目的 观察硫酸镁对小鼠肾缺血再灌注损伤的抗氧化保护作用.方法 雄性小鼠随机分为假手术组、模型组和硫酸镁高、低剂量(120、60 mg/kg)组,无创动脉夹夹闭左肾蒂45 min和再灌注3 h制备急性肾缺血再灌注损伤模型,检测肾脏指数、血清尿素氮(BUN)和肌酐含量、肾组织丙二醛(MDA)含量以及超氧化物歧化酶(SOD)和谷胱甘肽过氧化物酶(GSH-Px)活力,HE染色观察肾组织学变化.结果 硫酸镁高、低剂量组小鼠肾指数分别为(0.72±0.05)和(0.74±0.07)、血清BUN含量为(12.36±2.24)和(15.58±1.92)mmol/L、血清肌酐水平为(98.23±4.37)和(114.63±6.24)μmol/L、肾组织MDA含量为(2.11±0.24)和(2.27±0.21)nmol/(mg.prot),肾组织SOD活力为(4.03±0.68)和(3.51±0.58)U/(mg.prot),硫酸镁高剂量组肾组织GSH-Px活力为(323.90±23.50)U/(mg.prot),与模型组比较,差异均有统计学意义(P<0.05),且肾组织病理变化较轻.结论 硫酸镁对小鼠急性肾缺血再灌注损伤具有保护作用,其机制可能与抑制脂质过氧化反应有关.  相似文献

BACKGROUND/OBJECTIVES

This study was performed to investigate the in vitro antioxidant and cytoprotective effects of fermented sesame sauce (FSeS) against hydrogen peroxide (H2O2)-induced oxidative damage in renal proximal tubule LLC-PK1 cells.

MATERIALS/METHODS

1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (OH), and H2O2 scavenging assay was used to evaluate the in vitro antioxidant activity of FSeS. To investigate the cytoprotective effect of FSeS against H2O2-induced oxidative damage in LLC-PK1 cells, the cellular levels of reactive oxygen species (ROS), lipid peroxidation, and endogenous antioxidant enzymes including catalase (CAT), superoxide dismutase (SOD), and glutathione peroxidase (GSH-px) were measured.

RESULTS

The ability of FSeS to scavenge DPPH, OH and H2O2 was greater than that of FSS and AHSS. FSeS also significantly inhibited H2O2-induced (500 µM) oxidative damage in the LLC-PK1 cells compared to FSS and AHSS (P < 0.05). Following treatment with 100 µg/mL of FSeS and FSS to prevent H2O2-induced oxidation, cell viability increased from 56.7% (control) to 83.7% and 75.6%, respectively. However, AHSS was not able to reduce H2O2-induced cell damage (viability of the AHSS-treated cells was 54.6%). FSeS more effectively suppressed H2O2-induced ROS generation and lipid peroxidation compared to FSS and AHSS (P < 0.05). Compared to the other sauces, FSeS also significantly increased cellular CAT, SOD, and GSH-px activities and mRNA expression (P < 0.05).

CONCULUSIONS

These results from the present study suggest that FSeS is an effective radical scavenger and protects against H2O2-induced oxidative damage in LLC-PK1 cells by reducing ROS levels, inhibiting lipid peroxidation, and stimulating antioxidant enzyme activity.  相似文献

