In the present work we have evaluated the levels of polychlorinated biphenyls (PCBs) and organochlorine pesticides (OCPs) in 61 commercially available brands of cheese (54 conventional and 7 organic) to estimate their relevance as a source of organochlorines. Our results showed that hexachlorobenzene, α-HCH, dieldrin, p,p′-DDE, and PCBs 153 and 180 were present in most of the samples independent of the cheese type. The concentration of OCPs was low for both types of cheese, although organic had lower concentrations than conventional. The estimated daily intake (EDI) of OCPs was lower than the tolerable daily intake (TDI). The levels of PCBs in cheese were also low; however, there were higher levels of PCBs in organic than in conventional brands. Levels of dioxin- like PCBs (DL-PCBs) in both types of cheese reached concentrations in the 75th percentile higher than 3 pg WHO–TEQ/g fat, and above 100% of the levels established by the EU. People consuming the most contaminated brands could have an EDI well above the recommended TDI (2 pg WHO–TEQ/kg bw/day). These results are of concern as the adverse health effects exerted by dioxin-like compounds are well known.  相似文献

Abstract

Polycyclic aromatic hydrocarbons (PAHs) are a group of pollutants which occur in considerable amounts in the environment and food. In this study, a meta-analysis study was conducted on PAHs concentrations in fish in different parts of the world. The results showed that the PAHs were observed and quantified in fish in the most considered studies. The maximum and minimum concentration of PAHs was found in Cynoglossus Bilineatus fish of Persian Gulf (3970?ng/kg) and Cyprinus Carpio fish of Caspian Sea (0.004?ng/kg), respectively. These values are below the maximum value currently allowed by European Union regulations (12.0?μg/kg wet weight).  相似文献

A gas chromatography coupled with tandem mass spectrometry (GC-MS/MS) method is developed to determine 18 representative polycyclic aromatic hydrocarbons (PAHs) in cosmetics, including Benzo[a]pyrene (BaP) and others. The method offers high sensitivity and selectivity under selected reaction monitoring (SRM) mode to satisfy the requirements of both quantitation and qualitation. The extraction solvent system used in this study is acetone/hexane 1:1 (v/v) and other purification procedure is unnecessary. The linearities of 18 PAHs are validated in different concentration in the range of 0.25–20 ng/mL individually with coefficient correlation (r) higher than 0.996. The recoveries for spiking 3 different concentrations are from 87.40% to 120.44% for 18 PAHs and the coefficient of variation (CV) are below 12.32%. Limit of quantification (LOQ) of 18 PAHs is in the range of 0.05–0.2 mg/kg. A matrix enhancement effect is observed and can be compensated with deuterated internal standard. The method has been successfully applied to 73 samples, over 40 of them are lipsticks. The results show none of the samples detect Benzo[a]pyrene (BaP) and Dibenzo[a,h]anthracene (DBA), both are classified as the most carcinogenic. 8 PAHs are detected and the average value between 0.08 and 0.27 mg/kg. This study offers a sensitive and simple method to analyze 18 representative PAHs successfully and can be applied to cosmetic products and raw materials.  相似文献

The concentrations of 16 polycyclic aromatic hydrocarbons (PAHs) were determined in samples of foodstuffs widely consumed by the population of Catalonia, Spain. All samples were randomly acquired in 12 cities of Catalonia between March and June of 2006, and analyzed by gas chromatography-mass spectrometry. The dietary intake of total and carcinogenic PAHs was estimated for various age/gender groups of population. In order to determine the temporal trend on the dietary exposure to PAHs, the current results were compared with those of a previous survey (2000). The highest individual PAH levels corresponded to phenanthrene (29.66mug/kg), naphthalene (25.87mug/kg) and fluoroanthene (13.66mug/kg), while the lowest levels were benzo[a]pyrene (1.28mug/kg), benzo[k]fluoranthene (1.31mug/kg) and indeno[1,2,3-c,d]pyrene (1.44mug/kg). According to food groups, the highest levels of total PAHs were detected in meat and meat products (25.56mug/kg), oils and fats (23.48mug/kg), and cereals (20.44mug/kg). For an average male adult (70-kg body weight), the current dietary intake of the sum of PAHs was higher (12.0mug/day) than that found in our 2000 survey (8.4mug/day).  相似文献

