The aim of the current study is to examine the improving effect of Sasa borealis stem (SBS) extract extracts on high-fat diet (HFD)-induced hepatic steatosis in rats. To determine the hepatoprotective effect of SBS, we fed rats a normal regular diet (ND), HFD, and HFD supplemented with 150 mg/kg body weight (BW) SBS extracts for five weeks. We found that the body weight and liver weight of rats in the HFD + SBS group were significantly lower than those in the HFD group. Significantly lower serum total cholesterol (TC) and triglyceride (TG) concentrations were observed in the SBS-supplemented group compared with the HFD group. We also found that the HFD supplemented with SBS group showed dramatically reduced hepatic lipid accumulation compared to the HFD alone group, and administration of SBS resulted in dramatic suppression of TG, TC in the HFD-induced fatty liver. In liver gene expression within the SBS treated group, PPARα was significantly increased and SREBP-1c was significantly suppressed. SBS induced a significant decrease in the hepatic mRNA levels of PPARγ, FAS, ACC1, and DGAT2. In conclusion, SBS improved cholesterol metabolism, decreased lipogenesis, and increased lipid oxidation in HFD-induced hepatic steatosis in rats, implying a potential application in treatment of non-alcoholic fatty liver disease.  相似文献

The purpose of this work was to study the effect of early long-term high-fat feeding on energy balance and liver oxidative activity. To this end, rats aged about 30 d were fed a high-fat or a low-fat diet for 15, 30 or 60 d. Full energy balance and energy partitioning measurements were carried out. In addition, we measured hepatic mitochondrial and peroxisomal oxidative capacities. Serum levels of free triiodothyronine (T3) and leptin were also determined. Rats fed a high-fat diet showed an increase in metabolizable energy intake as well as in energy expenditure, while lipid gain over the whole period was lower than that expected due to a decrease in metabolic efficiency. An increase in serum free T3 levels was also found in rats fed a high-fat diet after 15 and 30 d. Statistically significant correlations between serum leptin levels and body fat mass were found after 15, 30 and 60 d of high-fat feeding. Finally, no variation in hepatic mitochondrial and peroxisomal fatty acid oxidation capacity was found in rats fed a high-fat diet for 15, 30 or 60 d. In conclusion, the results of the present study show that young Wistar rats fed a high-fat diet for up to 60 d are able to counteract, at least in part, obesity development.  相似文献

The changes in metabolic efficiency, body composition, and nutrient partitioning induced by high-fat feeding were evaluated in adult rats (90 d of age). The alterations in serum free triiodothyronine, insulin, and leptin levels, as well as in hepatic and skeletal muscle metabolism, were also assessed. Rats were fed either a low- or a high-fat diet for 2 weeks. Relative to the low-fat feeding, energy intake and expenditure, as well as body-energy gain, lipid gain, and energetic efficiency, were increased by the high-fat feeding. Increased serum leptin levels accompanied these variations. A positive correlation between serum leptin levels and percentage of body fat was found in the rats fed the low- or high-fat diet, with a significant divergence between the slope of the regression lines. Furthermore, a negative correlation between serum leptin level and energy intake was found in the rats fed the low-fat diet, while a positive correlation was found in the rats fed the high-fat diet. Finally, the high-fat feeding decreased the hepatic and skeletal muscle mitochondrial oxidative capacity. It is concluded that, in adult rats, a nutritional factor such as a high level of fat in the diet induces obesity, leptin resistance, and impairment of mitochondrial capacity, all phenomena typical of unrestrained aged rats.  相似文献

