We studied the in vitro antirhinovirus activity of a soluble form of intercellular adhesion molecule-1 (sICAM-1). sICAM-1 inhibited the cytopathic effect of 10 representative human rhinovirus (HRV) serotypes of the major receptor group with, 50% effective concentrations (EC50s) of 0.1 to 7.9 micrograms/ml. Cell type-dependent variation in the inhibitory activity of sICAM-1 was observed for two major receptor group serotypes in HeLa cells (EC50, greater than 32 micrograms/ml), and no inhibitory effect was observed for two serotypes which use different cell receptors. Yield reduction assays showed that sICAM-1 inhibited the replication of HRV serotype 39 (HRV-39) in human adenoid explants in a concentration-dependent manner. No direct inactivation of infectivity of HRV-39 (EC50, 0.5 microgram/ml) was observed after incubation with sICAM-1 (32 micrograms/ml) for up to 24 h. Single-cycle-of-replication experiments with the addition of sICAM-1 at 10 micrograms/ml at different times showed that the inhibitory effect occurs only when sICAM-1 is added within 30 min after infection. In experiments in which absorption was carried out at 4 degrees C and then a single cycle of replication incubation was carried out at 33 degrees C, it was found that sICAM-1 at 10 micrograms/ml was inhibitory only when it was present during the absorption period. Our data show that sICAM-1 is inhibitory for representative major receptor group serotypes of HRV in two cell lines and human respiratory epithelium, that the interaction of sICAM-1 with HRV is readily reversible by dilution, and that the inhibitory effect of sICAM-1 on virus replication is present early in the infection cycle.  相似文献

The antiviral potency of soluble intercellular adhesion molecule-1 (ICAM-1; the major receptor for human rhinoviruses) was determined for a subset of American Type Culture Collection reference serotypes and field isolates of rhinovirus. The results indicate that soluble ICAM-1 exhibits a broad spectrum of activity against rhinoviruses and that field isolates have a sensitivity indistinguishable from that of laboratory strains.  相似文献

目的进一步探讨可溶性细胞间黏附分子-1(sICAM-1)在类风湿关节炎(RA)中的作用。方法采用放射免疫测定法对34例RA患者及30例健康体检者血清、关节液中sICAM-1进行检测。结果所测静脉血清sICAM-1浓度,RA组显著高于健康对照组,差异有统计学意义(P〈0.001);RA未治疗组显著高于治疗后未复发组,差异有统计学意义(P〈0.001),但与治疗后复发组比较,差异无统计学意义(P〉0.05);RA患者关节液中sICAM-1明显高于血清中的水平,二者差异有统计学意义(P〈0.05),而未治疗组与治疗后复发组关节液中sICAM-1水平差异无统计学意义(P〉0.05)。结论sICAM-1在RA发病中起着重要作用,可作为判断病情严重性的指标用于监测RA的活动及疗效。  相似文献

The aim of our study was to investigate the influence of single low-density lipoprotein apheresis (heparin extracorporeal low-density lipoprotein precipitation [HELP]procedure) on plasma concentrations of soluble adhesion molecules (sAMs) such as soluble vascular cellular adhesion molecule-1 (sVCAM-1), soluble intercellular adhesion molecule-1 (sICAM-1), and P-selectin in patients with familial heterozygous hypercholesterolemia and documented coronary artery disease enrolled in a chronic weekly HELP apheresis. Before HELP apheresis, the mean plasma concentration of sVCAM-1 was 515 +/- 119 ng/ml, 204 +/- 58 ng/ml for sICAM-1, and 112 +/- 45 ng/ml for P-selectin. After single HELP apheresis, plasma concentrations of sAM declined significantly by 32 +/- 7%, 18 +/- 15%, and 33 +/- 25% for sVCAM- 1,sICAM-1 and P-selectin, respectively. After a 1 week interval, sAM concentrations rose to approximately the initial values. The concentrations of all sAMs studied were significantly lower in the plasma leaving than entering the filter. Due to filtration, the decline in plasma level of sVCAM-1, sICAM-1, and P-selectin was 62 +/- 19%, 51 +/- 39%, and 67 +/- 22%, respectively. In addition to lipid reduction, single HELP apheresis significantly lowers plasma concentrations of sVCAM-1, sICAM-1, and P-selectin.  相似文献

