BACKGROUND: The exact etiologic mechanisms leading to the formation of nasal polyps have remained largely obscure. A key phenomenon of this specific type of chronic inflammatory disease in nasal respiratory mucosa is remarkable edema. Vascular permeability/vascular endothelial growth factor (VPF/VEGF) plays an important role in inducing angiogenesis and modulating capillary permeability. OBJECTIVE: To study the expression and localization of VPF/ VEGF as a putative key factor in nasal polyp development. METHODS: Specimens of nasal polyps (n = 12) were harvested during endonasal sinus surgery in patients with polypous chronic rhinosinusitis. Specimens of healthy nasal respiratory mucosa (n = 12) served as controls and were obtained from inferior turbinates of patients undergoing surgery for nasal obstruction without signs and symptoms of inflammatory disease. Frozen sections were immunohistochemically stained for VPF/VEGF and quantitatively analyzed, using computer-based image analysis. RESULTS: The expression of VPF/VEGF in specimens of nasal polyps was significantly stronger than in specimens of healthy nasal mucosa of controls. VPF/VEGF in polypous tissue was mainly localized in vascular endothelial cells, in basal membranes and perivascular spaces, and in epithelial cells. CONCLUSION: The markedly increased expression in nasal polyps as opposed to healthy nasal mucosa suggests that VPF/VEGF may play a significant role in both the formation of nasal polyps and in the induction of heavy tissue edema. This finding is discussed with respect to the differential expression of cyclooxygenase (COX) isoenzymes-1 and -2 (COX-1 and COX-2) in nasal polyps was significantly stronger than in specimens of healthy nasal mucosa of controls. VPF/VEGF in polypous tissue was mainly localized in vascular endothelial cells, in basal membranes and perivascular spaces, and in epithelial cells. 目的 :探讨转化生长因子β1 (TGF-β1 )在鼻息肉组织中的表达及意义。方法 :将 34例鼻息肉标本及30例中鼻甲粘膜标本行 TGF-β1 免疫组化染色 ,光镜观察。结果 :1TGF-β1 在鼻息肉组织的细胞外基质和固有层浸润细胞的表达明显高于中鼻甲组织 (P <0 .0 1)。 2鼻息肉组织中 TGF-β1 阳性细胞的形态及分布相似于嗜酸性粒细胞 ,二者阳性程度显著相关 (P <0 .0 5 )。 3TGF-β1 阳性表达与鼻息肉的临床分型无关 (P >0 .0 5 ) ,嗜酸性粒细胞浸润程度与鼻息肉临床分型密切相关 (P <0 .0 5 )。结论 :1TGF-β1 可能直接参与鼻息肉的病理变化 ,导致上皮基底膜增厚和基质纤维化。 2嗜酸性粒细胞可能为鼻息肉中 TGF- β1 的主要来源。 3嗜酸性粒细胞在鼻息肉的发病及复发过程中可能发挥重要作用。  相似文献

OBJECTIVE: To study the distribution and expression of aquaporion-2 (water channel protein; AQP-2) in nasal polyps and to evaluate the role of AQP-2 in the formation of nasal polyps. METHODS: Eleven samples of normal inferior turbinates and forty-six samples of nasal polyps were used. In the group of nasal polyps, there were ten cases of type II phase 1, twelve type II phase 2, ten type II phase 3 and fourteen type III. The expression of AQP-2 in both groups was studied with immunohistochemical technique. RESULTS: 1. Both the number of AQP-2 positive cells and the area density in the epithelial cell layer from nasal polyps were higher than those from inferior turbinates (P < 0.01, P < 0.05 respectively); In the group of nasal polyps, the area density from type III, type II phase 2 and type II phase 3 was significantly higher than that from type II phase 1 (P < 0.05); 2. Both the number of AQP-2 positive cells and the area density in the subepithelial tissue from type III, type II phase 2 and type II phase 3 nasal polyps were higher than those from type II phase 1 nasal polyps and inferior turbinates(P < 0.05), whereas the difference between the latter two was not significant(P > 0.05). CONCLUSION: The high expression of AQP-2 protein in nasal polyps may contribute to the genesis and development of nasal polyps.  相似文献

