In the present study, the effets of VIP on the growth of two human pancreatic carcinoma cell lines PU-PAH-1 and PANC-1 were determined using tritiated thymidine incorporation, VIP receptors, intracellular cAMP and polyamings were investlsa.ted, The results indicated that VIP at a concentcation of 10^-8mol/L to 10^-7 mol/L can significantly Stimulate the growth of PU-PAN-1 cells but not PANC-1 cells, This effect is dose-dependent and abolished by VIP receptor antagonist, [4-CI-Phe^6 , Leu^7] VIP, suggesting VIP receptors in PU-PAN-1 cells maymediate this effect. VIP can markedly elevate the levels of intracellular cAMP and polyammes in PU-PAN-1 cells,indicating that the growth-promoting effect stimulated by VIP may be via a rapid increase in the biosynth~es of cAMP and polyamines. In addition, the VIP-antibody ir2Libited the growth of PU-PAN-1 cells in serum-free culture medium. The results above suggested that VIP has an autoctine regulatory effect on this pancreatic carcinoma cell line(PU-PAN-1).  相似文献

Background and purpose: Vasoactive intestinal peptide (VIP) appears to play an important role as a neurotransmitter or neuromediater in ischemic cerebrovascular diseases (ICVD). The effect of acupuncture, which is used in treatment of ICVD with good efficiency, on VIP has not been known. For finding the mechanism of acupuncture in treatment of ICVD and the effect of electro-acupuncture on VIP, the present study was performed. Materials and methods: 59 patients with acute ICVD were randomly divided into two groups. Electro-acupuncture and routine treatment were given in Group 1 (n= 29), and routine treatment was used alone in Group 2 (n= 30). The cerebrospinal fluid (CSF) and blood were taken before the beginning of treatment and after a course of treatment in both groups. The control group consisted of 38 cases of non-ICVD. VIP was measured by radioimmunoassay. Results: The level of CSF VIP in patients with acute ICVD was significantly lower than that in the controls, while the levels of plamsa VIP showed no significant difference between the ICVD and control groups, and the level of CSF VIP was not significantly correlated with the level of plasma VIP. After acupuncture treatment, the level of CSF VIP was increased and showed no significant difference as compared with the control group. Conclusion: Acupuncture might alleviate the disturbance of metabolism of VIP in CNS.  相似文献

The levels of vasoactive intestinal peptide(VIP)in three regions of rat brain were assayed in 62 rats.Bilateral common carotid artery ligation was donein 50 rats.Half an hour before ligation 26 rats weregiven 10 g/kg of Radix Salviae Miltiorrhizae(RSM);24 rats were given same volume of normalsaline as controls.A sham operation wasdone in 12 rats.Half an hour(n=30)and 3hours(n=32)after operation,the rats were quicklydecapitated.VIP levels were assayed in cerebralcortex,hippocampus and caudate nucleus,in salin-treated animals,VIP levels of cerebral cortex andcaudate nucleus at 3 hour group were significantlydecreased compared with the sham-operated group.No significant difference was found between RSM-treated and sham-operated groups.The preliminaryresults suggest that VIP may be involved in thepathophysiological procedures of cerebral ischemiaand RSM may attenuate the dysfunction of VIPduring cerebral ischemia.  相似文献

本文研究乙双吗啉(AT1727)和γ射线联合应用对动物小肠上皮细胞的作用。腹腔注射AT1727 160mg/kg,在放射前30min给药时,其斜率和单纯放射组相仿,DMF值为1.09;放射后30min给药时,D_o值增加1.3倍,DMF值为1.05。在使用相等分割剂量做分割放射时,显示出他们之间明显的相互作用。结果显示:联合应用增加了辐射生物效应,且和所用药物浓度有关,和二者之间应用次序和相隔时间也有关。  相似文献

