Aim of the study

Sceptridium ternatum Lyon (ST), a common Chinese herb, has been used in treatment of allergic asthma and whooping cough. In the present study, we investigated the Th1/Th2 ratio of peripheral blood and mRNA levels of leukotriene receptors after the treatment of ST in allergic asthma mouse model.

Materials and methods

Mouse asthma model was developed by ovalbumin (OVA) sensitization followed by the inhalation of aerosol allergen. Montelukast (10 mg/kg), as a positive control drug, and ST were administrated six days before the OVA sensitization for ten days. Airway responsiveness was evaluated by the Medlab 12.0 biological signal processing system. The ratio of Th1/Th2 cells was determined by flow cytometry. The expression level of Cyslt1 was measured by PCR. Pathological changes of lung tissues were examined by H&E staining.

Results

ST significantly reduced the airway responsiveness, elevated the ratio of Th1/Th2, and decreased Cyslt1 mRNA level in a dose-dependent manner. High-dose ST distinctly prevented the pathological changes of lung tissues.

Conclusion

High-dose ST had the same efficacy as Montelukast in a mouse asthma model, and ST could be a potential anti-asthmatic agent.  相似文献

雾化吸入银杏叶总黄酮对哮喘小鼠Th1/Th2失衡的影响   总被引:1,自引:0,他引:1  
目的:研究银杏叶总黄酮(total ginkgo flavone glycosides, TFG)对哮喘小鼠辅助性T细胞1/辅助性T细胞2(Th1/Th2)失衡的影响。方法:48只BALB/c小鼠随机分为正常组、哮喘组、TGF雾化吸入组以及地塞米松组,以卵蛋白(OVA)、氢氧化铝免疫系统致敏和局部激发的方法建立哮喘模型。自实验第21天起给予TFG治疗组连续雾化吸入50 g·L-1 TFG溶液30 min,地塞米松组腹腔注射地塞米松1 mg·kg -1,连续10 d。第32天酶联免疫吸附实验(enzyme-linked immunosorbent assay, ELISA)法测定肺细胞灌洗液(bronchalveolar lavage fluid, BALF)中细胞因子γ-干扰素(interferon-γ, IFN-γ)、白介素-4(interleukin-4, IL-4)和血清中总免疫球蛋白E(immunoglobulin E, IgE)含量,观察各组肺组织的病理变化,免疫组织化学染色技术检测肺组织中GATA连接蛋白-3(GATA binding protein-3, GATA-3)的表达情况。结果:与模型组比较,TFG组BALF中IL-4含量(49.30±7.95) ng·L-1降低(P<0.01)、IFN-γ水平升高(49.08±5.46) ng·L-1(P<0.01)、血浆总IgE水平降低(9.47±1.52) μg·L-1(P<0.01);模型组支气管平滑肌肥厚,黏膜充血、水肿,管腔内可见黏液栓,并有大量炎性细胞浸润;而TFG组和地塞米松组小鼠的上述炎症性改变明显减轻;模型组GATA-3表达显著,TFG治疗组则明显减轻。结论:雾化吸入TFG对哮喘小鼠有明显的治疗作用,其机制可能与阻断GATA-3的表达、恢复Th1/Th2平衡有关。  相似文献

目的探讨小青龙颗粒对支气管哮喘患者外周血Th17/Treg平衡的影响。方法将60例支气管哮喘急性发作期患者随机分为2组:治疗组给予小青龙汤治疗,对照组给予西医常规治疗组,治疗7 d后评定疗效。治疗前后采集标本进行Th17细胞、Treg细胞检测。结果 2组治疗后外周血Th17细胞百分率和Th17/Treg细胞的比值均下降,Treg细胞的百分率均上升;治疗后治疗组外周血IL-17细胞百分率、IL-17/TGF-β1的比值均低于对照组,TGF-β1细胞百分率高于对照组。结论小青龙颗粒可有效调控Th17/Treg失衡,在支气管哮喘发作期治疗和调节免疫上有应用价值。  相似文献

