目的:探讨fms样酪氨酸激酶(Flt-1)基因多态性与低氧训练敏感性的关联性。方法:选取70名中国北方平原地区汉族男子进行4周高住高练低训(HiHiLo),方案为每日在低氧环境(O2浓度为14.5%~14.8%,大约相当于海拔3000米)中休息10h,每周进行3次75%VO2max低氧训练(O2浓度为15.4%,大约相当于海拔2500米),其余时间在常氧下训练。测试其HiHiLo前后的VO2max及训练期间的血象指标。用PCR-RFLP法分析Flt-1基因SNP/A193019G多态性。结果:(1)4周HiHiLo后,AA基因型VO2max增加幅度高于AG基因型57.54%,但无显著性差异。(2)在HiHiLo期间(除第23天外),AA基因型Hb的变化幅度始终高于AG基因型,但无显著性差异。结果提示,与AG基因型者相比,AA基因型者可能有对低氧训练更敏感的趋势。  相似文献

肌红蛋白基因多态与有氧训练效果的关联性分析   总被引:1,自引:0,他引:1  
目的:探讨肌红蛋白第2外显子的单核苷酸多态(A79G)与有氧耐力训练效果的关联性。方法:对中国北方新征入伍的104名汉族士兵进行每次5000米、每周3次、训练强度为95~105%通气无氧阈(VT)、为期20周的耐力训练。递增负荷运动实验测定受试对象训练前后最大摄氧量(VO2max)和通气无氧阈(VT),PCR-RFLP法测定基因多态性,分析基因多态性与训练效果之间的关联性。结果:3种基因型的分布频率分别为AA(0.52)、AG(0.45)、GG(0.03),分布频率符合Hardy-weinberg遗传平衡定律;训练后3种基因型VO2max的组间比较均无显著性差异(P>0.05);训练后GG组和AG组VT显著提高(P<0.05),AA组达到VT时仅HR显著升高,其余指标均无显著变化(P>0.05)。在VO2max和VT的变化率(Δ%)方面,GG组受试者提高最多,AG组居中,AA组提高最少。提示肌红蛋白基因第2外显子的单核苷酸多态(A79G)与有氧耐力训练效果有一定关联,79G等位基因携带者对有氧耐力训练可能更敏感。  相似文献

目的:探讨瘦素受体(OB-R)基因第6外显子Gln223Arg多态性与有氧耐力及有氧耐力训练效果的关联性。方法:对新征入伍的102名中国北方汉族士兵进行为期18周、每周3次、每次5000米、强度为95~105%通气阈(VT)的耐力训练。采用跑台逐级递增负荷运动实验测定受试者训练前后最大摄氧量(VO2max),并在12km/h跑速时测定跑节省化(RE)时的心率(HR)、通气量(VE)和摄氧量(VO2);利用限制性片断长度多态(PCR-RFLP)法进行OB-R基因Gln223Arg多态性解析。结果:(1)OB-R基因Gln223Arg多态性在中国北方汉族人群中的分布频率分别为AA:3%、AG:25%、GG:72%,符合Hardy-Weinberg遗传平衡定律;(2)按基因型分组后,A等位基因携带者(AA+AG)训练前的最大摄氧量相对值明显高于未携带者(GG),其他指标训练前后在两组之间无显著性差异。结论:OB-R基因第6外显子的Gln223Arg多态性与有氧能力可能存在关联,而与有氧耐力训练效果不关联。  相似文献

目的:探讨中国北方汉族男性血管内皮生长因子(VEGF)基因C936T单核苷酸多态性与HiHiLo后心功能变化的关联性。方法:选取62名中国北方平原地区汉族男性进行为期30天高住高练低训(HiHiLo),每日在低氧房(O2浓度为14.8%~14.3%,海拔约2800~3000米)休息10小时,每周进行3次75%VO2max强度的低氧训练(O2浓度为15.4%~14.8%,海拔约2500~2800米),运动时间为30mim/次,其余时间在常氧环境下训练。使用Doppler超声心动图仪测试HiHiLo前后左心室结构和功能。采用PCR-RFLP法分析VEGF基因C936T单核苷酸多态性。结果:VEGF基因C936T多态性与HiHiLo后的LVM、LVMI、SV以及SI的变化有关联。其中,CC基因型LVM、LVMI、SV及SI增加幅度均显著高于CT基因型。结论:中国北方汉族男性VEGF基因C936T多态性的CC基因型者在HiHiLo训练后产生了更好的心脏适应性变化,可以作为预测HiHiLo后部分心功能(SV、SI)变化的分子遗传学标记。  相似文献

