To improve the efficiency of hepatic intra-arterial (h.i.a.) chemotherapy, we selected pirarubicin (THP) because it shows good properties for h.i.a. chemotherapy, such as fast and efficient cellular uptake, and used it for h.i.a. chemotherapy in rabbits with V x 2 tumour implanted in the liver. The anti-tumour effect of THP upon h.i.a. administration was compared with that upon intravenous (i.v.) injection and also with the anti-tumour activity of epirubicin (EPI) upon h.i.a. injection using optimal and maximal tolerated doses of each drug. When tumour growth rates and morphometric examinations were evaluated, it was found that THP and EPI were effective against V x 2 tumour when injected via the h.i.a. route. The activity of THP was stronger than that of EPI. As regards h.i.a. injection-related complication, plasma transaminase levels were temporarily elevated. To demonstrate higher anti-tumour activity and other advantages of h.i.a. injection of THP, plasma and tumour drug concentrations were determined by high-performance liquid chromatography after THP or EPI was administered at an equal dose to the rabbit V x 2 model. Hepatic intra-arterial injection of THP accomplished a selective and higher uptake into the tumour and lower effusion into the plasma than i.v. injection of THP or h.i.a. injection of EPL. Our findings indicate that THP is the better candidate of the two drugs tested for the h.i.a. chemotherapy because of its greater anti-tumour activity and the lower systemic drug exposure achieved upon h.i.a. injection.  相似文献

目的 探讨Axl 及其配体Gas6蛋白在乳腺癌组织中的表达及其与临床病理特征的关系。方法 收集55例女性乳腺癌患者的新鲜癌组织及配对癌旁组织,另收集新鲜良性乳腺增生组织标本20例作为对照。Western blotting及免疫组化PV-9000法分别检测上述标本中Axl及其配体Gas6的表达情况,分析其表达在良恶性组织间的差异及与腋窝淋巴结转移的关系。结果 Axl在乳腺癌组织中的相对表达量和阳性表达率均高于癌旁组织及良性乳腺增生组织(0.272±0.052、0.134±0.008和0.116±0.007,60.0%、36.4%和40.0%),差异有统计学意义(P<0.05);Gas6在乳腺癌组织中的相对表达量和阳性表达率均高于癌旁组织及良性乳腺增生组织(1.147±0.078、0.490±0.078和0.474±0.078,58.1%、21.8%和20.0%),差异有统计学意义(P<0.05);癌旁组织及良性乳腺增生组织间Axl及Gas6蛋白表达的差异无统计学意义(P>0.05)。淋巴结转移组中Axl和Gas6蛋白的相对表达量和阳性表达率均高于无淋巴结转移组(0.408±0.007 vs. 0.298±0.041,1.277±0.195 vs.0.880 ±0.037;80.0% vs.48.0%,76.7% vs.40.0%),差异有统计学意义(P<0.05)。乳腺癌组织中Axl 与Gas6蛋白表达呈正相关(r=0.612,P=0.041)。结论 乳腺癌组织中Axl 及Gas6蛋白表达与乳腺癌的发生、发展有关,两者表达促进了乳腺癌腋窝淋巴结转移。  相似文献

