目的 研究Ras相关结构域家族1A基因(RASSF1A)启动子区域在妇科恶性肿瘤细胞中的甲基化状态及其与RASSF1A基因表达的关系.方法 采用RT PCR及甲基化特异性PCR检测9种妇科恶性肿瘤细胞系中RASSF1A基因mRNA表达水平及其启动子区域的DNA甲基化状态.结果 A2780、Anglne、CaOV3、SKOV3、HEC-1B和Ishikawa细胞系中RASSF1A基因启动子区域DNA呈现高甲基化,而相应的mRNA表达缺失或低表达,COC1、Hela和Siha细胞系中RASSF1A启动子区域DNA仅部分甲基化,可检测到这些细胞系RASSF1A基因的表达.结论 妇科恶性肿瘤细胞系普遍存在RASSF1A基因启动子区域DNA的高甲基化,这可能在妇科恶性肿瘤的发生发展中起作用.  相似文献

目的 探讨非小细胞肺癌中潜在抑癌基因DAL-1的表达与其启动子甲基化的关系.方法 10例非小细胞肺癌初诊患者,每例在治疗前抽取胸水并分离培养肿瘤细胞成系.分别采用逆转录-聚合酶链反应(RT-PCR)、western blot检测DAL-1的mRNA水平和蛋白水平的表达,用甲基化特异性PCR检测DAL-1基因启动子的甲基化.结果 10个细胞系中有9个完全不表达DAL-1mRNA和蛋白,仅有1个表达与人支气管上皮细胞株HBE水平相当.9个不表达DAL-1的细胞系有6个检出启动子CpG岛甲基化.结论 DAL-1在大多数非小细胞肺癌患者胸水分离细胞系中表达缺失.而DAL-1启动子甲基化是造成这种现象的主要原因.  相似文献

目的: 探讨乳腺癌细胞系中雌激素受体α(ERα)启动子甲基化状态的改变对其MTA1 mRNA和蛋白表达水平的影响,阐明ERα启动子去甲基化对MTA1基因表达的作用。方法: 用去甲基化药物5-Aza-dC处理ERα启动子高甲基化状态的MDA-MB-435s和低甲基化状态的MDA-MB-231乳腺癌细胞系为5-Aza-dC处理组,同时设 2种细胞系的未处理组为对照组。甲基化特异性PCR(MSP)法检测各组细胞ERα启动子甲基化状态。Western blotting法检测各组细胞MTA1蛋白表达水平,RT-PCR法检测各组细胞MTA1 mRNA表达水平。结果: 5-Aza-dC逆转了MDA-MB-435s和MDA-MB-231细胞系ERα启动子甲基化状态; MDA-MB-435s乳腺癌细胞系中,与对照组比较,5-Aza-dC处理组MTA1 mRNA及蛋白表达水平均下调(P<0.05);MDA-MB-231乳腺癌细胞系中,与对照组比较,5-Aza-dC处理组MTA1 mRNA及蛋白表达水平亦均下调(P<0.05)。结论: 改变MDA-MB-435s和MDA-MB-231乳腺癌细胞系ERα基因启动子甲基化状态后,MTA1 mRNA和蛋白表达均下降,提示ERα启动子去甲基化后可以下调MTA1基因的表达,二者之间存在调控关系。  相似文献

目的:检测大肠癌患者肿瘤组织及其相应的癌旁组织中Runx3基因表达和启动子区域甲基化状态,探讨甲基转移酶抑制剂5-aza-dc对结肠癌细胞系中Runx3表达的作用。方法:将手术切除的37例大肠癌组织和对应癌旁组织提取RNA和基因组DNA,采用RT-PCR检测Runx3基因的表达,甲基化特异PCR方法(MSP)检测Runx3基因启动子区域甲基化状态,用5-aza-dC处理结肠癌细胞系后,RT-PCR方法检测Runx3基因的表达。结果:37例大肠癌和对应癌旁组织中Runx3基因表达阳性率分别为100%(37/37),2.7%(1/37)。癌组织中Runx3基因表达明显降低(P<0.01)。MSP检测发现11例(30%)大肠癌标本及1例(3%)癌旁正常组织中Runx3基因呈甲基化状态。癌组织中Runx3基因甲基化明显高于癌旁组织(P<0.05)。结肠癌细胞系HT29 5-aza-dC处理后Runx3基因表达水平增加。结论:Runx3基因启动子甲基化和Runx3基因表达在大肠癌的癌旁组织和原发癌灶间存在显著性差异,可能与结肠癌的发生发展有关。  相似文献

