Aim of the study

Evaluate the potential inhibition of cough in a mouse model induced by sulfur dioxide gas of methanol and chloroform extracts of aerial parts of Chamaedorea tepejilote.

Material and methods

The antitussive effect was examined using the Ogyhara method. The extracts and theobromine were orally administered to mice. Each extract was administered to one group (400 mg/Kg) and theobromine (2 mg), and negative control received PVP. The frequency of cough was measured at 60, 90 min after the administration of chloroform extract.

Results

The water extract of C. tepejilote had no effect on the pharmacological model used in this study. The effects of the methanol and chloroform extracts of this plant on sulfur-dioxide-induced cough in experimental animals show activity dose-dependently in the range between 100 to 500 mg/Kg. At doses of 400 mg/kg, the methanol and chloroform extracts inhibited cough by 41.72% and 49.1%, respectively; these results are comparable to the effect produced by the antitussive agent theobromine (53.4% inhibition).

Conclusions

Both extracts exhibited significant antitussive activity, and the effect of the chloroform extract was dose dependent. The antitussive activity of this extract was comparable to that of theobromine. The effect of chloroform extract on the trachea was also tested; it did not relax pre-contractions in guinea pig trachealis induced KCl, carbachol or histamine.  相似文献

Barleria prionitis Linn. (Family: Acanthaceae), one of the important Ayurvedic medicinal plant in India, has long been used to treat variety of ailments including swellings, gout, arthritic and rheumatic disorders, nervine and skin diseases, and also acts as immunorestorative.

Aim of the study

The present study was aimed to explore in vitro and in vivo immunomodulatory activities of the iridoids fraction i.e. n-butanol fraction of methanol extract from Barleria prionitis aerial parts (IFBp).

Materials and methods

IFBp was studied for in vitro [nitroblue tetrazolium (NBT) test and neutrophils candidacidal assay] and in vivo immunomodulatory activity on cellular and humoral immune responses to the antigenic challenge by sheep red blood cells (SRBCs) and by neutrophil adhesion test, phagocytic activity and cyclophosphamide-induced myelosuppression. The study comprised the preliminary phytochemical screening, HPTLC standardization and maximum tolerable dose determination of IFBp.

Results

IFBp (50, 100 and 200 μg/ml) significantly (P ≤ 0.01) increased the intracellular killing activity of stimulated neutrophils assayed by in vitro NBT reduction test and neutrophils candidacidal assay. Pretreatment of IFBp (100 and 200 mg/kg; p.o.) evoked a significant increase in percent neutrophils and neutrophils adhesion to nylon fibres. Oral administration of IFBp augmented the humoral immune response to SRBCs, evidenced by increase in antibody titres and dose dependently potentiated the delayed-type hypersensitivity reaction induced by SRBCs in mice. IFBp potentiated significantly (P ≤ 0.01) the macrophage phagocytic activity and ameliorated the red blood cells, total white blood cells and platelets count and haemoglobin concentration, and also restored the myelosuppressive effects induced by cyclophosphamide. The content (% w/w; mean ± SD, n = 3) of main iridoids i.e. shanzhiside methyl ester and barlerin was found to be 21.55 ± 2.40 and 10.03 ± 1.69 in IFBp of BP, respectively.

Conclusion

The present investigation reveals that IFBp is a potent immunostimulant, stimulating both the specific and non-specific immune mechanisms.  相似文献

Nelumbo nucifera Gaertn. (Nymphaeaceae) is a well-known aquatic plant which has been used for the treatment of several disorders including skin disease, cough, inflammation, fever and many other disorders.

Aim of the study

To explore the immunomodulatory activity of extract of rhizome (NNRE) and seed (NNSE) of the plant.

Materials and methods

The immunomodulatory activity of NNRE and NNSE was evaluated using various in vivo models including the total and differential leukocyte count (TLC and DLC), nitroblue-tetrazolium reduction (NBT) test, neutrophil adhesion test, phagocytic response and delayed type hypersensitivity (DTH) reaction. Sheep red blood cells (SRBC, 5 × 109 cells/ml) were used to immunize the animals. NNRE and NNSE at the doses of 100 and 300 mg/kg were administrated.

