侧脑室注射生长抑素对大鼠痛阈和电针镇痛作用的影响   总被引:6,自引:0,他引:6  
观察脑内生长抑素对痛阈和针刺镇痛的影响。以钾离子透入引起大鼠甩尾为测痛方法,侧脑室注射生长抑素,生长抑素的耗竭剂半胱胺和抗生长抑素血清,电针刺激大鼠足三里。侧脑室注射SS使大鼠痛阈升高并使电针镇痛的效应增强,侧脑室分别注射CSH和ASSS使大鼠前阈降低并使电针镇痛的效应减弱。  相似文献

目的观察鞘内注射抗生长抑素血清(ASSS)对佐剂性关节炎(AA)大鼠痛阈、电针(EA)镇痛及中枢β-内啡肽(-βEP)水平的影响,探讨其可能的镇痛机制。方法以AA大鼠为炎症痛模型,以痛阈为指标观察鞘内注射ASSS对痛阈和EA镇痛的影响;以放免法测定AA大鼠下丘脑、脊髓中-βEP含量,观察鞘内注射ASSS及EA镇痛对其影响。结果鞘内注射ASSS可显著降低AA大鼠痛阈,并使EA镇痛效应明显降低;EA镇痛可提高下丘脑和脊髓-βEP含量,鞘内注射ASSS可使中枢-βEP水平进一步显著升高。结论内源性生长抑素在痛觉调制中起重要作用,有显著的镇痛效果并与增强EA镇痛作用密切相关;其镇痛机制可能与中枢-βEP水平相关。  相似文献

大鼠中缝大核微量注射L-精氨酸对痛阈及电针镇痛的影响   总被引:1,自引:0,他引:1  
目的观察延髓中缝大核 (NRM )微量注射L -精氨酸 (L -Arg)对痛阈及电针镇痛作用的影响 ,以探讨NRM中一氧化氮 (NO)在痛觉调制和针刺镇痛过程的作用和作用机理。方法采用大鼠热水刺激甩尾测痛模型 ,以 5 0± 0 .5℃热水刺激大鼠尾部引起甩尾反应的潜伏期为痛阈指标 ,测定大鼠的痛阈。结果NRM微量注射 1mmolL -Arg对大鼠痛阈无明显影响 ;2mmol、4mmol、8mmolL -Arg均能降低痛阈 ,其作用呈剂量依赖性。NRM预先注入 8mmolL -Arg再电针 3 0分钟 ,大鼠痛阈明显低于生理盐水电针组。结论NRM内NO参与痛和电针镇痛的病理生理过程 ,NRM内NO含量增加明显降低痛阈 ,削弱电针镇痛效应  相似文献

目的:通过建立大鼠足底切口痛模型,拟从大鼠行为学方面来观察应用针刺夹脊穴和夹脊穴持续输注镇痛药物治疗术后疼痛的效果。方法60只健康雄性SD大鼠,随机分为健康对照组、模型组、皮下输注组、夹脊穴电针组、夹脊穴输注组,每组12只。按照Brennan法建立大鼠足底切口疼痛模型。对5组大鼠于术前1h、术后6、24、48h,测定机械缩足反射阈(MWT)和热退缩潜伏期(TWL)值。结果MWT值的变化:5组大鼠实验前MWT值的基础值组间差异无统计学意义(P>0.05);健康对照组各时间点的MWT值比较差异均无统计学意义(P>0.05);模型组、皮下输注组、夹脊穴电针组、夹脊穴输注组与健康对照组比较,在术后6、24、48hMWT值明显减少(P<0.05);皮下输注组、夹脊穴电针组、夹脊穴输注组与模型组比较,在术后6、24、48hMWT值明显增加(P<0.05);夹脊穴输注组与皮下输注组、夹脊穴电针组比较,在术后6、24、48hMWT值明显增加(P<0.05);TWL值的变化:与健康对照组比较,模型组大鼠在术后6、24、48h明显缩短(P<0.05);皮下输注组、夹脊穴电针组、夹脊穴输注组与模型组比较,在术后6、24、48hTWL明显延长(P<0.05);夹脊穴注射组与皮下注射组、夹脊穴电针组比较,在术后6、24、48hTWL明显延长(P<0.05)。结论电针夹脊穴和夹脊穴持续输注镇痛药物对切口痛大鼠具有明显镇痛效应;夹脊穴输注的镇痛效果好于皮下镇痛。  相似文献