目的观察促甲状腺激素受体(TSHR)在乳头状甲状腺癌(PTC)中的表达,分析其基因启动子区甲基化与肿瘤发生的关系。方法选择50例PTC和32例良性甲状腺肿瘤患者(对照组,包括20例结节性甲状腺肿和12例甲状腺腺瘤患者),对两组患者手术获取的标本提取RNA后,逆转录为cDNA,进行PCR,检测鸭HR基因的表达情况;运用甲基化特异性PCR(MSP)法检测上述组织中TSHR基因启动子区甲基化的情况。结果PTC组患者中,有34例(68.0%)TSHR基因启动子发生甲基化,16例(32.0%)TSHR基因mRNA表达缺失;对照组患者中,有7例(21.9%)TSHR基因启动予发生甲基化,4例(12.5%)TSHR基因mRNA表达缺失,PTC组织TSHR基因启动子区甲基化率及TSHR基因mRNA表达缺失率均显著高于对照组,差异有统计学意义(X^2值分别为16.61和4.02,P〈0.05)。34例发生了TSHR基因启动子区甲基化的PTC患者中,有14例(41.2%)TSHRmRNA表达缺失;16例未发生甲基化的PTC患者中,2例(12.5%)发生了mRNA表达缺失,二者mRNA表达缺失率间差异有统计学意义(X^2=4.11,P〈0.05)。结论PTC患者TSHR基因启动子甲基化发生率显著高于良性甲状腺肿瘤患者,而TSHR基因mRNA的表达则较良性甲状腺肿瘤降低;推测TSHR基因启动子区甲基化可能与PTC的发生、发展相关。  相似文献

目的:Eph基因和配体在多种肿瘤中呈高表达,并可能在肿瘤的发生、发展及预后中发挥重要作用。作者探讨EphA7基因在大肠癌细胞系和结直肠癌组织及其正常黏膜中的表达情况,以及EphA7基因在大肠癌发生、发展中的作用。方法:定量RT—PCR检测大肠癌细胞系和原发性结直肠癌标本中EphA7的表达。经甲基化特异性PCR(MSP)和亚硫酸氢钠处理后,用DNA测序等方法分析EphA7基因启动子区CpG岛甲基化状况。构建pEGFPN3-EphA7质粒,并转染至EphA7表达丢失的大肠癌细胞系中,对转染子进行mRNA基因表达芯片分析。结果:大肠癌细胞系DLD1、HT29、HCT116、SW480和SW620中Epha7基因低表达。59例进行定量RT-PCR测定的原发性结直肠癌患者中,29例患者癌组织中EphA7基因的表达水平低于来源相同患者的正常黏膜(P=0.008)。经MSP、亚硫酸氢钠处理后DNA测序等发现,EphA7基因启动子区CpG岛高甲基化。EphA7的高甲基化与肿瘤分化、性别以及肿瘤发生部位等有关。对EphA7质粒转染子mRNA基因芯片分析,观察到多种基因表达改变。结论:EphA7基因启动子区CpG岛甲基化是EphA7在大肠癌中低表达的机制。Epha7基因可能在结直肠癌的肿瘤发生、发展中起重要作用。  相似文献

目的:检测胶质瘤组织和细胞系中 Nanog 基因启动子区甲基化状态与 Nanog 基因的表达,并探讨其在胶质瘤发展中的作用。方法采用甲基化特异性 PCR(MSP)法分别检测正常脑组织、胶质瘤组织、胶质瘤癌旁组织、胶质瘤细胞系 U87和 U251中 Nanog 基因启动子区甲基化状态,实时定量 PCR(RT-PCR)法检测相应组织和细胞系中 Nanog 表达情况。结果 MSP 法检测胶质瘤组织 Nanog 基因启动子区呈非甲基化状态,且非甲基化检出率(58.82%)高于癌旁组织(13.63%),差异有统计学意义(χ2=14.852,P <0.01)。对MSP 法检测结果行灰度值分析,高级别胶质瘤中 Nanog 基因启动子区非甲基化程度高于低级别组。 U87和 U251细胞系中 Nanog 基因启动子区呈非甲基化状态。 RT-PCR 结果显示高级别胶质瘤中 Nanog mRNA 相对表达量高于低级别组, U87和 U251细胞系中 Nanog mRNA 的转录明显增多。结论胶质瘤中 Nanog 基因启动子区呈非甲基化状态,且可能促进 Nanog 基因的转录,影响胶质瘤的发生与发展。  相似文献

目的检测白血病患者标本及细胞系中分泌型卷曲相关蛋白5(SFRP5)基因启动子区域的甲基化状态及蛋白表达情况。方法甲基化特异性PCR(MSP)检测白血病患者及细胞系中SFRP5基因启动子区甲基化状态,亚硫酸盐测序PCR(BSP)方法检测KG1a细胞中SFRP5基因甲基化状态,Western blot方法检测SFRP5蛋白在白血病患者及细胞系中的表达。结果 12例白血病患者标本中有7例发生了SFRP5基因甲基化,6例正常对照标本均未发生SFRP5基因甲基化。4种白血病细胞系(HL-60、Raji、U937、KG1a)中均发生SFRP5基因甲基化。KG1a细胞中SFRP5基因启动子区-394 bp到+99 bp位点的CpG总甲基化频率为82.5%。白血病患者标本中有7例存在SFRP5蛋白表达,6例正常标本存在SFRP5蛋白表达,4种白血病细胞系中未检测到SFRP5蛋白表达。使用去甲基化试剂DAC处理4种白血病细胞系,结果均恢复SFRP5蛋白表达。结论白血病细胞中存在SFRP5基因启动子区甲基化,并导致SFRP5蛋白表达缺失或下调。  相似文献