目的:探讨葡萄糖转运蛋白1(glucose transporter 1)基因第2内含子上G22999T单核苷酸多态性(SNP)与HiHiLo(living high-exercise high-training low)对递增负荷运动中左心室结构和动态心功能的影响的关联关系。方法:采用关联分析(Association-Study)研究方法,聚合酶链反应-限制片段长度多态性(PCR-RFLP)技术,选取72名健康受试者,在模拟海拔2500米高度进行4周低氧训练(低氧暴露10h/d,3次75%VO2max低氧训练/周,共4周)。实验前后,运用二维及Doppler超声心动图技术,分别于HiHiLo前后测量72名受试者在递增负荷全过程中的心脏结构指标、收缩功能指标及舒张功能指标。结果:(1)GLUT1基因SNP/G22999T经扩增、酶切后得到GG、GT和TT三种基因型,符合Hardy-Weinberg遗传平衡定律,具有群体代表性。2)GLUT1基因SNP/G22999T多态位点T等位基因携带者短轴缩短率(SF)减小量、递增负荷实验100W(负荷下ESV减小量和50W、100W负荷下的EDT增加量均明显高于未携带T等位基因者。结论:SNP/G22999T多态位点T等位基因携带者较非携带者对HiHiLo训练更敏感,但将该多态位点作为筛选HiHiLo对左心室结构和功能影响效果的分子标记还需加大样本进一步研究。  相似文献

目的:探讨中国北方平原地区汉族男子过氧化物酶体增殖受体γ辅激活因子α(PPARGC1)基因多态性分布特点以及与耐力训练效果的关联性。方法:102名无训练史的健康男子进行18周有氧耐力训练,测试训练前后的最大摄氧量、跑节省化。用PCR-RFLP法分析PPARGC1基因的G482S、A2962G和T394T多态性。结果:(1)3个多态位点分布频率均符合H-W平衡,且有显著的种族差异。(2)A2962G多态性与ΔVO2max(ml/min/wt)可能关联,GG基因型群体显著性高于AG和AA,但GG基因型频率仅为3%;未发现其他单点多态性和单体型与训练敏感性有较强的关联。结论:PPARGC1基因的G482S和T394T多态性不是预测个体耐力训练敏感性的基因标记;对于A2962G,则有必要加大样本进一步确认。  相似文献

目的:探讨过氧化物酶体增殖受体γ辅激活因子1α(PGC-1α)基因多态性位点与优秀耐力运动员运动能力的关联性及其作用机制。方法:应用Case-Control实验设计,分析42个单核苷酸多态性位点在优秀耐力运动员组(235人)和对照组(504人)中的分布特征。采用双荧光素酶报告基因方法分析阳性关联多态性位点的生物学功能。结果:(1)8个单核苷酸多态性位点的基因型/等位基因在优秀运动员组与对照组中的分布频率有显著差异。(2)rs6821591的TT基因型、rs12650562的TT基因型、rs12374310的CC基因型、rs4452416的GG基因型和rs4697425的AA基因型在运动员组的频率显著高于对照组,相应的相对荧光素酶活性也显著高于对照组,但rs4452416的GG基因型频率较低。结论:PGC-1α基因的rs6821591、rs12650562、rs12374310和rs4697425四个多态性位点与优秀耐力运动员运动能力状态关联,并且上调其基因转录。  相似文献

