OBJECTIVE To know cardioprotective effect of U.lobataleaves extract on diabetic rat.METHODS This study uses control group post test only with male sprague dawley rats.Diabetic rats was induced by high fructose diet(HFD)and single dose streptozotocin 25mg·kg-1 bw intra peritoneal.The rat was administrated orally with water extract of U.lobataleaves in concentrations of 250,500 and 1000mg·kg-1 bw for 4 weeks.After scarifying,heart organ were collected and then superoxyde dismutase(SOD)heart level,malondialdehyda(MDA)and tumor necrosis factor-alpha(TNF-α)were examined.The data was analyzed using ANOVA test continued with LSD test(P<0.05).RESULTS The oral administration of U.lobataleaves extract 250,500,and 1000mg·kg-1 bw were able to increase SOD heart level about 40%,50% and 70% respectively compared to diabetic group(P<0.05),while the MDA heart level was decreased by 60%,90% and 110%(P<0.05)respectively.The supplementation of water extract from U.lobatain dose of 250,500 and 1000mg·kg-1 bw were also decrease TNF-αheart level approximately 20%,40% and 60% compared to control group(P<0.05).In diabetic groups,SOD heart level was decreased compared to normal group(P<0.05)while the MDA and TNF-αwere increased(P<0.05).CONCLUSION U.lobataleaves extract acts as cardioprotector on diabetic rats by increasing of SOD heart level,decreasing of MDA heart level and TNF-α.This effect may be related to active compounds that act as an antioxidant and anti-inflammatory in U.lobata extract.  相似文献

OBJECTIVE To investigate the potency of Urena lobata leaves extract on the inhibition of hepatic complication on diabetic rats.METHODS This study uses control group post test only with male Sprague dawley rats.Diabetic rats was induced by high fructose diet(HFD)and single dose streptozotocin 25mg·kg-1 bw intra peritoneal.The rat was administrated orally with water extract of U.lobataleaves in concentrations of 250,500 and 1000mg·kg-1 bw for 4 weeks.After scarifying,liver organ and blood were collected and then superoxyde dismutase(SOD)hepar level,malondialdehyda(MDA),tumor necrosis factor-alpha(TNF-α),serum glutamic oxaloacetic transaminase(SGOT)and serum glutamic piruvic transaminase(SGPT)were examined.The data was analyzed using ANOVA test continued with LSD test(P<0,05).RESULTS The oral administration of U.lobataleaves extract 250,500 and 1000mg·kg-1 bw were able to increase SOD hepar level about 90%,100% and 120%respectively compared to diabetic group(P<0.05),while the MDA hepar level was decreased by 40%,50% and 70% respectively(P<0.05),whereas the TNF-αhepar level was decreased by 30%,50% and 70%respectively(P<0.05).The supplementation of water extract from U.lobatain dose of 250,500 and 1000mg·kg-1 bw decrease SGOT level approximately10%,30% and 50% compared to control group(P<0.05),while the SGPT level was decreased by 10%,20% and 40%respectively(P<0.05).In diabetic groups,SOD heparlevel was decreased compared to normal group(P<0.05)whereas the MDA and TNF-αwere increased(P<0.05).Meanwhile SGOT level and SGPT were increased in diabetic group(P<0.05).CONCLUSION U.lobataleaves extract could inhibit hepatic complication on diabetic rats by increasing of SOD hepar level,decreasing of MDA hepar level,TNF-α,SGOT and SGPT.This effect may be related to active compounds that act as an antioxidant and anti-inflammatory in U.lobata extract.  相似文献

OBJECTIVE To investigate the effects of Cymbopogon citratus Stapf water extract on lipid profile and antioxidant status in streptozotocin-induced diabetic rats.METHODS Diabetes was induced by injection of streptozotocin(STZ,50mg·kg-1)intraperitoneally.Diabetic rats were divided into 5groups,consisting of 6rats.GroupⅠ,reserved as diabetic control,was administered distilled water and groupⅡ,reserved as positive control,was administered glibenclamide(10mg·kg-1·d-1)throughout the duration of the experiment.Those in groupⅢ,ⅣandⅤ were administered 250,500 and 1000mg·kg-1·d-1 of the extract,respectively for 28 d.RESULTS Treatment with 500 and 1000mg·kg-1·d-1 of the extract resulted in reduction of serum AST,ALT,serum cholesterol,triglycerides and LDL,whereas HDL was found to be increased compared with diabetic control rats(P<0.05).Moreover,increased serum activities of superoxide dismutases and catalase were found in diabetic rats treated with the extract whereas serum thiobarbituric acid reactive substance was decreased,in comparison with diabetic control rat(P<0.05).CONCLUSION Cymbopogon citratus Stapf water extract provides a benefit effect on serum lipid and antioxidant effect in diabetic rats.Thus,the extract may lower cardiovascular disease risk and others complications related to hyperlipidemia and oxidative stress in diabetic patients.  相似文献