目的研究不同剂量的槲皮素(Que)对大鼠高尿酸血症肾损伤的防治作用。方法将SD大鼠分为10组,空白组、模型对照组、别嘌醇组和槲皮素剂量组(50、40、30、20、10、5、2.5mg/kg)。利用腺嘌呤加盐酸乙胺丁醇灌胃法诱导大鼠制备高尿酸血症和肾损伤模型,Que预防治疗3周,测定与高尿酸血症和肾损伤相关的各项组织器官和血液生化指标。结果与模型组相比,槲皮素各剂量组均能降低血清尿酸(SUA)、尿素氮(SUN)、肌酐(SCr)、丙二醛(MDA)及肝脏黄嘌呤氧化酶(XOD)的水平;并能提高血清超氧化物歧化酶(SOD)的活性。而槲皮素降尿酸及对大鼠肾脏的保护作用较好的剂量为10~20mg/kg。结论槲皮素有干预腺嘌呤加盐酸乙胺丁醇诱导高尿酸血症和肾损伤的作用。可能的机制是通过抑制肝脏XOD的活性,增加机体清除氧自由基的能力、减少脂质过氧化来降低血清尿酸水平,起到对肾脏的保护作用。  相似文献

BACKGROUND: Sepsis is one of the major causes of death reported in intensive care units. A daily supplement of sesame oil for 1 week significantly attenuates oxidative stress-associated hepatic injury in septic rats. However, the excess intake of sesame oil may be associated with a health risk. This study investigates the effect of accumulative sesame oil on oxidative stress-associated hepatic injury after cecal ligation and puncture in rats. METHODS: Sesame oil was administered daily (4 mL/kg/d, orally) to rats, and the total intake of sesame oil ranged from 0 (control) to 140 mL/kg before cecal ligation and puncture in 9 groups of rats. Oxidative stress was examined by determining the levels of lipid peroxidation and glutathione. Hepatic injury was evaluated by measuring serum levels of aspartate aminotransferase and alkaline phosphatase. RESULTS: Rats that received sesame oil for 4 and 5 weeks had a lower body weight gain compared with those that received saline. Lipid peroxidation was decreased in the 20-mL/kg and 28-mL/kg groups, but it was increased in the 140-mL/kg group compared with the control group. Glutathione levels were increased in the < or =28-mL/kg groups compared with the control group. Serum levels of aspartate aminotransferase and alkaline phosphatase were reduced in the < or =28-mL/kg groups compared with the control group. Conclusion: Sesame oil does not demonstrate accumulatively enhanced protection against oxidative stress-associated hepatic injury after cecal ligation and puncture in rats.  相似文献

目的 研究RIFLE标准的急性肾损伤(AKI)分期与连续性肾脏替代治疗(CRRT)感染导致的多器官功能障碍综合征患者预后的关系,从而找出急性肾损伤患者最合适的治疗时机.方法 采用前瞻性随机对照研究方法,收集医院2008年1月-2011年1月121例严重感染导致的AK1患者,按照急性肾功能衰竭的RIFLE诊断标准分为Ⅰ、Ⅱ、Ⅲ期组;比较3组患者的住院死亡率和肾功能恢复率,同时比较不同组患者CRRT治疗前和治疗48 h的SOFA评分、APACHEⅡ评分、需要血管活性药物例数、降钙素原(PCT)、平均动脉压、氧合指数.结果 AKI Ⅰ、Ⅱ、Ⅲ期患者的住院死亡率分别为21.4%、25.6%和47.5%,Ⅲ期患者的死亡率明显比Ⅰ、Ⅱ期高;AKIⅠ、Ⅱ、Ⅲ期患者肾功能恢复率分别为76.2%、53.8%和22.5%,Ⅰ期和Ⅱ期患者的肾功能恢复率明显比Ⅲ期高,CVVH治疗48 h后,APACHEⅡ评分、SOFA评分和PCT在AKI Ⅰ期和Ⅱ期组患者显著降低,差异均有统计学意义(P<0.05);在AK1Ⅲ期患者中则变化不明显(P>0.05).结论 CRRT是治疗感染导致的多器官功能障碍综合征患者的有效手段,CRRT在RILFE标准早期(AKIⅠ、Ⅱ期)可明显改善感染导致的多器官功能障碍综合征患者的预后,而在RIFLE标准晚期(AKIⅢ期)行CRRT则对患者预后影响不大,故临床上必须重视CRRT时机的选择.  相似文献