The presence of polycyclic aromatic hydrocarbons (PAHs) in three brands of smoked marc spirits, the only actually on the market, was studied and compared with literature PAHs concentrations reported for whiskies. In two brands of smoked marc spirit the higher-molecular-weight PAHs concentrations (5 and 6 rings PAHs) resulted higher (0.09-0.3 microg L (-1)) than in whiskies (0.00-0.02 microg L(-1)) and consequently higher resulted the carcinogenic potential expressed using the toxicity equivalent factor (TEF).  相似文献

In this study, the dietary intake of 16 polycyclic aromatic hydrocarbons (PAHs) by the population of Tarragona County (Catalonia, Spain) was assessed using the duplicate diet approach. Duplicate diet samples, prepared as per consumption, were collected during September 2010 in various restaurants offering a variety of daily menus (breakfast, lunch, and dinner). For analysis of PAHs, a total of 90 composite samples were prepared. Analytical procedure of PAHs was performed by means of gas chromatography/mass spectrometry. Intake calculations were made for the standard male adult population. The highest intakes corresponded to acenaphthylene (12.7 μg/day), acenaphthene (12.4 μg/day), and fluorene (11.9 μg/day), while the lowest intake corresponded to dibenz[a,h]anthracene (0.12 μg/day), being also comparatively low those of pyrene, benzo[b]fluoranthene + benzo[j]fluoranthene, benzo[a]pyrene and benzo[ghi]perylene (0.13 μg/day in all cases). The results were compared with data from previous total diet studies (TDS) recently performed in the same geographical area. In the present study, the estimated mean dietary intake for a standard male adult living in Catalonia was 59.2 μg/day, a value notably higher than that found in our recent TDS (6.72 μg/day). However, it is essential to remark that important methodological differences exist between both surveys, reflecting that calculation methods should be similar when the purpose is to compare results from different surveys. In general terms, we conclude that for PAHs, duplicate diet studies may be a good alternative to total diet studies, especially when there are important economical limitations to perform a suitable TDS. The costs associated to the former may be notably lower, as they do not require such an extensive number of samples for chemical analysis. Moreover, a duplicate diet approach may even be more realistic, as cooked foodstuffs are used for dietary exposure assessment.  相似文献

The formation of condensed ring polycyclic aromatic hydrocarbons (PAHs) from the pyrolysis of ground tobacco in helium over the temperature range of 350-600 degrees C was investigated. PAH yields in the ng/g range were detected and the maximum yields of all PAHs studied including benzo[a]pyrene (B[a]P) and benzo[a]anthracene (B[a]A) occurred between 500 and 550 degrees C. The pathway to PAH formation in the 350-600 degrees C temperature range is believed to proceed via a carbonization process where the residual solid (char) undergoes a chemical transformation and rearrangement to give a more condensed polycyclic aromatic structure that upon further heating evolves PAH moieties. Extraction of tobacco with water led to a two fold increase in the yields of most PAHs studied. The extraction process removed low temperature non-PAH-forming components, such as alkaloids, organic acids and inorganic salts, and concentrated instead (on a per unit weight basis) tobacco components such as cell wall bio-polymers and lipids. Hexane extraction of the tobacco removed lipophilic components, previously identified as the main source of PAH precursors, but no change in PAH yields was observed from the hexane-extracted tobacco. Tobacco cell wall components such as cellulose, hemicellulose, and lignin are identified as major low temperature PAH precursors. A link between the formation of a low temperature char that evolves PAHs upon heating is established and the observed ng/g yields of PAHs from tobacco highlights a low temperature solid phase formation mechanism that may be operable in a burning cigarette.  相似文献