目的研究银杏提取物对糖尿病大鼠的抗氧化能力。方法将36只2型糖尿病模型Wistar大鼠按血糖分层随机分为模型对照组、银杏叶提取物高和低剂量(6和3 mg/kg)组,每组各12只。另取12只健康Wistar大鼠作为正常对照。连续给药4周。测定大鼠体质量、心脏/体质量比,血清及心脏丙二醛(MDA)、还原性谷胱甘肽(GSH)、超氧歧化酶(SOD)和糖基化终产物(AGEs)含量,以及血清糖化血红蛋白(HbA1C)。结果与正常对照组相比,模型对照组体质量、血清和心脏GSH、SOD含量显著降低(P<0.01或0.05),而心脏/体质量比、血清HbA1C以及血清和心脏体MDA、AGEs含量显著升高(P<0.01或0.05);与模型对照组相比,银杏叶提取物高和低剂量组大鼠体质量、血清和心脏GSH和SOD含量显著升高(P<0.01或0.05),而心脏/体质量比、血清HbA1C含量以及血清和心脏体MDA、AGEs含量显著降低(P<0.01或0.05)。结论银杏叶提取物可降低糖尿病大鼠的心脏氧化应激反应,此作用可能与银杏叶提取物预防糖尿病心脏病有关。  相似文献

OBJECTIVE To investigate anti-hyperglycemic effect of ethanolic leaves extract,hexane and water of Urena lobata on diabetic rat induced by high fructose diets and streptozotocin.METHODS This study uses control group post test only with male spraque dawley rats.Diabetic rats was induced by high fructose diet(HFD)and single dose streptozotocin 25mg·kg-1 bw intra peritoneal.The rat was administrated orally with ethanolic leaves extract,hexane and water of U.lobatain concentrations of 500mg·kg-1 bw for 4weeks.Blood sample were collected from the tail vein at 0,15,30,60 and 120min after oral glucose administration and then blood glucose level were measured using a commercially available glucometer.The integrated area under the postprandial glucose curves(AUCs)was calculated by the trapezoidal method.Insulin serum level examined by rat insulin elisa kits.The data was analyzed using ANOVA test continued with LSD test(P<0.05).RESULTS The oral administration of ethanolic leaves extract,hexane and water of U.lobata were able to decrease glucose AUC about 40%,20% and 60% respectively compared to diabetic group(P<0.05),whereas the insulin level was increased by 2,2and 4-fold(P<0.05),respectively.In diabetic groups,glucose AUC was increased approximately 70% compared to normal group(P<0.05)while the insulin level were decreased by 14-fold(P<0.05).CONCLUSION U.lobataleaves extract could control the increase of blood glucose level after glucose administration by improve of insulin secretion.This effect may be related to active compounds that act as an anti-hyperglycemic and antioxidant in U.lobataleaves extract.  相似文献

目的研究葛根素注射液(黄酮类药物)对自发性糖尿病(GK)大鼠脂代谢的影响和主动脉血管内皮生长因子(VEGF)表达的干预。方法GK大鼠和Wistar大鼠随机分为正常对照组、糖尿病组、葛根素治疗组。治疗16周,观察大鼠的血糖、甘油三酯、胆固醇、低密度脂蛋白、高密度脂蛋白、糖化血红蛋白、氧化低密度脂蛋白;分离主动脉,HE常规染色,免疫组化检测主动脉内膜VEGF表达。结果葛根素能降低糖尿病大鼠的甘油三酯、胆固醇、低密度脂蛋白、糖化血红蛋白、氧化低密度脂蛋白;升高高密度脂蛋白(P<0.05);葛根素可降低主动脉VEGF(均P<0.05)表达。结论葛根素可通过调节血脂、降低VEGF的表达,有主动脉保护作用。  相似文献

目的:观察红杉醇对2型糖尿病大鼠脂代谢的影响.方法:采用高糖高脂饮食加腹腔注射小剂量链脲佐菌素(Streptozotocin,STZ)诱导2型糖尿病大鼠模型35只,灌服红杉醇(50、25及12.5 mg·kg-1· d-1)6周,每天1次.全自动生化分析仪测定血清TG、TC、低密度脂蛋白胆固醇(Low density lipoprotein cholesterol,LDL-C)含量;ELISA法检测血浆游离脂肪酸(Free fatty acids,FFA)、肝脏载脂蛋白A1(Apolipoprotein A1,Apo-A1)和载脂蛋白B1(Apolipoprotein B1,Apo-B1)含量;放射免疫法测定肝组织瘦素(Leptin)含量;MASSON染色观察肝脏病理改变.结果:红杉醇能显著降低模型大鼠血脂、FFA含量,下调肝组织Apo-B1及Leptin水平,上调肝组织Apo-A1的含量,减轻肝细胞脂肪变性,减少肝细胞间质和血管旁胶原沉积.结论:红杉醇具有调节2型糖尿病大鼠脂代谢、减轻脂肪肝和肝脏胶原沉积的作用.  相似文献