背景:应用生物素一链亲和素系统建立的可溶性细胞间黏附分子1测定法,是一种敏感度高、可测范围广的检测方法.目的:建立可溶性细胞间黏附分子1生物素链亲和素系统时间分辨荧光免疫分析方法并应用于临床,探讨肺移植前后血清可溶性细胞间黏附分子1的变化及临床意义.方法:使用2株匹配的单克隆抗体分别作为捕获抗体和检测抗体,利用制备的铕标记链亲和素(SA-Eu")作为示踪物并与生物素化的检测抗体特异结合,建立双位点多层夹心法,通过对30例健康人血清可溶性细胞间黏附分子1检测和26例肺移植受者手术前后血清可溶性细胞间黏附分子1的变化测定,对可溶性细胞间黏附分子1生物素链亲和素系统方法学和在肺移植术前后的临床应用价值进行评价.结果与结论:30例健康对照测定结果为(348.63±69.12)μg/L.移植前可溶性细胞间黏附分子1与对照组无显著性差别;移植后,各组与对照组之间比较差异有显著性意义(P<0.05),急性排斥反应时血清可溶性细胞间黏附分子1升高,并发感染时降低,但各组之间比较无明显差别.表明可溶性细胞间黏附分子1生物素一链亲和素系统是一种新型的高灵敏度、宽范围的非放射性标记免疫分析法,对肺移植受者移植前后监测血清可溶性细胞间黏附分子1可作为辅助诊断急性排斥反应的免疫学指标.  相似文献

目的 探讨血清可溶性血管细胞黏附分子-1(sVCAM-1)和可溶性细胞间黏附分子-1(siCAM-1)在2型糖尿病大、小血管病变中的作用。方法 应用酶联免疫吸附试验(ELISA)法检测了62例2型糖尿病患者血浆sVCAM-1和siCAM-1水平,并与20例健康人作对照。结果 糖尿病各组血清sVCAM-1和siCAM-1水平明显高于健康对照组(P〈0.01),无血管病变组、微血管病变组和大血管病变组的含量逐步升高(P〈0.01);逐步多元回归分析表明sICAM-1水平与血浆假性血友病因子(vWF)、甘油三酯(TG)、收缩压(SBP)、舒张压(DBP)呈正相关(r=0.43、0.45、0.52、0.62,均P〈0.01);sVCAM-1水平与TG、胆固醇(TC)及尿白蛋白/肌酐(Alb/Cr)呈正相关(r=0.59、0.46、0.73,均P〈0.01);多因素logistic回归分析表明sVCAM-1与是否惠有微血管病变显著相关(β=2.48,P〈0.05),sICAM-1与是否惠有大血管病变显著相关(β=2.46,P〈0.05)。结论 sICAM-1和sVCAM-1参与了2型糖尿病血管病变的发生和发展,可作为早期2型糖尿病患者慢性血管并发症发生的预测及监测指标。  相似文献

目的探讨可溶性细胞间黏附分子-1(sICAM-1)含量与孕妇妊娠结局的相关性,及其在先兆早产及晚期先兆流产诊治中的意义。方法 2011年4月至2012年3月在中山大学附属第一医院产科规律产检并分娩单胎妊娠共73例,分为正常对照组32例,晚期先兆流产及先兆早产41例,其中足月分娩组24例,早产组17例。采用酶联免疫吸附法测定血清sICAM-1,比较组间血清sICAM-1含量差异及其与孕妇早产的关系。结果 (1)早产组sICAM-1浓度高于正常对照组及足月分娩组,早产组宫颈长度小于足月分娩组。(2)sICAM-1浓度、宫颈长度分别达到388.65 ng/ml、21.5 mm时,预测先兆早产及晚期先兆流产孕妇早产的敏感性、特异性、阳性预测值、阴性预测值分别为52.9%、95.7%、90.0%、74.2%,58.8%、91.7%、83.3%、75.9%。(3)以sICAM-1=388.65 ng/ml及宫颈长度21.5 mm为临界点,联合预测晚期先兆流产及先兆早产孕妇早产的敏感性、特异性、阳性预测值、阴性预测值分别为35.3%、87.5%、100%及84%。结论 (1)sICAM-1达到388.65 ng/ml时可预测先兆早产及晚期先兆流产孕妇早产的发生。(2)以sICAM-1=388.65 ng/ml及宫颈长度21.5 mm为临界点,两者联合预测晚期先兆流产及先兆早产孕妇早产可提高阳性预测值及阴性预测值。  相似文献