脑室内注射(icv)P物质(substance P,SP,1μg/10μl)可使痛阈明显升高,纹状体、下丘脑内甲啡肽样免疫活性物质(Ir-MEK)含量显著增多(P<0.05~0.01),而海马内Ir-MEK含量无明显变化 电解损毁中脑导水管中央灰质(PAG)后,SPicv升高痛阈和二脑区内Ir-MEK含量的作用显著降低(P<0.05~0.01)。 将SP注入PAG(0.4μg/4μl),电针“夹脊”穴(5Hz、3V)或腹腔注射吗啡(3mg/kg)均可使痛阈和纹状体、下丘脑内Ir-MEK含量显著升高。在SP注入PAG或腹腔注射吗啡时,海马内Ir-MEK含量也明显提高。将SP注入PAG加电针刺激增强上述效应的作用更为显著(P<0.01)。将SP注入PAG加腹腔注射吗啡增强上述效应的作用却未见进一步升高(P>0.05)。结果表明SP可能经PAG使脑内甲啡肽含量增加而发挥镇痛效应,SP注入PAG与电针具有协同作用。  相似文献

目的 探讨大鼠中缝大核(NRM)内神经降压素(NT)在电针镇痛中的作用,分析内源性NT和5-HT在电针镇痛中作用的相互关系。方法 以钾离子透入法引起大鼠甩尾反应的电流强度(mA)为痛反应指标,测定动物痛阈,观察中缝大核内神经降压素对痛和电针镇痛的影响。结果 大鼠中缝大核内微量注射神经降压素后,大鼠电针镇痛的效应明显增加;注入抗神经降压素血清或NT拮抗剂后,大鼠电针镇痛效应则明显降低。大鼠中缝大核内注入噻庚啶后,可阻断NT电针镇痛的效应。结论 大鼠中缝大核内的神经降压素在电针镇痛复杂病理生理过程中发挥着重要作用。至少其部分效应可能是通过内源性5-HT系统中介的。  相似文献

目的:观察低频脉冲电针对慢性神经病理痛大鼠背根神经节磷酸化环磷酸腺苷反应元件结合蛋白(p-CREB)表达的影响。方法:36只成年雄性SD大鼠,随机分为空白对照组、假手术组、模型对照组和电针治疗组,每组9只。模型对照组和电针治疗组采用右侧慢性坐骨神经结扎的方法,建立慢性神经病理痛模型。电针组大鼠给予低频电针刺激双侧"夹脊"穴和"足三里"穴,参数为波宽0.6 m s,频率2 Hz,电流强度分别为0.5、1和2 mA,各10 m in,共刺激30 m in。空白对照组、假手术组和模型对照组只陪同电针组抓取、固定,不做任何治疗。使用免疫印迹方法,观察各组大鼠右侧L4-6背根神经节p-CREB的表达变化。结果:模型对照组的热痛阈较空白对照组显著降低,并伴见p-CREB表达的显著增高(P〈0.01)。电针治疗组的热痛阈和p-CREB表达,较模型对照组均有显著变化(P〈0.05)。结论:低频脉冲电针的镇痛作用可能部分是通过抑制p-CREB的表达实现。电针可以通过改变初级传入神经元中基因的表达而对疼痛起到调控作用。  相似文献