目的 通过建立高脂血症大鼠模型,探讨中等强度运动联合大蒜素对高脂血症大鼠血脂代谢、肝组织氧化损伤的影响。方法:大鼠50只,分为普通饲料组(A组)、高脂饲料组(B组)、高脂饲料+大蒜素组(C组)、高脂饲料+运动组(D组)、高脂饲料+运动大+蒜素组(E组)。A组普通饲料喂养,B组、C组、D组、E组高脂饲料喂养,C组、E组灌服剂量为30mg/kg·d大蒜素,D组、E组进行10周运动。测定各组大鼠血TC、TG、LDL-C、HDL-C、AST、ALT,肝组织MDA、SOD、GSH-Px。结果:与A组相比,B组、C组、D组、E组TC、TG、LDL-C、AST、ALT、MDA升高,HDL-C、GSH-Px降低(P<0.05),B组、C组、D组SOD降低(P<0.05);与B组相比,C组、D组、E组TC、TG、LDL-C、AST、ALT、MDA降低,GSH-Px、SOD、HDL-C升高(P<0.05;);与C组相比,D组TC、TG、LDL降低(P<0.05),E组TC、TG、LDL降低,HDL-C升高(P<0.05);与C组、D组相比,E组AST、ALT、MDA降低,GSH-Px、SOD升高(P<0.05)。结论:中等强度运动和大蒜素均可降低高脂血症大鼠血脂,抑制肝组织脂质过氧化反应,减少肝细胞损伤,且两者具有协同效应。大蒜素对高脂血症大鼠血脂的调节效果不如中等强度运动。  相似文献

目的探究生命早期使用抗生素对生命后期高脂饲料负荷下的小鼠血糖、血脂等糖脂代谢指标的影响。方法将48只2周龄雌性BALB/c小鼠随机分为空白对照组、抗生素组、高脂组和联合作用组,每组12只,抗生素组和联合作用组以100 mg/kg头孢曲松灌胃,其余2组灌胃等量生理盐水,持续至4周龄后停止灌胃,接着高脂组和联合作用组以高脂饲料、其余2组以普通饲料喂养12周。最后1周测定空腹血糖并进行口服糖耐量实验,实验结束后测定血脂、肝脏脂质、胰岛素和瘦素水平,并计算胰岛素抵抗指数。结果与普通饲料相比,高脂饲料引起小鼠糖耐量受损,内脏脂肪、血糖、胰岛素抵抗指数、血脂、肝脏脂质和瘦素增加(P<0.05);与空白对照组相比,抗生素组小鼠糖耐量受损,内脏脂肪、血糖和肝脏甘油三酯水平增加(P<0.05);与高脂组相比,联合作用组小鼠的糖耐量受损、血糖、胰岛素抵抗指数和肝脏总胆固醇水平增加(P<0.05)。结论生命早期使用头孢曲松增加了机体内脏脂肪、血糖、胰岛素抵抗和肝脏脂质,且加剧了高脂饲料诱导的糖脂代谢紊乱,表明生命早期使用抗生素可能会提高机体对高脂饲料诱导糖脂代谢异常的易感性。  相似文献

目的比较两种不同高脂饲喂法致高脂血症大鼠模型的异同点,并对其使用特点进行初步评价。方法按体重、血清胆固醇水平将24只4周龄健康雄性SD大鼠随机分为正常对照组(A组)、普通高脂模型组(B组)和丙硫氧嘧啶模型组(C组),分别饲以基础饲料、未添加丙硫氧嘧啶的高脂饲料和添加0.2%丙硫氧嘧啶的高脂饲料。6周后处死大鼠,测定血脂指标、计算肝指数,并对肝脏进行组织病理学检查,采用单因素方差分析和LSD法对各指标进行分析比较。结果与A组相比,两模型组大鼠体重增加缓慢(P〈0.05),C组较B组增重更加缓慢(P〈0.01)。两模型组大鼠在实验期间表征有较大差异;两模型组TC、LDL-C水平均显著升高(P〈0.05),C组升高更为显著(P〈0.01)。两模型组TG、HDL-C水平变化差异无统计学意义(P〉0.05);病理组织学检查表明,两种高脂模型组大鼠肝脏均出现脂肪样变,肝细胞体积增大,细胞核被细胞内大脂滴挤向一侧。结论两种方法均能成功建立SD大鼠高脂血症模型,但普通高脂模型可用于食品、保健品等以预防为目的的降脂功效评估,而添加了丙硫氧嘧啶的高脂模型更适用于降脂药的临床前评价。  相似文献

