in which the mean frequencies of bleomycin-induced chromatid breaks (ctb) from three FA heterozygotes (X = 0.90, range 0.80-1.01) and 11 controls (X = 0.40, range 0.21-0.66) differ significantly (P<.001), indicating a high sensitivity to bleomycin of G(2) lymphocytes from these three FA heterozygotes. An increased sensitivity was not observed after exposure of G0 lymphocytes to 2 Gy of ionizing radiation.  相似文献

Although several cytogenetic biomonitoring studies on workers exposed to pesticides have been reported, there is only limited information on this topic from developing countries where pesticides have been widely used over the years. People in developing countries are at higher risk from exposure, due to poor working conditions and a lack of awareness of the potential hazards during manufacturing and application of the pesticides. The present study has assessed the genotoxic effects of pesticides on workers involved in the pesticide manufacturing industry. Subjects in the exposed group (29) were drawn from workers at a pesticide production plant in district Multan (Pakistan). The control group (unexposed) composed of 35 individuals from the same area but was not involved in pesticide production. Liver enzymes, serum cholinesterase (SChE), micronucleus assay and some haematological parameters were used as biomarkers in this study. A statistically significant (P < 0.001) increase in levels of alanine aminotransferase, aspartate aminotransferase and alkaline phosphatase was detected in exposed workers with respect to the control group. There was a significant (P < 0.001) decrease in the level of SChE in the exposed group. Exposed individuals exhibited cytogenetic damage with increased frequencies (P < 0.001) of binucleated cells with micronuclei and total number of micronuclei in binucleated lymphocytes in comparison with subjects of the control group. A decrease (P < 0.001) in cytokinesis block proliferation index similarly demonstrates a genotoxic effect due to pesticide exposure. The results indicate that the pesticide industry workers have experienced significant genotoxic exposure. This study highlights the risk to workers in the pesticide manufacturing industries of developing countries such as Pakistan and the need for implementation of suitable safety measures to prevent/limit exposure to harmful toxins.  相似文献

We report the first cytogenetic investigation of cranial chordoma. Three cranial chordomas were examined, two of which could be further histopathologically classified as chondroid chordomas. In addition, we have included a case of chordoma of a cervical vertebra to compare the cytogenetic abnormalities. Diagnosis was made at histological and immunohistochemical levels. The three cases of cranial chordoma showed a normal karyotype, while one vertebra showed 46,XY,t(6;11)(q12;q23). Chordomas, particularly those containing cartilage, have to be distinguished from chondrosarcomas of the skull base. Such a distinction is normally based on expression of epithelial markers which usually are lacking in chondrosarcoma. Cytogenetic investigation may eventually prove to be useful in the distinction of the two lesions, if chromosome anomalies are consistently absent in chordoma, although some chondrosarcomas may also present a normal karyotype. Such a distinction has clinical implications because chondroid chordomas show better survival, whereas chondrosarcomas show a propensity to infiltrate the surrounding tissues.  相似文献