Grilling muscle foods involves high temperatures that lead to production of cooking toxicants, such as heterocyclic aromatic amines (HAs) and polycyclic aromatic hydrocarbons (PAHs). To obtain realistic exposure levels of these two groups of mutagens analyses of the same samples using similar separation/detection techniques were performed. HAs and PAHs were quantified in well-done meat and fish samples grilled with wood and coconut shell charcoal at 200 °C. Quantitative HAs and PAHs profiles were different for beef and salmon using the same type of charcoal. Higher levels of HAs and PAHs were found in salmon samples. No significant differences were observed for HAs and PAHs in beef samples grilled with both charcoal types, whereas salmon grilled with coconut shell charcoal presented significantly lower amounts of HAs and PAHs than salmon grilled with usual wood charcoal. Continuous barbecuing with the same charcoal shown that combustion of fat that dropped along the grilling period contributed to higher formation of HAs and PAHs. Special attention must be given to the intake of barbecued foods since high amounts of HAs and PAHs can be taken in a single meal.  相似文献

The concentration of 16 polycyclic aromatic hydrocarbons (PAHs) was determined in traditional dry/fermented sausage along distinct stages of processing under two different technological procedures (traditional and modified processes). The influence of product’s position in the smoking room, on the variation of contaminants and in their migration dynamics from the outer into the inner part, was also followed up. Raw material mixtures presented expressive total PAH values, 106.17 μg kg−1 in wet samples and 244.34 μg kg−1 in dry mater (DM), expressing the frequent fire woods occurred in the regions pigs were extensively reared. Traditional processing produced a higher (p < 0.01) total PAH levels comparatively to modified/industrial procedures, with mean values reaching 3237.10 and 1702.85 μg kg−1 DM, respectively. Both, raw materials and final products, showed PAH profiles with light compounds representing about 99.0% of the total PAHs, mostly accounted by those having two rings (naphthalene-27.5%) or three rings (acenaphtene-16.9%; fluorene-27.1%; phenanthrene-19.5% and anthracene-3.9%). The benzo[a]pyrene (BaP) accumulated in traditional and modified processed products never surpassed the limit of 5 μg kg−1 established by the EU legislation. PAHs in products hanged in bars closer to heating/smoking source speed up their transfer from the surface/outer portion to the inner part of the product.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are one of the most widespread organic environmental pollutants that pose a potential risk to human health via marine biota. Toxic, carcinogenic and mutagenic properties of PAHs are studied in several researches. The toxicities (embryolethality) of 24 polycyclic aromatic hydrocarbons (PAHs) were determined in chick embryos using a 72-h test. The substances, dissolved in peanut oil, were injected into the air sacs of eggs preincubated for 7 days. LD50 values were determined for the four most toxic of the 24 compounds. Benzo [k] fluoranthene proved to be the most potent, with an LD50 of 14 g (56 nmol)/kg egg. Dibenz[a,h]anthracene, benz[a]anthracene and benzo[b]naphtho[2,3-d]thiophene were a few times less toxic [LD50=39 g (140 nmol)/kg, 79 g (349 nmol)/kg and 82 g (350 nmol)/kg, respectively]. The LD50 of benzo [k] fluoranthene was only about 5 times higher than that previously found for the most potent coplanar polychlorinated biphenyl (PCB), 3,3,4,4,5-pentachlorobiphenyl [LD50=3.1 g (9.4 nmol)/kg], in the same kind of test. The toxicities of 18 of the PAHs in this study have also been evaluated previously using a 2-week test in chick embryos. Dibenz[a,h]anthracene, which had not been studied earlier in the 2-week test, proved to be almost as toxic as previously found for benzo [k] fluoranthene in that test. Several of the PAHs studied induced EROD activity in chick embryos, and, in general, the most toxic PAHs were also the most potent inducers of EROD. The highest enzyme activities were found after treatment with indeno[1,2,3-c,d]pyrene (12 times the control value) and dibenz[a,h]anthracene (8 times the control value). However, due to the high toxicity of dibenz[a,h]anthracene, the dose used was 7 times lower than that of indeno [1,2,3-c,d]pyrene. Following injection of PAHs on day 7, the EROD activities on day 10 were considerably lower than those obtained after a corresponding treatment with coplanar PCBs in an earlier study. Of the PAHs studied, some exhibited very high embryotoxicity. The most toxic PAHs induced EROD activity, suggesting that their toxicity was at least partly mediated via binding to the Ah receptor.  相似文献