目的:观察化痰方(二陈汤)对高脂饮食大鼠血脂及胰岛素抵抗的影响。方法:30只健康成年雄性大鼠按照随机数字表法分为正常组、高脂组和化痰组各10只,正常组给予正常饲料喂养,高脂组和化痰组给予高脂饲料喂养14周,化痰组第11周起给予二陈汤灌胃4周。在第14周末进行葡萄糖耐量实验(OGTT)和胰岛素耐量实验(ITT),14周后处死大鼠,检测并比较各组血脂TC、TG的含量及第14周各组大鼠OGTT结果和ITT结果。结果:与正常组比较,高脂组大鼠血清中的TC和TG含量均明显升高(P〈0.01);与高脂组比较,化痰组大鼠血清中的TG含量明显降低(P〈0.01),TC有所降低,差异也有统计学意义(P〈0.05)。高脂组的空腹血糖明显高于正常组(P〈0.05),而化痰组空腹血糖与正常组的比较差异无统计学意义(P〉0.05);糖负荷后,高脂组的各个时间点的血糖均明显高于正常组(P〈0.05或P〈0.01),而化痰组除了糖负荷后120 min的血糖明显高于正常组外(P〈0.01),其余时间点与正常组的比较差异均无统计学意义(P〉0.05)。腹腔注射胰岛素后,化痰组各时间点血糖下降幅度均明显高于高脂组,30 min时高脂组血糖降低18%,而化痰组大鼠降低26%。结论:二陈汤能够降低高脂饮食大鼠血脂的含量,改善胰岛素抵抗,值得在临床上推广使用。  相似文献

BACKGROUND/OBJECTIVESTurmeric (Curcuma longa L.) has been reported to have many biological functions including anti-obesity. Leptin, peptide hormone produced by adipocytes and its concentration is increased in proportion to the amount of the adipocytes. In the present study, we examined the effects of Korean turmeric on the regulation of adiposity and leptin levels in 3T3-L1 adipocytes and rats fed a high-fat and high-cholesterol diet.MATERIALS/METHODSLeptin secretion, free fatty acid and glycerol contents in 3T3-L1 adipocytes were measured after incubation of cells with turmeric for 24 hours. Rats were divided into four experimental groups: a normal diet group (N), a high-fat and high-cholesterol diet group (HF), a high-fat and high-cholesterol diet group supplemented with 2.5% turmeric extracts (TPA group) and a high-fat and high-cholesterol diet group supplemented with 5% turmeric extracts (TPB group). Serum samples were used for the measurement of leptin concentration.RESULTSContents of free fatty acid and glycerol showed concentration dependent increase in response to turmeric extracts. Effects of turmeric extracts on reduction of lipid accumulation in 3T3-L1 cells were examined by Oil Red O staining. Treatment with turmeric extracts resulted in increased expression levels of adipose triglyceride lipase and hormone-sensitive lipase mRNA. The concentration of leptin from 3T3-L1 adipocytes was significantly decreased by turmeric. Proportional abdominal and epididymal fats weights of the turmeric 5% supplemented group, TPB has significantly decreased compared to the HF group. The serum levels of leptin in the TPA and TPB groups were significantly lower than those of the HF group.CONCLUSIONSBased on these results, we suggested that Korean turmeric may contribute to the decreasing of body fat and regulating leptin secretion.  相似文献

目的研究姜蒜片对高脂饲料饲养大鼠血脂水平的影响。方法将大鼠按基础血清总胆固醇水平随机分为4组:高脂对照组、低、中、高剂量实验组。实验期间,所有大鼠均饲以高脂饲料,低、中、高剂量组分别灌胃给予0.11、0.22、0.66g/kg.bw剂量的姜蒜片,对照组以蒸馏水灌胃,灌胃量1ml/100g.bw,连续30d后,测定血清总胆固醇(TC)、甘油三酯(TG)和高密度脂蛋白胆固醇(HDL-C)。结果0.11g/kg.bw剂量可显著降低大鼠血清TC、TG水平(P<0.05);各剂量对大鼠血清HDL-C水平无显著影响(P>0.05)。结论姜蒜片具有一定的调节血脂作用。  相似文献