Ligustrazine is an alkaloid isolated from the rhizome of Chuanxiong (Ligusticum chuanxiong Hort), which is known to possess antioxidant, anti-inflammatory, anti-fibrosis and immunomodulative effects. It is used clinically to treat asthma as an assistant therapy of glucocorticoid. The purpose of this study was to explore the effects of intraperitoneal ligustrazine on Th1/Th2 cytokines in a rat asthma model and the underlying mechanism. SD rats were sensitized and challenged with ovalbumin (OVA) to establish an asthmatic model. Within 24 hours after the last ovalbumin challenge, changes in airway histology were observed. The concentrations of IL-4 and IFN-gamma in bronchoalveolar lavage fluid (BALF) were measured by enzyme linked immunosorbent assay (ELISA). The protein expressions of GATA-3 and T-bet in lung were measured by Western blot. The results showed that an increase of Th2 cytokine and an inhibition of Th1 cytokine were accompanied by an increased expression of GATA-3 protein and a decreased expression of T-bet protein in rat asthmatic airways compared to those in normal control group. Intraperitoneal ligustrazine administration could significantly lower the level of IL-4 in BALF and the expression of GATA-3 protein in lung and also increase the level of IFN-gamma and T-bet in asthmatic rats, resulting in a decreased percentage of eosinophils (EOS) in BALF and ameliorated airway inflammatory cell infiltration. In conclusion, ligustrazine inhibits OVA induced airway inflammation by modulating key master switches GATA-3 and T-bet that result in reversing the Th2 cytokine patterns in asthma.  相似文献

目的:探讨小青龙汤联合阿奇霉素治疗喘息性支气管炎的临床疗效及对外周血辅助性T细胞(Th1/Th2)表达和肺功能的影响。方法:选取156例喘息性支气管炎患儿为研究对象。随机分为对照组和观察组,各78例。对照组给予阿奇霉素片治疗,观察组在对照组基础上给予小青龙汤治疗。结果:治疗后,两组中医证候积分均明显下降,观察组比对照组更明显(P<0.05)。治疗后,两组患儿IFN-γ水平明显降低,IL-10、FEV1及MVV指标水平均明显升高(P<0.05);观察组治疗后IFN-γ水平低于对照组,IL-10、FEV1及MVV指标水平均高于对照组(P<0.05);两组治疗期间均未出现明显不良反应,观察组总有效率94.87%(74/78)明显高于对照组76.92%(60/78)(P<0.05)。治疗后观察组生活质量评分显著高于对照组(P<0.05)。结论 :小青龙汤联合阿奇霉素可显著改善喘息性支气管炎患儿外周血IFN-γ、IL-10因子表达,提高肺功能,临床疗效显著且安全。  相似文献

OBJECTIVE: To investigate the effect of thorascopic administration of ginseng polysaccharides(GPS)plus dendritic cells(DC) on T helper cell type 1/T helper cell type 2(Th1/Th2) balance in patients with non-small cell lung cancer(NSCLC).METHODS: A total of 96 NSCLC patients were divided evenly into two groups. The control group was treated with DCs alone and the treatment group was treated with DCs plus GPS. After DCs and GPS were administered thoracoscopically, once a week,4 times for 30 days, the patients' quality of life was measured with the Functional Assessment of Can-cer Treatment-Lung(FACT-L) questionnaire before and after treatment. Serum interferon-γ(INF-γ), interleukin-4(IL-4), IL-2 and IL-5 were examined before and after treatments.RESULTS: The level of Th1 cytokines(INF-γ, IL-2)and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) increased in both treatment groups, while Th2cytokines(IL-4, IL-5) and FACT-L scores decreased(P〈0.01). Furthermore, after treatment Th1 cytokines(INF-γ, IL-2) and the ratio of Th1/Th2 cytokines(INF-γ/IL-4, IL-2/IL-5) were higher in the DCs +GPS group than in the control group(P〈0.05). Conversely, FACT-L scores and Th2 cytokines(IL-4, IL-5)were higher in the control group than in the DCs +GPS group(P〈0.05).CONCLUSION: The treatment regime of DCs plus GPS had a greater effect on NSCLC patients' immune function as compared with DCs alone. This was evident by increased expression of Th1 cytokines(INF-γ, IL-2) and the ratio of Th1/Th2(INF-γ/IL-4, IL-2/IL-5), as well as by decreased FACT-L scores and the expression of Th2 cytokines(IL-4,IL-5).  相似文献