目的评估所建立的可溶性细胞间黏附分子-1生物素-亲和素系统(BAS)时间分辨荧光免疫分析方法(sICAM-1 BSA-TRFIA)的技术指标及应用价值。方法建立sICAM-1 BSA-TR-FIA,分析其检测曲线的性质参数,研究该方法的灵敏度、精密度和回收率、特异性、稳定性、平行性,以及与酶联免疫吸附试验分析法(ELISA)间的相关性。结果此法特异性较高,同批试剂连续6个月应用分析结果稳定,灵敏度为1.32μg/L,批内和批间CV分别为4.24%和6.83%,平均回收率为99.87%,线性范围为(2.5~1933)μg/L,与ELISA所测结果比较高度相关(r=0.939),与TNF-α、IL-8、IL-1、IL-2和IL-6等免疫分子无交叉反应。结论sICAM-1 BSA-TRFIA是一种特异性强、稳定性好、灵敏度高、可测范围较广的检测方法,可以方便地应用于临床,为相关疾病的诊断和监测提供一项良好的实验诊断指标。  相似文献

Soluble intercellular adhesion molecule-1 (sICAM-1) level was measured in sera from 41 patients with Schistosoma mansoni schistosomiasis and compared with the sICAM-1 level in 41 healthy subjects. A significant increase in serum sICAM-1 was observed in patients with schistosomiasis compared with control subjects. As they were inhabitants of the French Antilles, the patients were, however, not settled in a malaria endemic zone, allowing this cause of sICAM-1 enhancement to be eliminated. No correlation was found between the level of sICAM-1 and the schistosomiasis serological titre. Such results favour the hypothesis of an activation of vascular endothelial cells due to egg deposition.  相似文献

目的:观察经中药复方制剂处理后损伤的人血管内皮细胞表达细胞间黏附分子水平的变化,分析其抗动脉粥样硬化的可能途径。方法:实验于2004-01/2005-12在龙华医院科研实验中心完成。①选择SD大鼠70只,按随机数字表法分为7组,即正常对照组、模型组、西药组、中药复方0.4g/mL,0.8g/mL,1.6g/mL及3.2g/mL组,每组10只。以上各组大鼠分别灌胃生理盐水、生理盐水、1g/L氟伐他汀、0.4g/mL,0.8g/mL,1.6g/mL及3.2g/mL中药复方(以黄芪为君,栝楼、薤白为臣)生药,2mL/(次·只),分别腹主动脉采血,分离血清。②人血管内皮细胞HMEC-1与大鼠分组情况相同。除正常对照组外,其余各组HMEC-1细胞均通过高脂血清处理细胞建立细胞损伤模型。各组细胞分别应用相应组别的大鼠血清处理72h,应用酶联免疫吸附法测定培养上清液中可溶性细胞间黏附分子1表达水平,反转录-聚合酶链反应检测各组HMEC-1细胞可溶性细胞间黏附分子1mRNA表达水平。结果:各组HMEC-1细胞可溶性细胞间黏附分子1及其mRNA表达水平比较:中药复方0.4g/mL,0.8g/mL,1.6g/mL及3.2g/mL组细胞培养上清可溶性细胞间黏附分子1水平显著低于模型组[(4.33±0.19,4.44±0.34,3.46±0.17,4.33±0.27,6.27±0.25)μg/L(P<0.01)],其中中药复方1.6g/mL组作用最为明显,而且显著低于西药组(4.55±0.20)μg/L(P<0.01);以上各剂量中药同时能明显下调损伤的血管内皮细胞可溶性细胞间黏附分子1mRNA表达水平,其中中药复方3.2g/mL组作用最为明显,而且显著低于西药组[0.63±0.12,1.03±0.12(P<0.01)]。结论:以黄芪为君,栝楼、薤白为臣的中药复方制剂可明显下调损伤的血管内皮细胞培养上清液中可溶性细胞间黏附分子1及其mRNA表达水平,从而减少细胞间黏附及血管内皮损伤,其抗动脉粥样硬化的作用可能通过这一途径实现。  相似文献