目的观察高脂饮食诱导的肥胖瘦素抵抗大鼠细胞因子信号转导抑制因子-3(SOCS-3)、过氧化物酶体增殖物激活受体γ(PPARγ)及乙酰辅酶A氧化酶(ACO)mRNA表达水平的变化。方法30只Wistar雄性大鼠,6只为对照组喂饲基础饲料,24只为高脂组喂饲高脂饲料,第8周末按体重增量从高脂组中筛选出8只大鼠作为肥胖组,测定对照组和肥胖组大鼠血清瘦素,附睾脂肪组织中SOCS-3,PPARγ以及ACO mRNA表达。结果肥胖组大鼠体重以及血清瘦素水平均显著高于对照组(P<0.05);肥胖组脂肪组织SOCS-3、PPARγ mRNA水平显著高于对照组(P<0.05),而ACO mRNA则显著低于对照组(P<0.05)。结论高脂饮食诱导的肥胖大鼠存在瘦素信号转导通路的抑制,脂肪酸氧化能力降低,脂肪合成能力增强。  相似文献

Dietary protamine can ameliorate hyperlipidemia; however, the protamine-derived active peptide and its hypolipidemic mechanism of action are unclear. Here, we report the discovery of a novel anti-obesity and hypocholesterolemic peptide, RPR (Arg-Pro-Arg), derived from protamine in mice fed a high-fat diet for 50 days. Serum cholesterol levels were significantly lower in the protamine and RPR groups than in the control group. White adipose tissue weight was significantly decreased in the protamine and RPR groups. The fecal excretion of cholesterol and bile acid was significantly higher in the protamine and RPR groups than in the control group. We also observed a significant decrease in the expression of hepatic SCD1, SREBP1, and adipocyte FAS mRNA, and significantly increased expression of hepatic PPARα and adipocyte PPARγ1 mRNA in the protamine group. These findings demonstrate that the anti-obesity effects of protamine are linked to the upregulation of adipocyte PPARγ1 and hepatic PPARα and the downregulation of hepatic SCD1 via SREBP1 and adipocyte FAS. RPR derived from protamine has a crucial role in the anti-obesity action of protamine by evaluating the effective dose of adipose tissue weight loss.  相似文献

Short-term, high-fat diets lower circulating leptin concentrations in rats   总被引:4,自引:0,他引:4  
BACKGROUND: Leptin is produced in proportion to body fat mass and can act on the brain to induce satiety and regulate adipose tissue mass; factors other than adipose tissue mass may influence circulating leptin concentrations. OBJECTIVE: We explored the possibility that short-term, moderately high-fat diets induce weight gain by producing inappropriately low circulating leptin concentrations. DESIGN: Female Hooded Wistar rats were fed either a moderately high-fat diet or control diet. Body weight, energy intake, body composition, and fasting plasma leptin were compared after 4 and 14 wk of dietary treatment. RESULTS: After 4 wk, abdominal fat mass was 38% greater in rats fed the high-fat diet than in those fed the control diet (P < 0.01). However, plasma leptin concentrations were 24% lower in animals fed the high-fat diet (P < 0.05), resulting in significantly lower plasma leptin concentrations per unit abdominal fat mass than in control animals (P < 0.005). From 4 to 14 wk, animals fed the high-fat diet gained twice as much weight and consumed 32 kJ/d more than controls (both P < 0.05). At 14 wk, plasma leptin concentrations per unit abdominal fat mass were 27% lower in rats fed the high-fat diet (P = 0.058) and there was a significant negative association between leptin concentrations per unit abdominal fat mass and body weight (r = 0.44, P < 0.05). CONCLUSIONS: In the short term, a moderately high-fat diet is associated with lower than expected circulating leptin concentrations, which correlate with a higher body weight. A high-fat diet may therefore contribute to weight gain by reducing leptin secretion in adipose tissue.  相似文献