Result

The TLC and lymphocyte count increased significantly but the neutrophil count was decreased for NNRE and NNSE treated groups compared to the control. A dose-dependent potentiation of DTH reaction induced by SRBC was observed from the extracts. The percentage of neutrophil adhesion to the nylon fiber was increased in NNRE treated groups (63.22 and 62.91%) compared to the NNSE treated group (54.86 and 54.23%). A potential phagocytic response was seen on treatment of the extracts, and significant changes were observed in the formation of formazone crystals.

Conclusion

This finding suggests that the extract of rhizome and seed Nelumbo nucifera stimulate defense system by modulating several immunological parameters.  相似文献

Ethnopharmacological relevance

Smilax canariensis is an endemic species of the Canary Islands, popularly known as “Zarzaparrilla sin espinas”. The rhizome, leaves and stem of this species has wide use in folk medicine practice on the islands, where they are habitually employed as diuretic.

Aim of the study

In this paper we report on the diuretic activity in experimental animals of several fractions of the methanol extract of this species.

Materials and methods

Four fractions of the methanol extract of the rhizomes, leaves and stem of Smilax canariensis (50 and 100 mg/kg), Furosemide and Hydrochlorotiazide (10 mg/kg), were orally administered to laboratory animals to evaluate their diuretic activity. Water excretion rate, pH, conductivity, and content of Na+, K+ and Cl were measured in the urine of saline-loaded mice.

Results

All the studied extracts showed an interesting increase of the diuresis, although the n-buthanol (27%; p < 0.05) and ethyl acetate extract (35%; p < 0.01), at 100 mg/kg p.o., showed the most interesting diuretic activity, which suggested that this diuretic effect is associated with the compounds contained in the fractions of intermediate polarity (ethyl acetate and n-butanol), decreasing in the most extreme apolar and polar sub-extracts (dichloromethane and methanol: water respectively). The increase in diuresis produced by these two extracts was very close to the values of Hydrochlorothiazide (32%) or Furosemide (39%), used as reference diuretics.

Conclusions

These data, together with previous results on the aqueous and methanol extracts, reaffirm assertions made regarding the effectiveness of the extracts of this plant against urinary pathologies in the Canary Islands folk medicine.  相似文献

The whole plant of Enicostemma axillare Raynal (Family: Gentianaceae) is used in variety of diseases in traditional Indian system of medicine including hepatic ailments.

Aim of the study

Swertiamarin isolated from Enicostemma axillare Raynal was evaluated for antioxidant and hepatoprotective activity.

Materials and methods

Swertiamarin was isolated from successive ethyl acetate extract of the plant Enicostemma axillare belongs to the family Gentianaceae. The concentration of swertiamarin was determined by high performance thin layer chromatography (HPTLC). The hepatoprotective and antioxidant activity of swertiamarin (100 and 200 mg/kg body weight) was carried out against d-Galactosamine (d-GalN) (200 mg/kg body weight intraperitoneally i.p.) induced liver injury in rats.

Results

Swertiamarin a secoiridoid glycoside was found to contain a major constituent of the extract. d-GalN caused significant hepatotoxicity by alteration of several hepatic parameters. It also caused significant lipid peroxidation and reduced the levels of antioxidant defense mechanisms. The treatment with swertiamarin at 100 and 200 mg/kg body weight when administered orally for 8 days prior to d-GalN caused a significant restoration of all the altered biochemical parameters due to d-GalN towards the normal, indicating the potent antioxidant and hepatoprotective nature of swertiamarin.

Conclusions

Swertiamarin isolated from Enicostemma axillare possesses significant antioxidant and hepatoprotective properties against d-GalN induced hepatotoxicity given at 100 and 200 mg/kg body weight orally for 8 days, which might be due to its in vitro antioxidant activity.  相似文献

Ethnopharmacological relevance

An ethnopharmacological investigation of medicinal plants traditionally used to treat diseases associated with fevers in Dharmapuri region of South India was undertaken. Twenty four plants were identified and evaluated for their in vitro activity against Plasmodium falciparum and assessed for cytotoxicity against HeLa cell line.