目的探讨检测病毒性脑炎(VE)患儿血清中可溶性细胞间黏附分子-1(sICAM-1)和可溶性P-选择素的作用。方法应用酶联免疫吸附双抗体夹心法分别检测26例VE患儿急性期、恢复期及20例正常儿童血清中sICAM-1[(372.6±113.6)μg/L]和可溶性P-选择素[(38.2±12.1)μg/L]的水平。结果 VE患儿急性期血清中的sICAM-1和可溶性P-选择素显著高于对照组[(277.4±87.8)μg/L和(19.6±4.2)μg/L],差异有统计学意义(P<0.01),恢复期患儿血清中sICAM-1[(305.2±99.1)μg/L]和可溶性P-选择素[(23.6±8.4)μg/L]与对照组比较差异无统计学意义(P>0.05)。结论 VE患儿sICAM-1和可溶性P-选择素水平与病程密切相关,检测血清sI-CAM-1和可溶性P-选择素有助于判断VE患儿病情轻重和评估预后。  相似文献

The adhesiveness of isolated canine cardiac myocytes for neutrophils is greatly increased by stimulation with cytokines such as tumor necrosis factor alpha (TNF alpha). Since this adhesion is significantly inhibited by an anti-CD18 MAb, experiments were performed to test the hypothesis that the newly expressed adhesion molecule on the cardiac myocytes was intercellular adhesion molecule-1 (ICAM-1). A newly developed MAb, CL18/6, was found to exhibit the functional and binding characteristics with canine neutrophils and canine jugular vein endothelial cells expected of an antibody recognizing ICAM-1. MAb CL18/6 also bound to isolated cardiac myocytes after stimulation of the myocytes with cytokines, and it blocked by greater than 90% the adhesion of neutrophils to stimulated myocytes. A partial cDNA clone for canine ICAM-1 was isolated, and ICAM-1 mRNA was found to be increased in both endothelial cells and cardiac myocytes after cytokine stimulation. Cytokines that both increased the CL18/6-inhibitable adhesion of neutrophils to myocytes and induced expression of ICAM-1 were IL-1 beta, TNF alpha, and LPS. These results are consistent with the conclusion that canine endothelial cells and cardiac myocytes express ICAM-1 in response to cytokine stimulation, and that ICAM-1 functions as an adhesive molecule for neutrophils on both cell types.  相似文献

目的观察西洛他唑对人脐静脉内皮细胞(HUVECs)血管细胞黏附分子1(VCAM-1)和细胞间黏附分子1(ICAM-1)mRNA表达的影响,探讨西洛他唑可能的抗动脉粥样硬化作用机制。方法将HUVECs用不同浓度的西洛他唑(0μg/L、0.05μg/L、0.1μg/L、1.0μg/L、10μg/L)溶液处理1小时后,用肿瘤坏死因子α(TNF-α)10μg/L诱导24小时。半定量复合逆转录聚合酶链反应(RT-PCR)测定黏附分子VCAM-1和ICAM-1mRNA的表达。结果 TNF-α能上调VCAM-1和ICAM-1的表达,西洛他唑在一定程度上可抑制上述作用,随着西洛他唑浓度的增加,ICAM-1mRNA表达水平逐步下降,分别为0.239±0.012、0.205±0.012、0.166±0.010、0.136±0.008,VCAM-1mRNA表达水平也逐步下降,分别为0.114±0.048、0.093±0.051、0.083±0.045、0.068±0.039。结论西洛他唑可抑制TNF-α诱导的HUVECs的黏附分子VCAM-1和ICAM-1mRNA表达,提示西洛他唑的抗动脉粥样硬化作用可能是通过阻止血单核细胞向血管内皮细胞聚集和黏附实现的。  相似文献