目的:观察化浊解毒消痈方对溃疡性结肠炎大鼠Th1/Th2平衡的影响,以探讨其作用机制。方法:将Wistar大鼠随机分为正常组、模型组、阳性对照柳氮磺胺吡啶组、化浊解毒消痈全方组、化浊方组、解毒方组、每组10只。以三硝基苯磺酸/乙醇法建立溃疡性结肠炎大鼠动物模型,造模成功后分别予化浊解毒消痈方31.8g/kg、化浊方19.3g/kg、解毒方19.3g/kg灌胃,SASP组予柳氮磺胺吡啶(0.5 g/kg)灌胃,模型组及正常组每日以蒸馏水10ml/kg灌胃,均连续14d。观察各组大鼠一般情况及疾病活动指数(DAI)评分、组织病理学、SOD含量、血清及结肠组织IFN-γ、IL-4、IFN-γ/IL-4变化情况。结果:化浊解毒消痈全方(31.8g/kg)、化浊方(19.3g/kg)、解毒方(19.3g/kg)和柳氮磺胺吡啶肠溶片(0.5g/kg)可改善大鼠的一般情况,降低DAI评分,提高SOD含量,同时能降低血液及结肠组织中IFN-γ水平,提高IL-4水平,降低IFN-γ/IL-4比值,其中以化浊解毒消痈全方组(31.8g/kg)作用效果更为显著。结论 :化浊解毒消痈方药能显著改善造模大鼠症状,通过下调Th1细胞因子分泌和上调Th2细胞因子水平,恢复Th1/Th2平衡,发挥抗炎治疗作用。  相似文献

目的:本研究通过玉屏风散治疗对卵白蛋白(OVA)致敏和激发的过敏性鼻炎小鼠模型,了解其对Th1和Th2及其平衡的影响。方法:8周龄Balb/c小鼠经OVA全身和鼻部局部致敏激发制作过敏性鼻炎小鼠动物模型,早期干预组和治疗组分别给予玉屏风散提取物2,500 mg/kg,7天后采用流式细胞术分别检测脾脏Th1和Th2细胞以及鼻部病理改变。结果:动物模型组Th2细胞表达显著增高(9.86±1.40);玉屏风散治疗组Th2表达显著下降(3.41±0.72,P<0.01);中药干预组也可保护OVA诱导的Th2细胞早期表达增高(6.27±1.21,P<0.05)。本研究表明玉屏风散可抑制Th2介导的上气道非特异性炎症反应(F=30.53,P<0.0001)。结论:本研究证明玉屏风散对Th2介导的上气道非特异性炎症反应有治疗作用,该方剂能提高Th1/Th2比值和抑制Th2细胞的过度表达。  相似文献

目的:研究六味补气方对慢性阻塞性肺疾病(COPD)模型大鼠气道炎症及免疫功能的影响及其作用机制。方法:将SD大鼠按随机数字表法分为正常对照组、模型组、桂龙咳喘宁组和六味补气方组,每组10只。除正常对照组外,其余各组采用烟熏加脂多糖(LPS)气管滴入方法建立COPD实验动物模型,造模成功后,分别给予0.9%氯化钠溶液及相应药物灌胃,连续干预14 d。于造模第4周和药物干预结束后检测肺功能; 苏木精-伊红(HE)染色法光镜观察肺组织的病理学改变; 酶联免疫吸附法(ELISA)检测血清及肺泡灌洗液(BALF)中白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)水平,并分析辅助性T细胞Th1/Th2平衡状态。结果:与正常对照组比较,模型组大鼠出现COPD症状和体征,肺功能降低,血清及BALF中IFN-γ水平明显下降,IL-4水平明显增加(均P<0.05),Th1/Th2平衡向Th2方向移动; 与模型组相比,六味补气方组和桂龙咳喘宁组大鼠一般情况及肺功能均有不同程度改善,血清及BALF中IFN-γ水平明显增加,IL-4水平下降(均P<0.05); 与桂龙咳喘宁组相比,六味补气方组大鼠COPD症状体征减轻,血清及BALF中IFN-γ水平明显增加,IL-4水平下降(均P<0.05),Th1/Th2平衡向Th1方向移动。结论:六味补气方可改善COPD模型大鼠肺功能,减轻气道炎症,其机制可能是通过调节Th1/Th2类细胞平衡,增加免疫功能实现的。  相似文献

目的:从基因及蛋白水平探讨中药槐耳调节T辅助淋巴细胞(Th)平衡的机制,为槐耳在临床上治疗缓解期哮喘提供理论依据。方法:将哮喘缓解期儿童外周血单个核细胞(PBMC)分为3组,即空白组、中剂量组和大剂量组,分别与不同浓度的槐耳菌质配液(6g/L)及培养液作用48h;然后测定细胞培养沉淀中白细胞介素-4(IL-4)及干扰素-γ(IFN-γ)mRNA的表达和细胞培养上清中IL-4及IFN-γ的量。结果:在中剂量组和大剂量组中,IL-4mRNA表达明显低于空白组(P<0.05);IFN-γ表达明显高于空白组(P<0.05);IFN-γ/IL-4比值的基因及蛋白水平表达均高于空白组;然而,对于以上4个指标的检测,大剂量组与中剂量组之间无明显差异(P>0.05)。结论:槐耳菌质能够明显增加IFN-γ及减少IL-4mRNA的表达,可调节哮喘儿童缓解期IFN-γ/IL-4比值的失衡,平衡Thl/Th2比值的漂移。  相似文献

