目的观察不同时问经皮穴位电刺激(TEAS)抗大鼠运动性疲劳作用以及对海马和下丘脑形态和5-羟色胺(5HT)含量的影响。方法采用7周递增负荷跑台法建立大鼠慢性运动性疲劳模型,第6周开始,分别在运动后即刻和3h后TEAS治疗,治疗方法采用经皮神经电刺激仪刺激大鼠足三里穴,治疗持续2周。HE染色观察大鼠海马、下丘脑组织形态学变化,免疫组化法观察海马、下丘脑5-HT的表达。结果TEAS可以明显减轻运动引起的大鼠海马、下丘脑组织形态损伤,降低大鼠海马、下丘脑5-HT表达,运动后即刻和3h后治疗的两治疗组间无显著差异。结论TEAS有促进疲劳恢复提高运动能力的作用。TEAS改善大鼠的运动性疲劳状态与降低中枢5HT表达、减少神经细胞损伤有关。  相似文献

目的:探讨电针足三里预防运动性疲劳的中枢5-HT机制。方法:应用跑台建立大鼠急性运动性疲劳模型,电针足三里于运动前介入,每日1次,于第6天,分别于运动后即刻和运动后3h处死大鼠,检测血浆乳酸(LAC)水平和海马、下丘脑5-HT及其代谢产物含量变化。结果:运动后即刻时,电针可下调疲劳大鼠血浆乳酸水平、显著降低海马5-HT和5-羟吲哚乙酸(5-HIAA)含量;运动后3h时,电针显著降低血浆LAC水平以及海马、下丘脑5-HT和5-HIAA含量。结论:降低海马、下丘脑5-HT水平并促进其代谢可能是电针预防运动性疲劳和促进疲劳恢复的中枢机制之一。  相似文献

目的:通过对运动性疲劳模型大鼠β-EP、血清BLA、下丘脑5-HT及力竭游泳时间变化的研究,探讨在不同时段针刺气海、后溪、承山穴对提高运动能力和消除运动性疲劳的作用。方法:通过建立运动性疲劳模型,观察SD大鼠β-EP及血清BLA、下丘脑5-HT的变化及电针的治疗作用。结果:不同时段的电针疗法可以明显降低运动性疲劳大鼠血浆β-EP和血清BLA、下丘脑5-HT的含量,提高下丘脑β-EP含量,且能明显延长大鼠的力竭游泳时间。结论:基本证实在力竭运动前后给予电针治疗对提高运动能力和消除运动疲劳有一定作用。  相似文献

目的 :从基因转录水平阐明运动性疲劳产生及药物调节的机理。方法 :采用生物素标记阿黑皮素原 (POMC)cRNA探针 (Bio cRNA)原位杂交法结合图像分析 ,观察中药复方体复康对运动性疲劳大鼠不同脑区POMCmRNA表达。结果 :对于慢性运动应激 ,体复康能增强POMC在海马CA1 区、下丘脑部位的基因表达 ,而能降低POMC在额叶皮层的基因表达。长时期 (共 7周 )大强度的跑台运动 (速度由 1 5m/min递增至 3 5m/min ,运动时间为每天 2 0~2 5min)的大鼠 ,末次运动后POMC基因表达在额叶皮层、海马CA1 下降 (前者至运动结束后 3h仍未恢复 ,后者至运动结束后 3 0min即恢复 ) ,在下丘脑则增强 ;体复康能增强末次运动后POMC在额叶皮层、海马CA1 区、下丘脑等脑区的基因表达 ,尤其是在运动结束后 3h最为显著。结论 :POMCmRNA表达在运动性疲劳大鼠不同脑区呈现不同的动态变化趋势 ,体复康对其POMCmRNA表达有重要动态调节作用。  相似文献