目的:探讨Tenascin(TN)在鼻息肉组织中的表达和分布特征及其在鼻息肉发生中的可能作用。方法:采用免疫组化链霉菌抗生物素蛋白—过氧化物酶(SP)法检测24例鼻息肉标本(鼻息肉组)和15例慢性肥厚性鼻炎下鼻甲标本(下鼻甲组)中TN的表达,并以5例健康者(对照组)下鼻甲黏膜作对照。结果:鼻息肉组和下鼻甲组黏膜上皮细胞及腺上皮细胞均表达TN;鼻息肉组TN的黏膜上皮阳性细胞表达的吸光度值显著高于下鼻甲组(P<0.01);鼻息肉组TN的腺上皮阳性细胞表达的吸光度值显著高于下鼻甲组(P<0.01);对照组下鼻甲组织中黏膜上皮及腺体几乎检测不到TN的表达;鼻息肉组腺体TN阳性率明显高于下鼻甲组(P<0.05)。结论:TN在鼻息肉组织中的高表达与鼻息肉的发生、发展相关;TN在鼻腔内的表达细胞是黏膜上皮细胞和浆液性腺上皮细胞。  相似文献

目的:探讨Fas和Bcl 2在鼻息肉组织中的表达及与鼻息肉发病的关系。方法:应用S P免疫组化法检测36例鼻息肉组织及19例下鼻甲组织中Fas和Bcl 2的表达情况。结果: (1)Fas在鼻息肉上皮及腺上皮的表达弱于对照组(P<0.05);(2)Bcl 2在鼻息肉上皮及腺上皮的表达强于对照组(P<0.05);(3)鼻息肉上皮细胞中Fas、Bcl 2蛋白的表达无相关性(P>0.05)。结论:(1)Fas和 Bcl 2在鼻息肉发生发展中起重要作用;(2)在NP上皮细胞中, Fas和Bcl 2无相关性,因此这两种凋亡基因在NP的形成过程中各起独立作用,协调抑制NP上皮细胞的凋亡。  相似文献

OBJECTIVE: To explore the role of tenascin (TN) and fibronectin (FN) in the pathophysiology of nasal polyp. METHODS: The expression of TN and FN in nasal polyps from 34 patients and in inferior turbinates from 20 patients with deviation of nasal septum was studied with immunohistochemical method. In patients with nasal polyps, the relations between expression and histopathologic characteristics, eosinophilias (EOS) infiltration, clinical staging and the size of nasal polyps were analyzed. RESULTS: (1) The gray score of TN and FN expression was 163.10 +/- 10.54 and 163.24 +/- 11.52 in nasal polyps respectively, whereas it was 175.49 +/- 9.29 and 173.93 +/- 7.92 in inferior turbinates respectively. The difference between two groups was significant(P < 0.01); (2) The expression of TN and FN in edematous type was significantly stronger than that in cystic fibrous and glandular type (P < 0.05); (3) The association between FN expression and EOS infiltration was significant(r = -0.60, P < 0.01); (4) The expression of TN and FN did not correlate with clinical staging and size(P > 0.05). CONCLUSION: Abnormal ECM may contribute to proliferation of epithelium, accumulation of EOS and edema formation, as a result, to enhance the development of nasal polyps.  相似文献

OBJECTIVE: To estimate the roles of CD43, CD20 and total IgE in the pathogenesis of nasal polyps. METHOD: Twenty-six cases of nasal polyps and 20 cases of mucosa of middle turbinates were used in present study. RESULTS: The number of CD43+ cells, CD20+ cells and plasma cells in nasal polyps were significantly more than that in middle turbinates (P < 0.01). The positive rate of local IgE+ cells in nasal polyps was significantly higher than in middle turbinate (P < 0.01). CONCLUSION: There were active cellular immunity and humoral immunity in nasal polyps. Local allergy may play a role in the pathogenesis of nasal polyps.  相似文献

目的:探讨转化生长因子β1(TGF-β1),基质金属蛋白酶1、7、9(MMP-1、7、9)及其组织抑制物-1(TIMP-1)在慢性鼻窦炎和鼻息肉以及正常下鼻甲黏膜组织中表达程度的差异。方法:采用酶联免疫吸附试验(ELISA)检测22例慢性鼻窦炎筛窦黏膜、21例鼻息肉和15例下鼻甲黏膜组织中TGF-β1,MMP-1、7、9以及TIMP-1的蛋白表达量,并比较他们在不同组织中表达程度的差异。结果:①TGF-β1、MMP-7、MMP-9及TIMP-1在慢性鼻窦炎和鼻息肉组织中的表达均比正常下鼻甲黏膜组织增高,差异具有统计学意义(均P<0.05);②慢性鼻窦炎组织中TGF-β1和TIMP-1相比鼻息肉组织中的表达增高,差异具有统计学意义(P<0.01和P<0.05);③鼻息肉组织中MMP-7的表达比慢性鼻窦炎组织中的表达增高,差异具有统计学意义(P<0.01)。结论:慢性鼻窦炎和鼻息肉组织显示出不同的TGF-β1、MMP-7、TIMP-1蛋白表达水平。  相似文献