Background Cryptotanshinone (CT) was originally isolated from the dried roots of Salvia militorrhiza, an herb that is used extensively in Asian medicine and the extracts of this herb have been used in the treatment of several pathologies, including cardiovascular diseases, hematological abnormalities, hepatitis, and hyperlipidemia, but no studies had been carried on the treatment for rheumatic diseases with it. This study aimed to investigate the effects of cryptotanshinone on immune functions in rats with adjuvant arthritis (AA). Methods Complete Freund's adjuvant was used to induce AA in rats. Thymus and spleen was aseptically taken from normal rats and the AA rats. Then a thymus lymphoid cell suspension, splenic lymphoid cell suspension and peritoneal macrophage cell suspension were prepared. After adding CT (0.1 ug/ml, 1.0 ug/ml, 10 ug/ml, 100 ug/ml, 1000 ug/ml) into the suspension, T and B lymphocytes proliferation was determined by 3-(4,5-2 dimethylthiazal-2yl)2,5- diphenyltetrazoliumbromide (MTT) assay. And the activities of interleukin-1 (IL-1) and IL-2 were measured by the mouse lymphocytes proliferation assay. Results Thymic T and splenic B lymphocyte proliferation of the AA rat was significantly lower, and could be stored through using CT in vitro. CT (100ug/ml and 1000ug/ml) increased T or B lymphocytes proliferation in vitro (P 〈0.01). In AA rats, the levels of IL-1 released by abdominal PMφ significantly increased whereas the level of IL-2 released by T cells decreased in vitro. CT (1000 pg/ml) decreased the production of IL-1 and promoted production of IL-2 in vitro (P 〈0.05). Conclusions CT can ameliorate the abnormal immunological functions in AA rats.  相似文献

目的:探讨电针(EA)对犬幽门扩约肌压力的调控作用,及其与血浆生长抑素(SS),一氧化氮(NO)的关系.方法:采用胃压测量仪及放免分析法、生化法同步监测电针不同穴位前后犬(n=5)幽门压力、频率,血浆中SS,NO的变化.结果:电针足三里穴后幽门扩约肌总压力、基础压(P<0.05)、频率下降(P<0.05),血浆SS含量下降(P<0.05),NO上升(P<0.01).电针上巨虚穴后幽门扩约肌总压力、基础压下降、血浆NO(P<0.05)含量上升,但变化趋势不如足三里组明显.其它组无显著变化.结论:电针可使犬幽门压力、频率下降,这可能为针刺调控胃肠功能的重要途径,这种作用与影响血浆中与幽门扩约肌压力相关的某些活性物质含量有关,并具有一定的经络和穴位特异性.  相似文献

目的 研究不同产妇性格分型对于针刺镇痛效果的影响.方法 将175例初产妇根据艾森克人格问卷神经质得分,分为稳定型性格、不稳定型性格;根据内外向得分,分为外向型性格、内向型性格;根据镇痛前后VAS评分比较,进行统计学分析,研究不同性格心理因素对针刺镇痛的影响.结果 性格稳定与否对镇痛效果以及产妇对镇痛满意度有显著影响.内外向性格对镇痛效果影响不显著,两者结合分析,将产妇分为外向稳定型、内向稳定型、外向不稳定型、内向不稳定型,镇痛效果由高到低依次为:内向稳定型、外向稳定型、外向不稳定型、内向不稳定型.结论 性格类型不同对针刺镇痛效果有显著性影响.测评产妇性格类型对于分娩镇痛方式的选择具有一定的指导意义.  相似文献

尼美舒利(Nime)0;6、3.0、15mg·kg~(-1)对佐剂性关节炎大鼠的原发性和继发性炎症反应均有明显的抑制作用,用药组继发性炎症反应的全身症状也明显减轻.而Nime对佐剂性关节炎大鼠低下的脾细胞增殖反应和腹腔巨噬细胞产生过高的IL-1活性却无明显影响,提示Nime对佐剂性关节炎大鼠无明显的免疫调节作用.本文结果表明,Nime对佐剂性关节炎大鼠有明显的治疗作用,这一作用可能与Nime有较强的抗炎作用有关.  相似文献