目的:观察逆电针“足三里”穴对急性跑台运动大鼠的抗疲劳效应及其中枢机制.方法:将清洁级雄性SD大鼠50只随机分为安静组(n=10)、模型组(n=20)和逆电针组(n=20),后两组又分为运动后即刻、运动后3h组(各10只).后两组大鼠完成适应性跑台运动后,接受急性跑台运动;逆电针组大鼠于跑台运动前先介入电针治疗(参数为双侧“足三里”穴、连续波、频率2 Hz、强度2mA、时间30 min、1次/d、连续6 d).分别于运动后即刻和运动后3h处死大鼠,检测血浆乳酸水平、下丘脑和纹状体多巴胺(DA)、5-羟色按(5-HT)含量变化,并计算DA/5-HT比值.结果:与安静组相比,模型组大鼠血浆乳酸和下丘脑5-HT水平呈升高趋势,下丘脑DA含量则显著升高(P<0.01);而纹状体DA和5-HT含量于运动后3h时显著降低(均P<0.01).运动后即刻时,与同期模型组相比,逆电针组大鼠下丘脑DA和5-HT含量显著升高(均P<0.01),而纹状体则明显下降(均P<0.01);运动后3h时,与同期模型组相比,逆电针组大鼠血浆乳酸水平、下丘脑5-HT含量均显著降低(P<0.05,P<0.01),而下丘脑DA/5-HT比值则明显升高(P<0.01).结论:逆电针“足三里”穴可促进急性跑台运动大鼠疲劳恢复,可能是通过降低跑台运动大鼠血浆乳酸堆积,提高下丘脑DA/5-HT比值发挥作用.  相似文献

目的观察人参皂甙Re和人参总皂甙对运动性疲劳模型大鼠中枢神经逆质乙酰胆碱(Ach)、多巴胺(DA)、5-羟色胺(5-HT)和γ-氨基丁酸(GABA)含量的影响。方法将40只雄性SD大鼠随机均分为人参皂甙Re组、人参总皂甙组、模型组和空白组,给药组和模型组大鼠在灌胃1h后进行中等运动强度的水平跑台运动,每天1次.连续14d。末次运动后即刻处死大鼠,迅速取下丘脑,色谱法测Ach、DA、5-HT和GABA含量。结果与模型组比较.人参皂甙Re和人参总皂甙组大鼠下丘脑Ach和DA的含量显著性升高,而5-HT和GABA的含量有显著性降低;与人参皂甙Re组相比较,人参总皂甙组大鼠下丘脑Ach、5-HT和GABA有显著性降低,而DA含量有显著性升高。结论人参皂甙Re及人参总皂甙均可对下丘脑神经递质含量产生影响,从而达到抗运动性疲劳效应,但人参总皂甙的效果更优,尤其能更好地改善运动性疲劳下中枢保护性抑制的状态。  相似文献

目的:观察抑郁模型大鼠海马及下丘脑五羟色胺(5-HT)的表达及眼针干预的影响。方法:雄性Wistar大鼠分为正常组、抑郁模型组和眼针组3组。采用慢性应激法建立抑郁模型并进行眼针干预。采用ELISA测定血清、海马及下丘脑的5-HT含量;采用免疫组化法测定海马及下丘脑5-HT表达。结果:与正常组相比,抑郁模型组海马中5-HT含量显著降低(P<0.01),血浆中5-HT含量显著升高(P<0.01);与抑郁模型组相比,眼针组海马和下丘脑5-HT含量显著升高(P<0.01),血浆5-HT含量显著降低(P<0.01)。结论:眼针能够调控抑郁模型大鼠海马和下丘脑5-HT的表达。  相似文献