目的:检测黏附分子E选择素在鼻息肉中的分泌水平与表达部位,借以探讨其与鼻息肉可能的病理机制的相关性。方法:采用酶联免疫吸附测定法检测25例鼻息肉(鼻息肉组)组织匀浆中E选择素水平,以9例鼻中隔手术患者之代偿肥大的下鼻甲后端黏膜作为对照(对照组);同时对上述标本进行苏木精伊红染色及E选择素免疫组织化学染色,观察其表达情况。结果:鼻息肉组大量嗜酸粒细胞浸润。E选择素浓度为(42.58±13.52)μg/L,对照组为(11.35±3.17)μg/L,差异有统计学意义(P<0.01)。E选择素主要表达于血管内皮、腺体上皮、间质和浸润炎症细胞上。鼻息肉组腺体上皮阳性表达率为92.0%,对照组为55.6%,差异有统计学意义(P<0.05)。鼻息肉组在血管内皮上阳性表达率为84.0%,对照组为44.4%;鼻息肉组在间质及浸润炎症细胞阳性表达率为92.0%,对照组为33.3%,差异均有统计学意义。Spearman相关性分析E选择素在血管内皮上的表达和在间质及浸润炎症细胞上的表达呈正相关(r=0.544,P<0.01)。结论:高表达的E选择素可能通过参与嗜酸粒细胞等炎症细胞的浸润、活化、增殖,从而在鼻息肉的形成过程中起重要作用。  相似文献

目的探讨血管内皮生长因子(Vascular endothelial growth factor,VEGF)、CD34在人各型鼻息肉组织中的表达及临床意义。方法采用免疫组织化学SP法,检测67例鼻息肉(Ⅱ型1期14例,Ⅱ型2期18例,Ⅱ型3期20例,Ⅲ型15例)和20例下鼻甲(对照组)组织中VEGF、CD34蛋白的表达情况,对CD34阳性微血管进行计数[微血管密度(microvessel density,MVD)]。结果鼻息肉Ⅱ型1期、Ⅱ型2期、Ⅱ型3期、Ⅲ型中,VEGF表达分别为17.35±5.29、19.13±4.85、36.76±4.38、38.49±6.07。CD34表达(MVD)分别为13.26±2.05、14.72±3.86、26.40±2.71、27.97±3.34。11型3期、Ⅲ型鼻息肉组织中VEGF、CD34的表达较Ⅱ型1期、Ⅱ型2期鼻息肉组织中显著升高(P〈0.05)。结论VEGF、CD34表达与鼻息肉发生、发展有关,对其检测有助于判断鼻息肉的发展趋势。  相似文献

BACKGROUND: The term nasal polyposis describes benign growth processes in the nasal and sinus mucosa, which are mainly located in the middle meatus and never in the inferior meatus. As a step to define the biochemical determinants relevant for growth regulation, we focused on endogenous lectins known for anti-apoptotic (galectin-3) and immunomodulatory (galectin-1) activities. DESIGN: Using computer-assisted microscopy, we performed an immunohistochemical investigation defining the quantitative parameters of expression of galectin-1 and galectin-3 in 10 nasal polyps, 10 middle turbinates, and 10 inferior turbinates, all of which were obtained from surgical resection. RESULTS: Our data show that galectin-3 expression is markedly (P<.001) higher in nasal polyps than in turbinates. No relation to the allergic status was discovered. Galectin-1 expression is higher in nasal polyps than in middle turbinates (P<.001) in nonallergic patients compared with allergic ones (in glandular epithelium, P =.009; in connective tissue, P =.006). The lowest galectin-1 expression was observed in the middle turbinate. CONCLUSIONS: These data are in line with a positive influence of galectin-3 on growth and an immunoregulatory role of galectin-1, mimicking an increased expression dependent on glucocorticoid.  相似文献