目的探讨经过重复性"饥饿/再投喂"饲喂方式处理的大鼠再摄取高脂食物时,与体重控制和能量平衡调节有关的瘦素(leptin)和一系列神经内分泌因子及激素的表达变化。方法设计"重复性饥饿1天/再投喂1天,持续6周"的饲喂方式(基础饲料)处理实验组(RFR),实验-高脂饲料组大鼠(RFR-LF/HF)另6周改为每天喂以高脂饲料。对照组随机分为3组,对照组喂基础饲料6周后处死,高脂饲料组改用高脂饲料,基础饲料组继续用基础饲料喂养至12周。比较各组大鼠体重、Lee’s指数和脂体比的差异,检测血脂、血清生长激素、甲状腺素、瘦素、胰岛素和血浆促肾上腺皮质激素含量,应用RT-PCR法检测下丘脑神经肽Y(NPY)、前阿黑皮素原(POMC)mRNA的表达水平。结果 RFR-LF/HF组Lee’s指数、脂体比、血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL)、瘦素和胰岛素水平均显著低于高脂饲料组而高于基础饲料组,下丘脑NPY mRNA表达水平显著高于高脂饲料组和基础饲料组,POMC mRNA表达水平显著低于高脂饲料组和基础饲料组。结论重复性"饥饿/再投喂"饲喂处理可降低高脂饮食诱导的肥胖程度,减轻瘦素抵抗和胰岛素抵抗,但会引起中枢神经内分泌肽表达的极度紊乱。  相似文献

Perinatal nutrition has a well-known influence on obesity susceptibility. We previously demonstrated the protective anti-obesity effects of perinatal leptin administration. Celastrol is a natural compound acting as a leptin sensitizer with anti-obesity effects when administered in adult animals. Here, we aimed to determine if perinatal treatment with leptin, celastrol, or their combination was able to improve metabolic health in animals fed an isocaloric high-fat (HF) diet. Leptin and/or celastrol or their vehicle were administered orally to rats during the suckling period. After weaning, animals were chronically pair-fed with an HF diet provided isocaloric to the intake of a normal-fat diet by control animals to avoid obesity. Isocaloric HF feeding in vehicle-treated animals resulted in metabolic features characteristic of the metabolically obese, normal-weight (MONW) phenotype, i.e., obesity-related disturbances without increased body weight. Leptin treatment prevented liver fat deposition and insulin resistance, induced greater insulin and leptin signaling capacity, decreased gene expression of orexigenic signals at the hypothalamic level, and induced browning in retroperitoneal adipose tissue. However, celastrol treatment did not provide any protective effect and resulted in greater size of the retroperitoneal adipose depot, higher circulating glucose and insulin levels, and decreased leptin sensitivity capacity in adipose tissue. The co-administration of leptin ameliorated the negative effects of celastrol on the retroperitoneal depot, inducing browning and decreasing its size. In conclusion, the perinatal administration of leptin, but not celastrol, provided protection against the consequences of dietary unbalances leading to an MONW phenotype in adulthood.  相似文献

目的 : 证实铬对肥胖基因表达和体重的影响及与血糖、血脂的关系。方法 : 将80只大鼠按体重随机分为 8组 ,分别为基础饲料对照组、基础饲料加铬组 (不同剂量的葡萄糖酸铬 )、高脂饲料对照组、高脂饲料加铬组 (不同剂量的葡萄糖酸铬 ) ,每周称重 1次 ,并于第 8周末断头取血 ,用放射免疫的方法检测血中的瘦素 ,用酶法检测血糖、血脂等指标 ,同时称取肝、脾、肾及睾丸等脏器 ,计算脏 /体比值。结果 : 加铬组与对照组相比 ,瘦素水平显著降低 (P<0 .0 5 )。血糖、总胆固醇 (TC)、甘油三酯 (TG)显著降低 (P<0 .0 5 ) ,高密度脂蛋白胆固醇 (HDL - C)显著升高 (P<0 .0 5 )。结论 : 铬对大鼠体重的影响不大 ,但可抑制肥胖基因的表达 ,同时具有降低血糖、TC、TG和升高 HDL- C的作用  相似文献