目的 研究可溶性细胞间黏附因子-1(sICAM-1)与P-选择素(P-selectin)在肝细胞癌(HCC)患者血清中的表达及与肝癌复发、转移的关系。方法 采用ABC—ELISA法对78例肝癌患者治疗前和治疗后第30天血清中sICAM-1和P-selectin的水平进行检测,同时测定甲胎蛋白(AFP)浓度,并用SPSS10.0统计软件及受试者操作特征(ROC)曲线进行分析,评价其临床应用价值。结果治疗前后血清中sICAM-1和P-selectin含量均高于健康对照组,并随临床TNM分期增加而升高,治疗有效组外周血中sICAM-1和P—selectin显著下降,而无效组则升高,治疗后二者的浓度转移和复发组均高于未复发组和转移组(P分别〈0.05及〈0.01)。sICAM-1和P-selectin与AFP的表达相关性表明,HCC患者血清sICAM-1、P—selectin的水平与AFP含量均呈正相关(分别为r=0.874,P〈0.05;r=0.892,P〈0.01)。对siCAM-1、P—selectin和AFP的ROC曲线分析提示,siCAM-1的诊断效能最高,AFP次之,P—selectin较差。结论sICAM-1和P—selectin与HCC生长关系密切,与HCC的侵袭、转移有关,血清sICAM-1和P—selectin有可能成为HCC辅助诊断、病情发展及疗效观察方面有价值的指标,可作为反映肝痛复发、转移潜能的生物学指标。  相似文献

Summary Previous reports have suggested soluble intercellular adhesion molecule-1 as a marker of disease activity in Hodgkin's disease. In the present study we investigated serum levels of intercellular adhesion molecule-1 at diagnosis in 104 patients with Hodgkin's disease and in 77 of these patients following the achievement of complete remission (within 12 months of diagnosis). Mean serum levels at diagnosis were significantly higher in patients than in controls (P<0.0001) and were related to advanced stages of disease (P=<0.0001) presence of “B” symptoms (P<0.0001), abnormality of laboratory indexes (P<0.0001), erythrocyte, sedimentation rate values (r=0.41,P<0.0001) and serum levels of soluble interleukin-2 receptor α chain (r=0.51,P<0.0001). Mean values in complete remission were significantly lower than at diagnosis (P=0.003). Lower mean values at diagnosis were detected in 30 patients with advanced disease who attained complete remission, compared with 6 patients who failed to attain complete remission with standard treatment. We conclude that in Hodgkin's disease, high serum levels of soluble intercellular adhesion molecule-1 are detectable at presentation and strictly correlate with some clinical features. Response to treatment is paralleled by reduced serum levels. Larger prospective studies are needed to evaluate the possible prognostic significance of serum levels of soluble intercellular adhesion molecule-1 at diagnosis.  相似文献

目的:检测鼻内应用肾上腺皮质激素(激素)治疗鼻息肉前后鼻息肉组织中的细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)和鼻分泌物中的可溶性细胞间黏附分子-1(soluble intercellular adhesion molecule-1,sICAM-1)表达水平的变化,探讨ICAM-1、sICAM-1两者间关系及其在鼻息肉发展过程中的作用及鼻内应用激素治疗鼻息肉的可能机制.方法:选择19例未用药的鼻息肉患者(未用药组)、14例经鼻内应用激素(氟替卡松)治疗后的鼻息肉患者(激素治疗组)、12名正常人(正常对照组),取其鼻息肉或鼻黏膜组织及鼻分泌物,用ELISA法检测其鼻息肉或鼻黏膜组织匀浆中ICAM-1和鼻分泌物中sICAM-1的含量并作比较.结果:未用药组鼻息肉组织匀浆中ICAM-1的含量及鼻分泌物中sICAM-1的含量,较激素治疗组及正常对照组为高(均为P<0.01),而激素治疗组与正常对照组鼻黏膜组织匀浆中ICAM-1的含量和鼻分泌物中sICAM-1的含量比较差异无统计学意义(P>0.05).未用药组鼻息肉组织匀浆中ICAM-1及鼻分泌物sICAM-1含量呈线性相关(r=0.843,P=0.000).结论:鼻内应用激素的治疗效果可能与其抑制ICAM-1的表达有关.在未受药物干扰的情况下,鼻分泌物中 sICAM-1的含量也能很好的反映鼻息肉组织中ICAM-1的表达情况.  相似文献