The factors involved in the development of chronic inflammation and edema in nasal polyps remain to be clarified. The expression of vascular endothelial growth factor (VEGF) has been described in plasma cells, suggesting that plasma cells may play a major role in the development of edema in nasal polyps through the production of VEGF. We performed immunohistochemical analysis using specific antibodies to VEGF and to the known VEGF receptors, VEGFR-1 and VEGFR-2, on paraffin sections of human nasal polyps ( n=11) and controls of human mucosa of the normal middle turbinates ( n=6). In normal turbinate mucosa, sporadic immunostaining for VEGF was observed throughout the endothelial cells of the small veins and arteries. VEGFR-1 and VEGFR-2 expression was faint in the healthy turbinates. In nasal polyp tissues, strong immunostaining for VEGF was found in the endothelium of blood vessels and in the infiltrating perivascular inflammatory cells. Fibroblasts also stained for VEGF. Strong immunolabeling to VEGFR-1 was evident in the vascular endothelium, whereas weak to moderate VEGFR-1-staining was generally confined to scattered mononuclear round cells. Mononuclear round cells and the endothelium of capillaries revealed immunoreactivity to VEGFR-2. These findings support a role for VEGF and its receptors, VEGFR-1 and VEGFR-2, in the development and perpetuation of edema and angiogenesis in nasal polyps.  相似文献

目的明确单核细胞趋化蛋白1(monocyte chemotactic protein 1,MCP-1)和血管内皮生长因子(vascular endothelial growth factor,VEGF)在鼻息肉组织中的表达及其相关性,初步探讨MCP-1与鼻息肉发生的关系。方法取40例鼻息肉组织和25例下鼻甲组织,应用原位杂交和免疫组织化学等方法检测MCP-1和VEGF mRNA及蛋白质的表达。结果鼻息肉组织中MCP-1和VEGF mRNA及蛋白质的表达均高于对照组下鼻甲组织(P值均〈0.01);鼻息肉组织中MCP-1和VEGF蛋白质的表达呈正相关(r=0.871,P〈0.05)。结论鼻息肉组织中MCP-1和VEGF表达增加,二者协同作用可能是鼻息肉形成的原因之一。  相似文献

目的 :检测鼻息肉组织和鼾症下鼻甲粘膜组织中的血管内皮生长因子 (VEGF) m RN A水平的表达 ,了解其在慢性炎症过程中的作用。方法 :取 6例行下鼻甲切除术的下鼻甲粘膜和 7例鼻息肉切除术的鼻息肉标本 ,用半定量的反转录 -聚合酶链反应 (RT- PCR)方法检测 VEGF的 m RNA表达。结果 :RT- PCR结果显示在鼻息肉组织中 V EGF的表达较鼾症患者下鼻甲粘膜组织明显升高。结论 :鼻息肉组织中 VEGF的表达显著升高 ,推测 VEGF在鼻息肉的形成、生长及复发过程中具有极其重要的作用。  相似文献

红霉素干预对人鼻息肉上皮细胞凋亡的影响   总被引:4,自引:0,他引:4  
目的观察红霉素对体外培养的鼻息肉上皮细胞B细胞淋巴瘤-2基因(B cell lymphomas-2,Bcl-2)和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)表达的影响,探讨上皮细胞凋亡在鼻息肉组织形成中的意义。方法将30例鼻息肉及15例下鼻甲组织均分成两组,分别对其上皮细胞进行原代培养,其中一组加入红霉素。培养第1、3、5天应用末端脱氧核苷酰转移酶介导的脱氧尿苷三磷酸原位缺口末端标记法(the TdT-mediated dUTP nick end labeling,TUNEL)检测细胞凋亡情况,并用免疫组化法检测细胞凋亡基因Bax、抑凋亡基因Bcl-2的表达。结果①鼻息肉红霉素组培养第1、3、5天的上皮细胞凋亡指数(用x±s表示,下同)分别为(33.23±6.50)%、(36.14±3.21)%和(52.63±7.86)%,对照组(不加红霉素组)为(31.02±5.60)%、(33.88±4.02)%和(39.64±7.48)%,培养第5天的上皮细胞凋亡指数红霉素组与对照组差异有统计学意义(t=4.480, P<0.05)。②鼻息肉上皮细胞Bcl-2和Bax表达均显著强于下鼻甲组织(P<0.01);培养第5天鼻息肉上皮细胞红霉素组Bax表达显著强于对照组(t=8.734,P<0.05),Bcl-2表达虽强于对照组,但差异无统计学意义;下鼻甲上皮细胞红霉素组与对照组的Bcl-2与Bax阳性表达差异均无统计学意义(P均>0.05);红霉素干预第5天鼻息肉上皮细胞显示明显的凋亡趋势(P<0.05)。结论红霉素能显著提高Bax蛋白的表达,有显著的促鼻息肉上皮细胞凋亡的作用。  相似文献