Effects and mechanisms of Cryptotanshinone on rats with adjuvant arthritis   总被引:1,自引:0,他引:1  
Background  Cryptotanshinone (CT) is the major active constituent of Salvia miltiorrhiza Bunge. The present study was carried out to investigate the effects of CT on rats with adjuvant arthritis (AA).
Methods  AA was induced by the metatarsal footpad injection with complete Freund’s adjuvant in male Sprague-Dawley rats. The secondary inflammatory reaction was evaluated by hind paw swelling and the polyarthritis index. Activity of interleukin-1 (IL-1) was detected by the concanavalin A-induced thymocytes proliferation assay. The lymphocytes proliferation and IL-2 production were assayed by 3-(4,5-2dimethylthiazal-2yl)2,5-diphenyltetrazoliumbromide (MTT) and activated mouse splenocytes proliferation, respectively.
Results  Intragastric administration of CT (50 and 100 mg/kg) significantly decreased secondary inflammatory reactions and increased the spleen and thymus index. There was a marked immunologic and inflammatory response in the AA model, which was accompanied by the decrease of thymocyte proliferation and IL-2 production as well as the increase of IL-1 production. CT apparently enhanced thymocyte proliferation and decreased IL-1 production in AA rats.
Conclusion  These results indicate that CT may exert its anti-inflammatory and immunoregulatory effects through inhibiting lymphocyte proliferation and production of pro-inflammatory mediators.

Objective To investigate the effects of oral administration of type Ⅱ collagen (CⅡ) on a djuvant arthritis (AA) in rats and its mechanisms, and to compare the effects of CⅡ with those of the Chinese traditional medicine Tripterygium Polyglycoside a dministered similarly.Methods Arthritis was induced in rats by immunization using Freund's complete adjuvant (FCA). After feeding rats either soluble CⅡ or Tripterygium Polyglycoside, chan ges in degree of articular swelling and articular histological findings were observed in AA rats. Some correlative immunological indexes were measured, includi ng delayed type hypersensitivity (DTH) reaction, anti-collagen and anti-Mycoba cterium tuberculosis (MT) antibody in serum, and levels of IFN-γ and TNF-α i n articular steep in rats.Results Oral administration of CⅡ was able to alleviate both distinctly articular and general symptoms in AA rats, suppress synovium hyperplasia and inflammatory cells infiltration in arthrosis capsule. The effects brought about by CⅡ were stronger than those by Tripterygium Polyglycoside. Oral administration of CⅡ inhibi ted antigen-specific immune response, such as DTH and antibody reaction to CⅡ . In addition, the expression of IFN-γ and TNF-α in joints were locally dow nregulated. Conclusions The therapeutic effect of oral administration of CⅡ is obvious on adjuvant art hritis in rats. Its remedial mechanisms are likely related to the downregulation of both IFN-γ and TNF-α, and the suppression of cell immunity.  相似文献

Objective:To study the effect of electro-acupuncture (EA) on pathomorphological changes of joints in rat model of chronic adjuvant arthritis.Methods: The rat chronic adjuvant arthritis model was established by subcutaneous injection of 0.1 ml of complete Freund’s adjuvant to the left hind sole. Forty Wistar rats were randomly divided into the model group, the low frequency (2 Hz) EA group, the high frequency EA (100 Hz) group and the body acupuncture group. After being modeled except the model group, the other three groups were treated with EA or body acupuncture in Yanglingquan points (bilateral) for 3 weeks, the left ankle joints and metatarsal joints of rats were taken for pathological examination by fixing with 10% formalin and embedding in paraffin, sectioning and staining with HE.Results: Obvious inflammatory cell infiltration, loosened synovial tissue, damage of articular cartilage and proliferation of synovial cells and granulation tissue were observed in the sections of joints in model rats. These pathological changes were significantly improved after treatment, and the effect in the high frequency EA group were significantly superior to that in the low frequency EA and body acupuncture group.Conclusion: High frequency EA could significantly improve the pathomorphological changes of joints in chronic adjuvant arthritis rat models.  相似文献

目的:探讨玉津药酒对大鼠佐剂性关节炎(AA)治疗效果及免疫学改变.方法:以完全弗氏佐剂诱导大鼠AA模型,灌胃服用玉津酒,观察AA大鼠病症改善情况及对免疫功能的影响.结果:在诱导AA模型前灌胃服用玉津酒,可推迟大鼠AA的发病时间,降低发病率,减轻关节炎症状;在大鼠发生AA后服用玉津酒,可明显抑制关节炎症程度;服用玉津酒可抑制AA大鼠T、B淋巴细胞增殖反应及IL-1活性.结论:玉津酒是通过机体免疫调节达到对AA的预防和治疗作用.  相似文献