目的:观察柴芍六君子汤对功能性消化不良(functional dyspepsia,FD)大鼠海马、下丘脑、胃组织及血清中五羟色胺表达的影响,探讨柴芍六君子汤治疗功能性消化不良的作用机制。方法:将60只SD大鼠随机分成空白对照组、模型组、多潘立酮组和柴芍六君子汤组,每组15只,除空白对照组外,其余3组用复合病因造模法造成FD模型,连续21d。灌胃14d后腹主动脉取血,离心制备血清样本,取胃、海马及下丘脑组织。测定胃排空率,采用ELISA法检测血清五羟色胺(5-HT)的含量,免疫组织化学法检测海马、下丘脑及胃组织中5-HT的表达。结果:模型组大鼠胃排空率显著低于空白对照组(P0.05);模型组大鼠血清、胃组织5-HT蛋白表达水平显著高于空白对照组(P0.05);模型组大鼠海马和下丘脑组织5-HT蛋白表达水平显著高于空白对照组(P0.01)。柴芍六君子汤组和多潘立酮组大鼠胃排空率显著高于模型组(P0.05);柴芍六君子汤组和多潘立酮组大鼠血清、胃组织中5-HT蛋白表达水平显著低于模型组(P0.05);柴芍六君子汤组和多潘立酮组大鼠海马和下丘脑组织中5-HT蛋白表达水平显著低于模型组,差异有统计学意义(P0.01)。结论:柴芍六君子汤能显著提高FD大鼠胃排空率,降低下丘脑、海马、胃组织及血清中的5-HT含量,减轻胃肠高敏状态,恢复胃肠正常动力,这可能是其治疗FD的机制之一。  相似文献

目的:观察电针五脏俞对失眠大鼠睡眠时间及脑内神经递质、细胞因子水平的影响,探讨电针五脏俞治疗失眠的机制。方法:SD大鼠随机分为对照组、模型组、电针组和地西泮组,每组10只。采用氯苯丙氨酸诱导构建失眠大鼠模型。电针组予以电针"肺俞""心俞""肝俞""脾俞""肾俞",频率60 Hz,强度1 mA,10 min/次,西药组予以地西泮灌胃治疗(0. 92 mg/kg),均每日1次,共治疗6 d。评估各组大鼠睡眠持续时间变化,HE染色检测治疗后大鼠海马病理学改变,ELISA法检测下丘脑5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)、肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β含量,免疫组织化学法、荧光定量PCR法和Western blot法检测海马组织外周型苯二氮卓受体(PBR)mRNA及蛋白表达。结果:与对照组比较,模型组大鼠昼夜节律紊乱,睡眠持续时间显著缩短、体质量下降(P<0. 05);与模型组比较,电针组及地西泮组大鼠睡眠持续时间增长、体质量增加(P<0. 05)。与对照组比较,模型组大鼠海马组织神经元数量减少,细胞排列不规则,下丘脑5-HT、5-HIAA、TNF-α和IL-1β表达水平均显著降低(P<0. 05),海马组织中PBR mRNA和蛋白表达水平显著升高(P<0. 05)。与模型组比较,电针组和地西泮组海马组织神经元数量增加,细胞排列逐渐规则,下丘脑中5-HT、5-HIAA、TNF-α和IL-1β表达水平均显著升高(P<0. 05),海马组织中PBR mRNA和蛋白表达水平显著降低(P<0. 05)。电针在增加睡眠持续时间、体质量、下丘脑5-HT和5-HIAA表达方面与地西泮疗效相当,在升高下丘脑TNF-α和IL-1β表达水平、降低海马组织PBR mRNA及蛋白表达方面优于地西泮(P<0. 05)。结论:电针五脏俞能够降低海马组织中PBR表达,促进下丘脑神经递质及细胞因子的合成和释放,缓解海马组织的病理损伤,改善失眠大鼠的症状。  相似文献

目的:观察灸法结合拔罐疗法对运动性疲劳运动员主观体力感觉及血液生化指标的影响。方法:将30例体操运动员随机分为2组,大强度运动后,治疗组15例采用温和灸加拔罐治疗,对照组15例休息30min,观察一次性大强度训练和周期性大强度训练前后运动员血清肌酸激酶(Creatine Kinase,CK)、血尿素氮(Blood Urea Nitrogen,BUN)、尿常规、主观体力感觉评分表(Rating of Perceived Exertion,RPE)和SCL-90症状自评量表的变化。结果:大强度训练后,治疗组升高的CK水平恢复明显快于对照组。SCL-90症状自评量表分析结果显示治疗组运动员的人际关系优于对照组。RPE分析表明治疗组运动员的疲劳感觉轻于对照组。结论:灸法结合拔罐疗法是消除运动性疲劳的一种有效的、简便可行的方法。  相似文献

Objective

To investigate the analgesic time-effect characteristics and changes in concentrations of rabbit’s hypothalamic 5-hydroxytryptamine (5-HT) and noradrenaline (NE) caused by buccal acupuncture in the rheumatoid arthritis (RA) rabbits, and to reveal the analgesic central mechanism of buccal acupuncture, thereby providing a theoretical basis for the treatment of pain by buccal acupuncture.

