Background  

The understanding of the molecular bases of blood groups makes possible the identification of red cell antigens and antibodies using molecular approaches, especially when haemagglutination is of limited value. The practical application of DNA typing requires the analysis of the polymorphism and allele distribution of the blood group genes under study since genetic variability was observed among different ethnic groups. Urban populations of Argentina are assumed to have a white Caucasian European genetic component. However, historical and biological data account for the influence of other ethnic groups. In this work we analyse FY and RH blood group alleles attributed to Africans and that could have clinical implications in the immune destruction of erythrocytes.  相似文献

Varroa destructor is an external parasitic mite that is a serious pest of honeybees and has caused severe losses of colonies worldwide. One of the feasible alternative treatments being used for their control is essential oils. The aim of this work was to evaluate the bioactivity of some essential oils on V. destructor and Apis mellifera in relation with their chemical composition and physicochemical properties. Lavender, lavendin and laurel essential oils showed linalool as main compound in their composition. 1,8-Cineole was also present as a predominant component in the laurel essential oil. However, thyme oil was characterized by a high concentration of thymol. Mites and bees toxicity was tested by means of complete exposure method. For mites, LC50 values for laurel, lavender and lavendin essential oil did not show significant variation throughout all observation times. However, the LC50 values for thyme oil at 48 and 72 h were lower than at 24 h. Bee mortality was evident only in treatment with thyme oil. At 48 and 72 h, lavender essential oil presented better selectivity indexes. In this research, all essential oils caused mite mortality without severe harmful effects on adult bees. The simultaneous evaluation of the physicochemical analysis of the essential oils, the characterization of the dosage response relationships among them, and the mortality effects on mite and bees, give us the possibility to obtain comparative results for future research in Varroa control.  相似文献

Varroa destructor is an ectoparasitic mite that affects colonies of honey bee Apis mellifera worldwide. In the last years, substances of botanical origin have emerged as natural alternative acaricides to diminish the population levels of the mite. In the present work, the bioactivity of propolis from different geographical locations of Pampean region from Argentina on V. destructor was evaluated. Fourteen propolis samples were organoleptic and physicochemically characterized and, by means topical applications, their activity was tested on mites. All propolis had a homogeneous composition and the bioactivity levels against mites were comparable among the different propolis samples. The percentage of mites killed by the treatments ranged between 60.5% and 90% after 30 s of exposure. Thus, V. destructor was highly susceptible to propolis. Moreover, the mites remained anesthetized during the first hours after topical treatment. The results suggest that propolis from Argentinean pampas could be incorporated in honey bee colonies as acaricidal treatment by spraying.  相似文献

Petasiger argentinensis sp. nov. is described from material found in the intestine of the Great Grebe, Podiceps major (Boddaert) and the White-tufted Grebe, Rollandia rolland Quoy et Gaimard (Podicipediformes, Podicipedidae), from Argentina. The new species is characterized by the head collar armed with 19 spines and by the testes arranged in tandem. These characters are shared with only two Neotropical species of the genus, P. novemdecim Lutz, 1928 and P. combesi Zamparo, Overstreet et Brooks, 2005. Petasiger novemdecim differs from the new species in having a larger body, longer collar spines, larger eggs, a cirrus-sac of similar size to the ventral sucker and almost entirely anterior to it, and vitelline fields confluent ventrally posterior to the ventral sucker. P. combesi differs from the new species in most metrical characters, the shape of the body (antero-posteriorly elongate) and the location of the genital pore (dextral) and the excretory pore (dorsal). This is the first record of a member of the genus Petasiger parasitizing birds from Argentina.  相似文献

Using molecular techniques, we documented the presence of Nosema ceranae in honeybees (Apis mellífera) from Argentina. Samples were collected from A. mellifera colonies in 38 districts of Buenos Aires province, Argentina. Molecular characterization was achieved with a multiplex PCR-based method, which allows parallel diagnosis of N. ceranae and N. osema apis. N. ceranae was identified in all the samples analyzed. Moreover, coinfections with N. apis were detected in Balcarce and Maipú districts. We identified three rRNA sequence variants of N. ceranae, which may represent diverse sources of bee importation. The results suggest that N. ceranae is widely distributed in Argentina and that the genetic variation observed between the different isolates could be related with the difference in the symptomatology found previously by our work group. Our results highlight the need to re-assess the health protocols currently in force so that they recognize N. ceranae as the main causal agent of Nosemosis in this country.  相似文献