Objective: Monochloroacetic acid (MCA) is corrosive to skin, and causes not only chemical injury but also fatal systemic poisoning. Little is known about the cause of death. We studied the acute toxicity of MCA before clinical symptoms appeared in fasting rats. Methods: Blood samples were analyzed 2 h after subcutaneous MCA injection (Ld90: 162 mg/ml kg body weight). Control rats were injected with saline. Results: Aspartate aminotransferase (AST) and alanine aminotransferase (ALT) were about 1.5-fold higher than in the controls, and mitochondrial AST (mAST) was 2-fold higher. Blood urea nitrogen and creatinine were significantly increased, while serum glucose was significantly decreased in the treated group. Lactate was 6-fold higher and pyruvate was 13-fold higher than in the controls. Conclusions: MCA caused injury to the liver and kidneys but these injuries were slight. However, the larger increase in mAST indicated that hepatocellular mitochondria were selectively targeted. Hepatocellular mitochondrial injury decreased gluconeogenesis and caused hypoglycemia and extremely high levels of lactate and pyruvate. Hypoglycemia and lactic acidosis were insidious before the critical symptoms appeared and this combination accelerated to death, affecting other organs such as the heart and brain. Nosotropic therapy of these abnormalities up to the appearance of symptoms may help to establish an early therapy for skin exposure to MCA.  相似文献

目的探讨神经导向因子-1(Netrin-1)联合肾损伤因子-1(Kim-1)在新生儿窒息后急性肾损伤(AKI)风险预测中的临床价值,为临床诊治提供参考依据。方法纳入中南大学湘雅二医院2015年5月-2017年5月收治的窒息新生儿96例。根据患儿AKI发生情况,将其分为发生AKI组、未发生AKI组,比较两组临床资料,通过绘制受试者工作特征曲线(ROC)明确Netrin-1联合Kim-1预测AKI的敏感度、特异度与最佳截断值,利用COX回归性分析确定AKI发生的独立影响因素。结果在92例患儿中,有42例(43.75%)发生AKI,54例(56.52%)未发生AKI;发生AKI组的重度窒息、尿液减少占比及Netrin-1、Kim-1水平较未发生AKI组更高,差异有统计学意义(均P<0.05);通过COX回归性分析发现,窒息程度、尿液减少、Netrin-1、Kim-1与新生儿窒息后AKI发生呈正相关(P<0.05)。结论Netrin-1联合Kim-1对新生儿窒息后AKI风险的预测效果较好,且两者均为AKI发生的独立危险因素。  相似文献

研究了大鼠腹腔注射1.0mgCd2+/kg体重的氯化镉和0.32mgPt2+/kg体重顺铂后,镉和铂在体内的分布及其肝、肾毒性。结果发现:镉主要分布于肝脏,随着染毒时间的延长,镉有向肾脏集中的趋势;铂主要分布于肾;氯化镉引起的肝损害发生在肾损害之前,氯化镉引起的肾损害与肝损害有关;顺铂引起的肝损害发生在肾损害之后,其肾损害与肝损害的发生无关。  相似文献

目的观察摄食地沟油对小鼠肝肾功能及脂肪代谢的影响。方法将72只小鼠平均分为6组,其中地沟油组与食用油组各3组,按照5、10和20μL/g剂量的地沟油或食用油连续隔天给予小鼠灌胃8周,比较两组小鼠灌胃前后丙氨酸氨基转移酶(ALT)、门冬氨酸氨基转移酶(AST)、血尿素氮(BUN)、血肌酐(Scr)、甘油三酯(TG)和总胆固醇(TC)水平。结果地沟油的水分含量、酸价和过氧化值指标均超标。连续灌胃8周后,5μL/g剂量地沟油组小鼠ALT明显高于食用油组(P0.05);10μL/g剂量地沟油组小鼠AST高于食用油组(P0.05)。5、10和20μL/g剂量地沟油组小鼠Scr均高于同剂量食用油组(P0.05);10和20μL/g剂量地沟油组小鼠BUN均高于同剂量食用油组(P0.05);但相同剂量地沟油和食用油组小鼠的TG和TC未见统计学差异(P0.05)。结论连续8周摄食地沟油对小鼠肝、肾功能均有明显损害作用。  相似文献

