目的 探讨吡柔比星对膀胱癌细胞增殖的影响及其作用机制。 方法 膀胱癌细胞T24和BIU-87分别用0.4、0.8、1.6、3.2 mg/L的吡柔比星处理24、48、72 h ,用四甲基偶氮唑盐(MTT)检测细胞增殖,流式细胞术检测细胞凋亡率,qRT-PCR、RT-PCR检测磷脂酶Cε(PLCε)mRNA及凋亡调控基因Bcl-2 mRNA的表达,蛋白质印迹法检测PLCε蛋白的表达。将膀胱癌细胞分为空白组、吡柔比星处理组、Ad-shPLCε处理组、吡柔比星联合PLCε干扰腺病毒载体(Ad-shPLCε)处理组,检测并比较各组细胞增殖和Bcl-2蛋白的表达量。 结果 在膀胱癌细胞T24、BIU-87中,吡柔比星对细胞的抑制率呈浓度、时间依赖性,即随着药物浓度增加、处理时间延长,细胞抑制率升高;吡柔比星促进T24和BIU-87细胞凋亡,并且抑制PLCε和Bcl-2表达。吡柔比星处理组、Ad-shPLCε处理组、Ad-shPLCε联合吡柔比星处理组均能抑制细胞增殖和Bcl-2表达,且Ad-shPLCε联合吡柔比星处理组的抑制作用高于吡柔比星处理组,差异有统计学意义 (P<0.05)。 结论 吡柔比星能有效抑制膀胱癌细胞的增殖,可能与其抑制PLCε癌基因及下游Bcl-2基因的表达有关。  相似文献

目的探讨百里醌对体外膀胱癌细胞株BIU-87生长的影响及其机制。方法百里醌单独作用人膀胱癌细胞株BIU-87及预处理NF-κB激活剂TPA后百里醌再作用BIU-87细胞,四甲基偶氮唑蓝(MTT)法检测细胞增殖;流式细胞术检测细胞凋亡;Western blotting检测膀胱癌细胞质中IκBα蛋白表达。结果与对照组相比较,百里醌可显著抑制体外膀胱癌细胞株BIU-87生长,并明显诱导细胞凋亡;预处理TPA可显著削弱百里醌对BIU-87细胞的增殖抑制和凋亡诱导作用;百里醌干预后膀胱癌细胞质中IκBα的表达明显上调。结论百里醌可显著抑制体外膀胱癌细胞生长,并诱导细胞凋亡,该作用可能通过促进其胞质中IκBα的表达而实现。  相似文献

目的 了解丹参酮ⅡA对人小细胞肺癌细胞(H446细胞)的凋亡诱导作用,并探讨其可能的分子机制。方法 采用MTT法检测不同浓度丹参酮ⅡA对人小细胞肺癌H446细胞增殖的影响结果;Hoechst 33258法检测不同浓度丹参酮ⅡA对人小细胞肺癌H446细胞凋亡的影响;qPCR法检测不同浓度丹参酮ⅡA对人小细胞肺癌H446细胞凋亡基因的影响;Westen blot法检测不同浓度丹参酮ⅡA对人小细胞肺癌H446细胞凋亡的影响。结果 不同药物浓度的丹参酮ⅡA (0.3、0.6、1.25、2.5、5和10μg/ml)抑制人小细胞肺癌H446细胞增殖且呈浓度和时间依赖性;随着药物浓度的提高,人小细胞肺癌H446细胞凋亡小体的形成数量增加,高浓度组差异有统计学意义(P<0.05);高浓度组丹参酮ⅡA促进凋亡基因(Bax,caspase-9、caspase-3)的表达,抑制凋亡抑制基因(Akt,Bcl-2)的表达且差异有统计学意义(P<0.05);高浓度组丹参酮ⅡA促进凋亡蛋白(Bax,caspase-9,caspase-3)的表达,抑制凋亡抑制蛋白(Akt,Bcl-2)的表达且差异有统计学意义(P<0.05)。结论 丹参酮ⅡA可能是通过Akt-Bax/bcl-2-caspase9-caspase3信号通路诱导人小细胞肺癌H446细胞的凋亡。  相似文献

目的 研究双氢青蒿素对前列腺癌 PC-3 细胞的凋亡诱导作用,并探讨其可能的机制。方法 MTT 法测定不同浓度双氢青蒿素对 PC-3 细胞增殖的影响,流式细胞仪 (FCM)、透射电镜 (TEM) 观察双氢青蒿素对 PC-3 细胞的凋亡诱导作用,免疫组化 (SP) 检测双氢青蒿素对 PC-3 细胞内 Bax、Bcl-2 蛋白阳性表达的影响。结果 双氢青蒿素对 PC-3 细胞有明显增殖抑制效应,能诱导 PC-3 细胞凋亡且具有浓度-时间依赖性,导致 PC-3 细胞线粒体肿胀、核碎裂、凋亡小体形成;随着双氢青蒿素浓度的增加,Bcl-2 蛋白阳性表达降低,而 Bax 蛋白阳性表达增加。结论 双氢青蒿素可能通过上调 Bax,下调 Bcl-2 蛋白表达诱导前列腺癌 PC-3 细胞凋亡。  相似文献