目的:探讨雄激素受体(AR)基因外显子1中(CAG)n多态在中国北方汉族男性中的分布特征及其与低氧训练后VO2max变化的关联性。方法:65名中国北方汉族健康青年男性进行为期30天HiHiLo(常压低氧)训练,晚上居住房间氧浓度为14.3~14.8%(模拟海拔2800~3000米),每周进行3次75%常氧VO2max的低氧训练,训练环境氧浓度约15.4%(模拟海拔约2500米)。实验前后测试体重和VO2max等指标,采用GeneScan和测序方法分析AR(CAG)n多态的等位基因(基因型)。结果:(1)共观察到(CAG)12、(CAG)16-28、(CAG)30共15种重复次数的等位基因(基因型),其中22次重复等位基因分布频率最高;(2)当分别以21和22次重复为分割点划分基因型时,短链组体重基础值均显著低于长链组;(3)当分别以21和22次重复为分割点划分基因型时,低氧训练后短链基因型携带者ΔVO2max和ΔrVO2max均显著高于长链组(P<0.01)。结论:中国北方汉族男性AR基因(CAG)n多态与HiHiLo(常压低氧)训练效果之间存在关联,重复次数较少基因型携带者的低氧训练敏感性较好。  相似文献

目的探讨核呼吸因子2(NRF2)基因多态性与中国北方平原地区汉族男性有氧能力的关联性。方法102名无训练史的健康男子进行18周有氧耐力训练,测试训练前后的最大摄氧量(VO2max)和12km/h跑速下的跑节省化(RE)。用PCR-RFLP分析NRF2基因的rs12594956、rs7181866和rs8031031多态性。结果(1)三个多态位点均符合H-W平衡。rs8031031和rs7181866强连锁不平衡(D’=0.966,r2=0.903);(2)三个单点多态性及其构成的单体型与VO2max的初始值及训练敏感性均不关联;(3)rs8031031多态性与RE/VO2(L/min)关联,CT基因型显著高于CC基因型;rs7181866和rs8031031的多态性与△RE/VO2(L/min,ml/min/LBM)关联,AG或CT基因型比相应的其他基因型有更高的训练敏感性;单体型分析表明携带ATG单体型群体的RE/VO2(L/min)训练敏感性最高(P=0.006),比其他群体高57.54%。提示NRF2基因的多态性可能预测部分中国北方地区汉族男性有氧能力的差异。  相似文献

目的:探讨中国北方平原地区汉族男子脑型肌酸激酶(CKB)基因多态性与耐力训练效果的关联性。方法:102名无训练史的健康男子进行18周有氧耐力训练,测试训练前后的最大摄氧量、跑节省化(RE)各指标。用PCR-RFLP法分析CKB基因启动子上rs3759582和rs3759584两位点的单核苷酸多态性。结果:二个多态位点均符合H-W平衡,不存在强的连锁关系;训练后rs3759582位点AA基因型最大摄氧量变化率、跑节省化变化率与AC基因型组间均无显著性差异(P>0.05);训练后rs3759584位点TT基因型△RE/VO2、△RE/rVO2、△RE/HR和△RE/VE显著低于CT和CC基因型(P<0.05),但是CC基因型频率仅为3%。结论:CKB基因的rs3759582多态性不是预测个体耐力训练敏感性的基因标记;rs3759584与个体的耐力训练敏感性相关,但需加大样本进一步确认。  相似文献

目的:探讨高住低训对女子跆拳道运动员运动能力和血象的影响。方法:13名优秀女子跆拳道运动员随机分为实验组(高住低训,n=8)和对照组(n=5),分别在模拟海拔2500m低氧舱(氧浓度15.4%左右)内和平原环境下进行为期4周的高住低训和跆拳道常规专项训练。分别于实验前、高住低训第28天和高住低训结束后10天进行有氧运动能力和无氧运动能力测试,并于实验前1天、高住低训期间每周及高住低训结束后第3、12天清晨空腹取血进行血细胞分析。结果:(1)实验组高住低训第28天和高住低训结束后10天VO2max和PWC170均显著高于实验前(P<0.01),4000m成绩显著优于实验前(P<0.01)。对照组变化不明显。(2)实验组高住低训结束后10天Wingate最高功率显著高于对照组(P<0.05)。实验组高住低训第28天和高住低训结束后10天Wingate平均功率均显著高于自身实验前(P<0.05和P<0.01),Wingate疲劳指数则分别显著低于自身实验前(P<0.05和P<0.01)。对照组无明显变化。(3)实验组高住低训第28天血红蛋白、红细胞数目、红细胞压积显著高于自身实验前(P<0.05),实验组在高住低训第25天和28天血红蛋白水平分别显著高于对照组(P<0.05和P<0.01)。结果表明,为期28天的模拟海拔2500m高度的高住低训能提高女子跆拳道运动员有氧运动能力,同时在一定程度上提高其无氧功和60秒踢靶成绩。另外,高住低训可以提高与跆拳道运动员机体载氧能力直接相关的血液指标水平,这种改变主要表现在实验第3、4周。  相似文献