本文收集广州地区6个月胎儿至6岁儿童新鲜大脑标本63例(126侧),其中5例作脑血管有机玻璃单体铸型,35例用碱性磷酸酶染色(包括7例墨汁灌注后复染),其余用于墨汁灌注法,在光镜暗视野全景放大观察和日立S-450扫描电镜观察,较完整地显示人脑海马内部血管动脉-微动脉-毛细血管前括约肌-毛细血管-微静脉-小静脉连续性立体构筑。此外还观察到微血管形态特征及微血管间存在多种吻合。  相似文献

目的:观察血管活性肠肽(vasoactive intestinal peptide,VIP)对离体逼尿肌肌条自发性收缩频率及收缩幅度的作用。方法:建立Wistar大鼠下尿路梗阻动物模型,6周后行充盈性膀胱测压,根据逼尿肌稳定与否将下尿路梗阻动物分为逼尿肌稳定组和逼尿肌不稳定组,用拉力传感器观察VIP对离体逼尿肌肌条的自发性收缩频率及收缩幅度的影响。结果:大鼠下尿路梗阻6周后膀胱逼尿肌不稳定发生率为62.1%,VIP能明显抑制逼尿肌的自发性收缩,降低逼尿肌肌条自发性收缩频率及收缩幅度(P<0.05),但梗阻后逼尿肌不稳定组,逼尿肌稳定组及对照组之间差异无显著性意义(P>0.05),结论:VIP能显著抑制正常和梗阻后逼尿肌自发性收缩,但其在逼尿肌不稳定发生中的作用仍需深入研究。  相似文献

观察重组人白细胞介素2(γ-IL2)体外诱导的腹腔巨噬细胞,在YPC3抗人胰腺癌单克隆抗体(YPC3mAb)介导下,通过抗体依赖性细胞介导的细胞毒性(ADCC)机制对胰腺癌细胞的杀伤作用。4h51Cr释放实验表明,在YPC3mAb作用下,活化腹腔巨噬细胞对Capan-2人胰腺癌细胞的杀伤活力明显增强,比单用巨噬细胞杀伤活力约提高70.0%,而对照1-F/7抗登革热病毒单抗无明显影响。γ-IL2作用的腹腔巨噬细胞和脾淋巴细胞具有类似的ADCC杀伤效应。结果提示,γ-IL2和YPC3mAb联合腹腔注射,可能为胰腺癌的局部治疗提供一个新方法。  相似文献

以免疫细胞化学与图像处理结果分析发现,针刺大鼠“足三里”不仅义叉上核中的血管活性肠肽免疫反应阳性神经元胞体数目显著增加,而且血管活性肠肽免疫反应性也显著增加。提示电针“足三里”所致的交叉上核血管活性肠肽免疫反应性的改变可能参与调节机体神经内分泌活动的昼夜节律。  相似文献

目的:探讨-氨基丁酸(GABA)在大鼠肠神经系统中与乙酰胆碱、一氧化氮、血管活性肠肽的共存关系。方法:应用免疫组织化学和免疫荧光双重染色技术对大鼠肠神经系统内谷氨酸脱羧酶(GAD)与乙酰胆碱酯酶(AchE)、一氧化氮合酶(NOS)、血管活性肠肽(VIP)的共存关系进行观察。结果:GAD样阳性神经元主要位于肌间神经丛,与AchE、NOS、VIP存在共存关系。双重标记神经元分别占GAD阳性神经元的5.6%、62.6%和51.1%,而占AchE、NOS、VIP阳性神经元的3.4%、19.8%和43.5%。结论:在肠神经系统中GABA可能通过调节其它神经递质的释放而发挥着间接的调控作用。  相似文献

应用免疫组织化学技术对48例大肠癌标本进行了雌激素受体(ER)检测。结果发现ER的阳性率为37.5%。48例大肠癌按组织学分型,ER阳性率分别为:高分化腺癌组52.3%;中分化腺癌组38.9%;低分化腺癌组11.1%。高分化腺癌与低分化腺癌组比较,ER阳性率有显著差别(p〈0.01)。48例大肠癌标本按部位、性别、年龄分组,19例结肠癌中,ER阳性率42.1%,29例直肠癌中,ER阳性率34.5%  相似文献