Aim of the study

This antimalarial in vitro study was planned to correlate and validate the traditional usage of medicinal plants against malaria.

Materials and methods

An ethnobotanical survey was made in Dharmapuri region, Tamil Nadu, India to identify plants used in traditional medicine against fevers. Selected plants were extracted with ethyl acetate and methanol and evaluated for antimalarial activity against erythrocytic stages of chloroquine (CQ)-sensitive 3D7 and CQ-resistant INDO strains of Plasmodium falciparum in culture using the fluorescence-based SYBR Green I assay. Cytotoxicity was determined against HeLa cells using MTT assay.

Results

Promising antiplasmodial activity was found in Aegle marmelos [leaf methanol extract (ME) (IC50 = 7 μg/mL] and good activities were found in Lantana camara [leaf ethyl acetate extract (EAE) IC50 = 19 μg/mL], Leucas aspera (flower EAE IC50 = 12.5 μg/mL), Momordica charantia (leaf EAE IC50 = 17.5 μg/mL), Phyllanthus amarus (leaf ME IC50 = 15 μg/mL) and Piper nigrum (seed EAE IC50 = 12.5 μg/mL). The leaf ME of Aegle marmelos which showed the highest activity against Plasmodium falciparum elicited low cytotoxicity (therapeutic index > 13).

Conclusion

These results provide validation for the traditional usage of some medicinal plants against malaria in Dharmapuri region, Tamil Nadu, India.  相似文献

Ethnopharmacological relevance

India is a rich source of medicinal plants and a number of plant extracts are used against diseases in various systems of medicine such as Ayurveda, Unani and Siddha. Only a few of them have been scientifically explored. One of such plants is Ammannia baccifera L., traditionally used as an antitumor agent.

Aim of the study

The objective of the present study was to explore the anticancer activity of the methanol extract of A. baccifera against Dalton's ascites lymphoma (DAL)-bearing Swiss albino mice and in vitro cytotoxicity against human cervical cancer cell line (HeLa).

Materials and methods

In vitro cytotoxic effects of A. baccifera was evaluated against HeLa and NIH 3T3 cells using MTT [3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide] method. Anticancer activity of the extract was evaluated in Swiss albino mice against DAL cell line at the doses of 100 and 200 mg/kg p.o. The extract was administered to animals for 14 consecutive days. After 12 h fasting from last dose, the mice were sacrificed and the anticancer effect of A. baccifera was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight, hematological parameters and certain antioxidant biochemical parameters against DAL bearing control group.

Results

The extract of A. baccifera was cytotoxic to the HeLa cancer cell line but relatively non-toxic to the normal cell line NIH 3T3. A. baccifera extract treatment resulted in significant decreases in tumor volume, viable cell count and tumor weight and enhanced the life span of DAL bearing mice. Hematological parameters such as RBC, hemoglobin and lymphocyte count reverted to normal level in A. baccifera treated mice. The extract also significantly (p<0.05) decreased the levels of lipid peroxidation and increased the activities of GSH, GPx, SOD and CAT. The anticancer effects of the A. baccifera extract is comparable with that of the standard drug 5-Fluorouracil.

Conclusion

The results showed that the methanol extract of A. baccifera is effective in inhibiting the tumor growth in ascetic models that is comparable to 5-fluorouracil. This plant has potential in the development of anticancer therapy and this study scientifically validated the folklore use of this plant.  相似文献

The traditional use of Punica granatum has been reported to regulate urine discharge and controls the burning sensation of urine.

Materials and methods

Animals model of calcium oxalate urolithiasis was developed in male rats by adding ethylene glycol 0.75% in drinking water. The Punica granatum chloroform extract (PGCE) and Punica grantum methanol extract (PGME) orally at 100, 200 and 400 mg/kg, respectively, were administered along with ethylene glycol for 28 days. On 28 day, 24 h urine was collected from individual rats and used for estimation of urine calcium, phosphate and oxalate. The serum creatinine, urea and uric acid levels were estimated in each animal. The kidney homogenate was used for the estimation of renal oxalate contents. The paraffin kidney sections were prepared to observe the CaOx deposits.