AIM: The aim of the study was to investigate the effect and mechanism of action of synthetic salidroside in an ovalbumin (OVA)-induced asthma model in mice. METHOD: BALB/c mice were sensitized with an intraperitoneal injection of ovalbumin (OVA) to induce a mouse model of asthma in paracmasis. The mice were treated with dexamethasone as the positive control. At the end of the study, respiratory reactivity was detected, the numbers of various kinds of white blood cells in the bronchoalveolar lavage fluid (BALF) were counted, and the levels of IL-4 and INF-γ in BALF were determined. Quantitative PCR was used to detect the mRNA contents of IL-4 and INF-γ in lung tissue. Histologic examination was performed to observe inflammatory cellular infiltration. RESULTS: Salidroside treatment virtually eliminated airway hyper-reactivity, markedly reduced the eosinophil percent, obviously reduced the levels of IL-4 and raised INF-γ in the bronchoalveolar lavage fluid (BALF) compared with the sham-treated group. Quantitative PCR on the mRNA content of IL-4 and INF-γ provided confirmation. Lung histologic observations showed that salidroside reduced inflammation and edema. These effects were equivalent to the effects of dexamethasone. CONCLUSION: Synthetic salidroside exhibits an anti-asthma effect which is related to the regulation of Th1/Th2 balance. This provides a new possibility for treatment of allergic asthma.  相似文献

目的观察补肺平喘汤对支气管哮喘大鼠Th1/Th2细胞因子失衡及JAK/STAT信号通路影响。方法将50只健康大鼠随机分为正常组,模型组,中药低剂量组,中药中剂量组,中药高剂量组5组,每组10只。除正常组外,其余各组给予卵白蛋白和氢氧化铝免疫反应制备支气管哮喘大鼠模型。造模4周后,中药低、中、高剂量组分别按照生药量10.8,21.6,32.4 g/kg进行灌胃给药,正常组和模型组给予等量生理盐水灌胃,灌胃4周后,分离大鼠血清和肺组织,Elisa法检测大鼠血清IFN-γ、IL-4水平变化,RT-PCR法检测大鼠肺组织JAK/STAT信号通路变化。结果与正常组比较,模型组大鼠血清IL-4水平显著升高(P0.01),而IFN-γ呈显著下降趋势(P0.01),IL-4/IFN-γ提示免疫调节显著向Th2水平失衡,肺组织JAK1、STAT6 mRNA水平明显升高;与模型组比较,中药中、高剂量组大鼠血清IL-4水平显著降低(P0.05,P0.01),IFN-γ水平显著升高(P0.05),免疫调节明显向Th1方向调节(P0.01),肺组织JAK1、STAT6 mRNA水平明显降低(P0.05,P0.01)。结论补肺平喘汤能有效改善支气管哮喘大鼠Th1/Th2细胞因子失衡,降低JAK1、STAT6 mRNA表达,调节Th1/Th2细胞因子平衡。  相似文献