The Varroa destructor mite has recently displayed an ever increasing resistance to new drugs, contributing to CCD proliferation. This work was aimed at determining new viable methods for identifying the pyrethroid resistance of V. destructor and DNA methylation in resistant and sensitive mites. DNA was extracted from Varroa mites. Nucleotide changes in the DNA of pyrethroid-resistant, pyrethroid-sensitive, and control mites were identified with polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) in the case of five mitochondrial gene fragments. More bands were observed in the drug-resistant mites than in the other two groups. Sequencing confirmed these observations. Decreased global DNA methylation levels were observed in the pyrethroid-resistant mites. There exists a previously undescribed mechanism of pyrethroid resistance development in Varroa mites. The PCR-SSCP methods can be considered and further developed as useful tools for detecting V. destructor resistance.  相似文献

A wild-caught adult male southern brown bandicoot (Isoodon obesulus) was presented for investigation of a pruritic skin condition that consisted of crusting, deep fissures, lichenification, alopecia, scale, and erythema, and affected the caudal dorsum, caudal, and medial thighs, distal hind limbs, tail, forepaws, and parts of the thorax. Examination of superficial and deep skin scrapings revealed large numbers of mites, which were identified as Sarcoptes scabiei. To the authors’ knowledge, infestation with this mite has not previously been reported in bandicoots.  相似文献

Causes of chromosomal differences such as mosaicism between embryos developed in vivo and in vitro may be resolved using animal models to compare embryos generated in vivo with those generated by different production systems. The aims of this study were: (1) to test a ZOO-FISH approach (using bovine painting probes) to detect abnormal chromosome make-up in the sheep embryo model, and (2) to examine the extent of chromosome deviation in sheep embryos derived in vivo and in vitro. Cytogenetic analysis was performed on day 6 in-vivo and in-vitro derived sheep embryos using commercially available bovine chromosome painting probes for sex chromosomes X–Y and autosomes 1–29. A total of 8631 interphase and metaphase nuclei were analyzed from 49 in-vitro-derived and 51 in-vivo-derived embryos. The extent of deviation from normal ovine chromosome make-up was higher (p < 0.05) in in-vitro-produced embryos relative to in-vivo-derived embryos (65.3% vs. 19.6% respectively) mainly due to diploid–polyploid mosaicism. Polyploid cells ranged from 3n to 8n with tetraploids most predominant among non-diploid cells. The proportions of polyploid cells per mixoploid embryo in in-vitro-produced embryos ranged from 1.4% to 30.3%, in contrast to less than 10% among the in-vivo-derived embryos. It was concluded that in-vitro-derived embryos are vulnerable to ploidy change compared to their in-vivo counterparts. The application of ZOO-FISH to domestic animal embryos is an effective approach to study the chromosome complement of species for which DNA probes are unavailable.  相似文献

This report describes the development of in-house real-time PCR assays using minor groove binding probes for simultaneous detection of the Bacillus anthracis pag and cap genes, the Francisella tularensis 23 KDa gene, as well as the Yersinia pestis pla gene. The sensitivities of these assays were at least 1 fg, except for the assay targeting the Bacillus anthracis cap gene, which showed a sensitivity of 10 fg when total DNA was used as a template in a serial dilution. The clinical value of the Bacillus anthracis- and Francisella tularensis-specific assays was demonstrated by successful amplification of DNA from cases of cow anthrax and hare tularemia, respectively. No cross-reactivity between these species-specific assays or with 39 other bacterial species was noted. These assays may provide a rapid tool for the simultaneous detection and identification of the three category A bacterial species listed as biological threats by the Centers for Disease Control and Prevention.  相似文献

Molecular characterization of methicillin-resistant Staphylococcus aureus isolates from three hospitals in Israel was the aim of the study presented here. We identified 11 distinct genetic clones by pulsed-field gel electrophoresis. Molecular typing identified four different SCCmec types—I, II, IV, and V−and nine spa types. In Argentina, Varroa destructor resistance to coumaphos has been previously reported. However, the status of mite susceptibility to other hard acaricides is still unknown. At present, high infestation levels of V. destructor are being detected in colonies of Apis mellifera after treatment with amitraz. The aim of the present study was to determine the LC50 of amitraz in V. destructor from three apiaries with high mite density after treatment with the acaricide. The LC50 values were 3.9, 3.5, and 3.7 μg/Petri dish for mites from three different apiaries. Significant LC50 differences were detected between resistant and susceptible mites. LC50 increased 35–39-fold when compared to the corresponding baseline, suggesting the development of resistance. These results are the first report of resistance to amitraz in V. destructor in Argentina and extend the knowledge according to the status of acaricides resistance in the country.  相似文献