Results

The ethylene glycol control (Gr.-II) had significant (P < 0.001 vs. normal) increase in levels of urine oxalate, calcium and phosphate, serum creatinine, urea and uric acid and renal tissues oxalates, as compared to normal (Gr.-I). The paraffin kidney sections show significant histopathological changes. The treatment of PGCE and PGME at 100, 200 and 400 mg/kg doses, significantly (P < 0.001 vs. control) decreased the urine oxalate, calcium and phosphate, renal tissue oxalates and serum creatinine, urea and uric acid, in EG induced urolithiasis after 28 days.

Conclusions

The PGCE and PGME at the doses of 400 mg/kg, found to be more effective in decreasing the urolithiasis and regeneration of renal tissues in male rats.  相似文献

Mangaba (Hancornia speciosa Gomez) is a medicinal plant frequently cited in ethnopharmacological inventories of the central region of Brazil against gastrointestinal disorders such as diarrhoea, ulcer, gastritis and stomachache.

Aim of the study

The hydroalcoholic extract (HE) and infusion (BI) of Hancornia speciosa bark were investigated for their ability to prevent and heal rodent gastric ulcer.

Materials and methods

The preventive and healing action of both preparations of Hancornia speciosa were evaluated in experimental models in rodents that simulated this disease in human gastric mucosa.

Results

BI did not exert gastroprotective effect, in contrast to HE (500 mg/kg, p.o.) that decreased (p < 0.05) the severity of gastric damage induced by HCl/ethanol (52%), indomethacin/bethanechol (51%), stress (52%) or pylorus ligature experiments (54%). HE increased (p < 0.05) the pH and decreased acid output of gastric juice. This extract promoted increase of mucus amount (3.62 mg/wt. tissue vs. 5.81 mg/wt. tissue), healing action (67%) and displayed anti-Helicobacter pylori effect.

Conclusions

The antiulcer action of Hancornia speciosa resulted in increase of gastric mucus formation and antioxidant properties of polymeric proanthocyanidins present in the bark composition of this medicinal plant.  相似文献

Aim of the study

Polygonum cuspidatum has long been used as a traditional medicine inducing wound healing. In this study, the extract from the Chinese medicinal herb Polygonum cuspidatum was investigated on its wound healing activity, in order to obtain an accurate elucidation of its traditional use value.

Materials and methods

After creating wound healing model on the back of rats, the extract from the Chinese medicinal herb Polygonum cuspidatum was applied. Wound healing rates were calculated at 3, 7, 14, and 21 days after the wounding, and tissues were harvested at 1, 3, 7, 14 and 21 days for histological and immunohistochemistry analysis. The stages of wound granulation tissues were evaluated histopathologically. The expression of TGF-β1 was determined by immunohistochemically.

Results

Wound healing rates were significantly higher at 3, 7, 14 and 21 days in the extract group than in the control (p < 0.05). Histological results showed more well-organized bands of collagen, more fibroblasts and hair follicle and less inflammatory cells in the extract group. The immunohistochemical results revealed that TGF-β1 increased in the extract group on day 1, 3 and 7 post-wounding (p < 0.05).

Conclusion

The present study has shown that the extract from the Chinese medicinal herb Polygonum cuspidatum possesses wound healing activity, and thus provided the evidence for its traditional use value.  相似文献

Ethnopharmacological relevance

It is well known that fragrance impacts behaviors and autonomic functions, and is increasingly used as relaxant, carminative, as well as sedative in aromatherapy. Kaempferia galanga L. is one of the popular traditional aromatic medicinal plants used in tropics and subtropics of Asia including China, Japan and Indochina.

Aim of the study

The aim of present study was to investigate sedative effects of hexane extract of Kaempferia galanga L.and 2 active aromatic compounds (compound 1: ethyl trans-p-methoxycinnamate and compound 2: ethyl cinnamate ) included in the extyract by means of inhalation in mice.

Materials and methods

Sedative activity was assessed by inhalation of the hexane extract and two major aromatic compounds.