The purpose of this study was to monitor general and individual changes in hematological variables during long-term endurance training, detraining and altitude training in elite Olympic distance triathletes. Over a period of three years, a total of 102 blood samples were collected in eleven (7-male and 4 female) elite Olympic distance triathletes (mean +/- SD; age = 26.4 +/- 5.1 yr; VO(2) max = 67.9 +/- 6.6 ml/min/kg) for determination of hemoglobin (Hb), hematocrit (Hct), red blood cell count (RBC), Mean corpuscular hemoglobin (MCH), Mean corpuscular hemoglobin content (MCHC), Mean corpuscular volume (MCV) and plasma ferritin. The data were pooled and divided into three periods; off-season, training season and race season. Blood samples obtained before and after altitude training were analyzed separately. Of all measured variables only RBC showed a significant decrease (p < 0.05) during the race season compared to the training season. Hematological values below the lower limit of the normal range were found in 46 % of the athletes during the off-season. This percentage increased from 55 % during the training season to 72 % of the athletes during the race season. Hemoglobin and ferritin values were most frequently below the normal range. There was a weak correlation between Hb levels and VO(2) max obtained during maximal cycling (r = 0.084) and running (r = 0.137) tests. Unlike training at 1500 m and 1850 m, training at an altitude of 2600 m for three weeks showed significant increases in Hb (+ 10 %; p < 0.05), Hct (+ 11 %; p < 0.05) and MCV (+ 5 %; p < 0.05). Long-term endurance training does not largely alter hematological status. However, regular screening of hematological variables is desirable as many athletes have values near or below the lower limit of the normal range. The data obtained from altitude training suggest that a minimum altitude (>2000 m) is necessary to alter hematological status.  相似文献

The purpose of this study was to investigate if strength training affects red blood cell variables in physically inactive men when taking into account seasonal variations. Seventy-four men aged 20-45 were randomly assigned to training (n = 52) and control (n = 22) groups. Training group underwent 20-week progressive strength training. Body composition and maximal voluntary contraction (MVC) during knee extension were measured before and after intervention. Fasting blood samples were analysed for haematocrit (Hct), count of red blood cells (RBC), haemoglobin (Hb), mean cell haemoglobin concentration (MCHC), and mean cell volume (MCV) at baseline, 10-week and 20-week follow-up. MVC and lean body mass increased in the training group. Hct, Hb and MCHC showed seasonal variation in the control group. The training group increased their Hct from 44.7 +/- 2.6 % to 45.4 +/- 2.5 % (p = 0.026) while the control group decreased their Hct from 44.3 +/- 2.2 % to 43.1 +/- 2.6 % (p = 0.037) after 20-week intervention. By contrast to the control group, the training group increased their Hct (p = 0.001), RBC (p = 0.005) and decreased their MCHC (p < 0.001) from 10-week to 20-week follow-up. We concluded that strength training could affect seasonal variation patterns of red cell variables. Unlike "sport anaemia" induced by endurance training, 20-week strength training elevated Hct.  相似文献