Agaricus bisporus (white button mushroom; WBM) contains high levels of dietary fibers and antioxidants including vitamin C, D, and B12; folates; and polyphenols that may provide beneficial effects on cardiovascular and diabetic diseases. The objective of this study was to examine the hypothesis that intake of the fruiting bodies of WBM regulates anticholesterolemic and antiglycemic responses in rats fed a hypercholesterolemic diet (0.5% cholesterol; 14% fat) and rats with type 2 diabetes induced by injection of streptozotocin (STZ) (50 mg/kg body weight), respectively. The STZ-induced diabetic male Sprague-Dawley rats fed the Agaricus bisporus powder (ABP; 200 mg/kg of body weight) for 3 weeks had significantly reduced plasma glucose and triglyceride (TG) concentrations (24.7% and 39.1%, respectively), liver enzyme activities, alanine aminotransferase and aspartate aminotransferase (11.7% and 15.7%, respectively), and liver weight gain (P < .05). In hypercholesterolemic rats, oral feeding of ABP for 4 weeks resulted in a significant decrease in plasma total cholesterol (TC) and low-density lipoprotein (LDL) (22.8% and 33.1%, respectively) (P < .05). A similar significant decrease in hepatic cholesterol and TG concentrations was observed (36.2% and 20.8%, respectively) (P < .05). Decrease in TC, LDL, and TG concentrations was accompanied by a significant increase in plasma high-density lipoprotein concentrations. It was concluded that A bisporus mushroom had both hypoglycemic and hypolipidemic activity in rats.  相似文献

Nutritional hepatic steatosis and mortality after burn injury in rats   总被引:2,自引:0,他引:2  
AIMS: To investigate the effects of diet composition and starvation on hepatic steatosis and mortality after severe burn injury in rats. Methods: Experiment 1: rats received either normal chow (55 cent of energy carbohydrates, 14 cent fat, 31 cent protein), a high-fat (40 cent carbohydrates, 40 cent fat, 20 cent protein), or a high-carbohydrate diet (81 cent carbohydrates, 4 cent fat, 15 cent protein) ad libitum for 6 days. Another three groups received these diets ad libitum for 6 days after 48|h starvation. Experiment 2: mortality after 60 cent total body surface area scald burn was determined in a control group of rats and a group with nutritionally induced hepatic steatosis. Hepatic steatosis was induced by feeding the rats a high-fat diet (40 cent carbohydrates, 4 cent fat, 15 cent protein) ad libitum for 6 days. RESULTS: Without starvation, liver triglyceride content (mg/g liver) increased in response to the high-fat diet (25.6'6.9) compared to normal chow (9.4'3.8; P < 0. 05); the high-carbohydrate diet had no influence on liver triglyceride content (12.4'3.7). Refeeding after starvation resulted in elevated (P < 0.05) liver triglyceride content in the high-fat (18.8'8.3) and the high-carbohydrate group (28.7'14.4 vs control 6. 7'3.7). Liver triglyceride content correlated (R2=0.72; P < 0.05) to non-protein energy intake but not to total energy intake. Burn caused 33 cent mortality in the hepatic steatosis group and no deaths in the control group (P < 0.05). CONCLUSIONS: Diet composition and preceding starvation independently manipulate hepatic fat content in rats. Hepatic steatosis increases mortality after burn injury. Thus, nutritional interventions to reduce hepatic fat accumulation may be beneficial.  相似文献

This study evaluated the anti-obesity effects of Phellinus baumii extract (PBE) in high-fat diet (HFD)-fed mice. 研究几丁聚糖(chitosan) 对饲以高脂饲料大鼠体重的影响及其与体内肥胖基因表达产物—瘦素和血脂水平的关系。将50 只大鼠随机分为5 组,分别喂饲基础饲料、高脂饲料及高脂饲料加低、中、高不同剂量的chitosan 共7 周,每周称一次体重,于第7 周末断头取血,采用放射免疫试剂盒法检测血浆中瘦素,酶法检测血清中甘油三酯,胆固醇和高密度脂蛋白胆固醇。结果显示chitosan 各剂量组大鼠体重与高脂对照组相比均显著降低(P< 0 .05 或P< 0 .01) ,前者的瘦素和血脂水平亦显著低于后者(P< 0 .05 或P< 0 .01) 。提示chitosan 不仅对高脂饲料诱导大鼠肥胖有明显的抑制作用,而且在降低体重的同时,有良好的调节血脂功能。通过体重与瘦素变化规律的分析,推测chitosan 对瘦素的分泌有促进作用,chitosan 抑制高脂饲料诱导大鼠肥胖的机理部分可能即在于此。  相似文献