Results

Inhalation of hexane extract at the doses of 1.5 and 10 mg showed significant reduction of locomotor activity, indicating considerable sedative and relaxant effects. Compound 1 and 2 were proved to possess sedative effects at 0.0014 mg and 0.0012 mg respectively.

Conclusions

These results suggest the promising application of Kaempferia galanga L. and its constituents in aromatherapy.  相似文献

Aim of the study

The present study was carried out to evaluate the immunomodulatory potential of Ziziphus mauritiana (Lamk.) seed extract to ascertain the folkloric claim as immunomodulator.

Materials and methods

The aqueous-ethanolic seed extract (100–400 mg kg−1) of Z. mauritiana was investigated for immunomodulatory potential in mice. The extract was standardized with HPLC using betulinic acid as a marker. Functions of various immunocytes in the form of humoral (development of anti-SRBC (sheep red blood cells) antibody titers) and cell-mediated immune response (delayed type hypersensitivity, nitroblue tetrazolium reduction, inducible nitric oxide synthase activity and bactericidal activity) was studied in SRBC immunized mice. The cytokine, IFN-γ (interferon-gamma) and IL-4 (interleukin-4) secretion was also measured quantitatively by ELISA as the expression of functions of Th-1 and Th-2 respectively. Levamisole (2.5 mg kg−1) was used as standard drug.

Results

The seed extract demonstrated significant (P < 0.05–0.001) up-regulation of cell-mediated, humoral immune response and Th-1 mediated cytokine IFN-γ and decline in Th-2 mediated cytokine IL-4. At higher dose of extract the results were comparable to that of the levamisole.

Conclusion

The immunostimulatory potential of this seed extract is likely to be mediated through its effect on macrophage function and Th-1 mediated immunity confirming the folkloric use of this plant.  相似文献

Ethnopharmacological relevance

Rhizome of Zingiber montanum has been extensively used as a folk medicine to ameliorate peptic ulcer at northern part of Bangladesh.

Aim of the study

To identify the antiulcer principle of the MeOH extract of the rhizome of Zingiber montanum by an ex vivo bioassay guided chromatographic separation and purification, and structure elucidation of the purified compound by spectroscopic methods.

Materials and methods

Dried powder of Zingiber montanum rhizomes was extracted with MeOH. The antiulcer activity of the crude extract and its chromatographic fractions were evaluated by the inhibition of 1 N HCl induced gastric lesions in Swiss albino mice. The pure compound was purified from the active fraction by crystallization with hexanes. Structure of the pure compound was elucidated by spectroscopic methods. The antiulcer activity of the pure compound was evaluated by the inhibition of 1 N HCl, 95% ethanol and indomethacin induced gastric lesions in mice.

Results

The MeOH extract of Zingiber montanum showed 61.97% and 83.10% inhibition of the 1 N HCl induced gastric lesions at doses of 200 mg/kg and 400 mg/kg, respectively, in mice. Chromatographic separation on silica gel of the extract was yielded seven fractions and the fraction 2 was found to have most potent antiulcer activity in mice. This fraction showed 77.46% inhibition of the 1 N HCl induced gastric lesions at a dose of 40 mg/kg in mice. Crystallization of the fraction yielded 1 (zerumbone, 180 mg). It showed statistically 45.77% and 92.25% inhibition of 1 N HCl induced gastric lesions in mice at doses of 20 mg/kg and 40 mg/kg, respectively. It also showed 29.07% and 45.35% inhibition of 95% ethanol induced gastric mucosal damage, and 64.76% and 72.38% inhibition of indomethacin induced gastric lesions in mice at doses of 20 mg/kg and 40 mg/kg, respectively.

Conclusion

Zerumbone (1) showed potent cytoprotective effect against necrotizing agent (HCl) and non-steroidal anti-inflammatory drug (indomethacin) induced gastric ulceration. It also exhibited moderate cytoprotective effect against noxious agent (EtOH) induced gastric lesions. It can be considered as a promising new antiulcer natural drug lead.  相似文献

Bunium persicum is an endemic plant to Iran which its seeds have a long history of medicinal uses.

Aim of the study

This work aimed to study the anticonvulsant effect of the essential oil and methanolic extract of the plant.