目的 研究大蒜素抑制人子宫内膜癌Ishikawa细胞生长的作用,并探讨其可能的作用机制。方法 以不同浓度大蒜素(12.5、25、50μg/ml)和顺铂(40μg/ml)分别干预对数生长期Ishikawa细胞,采用MTT法检测Ishikawa细胞增殖抑制;通过流式细胞术检测细胞周期和细胞凋亡状况,采用Western blot法检测Ishikawa细胞中caspase-3蛋白表达,RT-PCR法检测Ishikawa细胞中Bax mRNA、bcl-2 mRNA表达并计算Bax/bcl-2表达比值。结果 大蒜素能够显著提高子宫内膜癌Ishikawa细胞增殖抑制率、阻滞Ishikawa细胞周期于G2/M期,提高细胞凋亡率、上调caspase-3蛋白和Bax mRNA表达、下调bcl-2 mRNA表达、提高Bax/bcl-2表达比值,且上述作用均具有一定的剂量依赖性。结论 大蒜素对子宫内膜癌Ishikawa细胞增殖具有抑制作用以及促其凋亡的作用,从而表现出其抑制Ishikawa细胞生长的作用;作用机制可能与大蒜素能够改变Ishikawa细胞周期以及调节凋亡相关基因、蛋白表达有关。  相似文献

目的 研究虫草素对人骨肉瘤MG-63细胞凋亡的影响及其机制。方法 采用CCK-8实验检测不同浓度虫草素作用于MG-63骨肉瘤细胞24h和48h后对癌细胞增殖的抑制作用;采用流式细胞术分析虫草素对MG-63细胞周期分布及凋亡的影响;采用Western blot法检测虫草素对MG-63细胞凋亡相关蛋白Bax、Bcl-2、cleaved caspase-9、cleaved caspase-3及NICD1、Hes1蛋白表达的影响。结果 虫草素对人骨肉瘤MG-63细胞有明显的增殖抑制作用,导致MG-63细胞周期阻滞于G0/G1期并诱导细胞凋亡;虫草素可上调Bax、cleaved caspase-9和cleaved caspase-3蛋白的表达并下调Bcl-2,NICD1和Hes1蛋白的表达。结论 虫草素通过抑制骨肉瘤细胞增殖,诱导细胞周期阻滞于G0/G1期并通过线粒体凋亡途径诱导MG-63细胞凋亡从而发挥抗骨肉瘤作用,可能骨肉瘤细胞中Notch信号通路活性下调有关。  相似文献

[目的]研究萝卜硫素对人结肠癌HT-9细胞增殖和凋亡的影响及其可能的作用机制。[方法]采用CCK8试剂检测萝卜硫素对HT-9细胞增殖的影响;碘化丙啶(PI)染色检测萝卜硫素对HT-9细胞凋亡的影响;免疫印迹法检测PI3K/Akt信号通路蛋白及凋亡相关蛋白的表达。[结果]萝卜硫素可以明显抑制HT-9细胞的增殖,并呈时间和剂量依赖性;PI染色结果提示萝卜硫素可明显诱导HT-9细胞的凋亡,10、20、40 μg/mL萝卜硫素作用24 h后,HT-9细胞的凋亡率分别为(14.67±1.95)%、(27.95±2.53)%及(35.6±3.75)%,并且让细胞停留在G0/G1期;Western Blot结果提示萝卜硫素呈剂量依赖性的抑制PI3K、p-Akt及Bcl-2蛋白的表达,并诱导Bax蛋白的表达。[结论]萝卜硫素可明显抑制HT-9细胞的增殖并诱导其凋亡,其作用机制可能与抑制PI3K/Akt信号通路的活化有关。  相似文献

目的:探讨蛇床子素对人黑色素瘤A375细胞增殖和凋亡的影响。方法:人黑色素瘤细胞株A375细胞体外培养,将对数生长期的A375细胞以不同浓度的蛇床子素处理,MTT法检测A375细胞增殖;流式细胞术检测A375细胞凋亡;RTPCR法检测A375细胞Bcl-2 mRNA和Bax mRNA表达水平;Western blotting检测A375细胞IκB-α、p-NF-κB、NF-κB、Bcl-2和Bax蛋白表达。结果:蛇床子素可降低A375细胞增殖率以及Bcl-2 mRNA、p-NF-κB和Bcl-2蛋白表达水平,呈浓度依赖性;同时增加A375细胞凋亡率以及Bax mRNA、IκB-α、Bax蛋白表达水平,呈浓度依赖性,但对NF-κB表达无明显影响。结论:蛇床子素能诱导A375细胞增殖抑制和凋亡,呈浓度依赖性,其机制可能与抑制NF-κB信号通路激活有关。  相似文献