目的探讨尿中性粒细胞明胶酶相关脂质运载蛋白(NGAL)、肾损伤分子-1(KIM-1)和血清肌酐在成年人心脏手术后急性。肾损伤(AKI)早期诊断中的价值。方法收集65例行心脏外科手术患者术前及术后24、48、72h的血、尿标本,检测血清肌酐及尿NGAL、KIM-1,其中发生AKI(AKI组)34例,未发生AKI(非AKI组)31例,并与20例健康体检者(对照组)进行比较。采用Logstic回归模型判断血清肌酐及尿NGAL、KIM.1的早期诊断价值。结果心脏手术后AKI的发生率为52.31%(34/65)。AKI组和非AKI组术前及术后24、48、72h血清肌酐及尿NGAL、KIM.1均明显高于对照组[血清肌酐:(62.44±22.26)、(77.38±24.38)、(98.24±25.02)、(98.14±45.26)μmol/L和(56.67±21.68)、(59.25±15.58)、(55.40±22.91)、(40.10±23.45)¨mol/L比(16.43±5.01)斗mol/L,尿NGAL:(4.81±0.65)、(9.48±3.29)、(11.87±3.93)、(7.66±1.52)μg/L和(4.67±0.53)、(8.89±2.64)、(11.46±3.60)、(7.47±1.63)μg/L比(2.87±0.48)μg/L,尿KIM-1:(47.93±5.54)、(61.89±12.44)、(100.21±27.32)、(67.69±8.72)μg/L和(47.12±4.08)、(56.38±9.27)、(90.14±27.26)、(69.40±8.90)μg/L比(32.21±12.54)μg/L],差异有统计学意义(P〈0.01);AKI组和非AKI组术后24、48、72h尿NGAL、KIM-1明显高于术前,差异有统计学意义(P〈0.01);AKI组术后24h血清肌酐及尿NGAL、KIM.1均明显高于非AKI组,差异有统计学意义(P〈0.01)。术后24h尿NGAL、KIM-1诊断AKI的敏感度和特异度与血清肌酐比较差异无统计学意义(P〉0.05)。结论接受体外循环心脏手术的成年人术后AKI发生率高;术后24h对于传统诊断金标准的血清肌酐与早期生物学标记物尿NGAL、KIM-1在诊断AKI上具有相似的敏感度和特异度。  相似文献

目的 分析探讨影响脓毒症相关急性肾损伤(acute kidney injury, AKI)患者接受连续性肾脏替代治疗(continuous renal replacement therapy, CRRT)的相关因素。方法 选取我院于2017年6月—2018年12月间收治的278例脓毒症相关AKI患者为研究对象,根据患者是否选择接受CRRT将所有患者分为甲、乙2组,其中甲组为采取CRRT的患者,乙组则均为不接受CRRT的患者。记录2组患者的临床脏器功能评分和疾病严重程度评分,比较2组患者的一般资料及实验室检查结果,对影响患者接受CRRT的相关危险因素进行Logistic回归分析。结果 甲组患者的贫血程度重于乙组患者,凝血功能及肾功能均显著差于乙组患者,B型钠尿肽和乳酸水平均显著高于乙组患者(P均<0.05)。甲组患者的疾病严重程度重于肾脏、肝脏、肺脏损伤率等均显著高于乙组患者(P均<0.05)。Logistic回归分析结果显示,氧分压、二氧化碳分压、血清肌酐水平、急性生理与慢性健康评分、ICU住院时间等因素均是影响患者接受CRRT的相关危险因素。结论 氧分压、二氧化碳分压、血清肌酐水平、急性生理与慢性健康评分、ICU住院时间等均是影响脓毒症相关AKI患者接受CRRT的相关因素,具有较高的临床指导价值。  相似文献

