川芎嗪改善大鼠胰腺保存的机理 总被引:5,自引:0,他引:5
通过测定保存不同时间后未移植胰腺组织内6-酮-前列腺素F1α及血栓素B2含量,对川芎嗪改善胰腺保存效果的作用机制作出初步探讨。6-keto-PGF1α及TXB2测定结果显示:加川芎嗪对6-keto-PGF1α无明显影响,对TXB2则能显著性降低,6keto-PGF1α;TXB2的比值在加川芎嗪组最大,认为川芎嗪主要通过抑制TXA2合成,升高PGI2:TXA2的比2值,发挥PGI2的保护作用,抑制T 相似文献

本实验研究了川芎嗪加入本所自制的WMO-1号器官保存淮对大鼠胰腺保存效果的影响。结果表明,WMO-1号保存液对大鼠胰腺的确切保存时限为12小时,18小时保存存活率仅为1/3,加入川芎嗪要使保存时限延长至18小时,24小时保存存活率为1.3。证明川芎嗪确能改善胰腺保存。 相似文献

黄芪在犬肾离体灌注保存中的作用 总被引:1,自引:1,他引:0
目的研究黄芪提取液对犬肾离体低温灌注及保存中缺血再灌注损伤的影响。方法以常规的HC-A保存液为对照组,以高渗枸橼酸盐嘌呤溶液(HC-A)添加黄芪提取液为实验组,分别对离体犬肾进行低温灌注保存24、48、72 h。通过光镜、电镜观察肾组织结构变化,建立犬肾移植模型,检测移植前后生化指标变化,综合分析。结果实验组保存的犬肾组织超微结构损伤明显轻于对照组。病理组织检查评分,实验组低于对照组(P<0.05)。移植肾早期功能指标检测,移植后2 h实验组血肌酐值(98.25±36.76)μmol/L明显低于对照组(147.26±59.67)μmol/L;移植后24 h血肌酐值(221.55±62.13)μmol/L明显低于对照组(316.12±74.31)μmol/L,而内生肌酐清除值实验组(1.91±0.76)ml·min-1·kg-1明显高于对照组(0.87±0.82)ml·min-1·kg-1,差异均具有显著性(P<0.05)。结论黄芪用于肾脏低温灌注保存中,对缺血再灌注损伤的肾脏有一定保护作用。 相似文献

目的 研究川芎嗪(TMP)联合丹参延缓肾移植大鼠慢性移植肾肾病进程的作用及机制.方法 以Fisher 344大鼠和Lewis大鼠分别作为肾移植的供、受者进行原位肾移植,并建立CAN模型.按随机数字表法将受鼠分为5组:环孢素A(CsA)组(A组),TMP+ CsA组(B组)、丹参+ CsA组(C组)、TMP+丹参+CsA组(D组)及空白对照组(E组,术使用任何药物).分别于术后2、4、6、8和12周,处死每组5只受鼠,取移植肾进行移植肾组织病理学变化,采用免疫组织化学法检测移植肾组织转化生长因子β1(TGF-β1)的表达,采用荧光定量聚合酶链反应法检测移植肾组织TGF-β1 mRNA的表达.结果 术后空白对照组受鼠的存活时间均未超过2周.A组于术后4周时最早出现CAN病理改变,B组和C组出现CAN病理改变的时间较A组晚,D组出现CAN病理改变的时间最晚,且病理改变程度最轻.术后各组Banff评分均呈现明显的上升趋势,相同时间点,A组明显高于其他3组(P<0.05和P<0.0 1),D组明显低于与B组和C组(P<0.05),而B组与C组间无显著差异(P>0.05).随着术后时间的推移,各组TGFβ1表达强度呈逐渐增加的趋势,相同时间点,A组TGF-β1表达强度最高,与其他3组比较,差异均有统计学意义(P<0.05和P<0.01),D组明显低于B组和C组(P<0.05),B组与C组间无明显差异(P>0.05).各组TGF-β1 mRNA表达的变化趋势及组间差异与TGF-β表达强度的情况相一致.结论 TMP或丹参均具有通过下调TGF-β1 的表达延缓肾移植大鼠CAN进程的作用,当两者联合应用时作用更为明显. 相似文献

目的研究新型器官保存液Lifor液对猪肾脏的低温保存效果。方法 24只白色杂种猪,随机均分为Lifor液保存组和威斯康星大学保存液(UW液)组各12只,建立离体肾脏非循环灌注模型,供肾取出后分别以0~4℃的Lifor液和UW液灌注并低温保存,再根据保存时间随机分成2个亚组,分别为保存24h、48h组,然后行猪自体肾移植。比较两组肾脏低温保存结束后其病理学及肾皮质三磷腺苷(ATP)含量的改变,并观察自体肾移植后肾功能恢复情况。结果供肾离体保存24h、48h后,Lifor液组与UW液组的肾组织病理学改变基本一致,肾皮质ATP含量比较差异无统计学意义(P〉0.05)。移植后两组血清肌酐水平比较差异亦无统计学意义(P〉0.05)。结论 Lifor液低温保存肾脏的效果与UW液相当,且成本低廉,因此更具有临床应用前景。 相似文献