目的 探讨百里醌联合吉西他滨对胰腺癌BxPC-3细胞增殖和凋亡的影响,并探讨其作用机制。方法 采用CCK-8法检测细胞相对活性,Hoechst染色法及流式细胞术检测细胞凋亡情况,Western blot法检测凋亡相关蛋白(Bcl-2、Bax、XI-AP)、半胱天冬酶(cleaved-caspase-3、cleaved-caspase-9)、PTEN、Akt及phospho-Akt蛋白的表达.结果 百里醌呈浓度依赖性地抑制胰腺癌BxPC-3细胞增殖,诱导细胞凋亡.吉西他滨组(GEM)、百里醌组(TQ)、百里醌联合吉西他滨组(TQ+GEM)、百里醌与吉西他滨序贯给药组(TQ-GEM)相对细胞活性分别为83.7%±4.1%、51.8%±6.0%、48.25%±6.50%、33.3%±3.9%;早期凋亡率分别为12.2%±3.8%、30.4%±4.3%、43.5%±5.7%、58.3%±6.1%;各组相对细胞活性均明显低于对照组(P<0.05),而细胞凋亡率显著增高(P<0.05);与GEM组比较,TQ-GEM组与TQ+ GEM组相对细胞活性明显下调(P <0.001),凋亡率明显上调(P <0.001);TQ-GEM组较TQ+ GEM组出现更明显的增殖受抑(P <0.001),更高的细胞凋亡率(P<0.001).百里醌-吉西他滨序贯给药作用于胰腺癌BxPC-3细胞后,BxPC-3细胞中Bax蛋白表达明显下调,而Bcl-2、XIAP、cleaved caspase-3、cleaved caspase-9蛋白表达明显上调.百里醌可显著上调BxPC-3细胞PTEN的表达,明显抑制Akt磷酸化.结论 百里醌可明显增强吉西他滨对体外胰腺癌细胞生长抑制作用,可能是通过上调PTEN,Akt去磷酸化,促使Bcl-2、XIAP表达上调及Bax表达下调,活化caspase-3、caspase-9诱导细胞凋亡而实现。  相似文献

目的:探讨1-甲基-4-苯基-吡啶离子(1-methyl-4-phenyl-pyridinium,MPP+ )诱导的RSC96细胞凋亡与自噬功能障碍与TLR2/NF-κB信号通路的关系。方法:将RSC96细胞分为PBS组、MPP+ 组和MPP+ +CU-CPT22组;CCK-8检测不同MPP+ 浓度 (0.1、0.3、0.5、0.7、0.9 mmol/L)处理后细胞存活率;TUNEL检测细胞凋亡水平;RT-qPCR检测Toll样受体2(TLR2)mRNA转录水平;Western blot检测凋亡相关指标Bcl-2/Bax、cleaved caspase-3/caspase-3,自噬相关指标LC3Ⅱ/LC3Ⅰ和P62,以及TLR2、p-NF- κB/NF-κB蛋白表达水平。结果:与PBS组相比,MPP+ 组细胞存活率下降且呈浓度依赖性,TUNEL染色阳性细胞数增多,Bcl-2/ Bax蛋白比值水平下降,cleaved caspase-3/caspase-3比值增高。同时LC3Ⅱ/LC3Ⅰ比值下降,P62表达增加,p-NF-κB/NF-κB比值表达增加。RT-qPCR和Western blot结果显示,MPP+ 上调TLR2的表达。此外,与MPP+ 组相比,MPP+ +CU-CPT22组TUNEL阳性细胞数目减少,Bcl-2/Bax比值升高,cleaved caspase-3/caspase-3比值降低;同时,LC3Ⅱ/LC3Ⅰ比值上升,P62水平下降。结论: MPP+ 刺激诱导RSC96细胞凋亡和自噬水平失衡,发生机制可能与TLR2/NF-κB通路的激活有关。  相似文献