Materials and methods

The essential oil and methanolic extract of the plant were studied against pentylenetetrazole (PTZ) and maximal electroshock (MES) induced convulsions in mice in different doses. The neurotoxicity of the essential oil and methanolic extract was investigated using rotarod method.

Results

The essential oil and methanolic extract prolonged the onset of clonic and tonic seizures in PTZ. The tonic seizures were prevented by essential oil in both methods at dose of 1 mL/kg and higher doses. The methanolic extract inhibited PTZ-convulsions at dose 3 g/kg and was ineffective against MES induced convulsion.

Conclusions

The essential oil of the plant might be useful to control absence and grand mal seizures at dose 1 mL/kg. This activity might be due to its content of monoterpenes.  相似文献

Aim of the study

To evaluate the antidiarrhoeal properties of Alstonia congensis leaves claimed to be effective for the treatment of diarrhoea by traditional healers during our ethnopharmacological investigation conducted in Kinshasa, Democratic Republic of Congo.

Materials and methods

The aqueous extract (decoction), and the 80% hot methanol extract (Soxhlet extraction) were obtained. This last extract was fractionated. The antidiarrhoeal activity was evaluated using castor oil and magnesium sulphate-induced diarrhoea in animals. The potential antibacterial activity of all samples was also assessed in vitro.

Results

At all oral doses of 100 and 200 mg/kg body weight, all A. congensis samples showed significant and dose-dependent antidiarrhoeal activity in treated Wistar rats characterised by significant increase of onset time and decrease of all other diarrhoeal parameters at various degrees compared to untreated groups in both models. At the highest oral dose of 200 mg/kg bodyweight, the 80% hot methanol and aqueous extracts produced 79.8±2.1% and 78.6±0.5%, and 75.0±2.1% and 71.4±2.1% inhibition of defecation and diarrhoea respectively against castor oil-induced diarrhoea, and 75.0±1.2% and 73.3±1.2% inhibition of diarrhoea respectively against magnesium sulphate-induced diarrhoea. The 80% hot methanol and aqueous detannified extracts showed low activity (42–47% inhibition of defecation and/or diarrhoea in both tests) suggesting that tannins may be responsible for the observed activity. At the same oral doses, the total alkaloid extract, the chloroform soluble fraction rich in alkaloids, the 80% methanol and the alkaline aqueous soluble subfractions produced more than 50% inhibition of defecation and/or diarrhoea in both tests. From the antibacterial testing in vitro, results indicated that all A. congensis samples exhibited an antibacterial activity mainly against bacteria implicated in diarrhoea with MIC and MBC values in the range of 15.6–500 μg/ml. The most active samples were the aqueous (decoction) and the 80% hot methanol dried extracts, the chloroform subfraction rich in alkaloids and the total alkaloid extract (MIC: 15.7–125 μg/ml, MBC: 31.2–250 μg/ml). Proteus varibilis was found to be the most resistant microorganism.

Conclusion

These reported results can partly support and justify the traditional use of extracts from Alstonia congensis leaves for the treatment of diarrhoea in tradittional medicine.  相似文献

Ethnopharmacological relevance

Acacia nilotica is widely distributed in Asia. In India, it occupies an important place in the indigenous system of medicine against anti-inflammatory, antioxidant, cancers, and/or tumors.

Aim of the study

The purpose of this study is to investigate the inhibitory effect of Acacia nilotica leaves extract and γ-Sitosterol on cell proliferation, the apoptotic effect and cell cycle arrest in breast and lung cancer cells.

Materials and methods

GC–MS and HPLC were used to determine the chemical constituents of this extract and γ-Sitosterol respectively. Human MCF-7 and A549 cell lines were treated with Acacia nilotica extract and γ-Sitosterol. Cell viability was determined by MTT assay. Cell proliferation was determined by BrdU incorporation assay. Apoptosis was detected by cell morphologic observation through AO/EtBr staining, cell cycle analysis, and immunoblot analysis on the expression of protein associated with cell cycle arrest.