Methods

Forty rabbits were randomly divided into a normal group, a model group, a body acupuncture group, and a buccal acupuncture group, with 10 rabbits in each group. No model was established in the normal group, while equal dose of normal saline was injected at the matched site and time point; rabbits in other groups were subjected to the establishment of RA models using egg protein. From the 27th day of the experiment, rabbits in each group received the designated intervention. Rabbits in the normal group and the model group were fixed for 30 min every day using the same method as those in the other groups. In the acupuncture group, Dubi (ST 35) and Zusanli (ST 36) on bilateral hind limbs were selected. Perpendicular needling (using the needles with 0.25 mm in diameter and 25 mm in length) was performed with twirling manipulation for 15 s at intervals of 5 min. The needles were retained for 30 min and acupuncture was performed once a day. In the buccal acupuncture group, the knee point in the buccal acupuncture and needles with a diameter of 0.25 mm and a length of 15 mm were selected. Oblique needling was performed with twirling manipulation for 15 s at intervals of 5 min. The needles were retained for 30 min and acupuncture was performed once a day. The thermal pain thresholds at the 0, 5, 15, 30, 60, 120 and 240 min after the 1st and 10th acupuncture therapy were measured with a PL-200 thermal-inducing pain meter. After the 10th acupuncture therapy, rabbit’s hypothalamus was removed, and the 5-HT and NE concentrations in the hypothalamus at the peak point of the acupuncture pain threshold curve were determined by high performance liquid chromatography (HPLC).

Results

The analgesic effect was obvious at 5 min after buccal acupuncture started, peaked at 30 min, and decreased to the lowest value at 240 min. Rabbits in the body acupuncture group began to show significant analgesic effect at 15 min, which was peaked at 30 min, and began to decline at 60 min. The pain threshold at 240 min was still higher than that at 0 min. Compared with the model group, the concentrations of hypothalamic 5-HT in the buccal acupuncture group and the body acupuncture group was significantly increased, and the between-group differences were statistically significant (both P<0.05). The NE/5-HT ratios in hypothalamus in the buccal acupuncture group and the body acupuncture group were significantly lower than the ratio in the model group, and the differences were statistically significant (both P<0.05); difference in the decrease was statistically significant between the buccal acupuncture group and the body acupuncture group (P<0.05).

Conclusion

The analgesic effect of buccal acupuncture shows an obvious time-dependent curve. It is characterized by rapid onset of pain relief, rapid increase and decline in pain threshold. 5-HT and NE levels in rabbit’s hypothalamus can be affected by buccal acupuncture, with increased 5-HT concentration and reduced NE/5-HT ratio.

Ethnopharmacological relevance

Ding-Zhi-Xiao-Wan (DZ, also known as Kai-Xin-San) is a famous traditional Chinese medicine used for the treatment of emotional disease. Previously, we have found that in a variety of animal models of depression (such as tail suspension model, model of chronic fatigue and forced swimming model) DZ demonstrated significant antidepressant behavior and promoted the production of 5-hydroxytryptamine (5-HT). However, the mechanisms of 5-HT regulation are still unclear. Therefore, the current study is designed to further investigate the antidepressant effect of DZ by observing its influence on 5-HT synthesis, metabolism, transport and other key links, so as to clarify the molecular mechanism of its 5-HT regulation.