目的观察银杏叶提取物(GBE)对高脂血症大鼠血脂代谢及脂质过氧化反应的影响。方法高脂乳剂灌胃30d,同时给大鼠灌胃GBE(40,80,160mg·kg^-1),测定血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL—C)、高密度脂蛋白(HDL-C)的活性,测定血清一氧化氮(NO)、丙二醛(MDA)及超氧化物歧化酶(SOD)活性。结果GBE(80,160mg·kg^-1)组均能明显降低血清TC、TG、LDL—C及MDA的活性(P〈0.05或P〈0.01),并能显著升高血清HDL—C和NO、SOD的活性(P〈0.05或P〈0.01)。结论GBE能通过调节脂蛋白-胆固醇代谢,改善血管内皮功能、纠正自由基代谢紊乱、促进抗氧化酶活性等途径发挥调脂及抗氧化作用。  相似文献

The present study investigates the effect of oral administration of an aqueous Enicostemma littorale whole plant extract on some key carbohydrate metabolic enzymes and antioxidant defence in alloxan-induced diabetes in rats. Rats were rendered diabetic by alloxan (150 mgkg(-1) body weight) administration. Oral administration of E. littorale extract for 45 days increased the activity of hexokinase and decreased the activities of glucose 6-phosphatase and fructose 1,6-bisphosphatase significantly in the serum, liver and kidney of diabetic rats. The extract lowered the concentration of thiobarbituric acid reactive substances and lipid hydroperoxides significantly in brain and increased it significantly in heart in diabetic rats. E. littorale administration increased the concentration of reduced glutathione and the activity of glutathione peroxidase in diabetic rats. The activities of superoxide dismutase and catalase were increased significantly by E. littorale treatment in diabetic rats. The effect of a 2 g kg(-1) dose was greater than that of a 1 gkg(-1) dose. Insulin (6 units kg(-1)) normalized all the parameters in diabetic rats. Our study has provided evidence for the antidiabetic activity of E. littorale aqueous extract. This study can also be extrapolated to clinical studies in future.  相似文献

Hyperlipidaemia is an associated complication of diabetes mellitus. We recently reported that tetrahydrocurcumin lowered the blood glucose in diabetic rats. In the present study, we have investigated the effect of tetrahydrocurcumin, one of the active metabolites of curcumin on lipid profile and lipid peroxidation in streptozotocin-nicotinamide-induced diabetic rats. Tetrahydrocurcumin 80 mg/kg body weight was administered orally to diabetic rats for 45 days, resulted a significant reduction in blood glucose and significant increase in plasma insulin in diabetic rats, which proved its antidiabetic effect. Tetrahydrocurcumin also caused a significant reduction in lipid peroxidation (thiobarbituric acid reactive substances and hydroperoxides) and lipids (cholesterol, triglycerides, free fatty acids and phospholipids) in serum and tissues, suggesting its role in protection against lipid peroxidation and its antihyperlipidemic effect. Tetrahydrocurcumin showed a better effect when compared with curcumin. Results of the present study indicate that tetrahydrocurcumin showed antihyperlipidaemic effect in addition to its antidiabetic effect in type 2 diabetic rats.  相似文献

目的比较黄芩提取物中主要成分黄芩苷和汉黄芩苷在糖尿病大鼠和正常大鼠体内的药动学.方法腹腔注射链脲佐菌素建立糖尿病大鼠模型,灌胃黄芩提取物后取血分离血浆,采用HPLC-Uv法检测血浆中黄芩苷和汉黄芩苷浓度,以矩量法计算药动学参数,AUC(0-τ)的计算采用梯形法.采用黄芩提取物与大鼠粪便温孵,从肠道代谢研究黄芩苷和汉黄芩苷动力学改变的初步机制.结果与正常大鼠比较,糖尿病大鼠给予黄芩苷和汉黄芩苷后体内Cmax1明显增加[黄芩苷(6.07±0.95)vs.(17.01±3.60)μg·mL-1,P<0.01;汉黄芩苷(3.50±0.72)vs.(9.34±2.04)μg·mL-1,P<0.01],Cmax2明显增加[黄芩苷(1.61±0.18)vs.(7.39±3.04)μg·mL-1,P<0.01;汉黄芩苷(1.95±0.52)vs.(6.72±2.60)μg·mL-1,P<0.01],AUC(0-τ)也明显增加[黄芩苷(38.72±7.25)vs.(86.70±20.91)μg·mL-1·h,P<0.01;汉黄芩苷(39.20±12.10) vs.(69.40±24.20)μg·mL-1·h,P<0.01],粪便悬浮液中黄芩苷降解也加快(Ke0.087 vs.0.173 min-1).结论黄芩苷和汉黄芩苷在糖尿病大鼠与正常大鼠之间存在明显的药动学差异,在糖尿病大鼠肠道内代谢加快.  相似文献