There have been a number of reports describing the hematological indicators of Andean residents living at altitudes above 4,000 m, but several confounding factors have made the published results difficult to interpret. To clear up the effect of hypoxia on hemoglobin concentration (Hb, g/dL), hematocrit (Hct, %) and red blood cell concentration (RBC, cells/microL), this publication describes and analyzes these variables in children, men, and women from three large and homogeneous populations living at 4,355 m (n = 151), 4,660 m (n = 400), and 5,500 m (n = 273) in the Southern Peruvian Andes. Hb, Hct, and RBC increase with age in men (p < 0.001), as well as in women (p < 0.001) at the three altitudes of the study. In children (boys and girls) living at 5,500, Hb increases 11% when compared with children living at 4,355 m, and in adults, Hb increases 9.6% when comparing the same altitudes. The maximum percentage increase in Hb with age was 5.6% at 5,500 m, in men and 3.2% at 4,355 m, in women. The average percentage of difference for the Hb concentration between adult men and women is 6.6% at 4,355 m, 9.8% at 4,660 m, and 11.6% at 5,500 m. The differences in Hb concentration between men and women can only be seen after puberty. Finally, Hb is higher in older than younger women, which confirms the role of menopause in the development of erythremia. The result of this analysis reinforces the notion that Hb and Hct seem to be stable and useful parameters for acclimatization only at moderate altitudes; with aging or with increasing altitude, they may become excessive and lose their efficiency to protect the venous oxygen pressure.  相似文献

PURPOSE: Alterations of the red blood cell system and iron metabolism can influence physical performance. On the other hand, exercise can influence hematological variables. The purpose of this epidemiological study was to investigate the characteristics of the red blood cell system and the iron metabolism in athletes of different sporting disciplines and at different levels of performance. METHODS: We studied 851 male subjects (747 athletes, 104 untrained controls). Hemoglobin (Hb), hematocrit (Hct), red blood cell count (RBC), iron, transferrin, ferritin (Fer), and haptoglobin were analyzed in standardized blood samples, obtained after 2 d of rest, considering levels of performance (internationally, nationally, locally competitive, and leisure time), distinctive sporting category (endurance- (END), strength- (POW), and mixed-trained (MIX)), and, within endurance athletes, distinctive disciplines (cycling (CYC) and running (RUN)). RESULTS: No difference was found between athletes and controls in Hb and Hct. Reduced Hb, Hct, and RBC levels were observed in END compared with POW and MIX. These findings can mainly be attributed to exercise-induced plasma volume expansion, and only to a lesser degree and in selected athlete populations to hemolysis, as low haptoglobin is only observed in RUN, not in CYC, suggesting that not exercise itself but the "traumatic" movement of running might trigger the destruction of red blood cells. Physical activity of increasing duration and workloads (leisure time compared with competitive athletes) leads to decreased Fer levels in athletes, disregarding their discipline, but more pronounced in RUN. CONCLUSION: Physical training itself has no significant effect on selected hematological variables in athletes compared with untrained controls. The specific type and duration of exercise is of major importance in the adaptations of the blood cell system and the iron metabolism.  相似文献

Knowledge is sparse about the extent of potential dehydration due to prolonged strenuous cycling and its haematological acute effects on the haematocrit (Hct) in study populations credibly not taking any kind of doping. With increasing training load levels of Hct and haemoglobin (Hb) decrease in both amateurs and professionals as a long-term consequence due to expanded plasma volume (PV). On a short-term basis, however, counteracting dehydration potentially brought about by endurance exercise may cause a rise in Hct bringing competitive cyclists into conflict with the current condition regulations and Hct cut-off of 50 % set by the International Cycling Union (UCI) in its fight against erythropoietin (rhEPO) doping. On the other hand adequate and sufficient fluid substitution being substantial for a successful endurance performance should prevent any pronounced Hct rises. To study the haematological acute effects of prolonged strenuous cycling we measured Hct, Hb, red blood cell (RBC) count and plasma protein in a reliably 'clean' population of 38 well-trained male amateur cyclists before, immediately after and one day after an extraordinary ultramarathon. The pre-race levels of Hct, Hb and RBC count were placed in the lower range of normal distribution and well below the Hct cut-off limit of the UCI. Immediately post-exercise the mean levels of Hct, Hb, RBC count and protein remained unchanged. One day after race, however, all four parameters significantly dropped by 3 %, 6.7 %, 6.5 %, 9.9 % respectively (p < 0.001), indicating marked post-exercise PV expansion. The calculated percentage increase in PV was 11.9 %. No evidence for coexisting exercise-induced haemolysis was found. Our study shows that in "clean, rhEPO-free" amateur cyclists who involve in strenuous marathon cycling the haematological short-term effects of extraordinary marathon cycling consist in considerable PV expansion making Hct values fall on the following day. The findings - gained from amateurs though - suggest that despite all its disadvantages the UCI Hct cut-off represents an appropriate means to discourage from excessive rhEPO doping at least as long as the available direct methods for detecting this kind of misuse are not yet applied by the international sports federations.  相似文献