目的:总结高渗枸橼酸盐腺嘌呤(HC-A)肾保存液2086例临床应用经验。方法:1980年8月至2000年12月连续2086例肾移植,供肾采用HC-A肾保存液保存。按冷缺血时间分组观察移植后血肌酐,移植肾存活率和平均存活时间。结果:冷缺血时间<12h(573例)、12-24h(779例)和>24h(734例)组移植肾平均存活时间分别为9.32、10.93和7.23年,后者与前两组比较差异均有显著性意义(P<0.05);移植后血肌酐、移植肾存活率在各组间差异无显著性意义(P>0.05)。约3.16%的移植肾因保存问题出现肾功能异常。结论:应用HC-A肾保存液可取得较好的近期肾功能恢复和长期移植肾存活,冷缺血时间以24h内最佳。 相似文献

目的 探讨去甲肾上腺素对肾移植术患者肾功能的影响.方法 同种异体肾移植术患者32例,ASA分级Ⅲ或Ⅳ级,年龄22~64岁,随机分为多巴胺组(D组)和去甲肾上腺素组(N组),每组16例.麻醉诱导后D组静脉输注多巴胺1~10μg·kg-1·min-1,N组静脉输注去甲肾上腺素0.03~0.3μg·kg-1·min-1,持续至术毕,维持术中MAP波动幅度不超过基础水平的10%.分别于术毕及术后12 h时取中心静脉血样及尿样,测定血清半胱氨酸蛋白酶抑制因子C(Cystatin C)、β2-微球蛋白(β2-MG)及尿α1-微球蛋白(α1-MG)、β2-MG的浓度,并记录术中输液总量、肾血管开放至术毕时的尿量、术后12 h内的尿量及呋塞米的用量.结果 两组术中输液总量、尿量及呋塞米用量和各时点血清Cystatin C、β2-MG及尿α1-MG、β2-MG浓度比较差异无统计学意义(P>0.05);与术毕时比较,术后12 h时两组患者血清Cystatin C、β2-MG及尿α1-MG、β2-MG浓度均降低(P<0.05).结论 静脉输注去甲肾上腺素0.03~0.3μg·kg-1·min-1对移植肾功能无不良影响,可用于肾移植术患者. 相似文献

评价不同温度灌注和保存犬肺对肺保存效果的影响.方法 将24只犬随机分为实验组和对照组两组,每组供、受体各6只.实验组以23℃低钾右旋糖酐(LPD)液灌注后切取右肺保存于10℃LPD液,对照组以4℃LPD液灌注后切取右肺保存于4℃LPD液,其后两组均行左肺移植术.比较两组供体肺组织不同时间段的髓过氧化物酶(MPO)活性、丙二醛(MDA)含量及湿/干重量比(W/D),比较两组受体犬移植前后不同时间肺血流动力学参数以及血气分析指标,并比较病理结果.结果与对照组相比,实验组肺组织灌注后各时间段的MPO活性、丙二醛含量及W/D较低(P<0.05).实验组受体犬移植后的肺毛细血管楔压(PCWP)、平均肺动脉压(MPAP)较低,动脉血氧分压(PaO2)较高,差异均有统计学意义(均为P﹤0.05).病理学结果显示,实验组肺组织中性粒细胞浸润、肺泡水肿及损伤等病理变化较对照组减轻.结论 在本实验条件下,与4℃灌注+4℃保存相比,23℃灌注+10℃保存的温度对犬肺具有较好的保存效果,更适用于实验肺移植. 相似文献

Abstract Ischemia-reperfusion injury conditions short-term and long-term graft function. The effects of the inversion of K+ and Na+ concentrations and substitution with polyethylene glycol for hydroxyethyl starch in University of Wisconsin (K-UW) solution were evaluated in isolated perfused rat kidneys and in autotransplanted pig kidneys. In the rat model kidneys were cold-stored for 24 h in K-UW or Na-UW or Na-PEG UW solutions (IGL-1 solution). Fractional sodium reabsorption and glomerular filtration rate was better in kidneys preserved in Na-UW and IGL-1 solution than those preserved in K-UW solution. In the pig model the left kidney was harvested and preserved in K-UW or IGL-1 solution for 24 h and then transplanted. In the autotransplanted pig model, kidneys preserved in IGL-1 solution showed a better function and a significant reduction of MHC class II expression, cellular apoptosis and interstitial fibrosis. In conclusion, kidneys preserved in IGL-1 solution tolerated ischemia/ reperfusion injury better than those preserved in K-UW solution. 相似文献

The main goal of transplantation is to restore good renal function and to improve the quality of life of thousands of dialysis patients, something which can only be achieved by providing them with well functioning grafts. Delayed renal allograft function is a serious problem. It is important to prevent this complication because it makes the diagnosis of acute rejection in the early postoperative period difficult, increases the necessity for diagnostic procedures, introduces dialysis treatments and prolongs hospital stay. The aetiology of delayed graft function (DGF) is multifactorial, and factors including donor management, technique used for organ procurement and preservation, age, anatomical variations in the graft, ischemia periods, use of cyclosporine A (CyA) or recipient immunological reactions have been implicated. Using different preservation solutions DGF rates vary from 30% to 60%. Recent clinical data have demonstrated better preservation and improved renal function posttransplant with HTK and University of Wisconsin (UW) solutions compared to EuroCollins solution. In a randomized multicenter study in collaboration with the Eurotransplant organ exchange organization, the efficacy of the HTK solution in renal transplantation was compared to EuroCollins and UW solutions in two parallel prospective randomized trials.
