Altered expression of adipose differentiation-related protein gene in placental tissue of pre-eclampsia

Objective: To investigate the altered expression of lipid metabolism-related gene adipose differentiation-related protein (ADRP) in pre-eclampsia. Methods: Semi-quantitative RT-PCR and Western blotting were used to validate the altered expression of ADRP gene between pre-eclamptic placentas (pre-eclampsia group) and normotensive placentas (control group) respectively. In situ hybridization (ISH) was used to localize ADRP mRNA in pre-eclamptic placentas. Results: There was a significant difference in the levels of placental ADRP mRNA between pre-eclampsia group and control group (1. 98±0. 50 vs 1. 09±0. 20, P<0. 01). Western blotting showed that placentas both in pre-eclampsia group and control group expressed the special ADRP band at 48. 1 kD. The relative levels of ADRP protein in pre-eclampsia group were significantly higher than those of control group (0. 40 ±0. 19 vs 0. 19 ±0. 09, P<0. 01 ). ADRP mRNA was diffusely distributed in pre-eclamptic placentas. Their positive staining existed in cytoplasm of trophoblast. Conclusion: Abnormal expression of ADRP gene in pre-eclamptic placenta may be associated with the pathogenesis of pre-eclampsia.

The Effects of Magnesium Sulfate on Fetal Rats of FGR and the Expression of Caspase-3 in the Placenta of Maternal Rat

To investigate the effect of magnesium sulfate on the fetal rats of FGR and the expression of caspase-3 in the placenta of maternal rat ; To explore the mechanism of using magnesium sulfate to cure the FGR. Establish model of FGR by a way of passive smoking： giving the maternal rats different agent of magnesium sulfate by subcutaneous injection： low agent group （300 mg/kg）, high agent group （600 mg/kg）. Concentration of magnesium sulfate was monitored. The expression of caspase-3 was measured by RT-PCR and immunohistochemistry technology. Both of the concentrations of magnesium sulfate in high and low agents group are higher than the FGR group （P〈0.01）;the weight of the placenta and fetal rat in high agent group are higher than the FGR group （P〈0.05 and P〈0. 01） ; the expression of mRNA and protein of caspase-3 in the two agent group is higher than the FGR group （P〈0.05 respectively） ; concentration of magnesium sulfate in the maternal rat blood correlate to the weight of fetal rat （r=0. 899, P=0. 038） and the expression of caspase-3 in the placenta of maternal rat （r=-0. 747, P=0. 033; r=-0. 915, P=0. 001）. The research suggests that the weight of fetal rat could be increased by treatment of magnesium sulfate. Because it would imfrmove the placental function by depressing the expression of caspase-3.     

Combined use of Ad-hENDO-VEGI151 and Dexamethasone for prevention and treatment of keratoplasty rejection.an experimental study

Objective:To evaluate the probability and efficacy of endostatin-vascular endothelial growth inhibitor (VEGI) recombinant adenoviruses combined with dexamethasone on suppressing heterolamellar corneal transplantation rejection. Methods: Heterolamellar corneal transplantation models were established in 64 New Zealand rabbits, which were randomized into 4 groups of 16 rabbits each. After the transplantation, all the 64 right eyes were injected subconjunctively with 0. 2 ml saline (saline group), 0. 1 ml AdCA13-hENDO-VEGI151 plus 0. 1 ml saline (AdCA13-hENDO-VEGI151 group), 0. 1 ml Dexamethasone (DXM) plus 0. 1 ml saline (DXM group), 0. 1 ml AdCA13-hENDO-VEGI151 plus 0. 1 ml DXM (Ad-CA13-hENDO-VEGI151 combined with DXM group) with one time each 3 days for 10 times. Graft survival and ocular surface were observed for 6 weeks. The fusion protein expression was detected by immunohistochemistry 6 weeks after transplantation. Results: Both the CNV index, rejection index and the xenograft rejection rate in the AdCA13-hENDO-VEGI151 combined with DXM group were statistically lower than those in other groups. Immunohistochemical staining showed the fusion protein expressed mainly in corneal epithelium. Conclusion: The fusion protein expressed by the recombinant adenovirus has significant effect on inhibiting neovascularization after heterolamellar corneal transplantation. The topical application of AdCA13-hENDO-VEGI151 combined with DXM suppressed effectively the postoperative xenograft rejection rate of heterolamellar corneal transplantation.

Protective effect of panaxatriol on function of hypothalamus-pituitary-testes system in rats irradiated with large dose of X-rays

<正> The present study observed the protective effect of panaxatriol on the function of hy-pothalamus - pituitary - testes system after whole body irradiation with 5 Gy X -rays in male rats. The rats were given panaxatriol intraperitoneally once a day (5mg/day) for 14 days beginning from 24 h before the irradiation. The results showed that hypothalamic L-Enk (P<0. 01) and pituitary (3 - EP(P<0. 05) and testicular cAMP (P<0. 01) contents and serum testosterone level (P<0. 01) were all decreased significantly,while serum FSH level (P<0. 01) increased significantly in single irradiation groups compared with the normal control. However, the above - mentioned indices all approached to the normal control levels in irradiation plus panaxatriol groups. These suggest thhat panaxatriol may weaken the decrement of hypothalamic and pituitary opioid peptide contents, testicular cAMP content and serum TS level and the increment of serum FSH level after the rats being irradiated, and that panaxatriol may have protective fu     

Effect of BMPs on hematopoietic injury of acute radiation sickness in mice

Objective: To investigate the effect of bone morphogenetic proteins (BMPs) on hematopoietic injury of acute radiation sickness in mice. Methods: Mice were subjected to whole-body 60Co γ ray irradiation, then bpBMP was put into spatium intermusculare or rhBMP-2m, PBK/ hBMP-2 -NIH3T3 cells were injected into abdominal cavity. The effect of BMPs on hematopoiesis including some hematological parameters, the survival rate of 30 d and formation of bone marrow CFU-GM colony were detected at postradiation. Results: pbBMP (purified bovine bone morphogenetic protein) increased the formation of bone marrow CFU-GM colony (P<0. 05) on d 10 after irradiation. rhBMP-2m increased the survival rate of mice irradiated by 7. 5 Gys Mice in control group died in 30 days, while 10%, 15% and 35% mice survived when they were injected i. p. with 0. 5 mg, 1. 0 mg and 2. 0 mg of rhBMP-2m respectively.