 Cytochrome P450-dependent 7-ethoxyresorufin O-deethylase (EROD), 7-pentoxyresorufin O-dealkylase (PROD) and 7-ethoxycoumarin O-deethylase (ECOD) activities in 14-day-old chick embryo livers were determined 24 h after pretreatment with selected widespread aromatic environmental contaminants, including polychlorinated biphenyls (PCBs), polycyclic aromatic hydrocarbons (PAHs), hexachlorobenzene, and dialkylesters of phthalic acid, and compared with the inducing potencies of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD) and the coplanar and mono-o-chlorinated PCBs. The effects of other model inducers, i.e. phenobarbital and pyrazole, were also examined. Specificity of EROD induction was estimated with regard to contaminants frequently present in environmental samples and dose-response curves for EROD induction were determined. A strong induction (comparable with that by mono-o-chlorinated biphenyl treatment) by dibenzo[a,h]anthracene, benzo[k]fluoranthene or benzo[b]fluoranthene was found, but the maximal level of EROD activity inducible by TCDD was not achieved, partly due to the high toxicity of the tested PAHs. 3-Methylcholanthrene showed moderate inducing potencies; benz[a]anthracene, benzo[a]pyrene, chrysene and 2,2′,3,4,4′,5′-hexachlorobiphenyl appeared to be weak inducers. Other PAHs and PCBs tested, as well as hexachlorobenzene, dialkyl phthalates, phenobarbital and pyrazole had no marked effects on the EROD level. ECOD activities were increased non-specifically by TCDD, 3-methylcholanthrene, hexachlorobenzene and phenobarbital. A significant enhancement of PROD activity by TCDD and related inducers was observed, while phenobarbital induced the PROD activity only weakly; SDS-PAGE analysis showed that the chicken phenobarbital-inducible cytochromes P4502H with apparent molecular weights 50 kDa were not markedly induced by the TCDD- or 3-methylcholanthrene treatments. Inhibition of EROD and PROD by 9-hydroxyellipticine, a specific inhibitor of rat hepatic cytochrome P4501A1, revealed that PROD induction by TCDD and other P4501A-inducers was probably a result of a broader substrate specificity of chick embryo P4501A. Measurement of EROD activities in chick embryo liver is highly sensitive, specific and suitable for the determination of TCDD-type toxicity of new drugs, agrochemicals, and industrial pollutants. Polycyclic aromatic hydrocarbons (PAHs) are a group of over 200 different organic compounds that are mainly formed as the result of pyrolytic processes. For the general population, a number of studies have shown that, as for other environmental organic contaminants, the major route of exposure to PAHs is through food consumption. In this paper, the scientific literature concerning the concentrations of PAHs in food and the human dietary intake of these compounds, is reviewed. Unexpectedly, we have detected a considerable lack of scientific papers for countries such as Australia, Canada, Germany, or Japan, among many others, which seems rather unusual given the important scientific and social interest on the knowledge of the health risks of human exposure to environmental pollutants. The data from countries for which the human dietary intake of PAHs is available, show important differences among them, which would be explained by a number of factors, being probably the most important, the different methodologies used in the respective surveys. Regarding the influence of cooking on the levels of PAHs in foodstuffs, and the consequent intake, we conclude that it depends, not only on the particular cooking process, but even more on the specific characteristics of the respective food items.  相似文献

The aromatic hydrocarbon (AH) receptor mediates the induction of cytochromes P-450 (CYP) of the CYP1A subfamily caused by polycyclic aromatic hydrocarbons (PAHs). CYPIA induction by PAHs is accompanied by down-regulation of CYP2C11, the predominant CYP expressed constitutively in the liver of male rats. We performed a structure-activity relationship study with a series of PAHs of the anthracene class in order to determine if the AH receptor is involved in CYP2C11 down-regulation. Anthracene, benz[a]anthracene, dibenz[a,c]anthracene, dibenz[a,h]anthracene, 7,12-dimethyl[a]anthracene, as well as 2,3,7,8-tetrachlorodibenzo-p-dioxin and 3-methylcholanthrene decresed CYP2C11 immunoreactive protein levels to varying degrees in primary rat hepatocytes cultured on a laminin-rich extracellular matrix. The binding affinity of the PAHs for the rat liver cytosolic AH receptor correlated with the potency for transforming the cytosolic AH receptor to its DNA-binding form. In addition, the ability of the PAHs to suppress CYP2C11 correlated with both the AH receptor binding affinity and the AH receptor transformation potency. These results suggests the AH receptor plays a role in the down-regulation of CYP2C11 caused by PAHs.  相似文献