Clusters of pseudohyphae are commonly seen on Gram stain of blood cultures from patients with Candida albicans fungemia. Whether this morphologic feature is useful for differentiating C. albicans from other yeasts has not previously been systematically evaluated. Yeast morphology on Gram stain of blood cultures from consecutive patients with fungemia detected by the Bactec automated blood culture system was prospectively assessed and correlated with the final culture-based yeast identification. The distribution of yeast in 60 consecutive patients with fungemia included Candida spp. (C. albicans 43%, Candida glabrata 28%, Candida parapsilosis 8%, Candida krusei 7%, Candida tropicalis 5%, Candida dubliniensis 2%, and Candida lusitaniae 2%), Rhodotorula mucilaginosa 2%, and Cryptococcus neoformans 3%. Upon analysis of the first positive blood culture bottle per patient, the presence of clustered pseudohyphae on Gram stain had a sensitivity, specificity, positive predictive value, and negative predictive value of 85, 97, 96, and 89%, respectively, for C. albicans. The sensitivity and specificity of aerobic vs Myco/F blood culture bottles were 96 and 95% vs 25 and 100%, p < 0.001, respectively. Inter-rater agreement for ten separate observations among five reviewers was 100%. The presence of pseudohyphae clusters by Gram stain of blood cultures is useful in distinguishing C. albicans from non-albicans yeast. Additional studies are necessary to determine the clinical impact of these findings and their validity with other blood culture systems.  相似文献

The objective of this study was to investigate the incidence and density of Demodex folliculorum in the patients with rheumatoid arthritis (RA). Forty-one patients with RA and twenty-seven age and sex matched healthy controls were enrolled in this study. Disease Activity Score (DAS 28) was used for the assessment of disease activity. Out of 41 patients, 33 were females and 8 males. The mean disease duration was 10.9 ± 8.2 years. The mean DAS 28 was 3.8 ± 1.2. No statistically significant differences in the incidence and density of Demodex mites were found between patients with RA and controls. Although immunosuppression is thought to be a risk factor for the D. folliculorum infestation no such correlations could be found in the 41 immunosuppressed patients with RA, therefore, further studies with larger groups are needed.  相似文献

In this paper Steringotrema microacetabularis (Suriano et Martorelli, 1983) is redescribed and transferred to Bacciger Nicoll, 1924 in the Faustulidae Poche, 1926 based on newly collected material from the type-host, Paralichthys orbignyanus Valenciennes, 1839 and the type-locality, Mar Chiquita coastal lagoon, Buenos Aires Province, Argentina. A careful re-examination of the new specimens shows that some anatomical characteristics were ignored or incompletely described by the previous authors and they are included herein. The species is tentatively transferred to Bacciger with which it appears to have closest affinity. Despite the anatomical differences detailed in this paper, confirmation of this proposal must await further work, including molecular studies.  相似文献

Vibrio cholerae (V. cholerae) cholera toxin (CT), which causes a severe watery diarrheal illness, is secreted via the type II secretion machinery; it remains unclear, however, how this toxin is transported toward the machinery. In this study, we determined that the pH-dependent intrabacterial transport system correlates with the priming of CT secretion by V. cholerae. The secretion and production of V. cholerae treated at different pHs were examined by enzyme immunoassay. The localization of the CT was analyzed by immunoelectron microscopy. The CT secretion level rapidly increases in the alkaline-pH-treated V. cholerae but does so more slowly in neutral- and acidic-pH-treated V. cholerae. The CT was found to be densely localized near the membrane in the alkaline-pH-treated bacterial cytoplasm, suggesting that the CT shifts from the center to the peripheral portion of the cytoplasm following an extracellular rise in pH. The shift was observed in V. cholerae treated at alkaline pH for more than 10 min. The pH treatment did not enhance CT production at the same stage at which secretion and intrabacterial transport of the CT were enhanced. We propose that V. cholerae possesses a pH-dependent intrabacterial nanotransportation system that probably accelerates priming for CT secretion.  相似文献

Plasmodium, Haemoproteus, and leukocytozoon are the most important hematozoa in birds, which have been reported in different areas of the world. The present study was undertaken to find which blood protozoans exist in indigenous chickens in Shiraz, southern Iran and to evaluate hematological parameters in birds infected with hematozoas. Plasmodium and Aegyptianella were the two parasites found in 740 blood samples examined from indigenous chickens of which 29 (3.91%) were positive for Aegyptianella pullorum, 106 (14.32%) for Plasmodium gallinaceum, and 12 (1.62%) for A. pullorum and Plasmodium gallinaceum together. There was no significant difference between hematological parameters of non-infected and naturally infected chickens with Plasmodium gallinaceum, A. pullorum, and both (P > 0.05). Low infection of indigenous chickens with A. pullorum, Plasmodium gallinaceum, and both had no significant effects on hematological parameters (P > 0.05), which is probably due to low parasitemia rate and immunity against these two parasites.  相似文献

These mechanisms are in part under the control of two interacting signalling pathways, the serotonergic and the brain-derived neurotrophic (BDNF) pathways.