目的:探讨PAX5基因启动子甲基化作为前列腺癌生物标志物的临床价值。 方法:采用Real time RT-PCR检测前列腺细胞系(RWPE-1、LNCaP、PC-3、DU145)中PAX5 mRNA的表达,甲基化特异性PCR(MSP)法分析4株前列腺细胞的甲基化状态;去甲基化药物处理后,Real time RT-PCR法检测肿瘤细胞PAX5基因mRNA表达; Real time MSP法检测64例前列腺癌患者、22例前列腺增生患者与47例健康体检者血清中PAX5基因启动子甲基化水平,并统计分析血清PAX5基因启动子甲基化水平与临床病理参数的相关性;运用ROC曲线分析PAX5基因启动子甲基化在前列腺癌诊断中的价值。结果:Real time RT-PCR结果显示,与正常前列腺上皮细胞RWPE-1相比,PAX5 mRNA在3株前列腺癌细胞中均表达下调;LNCaP、PC-3、DU145前列腺癌细胞均可检测到PAX5基因启动子甲基化,而正常前列腺上皮细胞RWPE-1不存在PAX5基因启动子甲基化;去甲基化药物可恢复PAX5基因mRNA表达;Real time MSP结果显示,前列腺癌患者的血清PAX5基因启动子甲基化水平显著高于前列腺增生患者和健康体检者;前列腺癌患者的血清PAX5基因启动子甲基化率与Gleason评分和TNM分期有关;ROC结果显示PAX5基因的诊断价值优于PSA。结论:本研究结果提示启动子甲基化是前列腺癌细胞中PAX5基因表达下调的主要原因,血清PAX5启动子甲基化是一种潜在的前列腺癌诊断标志物。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Objectives To explore serum cytokines levels (including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v) and their significance in patients with acute coronary syndrome (ACS) and the subsequent follow-ups, with attempt to estimate the role of various serum inflammatory markers in the diagnosis and assessment of ACS.Methods The study population include 40 patients with acute myocardial infarction (AMI), 40 patients with unstable angina pectoris (UAP), and 40 controls. Among the 80 patients, 60 patients attended a follow up 4 months later. Serum inflammatory markers including IL-1 β, sIL-2R, IL-6, TNF-α, and IFN-v were measured by enzyme linked immunosorbent assay.Results Serum IL- 1 β, sIL-2R, IL-6, TNF-α were significantly higher in AMI group or UAP group compared to the control group and became significantly lower 4 months later in the follow-up patients. Serum levels of IFN-v shows no significant difference between AMI group or UAP group and controls, also showing no significant change when measured in follow up patients. There was no correlation between serum creatine kinase-MB isoenzyme levels and serum inflammatory markers either in UAP or AMI group. Furthermore, when divided into two subgroups using Wagner's QRS scoring system in the AMI group, there is no difference of each serum inflammatory marker between ≤ 6 scores group and > 6 scores group.Conclusion Serum levels of certain inflammatory markers may have some diagnostic value for ACS, and can be a useful marker reflecting disease stability.  相似文献

Objective:To explore the epidemiology and etiology for an outbreak of acute respiratory tract infection that occurred in one county of Jiangsu Province, China 2004. Methods: Only cases meeting the case definition were included in the study. We reviewed the medical records of the cases who were admitted to the local hospitals, interviewed cases by a standard questionnaire, and then described the epidemiotogic features and analyzed risk factors by means of a case-control study. We collected pharyngeal swab specimens and sent them to different laboratories for isolation and culture. The laboratory used different detection methods such as DIP, PCR, electron microscope examination and microneutralization assay, to identify and then type the positive specimens. Results:A total of 871 cases were reported during the period from April 18 to July 4,2004. The distribution of onset times presented two peaks, one in late May and another in middle June. The epidemic occurred mainly in the elementary and junior high schools in ten townships of one county, and the mean age of the cases was 12 years (range 7 months to 18 years). The course of the disease was acute, and was characterized by fever accompanied with sore throat and tonsillitis. The WBC count of cases was normal or elevated. The mean duration of illness was 5 days (range 2 to 12 days). No fatalities from illness were reported. A case-control study indicated that the possible risk factors were close contact with a case and/or poultry before onset and sharing of towels among members of the family. The typical CPE was observed through inoculating pharyngeal swab specimens into the HEP-2 cell cultures in different laboratories. An infection of adenovirus type 3 was verified by detecting positive specimens in different methods. Conclusion:This investigation demonstrated that the acute respiratory infection in cases was caused by adenovirus type 3. Cases occurred in over 70 schools in ten townships in 2004, and the route of transmission was possibly close contact with cases or droplet transmission.  相似文献