Objective: To evaluatel the value of D-dimers in patients with acute aortic dissection (AAD). Methods: This study consisted of 16 patients with AAD and 27 non-AAD patients. Serum D-dimets were measured by Sta-Liatest D-DI immunoturbidimetric assay. Results: D-dimer level was higher (P < 0.001) in patients with AAD(7.91 ± 5.52 μg/ml) than that in non- AAD group(1.57±1.24 μg/ml). D-dimer was positive (>0.4 μg/ml) in all patients with AAD and in 10 control group patients (37%). Among patients with acute AAD, D-dimers tended to be higher in Stanford A than in Stanford B (8.67 ± 4.31 μg/ml vs. 3.24±1.27 μg/ml, P <0.01). D-dimer values tended to be higher in more extended disease(3.84 ± 1.65 μg/ml, 8.57 ± 3.58 μg/ml and 11.87 ± 5.69 μg/ml in thoracic aorta, thoracic and abdominal aorta, thoracic and abdominal aorta and iliacal arteries, respectively, P < 0.05 for both 8.57 ± 3.58 and 11.87 ± 5.69 vs. 3.84 ± 1.65 ). Including the control group into the analysis, we found a sensitivity of 100%, a negative predictive value of 100%, and a specificity of 66% and a positive predictive value of 64% for D-dimer in diagnosis of AAD in our patients with suspected AAD. Conclusion: D-dimer was elevated in patients with AAD. A negative D-dimer test result could be useful in excluding AAD.  相似文献

Objective: To set up a simple and reliable rat model of combined liver-kidney transplantation. Methods: SD rats served as both donors and recipients. 4℃ sodium lactate Ringer's was infused from portal veins to donated livers,and from abdominal aorta to donated kidneys, respectively. Anastomosis of the portal vein and the inferior vena cava (IVC) inferior to the right kidney between the graft and the recipient was performed by a double cuff method, then the superior hepatic vena cava with suture. A patch of donated renal artery was anastomosed to the recipient abdominal aorta. The urethra and bile duct were reconstructed with a simple inside bracket. Results: Among 65 cases of combined liver-kidney transplantation, the success rate in the late 40 cases was 77.5%. The function of the grafted liver and kidney remained normal. Conclusion: This rat model of combined liver-kidney transplantation can be established in common laboratory conditions with high success rate and meet the needs of renal transplantation experiment.  相似文献

Shock wave lithotripsy (SWL) is a treatment of choice for upper urinary stones. However, this procedure is inappropriate for obese patients because the focus is often unable to reach the target owing to the limited focal distance in shock wave source. Although treating such patients in a blast path may increase the application length of shock wave source, it's difficult to find this path on the lithotripter monitor. Excess production of reactive oxygen species(ROS)of mitochondrion mediated by hyperglycemia is the common pathogenesis of angiopathic complications of diabetes.TCM holds that the damp from the dysfunction of spleen.kidney and liver is the causative factor of complications of diabetes.This is similar to the mechanism of Ros resulting in angiopathic complications of diabetes.When the angiopathic complications of type II diabetes mellitus(T2DM)are difierentiated as caused by turbid damp in TCM can be explained as ROS.Since the obstruction of pathogenic damp in channels and collaterals is said to be the main pathogenesis,the treating principle should be dissolving the damp to remove the obstruction.  相似文献

Objective To observe blood pressure change with age in salt-sensitive teenagers whose salt sensitivity were determined by repeated testing.Methods Salt sensitivity was determined through intravenous infusion of normal saline combined with volume-depletion by oral diuretic furosemide in 55 teenagers. After five years, salt sensitivity was re-examined and subject blood pressure was followed up. Blood pressure changes in salt-sensitive teenagers were compared to that of non-salt sensitive teenagers over five years.Results After 5 years, the repetition rate of salt sensitivity determined by intravenous saline loading is 92.7%. In teenagers with salt sensitivity on the baseline, both the systolic blood pressure increments and increment rates were much higher than non-salt sensitive teenagers (12.7±12.1 mmHg vs. 2.8±5.2 mmHg, P< 0.01; 12.2%± 12.0% vs. 2.5% ±4.4%, P< 0.001,respectively). There was a similar trend for diastolic blood pressure (8.4 ± 6.4 mmHg vs. 3.7 ± 6.4 mmHg, P = 0.052; 13.2% ±10.6 % vs. 6.8%± 10.1%, P = 0.053, respectively).Conclusions Salt sensitivity determined by intravenous saline loading showed good reproducibility. Blood pressure increments with age were much higher in salt-sensitive teenagers than non-salt sensitive teenagers, especially in terms of systolic blood pressure.  相似文献

Objective: To observe the therapeutic effects in acupunture treatment of primary dysmenorrhea combined with spinal Tui Na, and study its mechanism. Methods: Thirty cases of the treatment group were treated by acupuncture combined with spinal Tui Na, and thirty cases in the control group were treated by routine acupuncture. Results: The total effective rate was 93.3% in the treatment group, and 73.3% in the control group, with a significant difference between the two groups (P<0.05). Conclusions: Acupuncture combined with spinal Tui Na has good prospects for treatment of primary dysmenorrhea.  相似文献