Objective To study the effect of ginkgo biloba extract (GbE) on the expression of matrix metalloproteinase (MMP)-2 in kidneys of diabetic rates and to analyze the protecting mechanism of GbE on diabetic nephropathy. Methods Totally 24 SD rats of diabetic nephropathy induced by streptozotocin were randomly divided into two groups: diabetes group and treatment group with GbE, and 12 rats with placebo therapy were enrolled as the normal control group. Rats were killed after 4 ~ 8 weeks treatment and the expression of MMP-2 and type Ⅳ collagen were studies by immunohistochemistry and RT-PCR. Results In diabetic rats, blood sugar, weight and kidney/body weight ratio were decreased significantly and the excretion of 24 hour urinary protein was increased significantly compared to the diabetes group. But the excretion of 24 hour urinary protein was improved in GbE group ( P <0.05 ). The expression level of MMP-2 protein and mRNA was decreased significantly in renal glomduri of diabetic rats ( P < 0.05 ) but showed no changes in GbE treatment group compared to diabetes group( P > 0.05 ). The expression level of type Ⅳ collagen was significantly increased in diabetic rats( P <0.05 ) but showed no changes in GbE group( P >0.05). 目的 观察银杏叶提取物(GbE)对雄性SD糖尿病大鼠肾脏基质金属蛋白酶2(MMP-2)和Ⅳ型胶原表达的影响,从而探讨银杏叶对糖尿病肾病的保护机制.方法 24只链脲佐菌素诱导的SD大鼠完全随机分成糖尿病对照组(DM组)和GbE组,并以12只正常雄性SD大鼠作为正常对照组(NC组).分别于4、8周后,观察各组大鼠体重、肾重、随机血糖、尿蛋白排泄率等生化指标,并用免疫组化和逆转录聚合酶链反应方法 观察糖尿病大鼠肾脏MMP-2及Ⅳ型胶原的表达.结果 糖尿病大鼠的随机血糖、体重、肾脏/体重明显下降,尿白蛋白排泄率明显升高[DM组(31.59±4.59)mg/24 h比NC组(8.74±2.08)mg/24 h,P<0.05],GbE组糖尿病大鼠尿白蛋白排泄率[(21.38±3.86)mg/24 h]较DM组有所下降(P<0.05).8周时,与正常对照组相比,DM组大鼠肾小球MMP-2蛋白的表达水平明显下降[(0.49±0.07)mg/24 h比(0.15±0.04)mg/24 h,P<0.05],而GbE组DM组大鼠肾小球MMP-2蛋白的表达水平[(0.48±0.07)mg/24 h]较DM组无明显下降(P>0.05);DM组大鼠Ⅳ型胶原的表达水平较NC组明显升高[(3.48±0.54)mg/24h比(1.68±0.51)mg/24 h],而GbE组糖尿病大鼠较NC组无明显升高[(2.55±0.50)mg/24 h比(1.68±0.51)mg/24 h].结论 GbE可通过抑制糖尿病大鼠MMP-2过表达,进而对糖尿病肾病发挥保护作用.

目的 观察桑银降糖胶囊对实验性糖尿病大鼠脂质过氧化物酶水平的影响.方法 用微量血糖测定法测定链脲霉素(STZ)实验性糖尿病大鼠实验前后各组空腹血糖浓度.用放免法测定血清胰岛素和C肽.用黄嘌呤-鲁米诺化学发光法测定血液超氧化物歧化酶(SOD)的活性.用硫化巴比妥酸(TBA)比色法测定血清丙二醛(MDA)的含量.用HE染色方法 观察桑银降糖胶囊治疗前后胰腺、肝脏、肾脏的组织学变化.结果 桑银降糖胶囊可升高糖尿病动物模型SD大鼠胰岛素(36.08±11.40 μIU/ml)和C肽(0.10±0.04 μIU/ml)的浓度,具有明显降糖(6.33±1.66)mmol/L作用,对糖尿病动物模型SD大鼠具有提高血液SOD的活性(591.00±53.03 ug/g.HB),降低MDA水平的功效(6.3±0.85umol/L.对糖尿病动物模型SD大鼠的胰腺、肝脏、肾脏等组织有改善作用.结论 桑银降糖胶囊减少自由基对糖尿病动物模型SD大鼠的胰腺、肝脏、肾脏等组织的损伤并具有保护作用,纠正糖尿病动物模型SD大鼠的糖代谢紊乱,减少并发症的发生.  相似文献