One of the factors negatively influencing the outcome after kidney transplantation is ischemia-reperfusion (I/R) injury. Preoperative fasting is able to confer protection against I/R injury. We hypothesized that the protection imposed by preoperative fasting is mediated by increased levels of acylated ghrelin. Male C57BL/6 mice, 10 to 12 weeks old, were fasted for 1, 2, or 3 days, after which, acylated ghrelin levels were determined. Ad libitum fed mice were injected with acylated ghrelin or phosphate-buffered saline before renal I/R injury. Furthermore, mice were fasted for 3 days during which they were injected with a growth hormone secretagogue receptor antagonist, to block the effects of ghrelin, or a vehiculum. Bilateral renal I/R injury was induced by clamping the artery and vein of the left and right kidney simultaneously for 37 minutes. Kidney function was assessed by means of serum urea values determined at 24 and 48 hours after reperfusion. Fasting significantly increased acylated ghrelin serum levels. Ghrelin suppletion in ad libitum fed animals or ghrelin receptor blockade in fasted animals did not affect renal function after I/R injury. Our data suggest that the increased levels of acylated ghrelin induced by fasting do not mediate its protection against renal I/R injury.  相似文献

目的观察参芎葡萄糖治疗急性肾衰竭的疗效。方法将80例急性肾衰竭患者随机分为治疗组和对照组,于3周后分别进行血肌酐(SCr)、血尿素氮(BUN),纤维蛋白原等相关指标检测。结果治疗组治疗3周后血肌酐(SCr)、尿素氮(BUN),纤维蛋白原均较治疗前明显改善(P﹤0.05),且血肌酐、尿素氮,纤维蛋白原较对照组治疗后明显改善(P﹤0.05)。结论参芎葡萄糖注射液可以快速改善急性肾衰竭患者的肾功能。  相似文献

目的研究大蒜油对由对乙酰氨基酚造成的肝损伤的预防作用。方法将75只小鼠随机分组:对照组、模型组、大蒜油高、中、低剂量组,每组15只小鼠。大蒜油组灌胃相应剂量大蒜油,对照组和模型组灌胃等体积玉米油;2 h后除对照组外各组灌胃对乙酰氨基酚,饲养8h,禁食1 6h称重,摘除眼球取血,测定生化指标;摘取肝脏,做病理切片。结果模型组血清A L T、A ST水平与对照组相比显著升高,两者与对照组比较差异均有统计学意义;大蒜油各剂量组均能降低生化值。大蒜油组无肝坏死,肝脏颜色较为红润,包膜光滑。结论大蒜油对对乙酰氨基酚造成的肝损伤有一定的预防作用。  相似文献

目的研究小鼠肾小体发生发育过程中血管球和肾小囊体积的增长规律。方法光镜下应用体视学方法对胚胎和生后发育期小鼠肾血管球和肾小囊的绝对和体积进行测量。结果从胚龄16天到生后40天血管球体积增大约86倍,肾小囊体积增大约32倍。生后7天之前,血管球的相对体积增大迅速,而肾小囊的相对体积减小迅速,之后二者变化平缓。结论肾小体发育中血管球和。肾小囊的体积变化为肾小体的滤过功能的逐渐成熟提供了形态结构基础。  相似文献