Certain polycyclic aromatic hydrocarbons (PAHs) have been reported to induce cytochrome P450 (CYP) 1A1 and 1A2. In the present study, the effects of six well-known PAHs, such as benzo[a]pyrene, benz[a]anthracene, dibenz[a,h]anthracene, chrysene, benzo[k]fluorancene and benzo[b]fluorancene, on the activities of hepatic and pulmonary CYP enzymes were investigated in male ICR mice. When mice were treated intraperitoneally with 3, 10 and 30 mg/kg of individual PAHs for 3 consecutive days, the activities of ethoxyresorufin- and methoxyresorufin-O-dealkylases were significantly and differentially induced in both liver and lung. Moreover, other CYP isozyme-associated monooxygenase activities were also induced significantly in liver and lung with characteristic induction profiles. Our present results suggest that individual PAHs might have inductive effects on CYP isozymes, and that the characteristic inductive effects of individual PAHs on certain CYP isozymes would be developed as a marker for determining exposure to certain PAHs.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are persistent pollutants and chemically a class of structurally similar chemical compounds characterized by the presence of fused aromatic rings. This research was undertaken to find out immunotoxic effects produced by pyrene, phenanthrene and fluoranthene. These chemicals were injected into developing chicks at three dose levels (0.2, 2 and 20?mg per kg) through allantioc route to rule out possible mechanisms involved in immunotoxicity. DNA adduct produced by PAHs in immune organs were analyzed by DNA adduct enzyme-linked immunosorbent assay (ELISA) kit and DNA damage was assessed by comet assay. A significant increase in the DNA adduct levels was found in thymus and bursa in 2?mg and 20?mg dose levels of pyrene, fluoranthene and phenanthrene treated groups, whereas those in spleen simulated the value of controls. Comet assay indicated that PAHs especially pyrene, fluoranthene and phenanthrene were capable of inducing increased level of comet parameters in thymus at all the dose levels. Bursa of Fabricius and spleen also showed a gradual rise in comet parameters corresponding to all dose levels, but the increase was more marked as in thymus. Thus, it can be concluded that DNA adducts produced by PAHs lead to single-strand breaks and reduced DNA repair, which ultimately begin a carcinogenic process. Hence, this experiment can be considered as a strong evidence of genotoxic potential of PAHs like pyrene, phenanthrene and fluoranthene in developing chicks.  相似文献

Polycyclic aromatic hydrocarbons (PAHs) are one of the main components of urban air pollution. The level of concentration PAHs and effects are depended on seasonal variations and amount of exposure to PAHs. Nowadays, PAHs are one of the important pollutants that threat the public health. The purpose of this study was to evaluate the polycyclic aromatic hydrocarbons of the outdoor air in Ahvaz, southwest Iran using active sampling system during the warm and cold seasons of 2016. Data collection was performed daily during the study period in three stations in Ahvaz citizens at industrial, high traffic and residential areas. Omni sampler and equipped with PTFE filters (8*10) were utilized for sampling in this study. Gas chromatography with mass spectrometry (GC/MS) were utilized for detection of PAHs concentration (ngm?3). Statistical SPSS software (SPSS version 16) was used to determine the effects of PAHs during warm and cold season. According to result this study, the industrial and residential areas had the highest and the lowest PAHs concentrations. Based on result this study, PAHs concentrations in three main areas at industrial, high traffic and residential were 7.021?±?3.45, 5.162?±?2.63 and 4.187?±?1.76 ngm?3, respectively. The results showed that the average concentration of polycyclic aromatic hydrocarbons in three main areas outdoor air in Ahvaz were more than recommended scales by international organizations. According to this study, the mean of PAHs concentration in different areas was significantly higher than standard. Results showed that the short-term exposure to PAHs during cold season were higher than the levels of exposure during warm season. The levels of exposure to PAHs can increased risk of health endpoint and the oxidative stress.  相似文献