Materials and methods

Solitary rising combined with the chronic unpredictable mild stress (CMS) was used to establish the rat model of depression. The rats were given DZ for 3 weeks, the behavior change and the following items in hippocampus and prefrontal cortex were detected simultaneously: 5-HT, 5-hydroxyindoleacetic acid (5-HIAA), tryptophan hydroxylase (TPH), aromatic amino acid decarboxylase (AADC), monoamine oxidase (MAO) and 5-HT transporter (5-HTT) were observed.

Results

Our results showed that treatment with the DZ significantly improved the behavior and simultaneously increased the 5-HT level in the hippocampus, prefrontal cortex tissues and hippocampus extracellular of depressive rats. In future studies revealed that DZ could significantly increase the protein and mRNA expression of the key enzymes TPH during the 5-HT synthesis process in the hippocampus and prefrontal cortex of the depressed rats, and suppress the expression of 5-HTT protein and mRNA at the same time. But it had no effects on MAO-A and MAO-B activities.

Conclusion

We believe that antidepressant effect of DZ is caused by the increase of 5-HT synthesis and reduction of 5-HT re-uptake, and eventually increase the content of 5-HT in the brain and the synaptic gaps.  相似文献

目的:寻求足三里穴抗运动性疲劳的最佳刺激途径。方法:通过跑台法建立大鼠运动性疲劳模型,分别采用艾灸、经皮穴位电刺激(TEAS)、穴位注射等方法刺激大鼠"足三里"穴,观察毛刷刺激频率及血清LD、SOD的改变。LD检测采用酶法,SOD检测采用黄嘌呤氧化酶法。结果:第7周,各治疗组与模型对照组比较,所需毛刷刺激频率明显少;TEAS组第7周所需刺激频率明显少于第5周。治疗前,各跑台组与安静对照组比较,大鼠SOD浓度、LD浓度明显升高;治疗后与治疗前比较,各治疗组SOD浓度、LD浓度明显下降。结论:TEAS、穴位注射和艾灸刺激"足三里"穴均能起到良好的抗运动性疲劳的作用,TEAS疗效略优。  相似文献

目的:观察经皮穴位电刺激(TEAS)对力竭运动大鼠的抗运动性疲劳效应,初步探讨其抗氧化机制。方法:采用5级递增跑台运动建立力竭运动大鼠模型。27只雄性SD大鼠随机分为安静对照组(n=9)、运动组(n=9)和TEAS组(n=9)。TEAS治疗选穴"足三里",电针参数:施以连续波,频率2 Hz,强度5 mA,时间30 min,左右交替,每日1次,连续治疗7 d。力竭运动后,记录大鼠力竭运动时间,用乳酸氧化酶法测定血浆乳酸水平,黄嘌呤氧化酶法测定血浆超氧化物歧化酶(SOD)活性,硫代巴比妥酸法检测血浆丙二醛(MDA)含量。结果:①TEAS组力竭运动时间显著长于运动组(P<0.05)。②运动组血浆乳酸显著高于安静对照组(P<0.05)和TEAS组(P<0.05)。③与安静对照组比较,运动组大鼠血浆SOD活性无显著性变化,但MDA含量升高却十分显著,差异有显著性意义(P<0.01),其SOD/MDA比值显著降低(P<0.05);TEAS组SOD活性显著高于安静对照组和运动组(P<0.01),MDA含量显著低于运动组(P<0.05),其SOD/MDA比值明显高于运动组(P<0.01)。结论:TEAS"足三里"穴可有效抗运动性疲劳,可能是通过其减少血乳酸堆积、提高机体抗氧化能力、减少脂质过氧化反应实现的。  相似文献

Objective

To investigate the effect of moxibustion at Shenque (CV 8) on free radical metabolism in rat’s hippocampus after different degrees of exhaustive exercise.