目的 建立刀豆球蛋白A(ConA)诱导小鼠实验性肝损伤的模型。方法Balb/C小鼠40只,分别尾静脉注射ConA,根据注射剂量,小鼠被随机分为4组(每组10只):A组10mg/kg;B组20mg/kg;C组30mg/k;另设D组作为实验对照组,尾静脉仅注射生理盐水。给药后8h观察小鼠血清丙氨酸转氨酶(ALT)活性、死亡率和肝组织的病理改变。选择最佳ConA剂量,观察此剂量下不同时间点ALT活性、死亡率和肝组织的病理改变特点。结果经ConA处理后8h,A组与D组无小鼠死亡,B组死亡3只,C组小鼠全部死亡;各组血清AJJ值:A组为(215.55±70.19)IU/L,B组为(2516.14±764.69)IU/L,均显著高于对照组D组的(57.30±12.21)IU/L(分别t=-8.466、t=10.143,均P=0.000)。光镜下,A组可见肝细胞变性;B组和C组可见肝组织炎症细胞浸润和肝细胞变性、坏死,以C组为甚。20mg/kg Con A尾静脉注射,小鼠死亡率、ALT活性、肝组织炎症细胞浸润和肝细胞变性、坏死在一定时间范围内(24h)随时间增加而加剧。结论ConA小鼠实验性肝损伤模型建立,ConA剂量以20mg/kg、观察时间以8h为宜。  相似文献

目的:探讨硫酸镁和复方丹参联合应用对新生儿窒息(NA)心肌损伤的治疗作用及安全性。方法:NA合并心肌损伤患儿24例,随机分为对照组和治疗组各12例。对照组应用1,6-二磷酸果糖(FDP)治疗,剂量250mg.kg-1,静脉点滴连用7天。治疗组应用硫酸镁和复方丹参治疗:硫酸镁剂量200mg.kg-130 m in静脉滴注,继以10 mg.kg-1.h-1维持量用微量输液泵持续泵入,连用5天;复方丹参1 m l.kg-1静脉点滴,疗程7天。观察心肌钙蛋白I(cTnI)、肌酸激酶心肌同工酶(CK-MB)、肌酸激酶(CK)等指标的变化。结果:(1)治疗前治疗组和对照组cTnI分别为(1.83±2.01)ng.m l-1和(1.78±1.86)ng.m l-1;治疗后分别为(0.06±0.02)ng.m l-1和(0.15±0.11)ng.m l-1治疗后两组相比结果有显著性差异(P<0.05)。(2)治疗前治疗组和对照组CK-MB分别为(69.3±30.2)IU.L-1和(61.7±38.5)IU.L-1;治疗后两组分别为(31.3±12.5)IU.L-1和(43.2±16.8)IU.L-1,均较治疗前明显降低(P<0.05)。(3)治疗前治疗组和对照组CK分别为(968.3±580.2)IU.L-1和(897.5±502.7)IU.L-1;治疗后两组分别为(210.4±178.3)IU.L-1和(357.3±190.2)IU.L-1,与治疗前相比均有显著差异(P<0.05)。结论:硫酸镁和复方丹参对NA心肌损伤有明确治疗作用,无不良反应。  相似文献

目的:探讨复合氨基酸片(谷氨酰胺、精氨酸、甘氨酸和L-半胱氨酸)对四氯化碳(CCl4)肝损伤的保护作用。方法:将40只ICR小鼠随机分成阴性对照组、CCh模型组及490mg、980nag/(kgb.w)两个剂量组,剂量组每天灌胃给予复合氨基酸片,阴性对照组和CCIn模型组给予双蒸水。30d后,CCl4模型组及剂量组以5ml/(kgb.w)的剂量一次性经口给予色拉油配置的0.5%CCl4,24h后处死所有小鼠,测定血清丙氨酸转氨酶(ALT)、天冬氨酸转氨酶(AST)水平,取肝脏左叶进行组织病理学检查。结果:980mg/(kgb.w)剂量组的ALT、AST水平降低,与cch模型组比较,有极显著性差异(P〈0.01);980mg/(kgb.w)剂量组发生肝细胞坏死的病变程度减轻,肝细胞坏死评分降低,与CCl4模型组比较,有极显著性差异(P〈0.01)。结论:复合氨基酸片对CCl4肝损伤具有辅助保护作用。  相似文献