目的:建立单纯型和肾阳虚复合型2种不同变应性鼻炎模型,探讨小青龙汤对2种变应性鼻炎模型Th1/Th2失衡作用的异同。方法:将105只雄性豚鼠随机分7组,每组15只:空白对照组(A组),单纯模型对照组(B1组),小青龙汤干预单纯模型组(B2组),息斯敏干预单纯模型组(B3组),肾阳虚复合模型对照组(C1组),小青龙汤干预肾阳虚复合模型组(C2组),息斯敏干预肾阳虚复合模型组(C3组)。B1、B2、B3组进行单纯变应性鼻炎造模;C1、C2、C3组在单纯变应性鼻炎的模型上进行肾阳虚复合模型造模。造模成功后,模型组进行用药观察,记录各组局部过敏反应;ELISA法(酶联免疫吸附试验法)检测血清中IFN-γ、IL-4、IL-2和IL-10等细胞因子含量;荧光定量PCR法测脾组织中IFN-γmRNA、IL-4mRNA、T-bet mRNA、GATA-3 mRNA等转录因子的相对含量。结果:变应性鼻炎单纯模型和肾阳虚复合模型均出现IFN-γ降低(P0.01),IL-4升高(P0.01),IFN-γ/IL-4比值降低(P0.01),IFN-γmRNA降低(P0.01),IL-4 mRNA升高(P0.01),IFN-γmRNA/IL-4 mRNA比值降低(P0.01),T-bet mRNA降低(P0.01),但可使GATA-3 mRNA升高(P0.01),因而T-bet mRNA/GATA-3 mRNA比值也相对降低(P0.01)。小青龙汤可使单纯模型IFN-γ升高(P0.01),IL-4降低(P0.01),IFN-γ/IL-4比值升高(P0.01);升高IFN-γmRNA(P0.01),降低IL-4 mRNA(P0.01),使IFN-γmRNA/IL-4 mRNA比值升高(P0.01);使T-bet mRNA升高(P0.01),但可使GATA-3 mRNA降低(P0.01),从而使T-bet mRNA/GATA-3 mRNA比值也相对升高(P0.01)。小青龙汤可降低肾阳虚复合模型中IL-4的含量(P0.01),对IFN-γ作用不明显,升高IFN-γ/IL-4比值(P0.01);可使IFN-γmRNA升高(P0.01),但对IL-4 mRNA变化不大,IFN-γmRNA/IL-4 mRNA比值升高(P0.01);对T-bet mRNA作用不明显,但可使GATA-3 mRNA降低(P0.05),因而T-bet mRNA/GATA-3 mRNA比值也相对升高(P0.01)。结论:小青龙通过调节IFN-γ/IL-4比值、IFN-γmRNA/IL-4 mRNA比值和T-bet mRNA/GATA-3 mRNA比值,来调节Th1/Th2平衡,对单纯变应性鼻炎模型的过敏反应的改善更为明显。  相似文献

Ninjin-youei-to (NYT; ren-shen-yang-rong-tang in Chinese) is a traditional herbal formula, which is widely used in Japan, Korea and China to modulate physiological immunity. The effects of oral administration of NYT on cytokine production from splenocytes were investigated in both C57BL/6 and BALB/c mice in which Th1 and Th2 were dominant, respectively. Splenocytes from C57BL/6 and BALB/c mice, which took NYT orally forfour weeks, were cultured with anti-mouse CD3 mAb, and the supernatant was examined for cytokine production using enzyme-linked immunosorbent assay (ELISA). Administration of NYT to C57BL/6 mice, increased the production of interleukin-4 (IL-4) significantly, and slightly decreased interferon-y (IFN-gamma) production from splenocytes. In contrast, the same treatment significantly increased IFN-gamma secretion from splenocytes of BALB/c mice. No remarkable changes of IL-12 production from splenocytes were observed in either strain of mice. These results suggest that oral administration of NYT ameliorates the excessive inclination of Th1 and Th2 type cytokine production, and NYT may provide a beneficial effects for the treatment of diseases caused by a skewed Th1-Th2 balance in the immune system.  相似文献

[目的]观察银翘散对呼吸道病毒感染相关性哮喘(RVA)的临床疗效及对Th1/Th2细胞因子的影响。[方法]将68例RVA患者随机分为对照组32例,常规西药治疗;治疗组36例,在常规西药的基础上加用银翘散治疗。2组均治疗10 d。观察治疗前后患者的哮喘控制评分(ACT),肺功能[第1秒时间肺活量(FEV1)、一秒率(FEV1/FVC%)、呼气峰流速(PEF)]、中医证候疗效及Th1/Th2细胞因子[分泌性免疫球蛋白A(s Ig A)、白介素-4(IL-4)、γ干扰素(IFN-γ)、IFN-γ/IL-4]的变化。[结果]治疗后两组患者的ACT评分均较治疗前明显改善,且治疗组优于对照组(P0.05);治疗组对肺功能的改善优于对照组(P0.05)。治疗组中医证候有效率为94.44%(34/36),对照组中医证候有效率为71.87%(23/32)。治疗后,治疗组s Ig A水平高于对照组(P0.05)。IFN-γ及IFN-γ/IL-4低于对照组(P0.05),IL-4水平两组之间未见差异(P0.05)。[结论]银翘散联合常规西药治疗RVA的临床疗效优于单用常规西药治疗,可能与银翘散可调节患者Th1/Th2细胞因子,逆转因子间的失衡,进而减少气道炎症相关。  相似文献