 目的 明确心脏型肌球蛋白结合蛋白C(cardiac myosin binding protein c, cMYBPC)18443 A/G 多态对肥厚型心肌病(hypertrophic cardiomyopathy,HCM)临床表型的影响。 方法 选择226例无血缘关系的HCM患者,超声心动图显示原因不明的心脏左室壁厚度≥13 mm,或心尖部肥厚或右室肥厚,并排除长期高血压、缺血性心肌病及其他可引起继发性心室肥厚的疾病,并选择226例性别、年龄匹配的健康人作为对照组。抽取外周静脉血,分离白细胞,从白细胞中提取基因组DNA,设计特异引物,PCR扩增包含MYBPC3第30号外显子A/G18443多态的目的片断,根据PCR产物限制性酶切后的长度及ABI377 DNA测序仪测序来判定上述多态。 结果 cMYBPC基因18443A/G多态位点不同基因型在HCM患者和对照组的分布频率无明显差别。在cMYBPC基因18443位置存在A/G多态,携带GG基因型的HCM患者肥厚心肌的厚度(25.2±5.9)mm大于携带AG基因型和AA基因型(18.9±4.98)mm的HCM患者。 结论 cMYBPC不仅是HCM的致病基因,而且也可能是HCM的修饰基因。  相似文献

目的:观察世居高原的中长跑运动员进行平原训练时身体机能的变化。方法:以12名世居高原男子中长跑运动员为研究对象,系统测定其平原训练前、平原训练1周后、3周后的促红细胞生成素(EPO)、红细胞(RBC)、血红蛋白(Hb)、网织红细胞(RC)、红细胞压积(Hct)、血清睾酮(T)、皮质醇(C)等血液生化指标,以及平原训练1周后、3周后的VO2max等生理指标。结果:世居高原的男子中长跑运动员在平原训练期间,血清EPO水平逐渐下降,且3周后下降显著(P<0.05);平原训练1周后和3周后,RBC和Hb含量均显著下降,Hb在3周后下降幅度更大;RC和Hct变化不明显;血清T、C、T/C值在下高原后有不同程度的降低,但无显著性差异;VO2max等相关心肺功能指标在平原训练3周后均低于1周后的值,但呼吸商(R)有所升高,均无统计学意义。结果表明,世居高原男子中长跑运动员平原训练期间身体机能有所下降,且3周后的下降程度比1周后更加明显。提示世居高原男子中长跑运动员到平原参加比赛时最佳时间应在1周内。  相似文献

目的 探讨局部进展期直肠癌X射线修复交叉互补基因1(XRCC1 rs25487)多态性与新辅助放化疗相关性。方法 使用前瞻性队列研究,纳入于2018年8月至2019年7月在贵州省肿瘤医院腹部肿瘤科就诊的55例局部进展期直肠癌患者,对患者进行新辅助同步放化疗,于同步放化疗前采血并进行DNA测序确定XRCC1 rs25487基因型。采用调整混杂因素的logistic回归分析研究肿瘤新辅助放化疗后T分期、N分期降期情况与患者XRCC1 rs25487基因多态性的关系并分层分析探究各基因型与中性粒细胞淋巴细胞比值(NLR)等临床特征的交互作用。结果 各基因型频率均符合Hardy-Weinberg平衡。调整混杂因素以后,相较于AA基因型患者,GG型患者新辅助放化疗后T降期率减低(OR=0.1,P<0.05)。而GA型患者新辅助放化疗后无论是T降期率还是N降期率,与AA基因型患者差异均无统计学意义(P>0.05)。并且AA/GA基因型与NLR存在交互作用而影响放疗后T降期率。结论 XRCC1 rs25487基因多态性与局部进展期直肠癌患者新辅助放化疗的疗效相关,且与NLR值有交互作用,可能成为新辅助放化疗疗效的预测因子。  相似文献