Results

Experimental results of bioactive compound analysis indicate that γ-Sitosterol, bioactive ingredients of Acacia nilotica extract. The IC50 value of extract on MCF-7 and A549 cancer cells was 493.3 ± 15.2 and 696.6 ± 11.5 μg/ml, respectively. Acacia nilotica extract and γ-Sitosterol were inhibited the cell proliferation by 54.34 ± 1.8 and 42.18 ± 3.9% for MCF-7 and 58.26 ± 1.5 and 44.36 ± 3.05% for A549 cells. The percentage of apoptotic cells observed in the MCF-7 and A549 cell lines were increased to 42.46 and 36.8% of extract; 46.68 and 43.24% for γ-Sitosterol respectively. Flow cytometric analysis results demonstrate that cells were arrested at the G2/M phase and decrease the c-Myc expression.

Conclusions

This study demonstrates in vitro results, which support the ethnomedical use of γ-Sitosterol against cancer. Experimental results of this study suggest that γ-Sitosterol exerts potential anticancer activity through the growth inhibition, cell cycle arrest and the apoptosis on cancer cells.  相似文献

Ethnopharmacological relevance

Lithrea molleoides (Vell.) Engl. (Anacardiaceae) is a medicinal plant commonly used in traditional medicine in South America.

Aim of the study

In the present study, the in vivo antinociceptive effect of L. molleoides' aqueous extract and its isolated compounds has been investigated.

Materials and Methods

Antinociceptive activity was evaluated through writhing, formalin and hot plate tests in mice. The phytochemical analysis was performed.

Results

The extract produced significant inhibition on nociception induced by acetic acid (ED50: 8.7 mg/kg, i.p.) and formalin (ED50: 7.7 mg/kg, i.p.) administered intraperitoneally and also orally. Yohimbine diminished the activity of the extract in the acetic acid test meanwhile haloperidol enhanced its effect. Two majority compounds, shikimic and vanillic acid were active in chemical nociceptive models used in this work, producing the highest inhibition of the writhing response at a dose of 30 mg/kg i.p. (55.4% and 57.1%, respectively) meanwhile at 100 mg/kg p.o. produced a slight response (23.3% and 23.9%, respectively).

Conclusions

These results suggest that L. molleoides' aqueous extract produced antinociception possibly related to the presence of shikimic and vanillic acid. The adrenergic and dopaminergic systems seem to be involved in the mechanism of antinociception of the extract.  相似文献

Ethnopharmacological relevance

Anthocephalus cadamba (Roxb.) Miq. (Family: Rubiaceae) is commonly known as “Kadamba” in Sanskrit and Hindi in India. Various parts of this plant have been used as a folk medicine for the treatment of tumor, wound healing, inflammation and as a hypoglycemic agent.

Aim of study

The purpose of this investigation was to evaluate the antitumor activity and antioxidant status of defatted methanol extract of A. cadamba (MEAC) on Ehrlich ascites carcinoma (EAC) treated mice.

Materials and methods

In vitro cytotoxicity assay has been evaluated by using the trypan blue method. The determination of in vivo antitumor activity was performed by using different EAC cells (2×106 cells, i.p.) inoculated mice groups (n=12). The groups were treated for 9 consecutive days with MEAC at the doses of 200 and 400 mg/kg b.w. respectively. After 24 h of last dose and 18 h of fasting, half of the mice were sacrificed and the rest were kept alive for assessment of increase in life span. The antitumor potential of MEAC was assessed by evaluating tumor volume, viable and nonviable tumor cell count, tumor weight, hematological parameters and biochemical estimations. Furthermore, antioxidant parameters were assayed by estimating liver and kidney tissue enzymes.

Results

MEAC showed direct cytotoxicity on EAC cell line in a dose dependant manner. MEAC exhibited significant (P<0.01) decrease in the tumor volume, viable cell count, tumor weight and elevated the life span of EAC tumor bearing mice. The hematological profile, biochemical estimations and tissue antioxidant assay were reverted to normal level in MEAC treated mice.

Conclusion

Experimental results revealed that MEAC possesses potent antitumor and antioxidant properties. Further research is going on to find out the active principle(s) of MEAC for better understanding of mechanism of its antitumor and antioxidant activity.  相似文献