Methods

A total of 72 male Sprague-Dawley (SD) rats were randomly divided into a normal group (n=8), a model group (n=32) and a moxibustion group (n=32). According to the times of modeling or treatment, the model group was further randomly divided into different subgroups of a 1-time model subgroup, a 4-time model subgroup, a 7-time model subgroup and a 10-time model subgroup (n=8); the moxibustion group was also further randomly divided into different subgroups of a 1-time moxibustion subgroup, a 4-time moxibustion subgroup, a 7-time moxibustion subgroup and a 10-time moxibustion subgroup (n=8). Rats in both model and moxibustion subgroups were subjected to establishing the swimming exhaustive model. Rats in each moxibustion subgroup received mild moxibustion at Shenque (CV 8) for 15 min immediately after modeling, once every other day. The concentration of malic dialdehyde (MDA), as well as the activities of superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and total antioxidant capacity (T-AOC) in rat’s hippocampus in each group were detected 24 h after the exhaustive exercise.

Results

Compared with the 1-time model subgroup, the exhaustive swimming time of rats was significantly prolonged in the 4-time model subgroup (P<0.01), while it was significantly shortened in the 7-time and 10-time model subgroups (both P<0.01). Compared with the matched model subgroup, the exhaustive swimming time of rats in the 7-time and 10-time moxibustion subgroups was significantly prolonged (both P<0.01). Compared with the normal group, the MDA concentration was increased significantly (P<0.01), and the activities stress response of SOD and T-AOC were increased in the 1-time model subgroup (both P<0.05); the MDA concentration was increased (all P<0.01), and the activities of SOD, GSH-Px and T-AOC were decreased differently (P<0.05 or P<0.01) in the 4-time, 7-time and 10-time model subgroups. Compared with the matched model subgroup, the concentration of MDA was significantly reduced (P<0.05 or P<0.01), and the activities of SOD, GSH-Px and T-AOC were significantly increased in the 4-time, 7-time and 10-time moxibustion subgroups (all P<0.01).

Conclusion

Moxibustion at Shenque (CV 8) can improve the fatigue status of the body after long-term exhaustive exercise by regulating free radical metabolism in rat’s hippocampus. To some extent, this provides an experimental basis for moxibustion at Shenque (CV 8) against exercise-induced fatigue.

Objective: To observe the clinical efficacy of aerobic exercise combined with auricular point sticking for insomnia in college students. Methods: A total of 128 cases from Zhejiang Economic and Trade Polytechnic were randomly allocated into an observation group and a control group. The 64 cases in the observation group were treated with auricular point sticking plus aerobic exercise, while the 64 cases in the control group were treated with auricular point sticking alone. The clinical efficacies in the two groups were compared after treatment. Results: The total effective rate in the observation group was 96.9%, versus 70.3% in the control group, showing a statistically significant difference (P〈0.05). Conclusion: Combining aerobic exercise and auricular point sticking has accurate effect on insomnia in college students and is better than auricular point sticking alone.  相似文献

目的:探讨推拿功法《易筋经》练习对膝骨关节炎(Knee Osteoarthritis,KOA)患者股四头肌肌力的影响,为推拿功法《易筋经》干预KOA提供科学依据。方法:将90例KOA患者随机分为观察组和对照组,每组45例。观察组进行《易筋经》锻炼,每日1次;对照组不予任何治疗。观察8星期后进行疗效评定。结果:观察8星期后,观察组患者股四头肌等速肌力较治疗前有显著改善,对照组患者股四头肌等速肌力无明显变化。结论:推拿功法《易筋经》练习对改善KOA患者股四头肌肌力具有一定作用,是干预该疾病的有效运动方法。  相似文献

目的:观察针刺治疗慢性疲劳综合征(Chronic Fatigue Syndrome,CFS)患者的临床疗效及对其焦虑状态的影响。方法:将90例符合纳入标准的患者随机分为2组,治疗组45例,采用针刺治疗;对照组45例,采用药物治疗。采用焦虑自评量表(Self-ratingAnxietyScale,SAS)对两组患者治疗前后的焦虑状态进行评价。结果:两组患者治疗后SAS评分较同组治疗前均有降低(P<0.01)。两组治疗后SAS评分比较,差异无统计学意义(P>0.05)。结论:针刺和药物治疗均能改善CFS患者的焦虑症状。  相似文献