Objective The effects of BMI and physical activity on the risk of pancreatic cancer were investigated in a large population-based cohort study in Japan (JPHC study). In particular, we explored how these effects were influenced by smoking status and a history of diabetes. Methods In total, 99,670 participants (47,499 men, and 52,171women) were followed for an average of 11 years through the end of 2003. Hazard ratios (HR) and their 95% confidence intervals (CI) were estimated by Cox proportional hazards models. Results A total of 224 incident cases (128 men, 96 women) of pancreatic cancer were identified. A statistically significant excess risk of pancreatic cancer was associated with current smoking (HR = 1.8, CI 1.1–3.0) and a history of diabetes (HR = 2.1, CI 1.3–3.5) among men. A similar increase in risk associated with current smoking (HR = 2.0, CI 0.9–4.4) and diabetes (HR = 1.5, CI 0.7–3.5) was also seen among women. BMI was inversely associated with risk of pancreatic cancer among men, especially among current male smokers or diabetes patients, but no association was found among women. Leisure-time physical activity was not associated with risk in either men or women. Conclusions Our study confirms the association between cigarette smoking, history of diabetes and increased risk of pancreatic cancer. However, our data suggest that the association between BMI and risk of pancreatic cancer in this Japanese population may be different from that in Western populations. PURPOSE: 5-Iodo-2-pyrimidinone-2'-deoxyribose (IPdR) is an oral prodrug of 5-iodo-2'-deoxyuridine (IUdR), an in vitro/in vivo radiosensitizer. IPdR can be rapidly converted to IUdR by a hepatic aldehyde oxidase. Previously, we found that the enzymatic conversion of IPdR to IUdR could be transiently reduced using a once daily (q.d.) treatment schedule and this may affect IPdR-mediated tumor radiosensitization. The purpose of this study is to measure the effect of different drug dosing schedules on tumor radiosensitization and therapeutic index in human glioblastoma xenografts. EXPERIMENTAL DESIGN: Three different IPdR treatment schedules (thrice a day, t.i.d.; every other day, q.o.d.; every 3rd day, q.3.d.), compared with a q.d. schedule, were analyzed using athymic nude mice with human glioblastoma (U251) s.c. xenografts. Plasma pharmacokinetics, IUdR-DNA incorporation in tumor and normal proliferating tissues, tumor growth delay following irradiation, and body weight loss were used as end points. RESULTS: The t.i.d. schedule with the same total daily doses as the q.d. schedule (250, 500, or 1,000 mg/kg/d) improved the efficiency of IPdR conversion to IUdR. As a result, the percentage of IUdR-DNA incorporation was higher using the t.i.d. schedule in the tumor xenografts as well as in normal small intestine and bone marrow. Using a fixed dose (500 mg/kg) per administration, the q.o.d. and q.3.d. schedules also showed greater IPdR conversion than the q.d. schedule, related to a greater recovery of hepatic aldehyde oxidase activity prior to the next drug dosing. In the tumor regrowth assay, all IPdR treatment schedules showed significant increases of regrowth delays compared with the control without IPdR (q.o.d., 29.4 days; q.d., 29.7 days; t.i.d., 34.7 days; radiotherapy alone, 15.7 days). The t.i.d. schedule also showed a significantly enhanced tumor growth delay compared with the q.d. schedule. Additionally, the q.o.d. schedule resulted in a significant reduction in systemic toxicity. CONCLUSIONS: The t.i.d. and q.o.d. dosing schedules improved the efficiency of enzymatic activation of IPdR to IUdR during treatment and changed the extent of tumor radiosensitization and/or systemic toxicity compared with a q.d. dosing schedule. These dosing schedules will be considered for future clinical trials of IPdR-mediated human tumor radiosensitization.  相似文献

Caffeine was injected s.c. every12 h at the maximum tolerated dose (m.t.d.) of 120 mg/kg bodyweight, half the m.t.d., and one quarter the m.t.d. from 12to 72 h before and 0 to 72, 72 to 132, and 0 to 132 h after4-HAQO treatment. Post-treatment with caffeine from 0 to 132h had a dose-dependent biphasic effect on pancreatic tumorigenesis:post-treatment with the m.t.d. of caffeine decreased the totalnumber of nodules, whereas treatment with one quarter the m.t.d.of caffeine increased their number. Decrease in the number ofnodules was also observed on post-treatment with the m.t.d.of caffeine from 0 to 72 or from 72 to 132 h. Pre-treatmentwith the m.t.d. of caffeine had no significant effect on thenumber of nodules. Recovery of pancreatic DNA synthesis wasslower after simultaneous treatment with the m.t.d. of caffeineand 4-HAQO than after treatment with 4-HAQO alone. The possiblemechanism of the effect of caffeine on pancreatic tumorigenesisinduced by 4-HAQO in rats is discussed.  相似文献
